1.In vitro culture and phenotype analysis of monocyte-derived dendritic cell from peripheral blood of patients with oral carcinoma

Objective: To compare the phenotype of induced dendritic cell (DC) derived from peripheral blood of the healthy individuals and the patients with oral carcinoma. Methods:15 patients with oral carcinomal at stages II or above were assigned to study group and 15 cases of healthy volunteers were assigned to control group. Leukocyte were isolated by density centrifugation. Adherent fractions were incubated in present of GM-CSF,IL-4 and TNF-? for 7 days in vitro. The morphological change of the cultured cells was observed by light microscopy and electronic microscopy. The phenotype expressions of CD83,CD1a,CD86, CD80, HLA-DR were analysized by flow cytometry. Results:Typical DCs were obtained from two groups after 7 days culture. The expression rate of CD83,CD1a on DC from healthy volunteers was significantly higher than that from patients (P0.05). Conclusion:DC derived from oral cancer patient' s peripheral blood monocyte might be more matured and is a good cell model for further immunotherapy and research.

8.Influence of Compound Glycyrrhizin Injection on Hormone Withdrawal Syndrome in SARS Patients

OBJECTIVE:To summarize the effects of compound glycyrrhizin injection(GL)on hormone withdrawal syndrome(HWS)in SARS patients.METHODS:Using contrast design,the HWS of 134 cases of SARS who had received hormone theropy was retrospectively analysed.RESULTS:Administration of GL could descend the incidence rate of symptoms of HWS,of which the incidence rates of short breath and chest distress,the main symptoms of SARS patients,dropped from 24.5%and 22.6%to 3.6%(P≤0.01)and 7.1%(P≤0.05)and the incidence of asthenia,musculoarthragia and headache decreased from 13.2%,15.1%and 10.4%to 7.2%,14.3%and 0%respectively.The number of patients with elevated ALT reduced(P≤0.01).CONCLUSION:GL can decrease the incidence of HWS.
9.Immunogenicity study of human papillomavirus type 68b virus-like particles expressed in a Hansenu-la polymorpha strain

Objective To evaluate the immunogenicity of the recombinant human papillomavirus type 68b (HPV68b) virus-like particles(VLPs)in a mouse model.Methods The L1 protein of HPV type 68b was successful expressed in the Hansenula polymorpha strain (NVSI-68b-1).Processes including purifi-cation and reconstitution were performed to achieve pure HPV 68b VLPs.The purity, morphology and immu-nogenicity of the purified HPV 68 b VLPs were further analyzed .The BALB/c mice were immunized with HPV68b VLPs formulated on aluminum adjuvant .Pseudovirus-neutralizing antibody ( PsV NAb) assay was performed to detect the neutralizing antibodies in serum samples .Results The HPV 68 b L1 VLPs were ob-tained as indicated by the results of SDS-PAGE, Western blot assay , HPLC, electron microscopy and dy-namic light scattering with a high purity of 95%.Transmission electron microscopy and dynamic light scat-tering analysis revealed that the HPV68b L1 VLPs resembled the native virus with an average particle diame-ter of 50 nm.High levels of HPV68b-neutralizing antibodies were detected in serum samples from the mice immunized with HPV68b L1 VLPs.Moreover, a cross-protective efficacy of HPV68b L1 VLPs for HPV68a was observed .Conclusion This study suggested that the recombinant HPV 68 b VLPs expressed in a Han-senula polymorpha strain might be used as a potential candidate for the development of HPV vaccine .
10.Clinical analysis on pregnant outcomes of gravidas with positive urine glucose

Objective To study the effects of positive urine glucose on maternal pregnancy.Methods A total of 1 338 pregnant women were tested urine glucose.On the basis of their urine-glucose level,two groups were devided:experimental,68 cases of posi-tive urine-glucose gravidas,control group,199 cases of negative urine-glucose gravidas.Analyze on the outcome of pregnancy of the two groups.Results Compared between positive group and control group,the incidence of died(abnormal)fetus,gestational hyper-tension,abnormal amniotic fluid,fetal distress,and fetal macrosomia were not statistically different(P >0.05),but the incidence of premature rupture of membranes was statistically different (P <0.05 ).Conclusion Positive glucose urine of gravidas might in-crease the risk of premature rupture of membranes,positive urine glucose detected during pregnancy should be highly valued.
