1.Research progress of CTGF/CCN2 in the development and targeted therapy of malignant tumor

The current incidence of malignant tumors is increasing.Although we have been exploring the mechanism of tumor development and its treatment,its efficacy is little.Malignant tumor development mechanism is very complex,and a variety of proteins and genes involved.Connective tissue growth factor(CTGF,also known as CCN2)is a secreted protein,a member of the CCN family,which plays a very important role in the development and progression of tumors.This article summarizes the structure and function of CTGF /CCN2 protein,the mechanism of action in the development of malignant tumors and the latest research progress in targeted therapy.

Conclusion Automatic potentiometric titration was easy to grasp, rapid and accurate, which was a new analytical method worthy to be applied extensively.

Objective To investigate the roles of hypoxia regulated genes/protein in chemical hypoxia preconditioning in differentiated SH-SY5Y cells.Methods Differentiated SH-SY5Y cells were randomly divided into control group,chemical hypoxic preconditioning group(50 ?mol/L CoCl2 preconditioning for 3 h,normal culture for 1 h,then in 2%O2 for 28 h) and hypoxia group(in 2%O2 for 28 h).RT-PCR was used to examine the mRNA expressions of VEGF,GLUT-1,EPO,LDH-A.Further evaluation of the potential neuroprotective effect of VEGF was done by investigating the effect of recombinant human VEGF on subsequent hypoxia injury.Results The mRNA levels of GLUT-1,EPO increased in the preconditioning group as compared with those in the hypoxia group(P
4.A Study on Treatment of Acute Hemorrhagic Shock at High Altitude with Hyperosmotic Sodium Chloride in Rabbits

Eleven male rabbits were divided into 2 groups. Normal saline(NS) group consisted of 5 animals and was treated with 0.9% NaCl solution, and hyperosmotic saline (HS) group consisted of 6 rabbits and was treated with 7.5% NaCl. The animals were exposed to hypobaric hypoxia at a simulated altitude of 4000 m and hemorrhagic shock was inflicted to them through femoral artery bleeding. The mean arterial pressure (MAP) was brought to 6.0 kPa over 10 minutes and kept at that level for 70 minutes. A certain amount of 0.9% NaCl (300 mmol/L) or 7.5% NaCl (2400 mmol/L) was intravenously infused over a 10-minute interval. The volume infused was equivalent to 10% of the total blood shed. It was found that MAP, left ventricular systolic pressure, and LV dp/dt were significantly higher in HS group than in NS group (from 3rd to 20th minute after infusion). 5.Effect of early normobaric hyperoxia on cerebral oxygenation in patients with extremely severe craniocerebral injury

Objective To observe the changes of brain oxygenation in patients with extremely severe craniocerebral injury and the therapeutic effect of early normobaric hyperoxia.Methods Sixtyeight patients with extremely severe craniocerebral injury treated from January 2011 to January 2013 were assigned to two groups according to the random number table:50% oxygen breathing for one week in control group (34 cases) and 80% oxygen breathing for one week in treatment group (34 cases).Blood samples from jugular vein and radial artery were collected at 1,3,5,and 7 days to measure indices of blood gas analysis,i.e.,PaO2,PjvO2,CaO2,CjvO2,Da-jvO2,CERO2 and Djv-a Lac.GCS and content of neuron-specific enolase (NSE) were recorded as well.Results Values of PaO2 at each time point and GCS at 5 and 7 days were significantly higher in treatment group than in control group (P <0.05).Djv-a Lac at 3,5 and 7 days and NSE at 7 days revealed significantly higher levels in treatment group than in control group(P < 0.05).Whereas at each time point,there were no significant differences between the two groups in aspects of PjvO2,CaO2,CjvO2,Da-jvO2 and CERO2 (P > 0.05).Conclusion Early use of 100% oxygen in patients with extremely severe craniocerebral injury may be beneficial to the prognosis.
6.Characterization of ?-Ketoacyl-acyl Carrier Protein Synthase Ⅱ Homologues in Enterococcus faecalis

FabB(?-ketoacyl-acyl carrier protein synthase Ⅰ)and FabF(?-ketoacyl-acyl carrier protein synthase Ⅱ)are two key enzymes of fatty acid biosynthesis in E.coli.The Gram-positive pathogenic bacterium Enterococcus faecalis has a fatty acid composition very similar to that of E.coli.Bioinformatic analysis reveals that though E.faecalis has two fabF homologues,there is no recognizable fabB homologue in the genome of E.faecalis.Two fabF homologues(fabF1 and fabF2)were amplified by using E.faecalis V583 genomitic DNA as template,and two plasmids,pHW13(fabF1)and pHW14(fabF2),were constructed.The results of experiments in vivo and in vitro have shown that fabF1 gene could complement E.coli fabB mutation and FabF1 possessed ?-ketoacyl-acyl carrier protein synthase Ⅰ(FabB)activity,while fabF2 gene could complement E.coli fabF mutation and FabF2 had ?-ketoacyl-acyl carrier protein synthase Ⅱ(FabF)activity.Meanwhile the data also shown that FabF2 possessed partial function of ?-ketoacyl-acyl carrier protein synthase Ⅰ(FabB),and it could make E.coli fabB mutation synthesized low amount of unsaturated fatty acid.From these data it is clear that FabF species enzymes could have activity of ?-ketoacyl-acyl carrier protein synthase Ⅰ(FabB).
7.Study on three-dimensional echocardiography for quantifying left ventricular function in conotruncal defects

Objective To evaluate the accuracy and value of ventricular function in conotruncal defects(CTD)by three-dimensional echocardiography(3DE).Methods Fifty-two children with CTD and forty-three children with normal hearts were examined by 3DE.The measurements of left ventricular volume and wall mass were outlined and calculated.The measurements of left ventricular volume and wall mass of 3DE in preoperative CTD were compared with those of 2D biplane Simpson-method in preoperative CTD,3DE in control group and 2D biplane Simpson-method in control group.The results of 3DE and 2DE preoperative measurements were also compared with their postoperative clinical cardiac conditions.Results From the findings of analysis of variance and Student-Newman-Keuls(SNK)test,there was no significant difference between 3DE and 2DE measurements of left ventricular function in normal children,but there was significant difference between 3DE and 2DE measurements of left ventricular function in children with CTD.2DE measurements of left ventricular volume and wall mass in the CTD group were significant underestimated and less correlated with their postoperative clinical cardiac conditions(r=0.20,P=0.2086)than 3DE measurements(r=0.39,P=0.0090).Furthermore,left ventricular function in the CTD group was poorer than that in the control group.Stroke volume and effective function were reduced due to increasing end-systolic volume.while left ventricular wall mass was increased due to the possible compensation of its ventricular muscle.With the lower end-diastolic volume,stroke volume and effective function,patients were more likely to have the occurrence of postoperative low cardiac output.Conclusions By making a precise quantitative assessment,3DE is very useful in providing more information for preoperative diagnosis in CTD and predicting such postoperative prognoses as the occurrence of postoperative low cardiac output.
8.Assessment of longitudinal systolic ventricular function in children with single ventricle using velocity vector imaging

Objective To investigate the longitudinal systolic ventricular function of children with single ventricle, and to appraise the feasibility and clinical value of velocity vector imaging (VVI) on assessing it. Methods The study group consisted of 30 patients with functional single ventricle. The control group consisted of 30 age- and gender-matched normal children. Using Sequoia C512 echocardiography machine,the peak velocity(V),peak displacement(D) ,strain(S) ,and strain rate(SR) were measured. Results Basel and median velocities,as well as basel and median displacements of rudimentary chamber side(RCS), were lower than those of septum[Basel velocity (1. 93 ± 0. 71)cm/s vs (3. 53 ± 1. 07)cm/s;Median velocity (1.19±0. 57)cm/s vs (2. 03 ± 0. 90) cm/s; Basel displacement (2. 53 ± 1.65) mm vs (6.21 ± 2. 12)mm;Median displacement(1. 26 ± 1. 06)mm vs (3. 21 ± 1.37)mm]. The values of strain of all the six segments of single ventricle were significantly lower than Corresponding segments of the control group [ basel nonrudimentary chamber side(NRCS) ( - 16. 17 ± 4. 37) % vs ( - 19. 66 ± 3. 47) % ; Median NRCS ( - 15. 23 +4.36)% vs (-19.64± 4. 75)%; Apical NRCS (-13.84 ± 5.79)% vs ( - 16. 7 ± 4. 15)%; Basel RCS(-10. 54±5.35)% vs (-19.49±3. 74)%;Median RCS ( - 10. 16 ± 5. 26)% vs ( -20. 83 ± 3. 82)% ;Apical RCS ( - 10. 97 ± 5. 22) % vs C - 18. 11 ± 4.43) %]. Aside from basel strain rates of NRCS, strain rates of all the other five segments of single ventricle were significantly lower than Corresponding segments of the control group [Median NRCS ( - 1. 21 ± 0. 42)s-1 vs (- 1.49 ± 0. 24)s-1 ;Apical NRCS ( - 1.10 ±0.41)s-1 vs (-1.47 ± 0.24)s-1;Basel RCS ( - 1.07 ± 0. 35) s-1 vs (- 1. 49 ± 0. 22) s-1; Median RCS (-0.97± 0.34)s-1 vs ( -1.48 ± 0. 20)s-1 (Apical RCS ( - 0. 93 ± 0. 39)s-1 vs ( - 1. 48 ± 0. 22)s-1]. Conclusions Longitudinal systolic ventricular function is impaired in children with functionally single ventricle. VVI can be used to asses ventricular systolic performance in children with functionally single ventricular.
9.Multi-factorial analytical study on real-time three-dimensional echocardiographic examination and image processing in children

Objective To evaluate the importance of factors affecting efficiency of image display quality of realtime three-dimensional echocardiography (RT-3DE) and establish an optimized method for RT-3DE examination and image processing in children. Methods Based on corresponding cardiac acoustic characteristics in children, 87 healthy children (46 boys, 41 girls, mean 36.2 ± 44.2 months) were selected. Three aspects of seven factors were selected, including acquisition windows, gain, compress, post process, smoothing, frequency fusion and 3D vision for RT-3DE examination and analysis using orthogonal test design method. The principal effectiveness analysis of orthngonal test design method and its table L_(18)3~7 were used. The efficiency rate of 3D image display quality was used as the inspection index for orthogonal test. Results According to the average deviation orders of the seven factors, the maximal average deviation was gain. The aspects with maximal average deviation of each factor were at via-subcostal and via-apical acoustic windows, 90 of gain, 60 of compress, E of post process, 5 of smoothing, F3 of frequency fusion and A to B of 3D vision, respectively. Conclusions The optimized method for RT-3DE examination and image processing could be achieved by setting acquisition windows, gain, compress, post process, etc. It is helpful to promote clinical application of RT-3DE and benefits the design of multi-parameter presetting of ultrasound system to optimize RT-3DE examination in children.
10.Determination of scopoletin in Cortex Mori from different sources

Object To study the contents and the determinating method of scopoletin in Cortex Mori from different sources.Methods Scopoletin was determined by HPLC.The mobile phase was MeOH-H 2O-HAc (40∶60∶0.25).Detection wavelength was at 354 nm.Results The average recovery was 98.88% with RSD=1.91% and the linear range was 0.025 6—0.128 ?g.The highest content of scopoletin was 0.017 3% and the lowest content was 0.002% from different producing areas.Conclusion The method is easy, accurate, reliable with good reproducibility and high sensitivity.
