1.Uric acid, hyperuricemia and ischemic stroke

Uric acid is an end product of purine metabolism.Hyperuricemia is defined as serum uric acid level >420 μmol/L in man,and 360 μmol/L in woman.Several mechanisms can cause elevated serum uric acid levels.Systematic reviews and Meta-analyses have shown that hyperuricemia is significantly correlated with the increased morbidity and mortality in patients with ischemic stroke.However,as a powerful antioxidant and a radical scavenger,uric acid seems to have a neuroprotective effect in ischemic stroke.Uric acid administered early can enhance the effect of recombinant tissue plasminogen activator (rtPA) thrombolytic therapy in rats with thromboembolic stroke.Therefore,it is necessary to conduct a randomized controlled trial for the clinical effects of rtPA thrombolysis in combination with uric acid in patients with acute ischemic stroke.A phase 3 clinical trial is ongoing,and its results are worth waiting for.
2.Study on the relationship between sex hormone and lipid metabolism in overweight and obese boys aged 9 to 12 years

Objective:To explore the specific changes and correlations of sex hormones and lipid metabolism in overweight and obese boys aged 9-12 years.Methods:Seventy-two male subjects (9 to 12 years old) were divided into normal weight group ( n=42), overweight group ( n=15), and obese group ( n=15). Plasma sex hormone levels and lipid levels were detected by chemiluminescence immunoassay and ultra performance liquid chromatography-quadrupole time-of-flight mass spectrometry (UPLC/QTOF/MS). All data were analyzed by multivariate statistics analysis. Results:(1) The serum concentration of sex hormone-binding globulin (SHBG) in obese group and overweight group was significantly lower than that in normal weight group, and the serum concentration of SHBG in obese group was significantly lower than that in overweight group ( P<0.05). The serum concentrations of dehydroepiandrosterone (DHEA) in overweight and obese group were significantly higher than those in normal weight group ( P<0.05). (2) There were 171 kinds of differential lipid compounds between normal weight group and overweight group, 218 species of differential lipid compounds between normal weight group and obese group, and 34 species of differential lipid compounds between overweight and obese group. Among them, 150 kinds of lipid compounds in normal weight group were significantly different from those in overweight and obese group. (3) SHGB was positively correlated with phosphatidyl cholines (PC) (20∶0/0∶0) and PC[18∶1(9E)/0∶0]( r=0.6, P<0.05), DHEA was positively correlated with diacylglycerol (DG)[17∶2(9Z, 12Z)/22∶0/0∶0]( r=0.5, P<0.05). Conclusions:9-12 years old overweight and obese boys had early growth and development, but sex hormone levels were not synchronized with growth and development. The serum DHEA level in overweight and obese boys was significantly increased, while the level of DG [17∶2 (9Z, 12Z)/22∶0/0∶0] was significantly increased, and the serum SHGB concentration was significantly decreased, while the vast majority of PC was significantly down-regulated. The mechanism of these changes needs to be further studied.
3.Inhibitor of growth 2 and tumors

Inhibitor of growth 2 (INC2 ) is a member of INC family, and cooperates with p53 in regulating cell cycle, apoptosis, and aging. It can also facilitate chromatin remodeling and transcription regulating through histone modifications. Researches suggest that INC2 protein is an important factor in the genesis, metastasis and prognosis of many kinds of tumors. Resent studies indicate that INC2 can not be simply classified as oncogene or tumor suppressor. At present, the function of INC2 in tumorigenesis and tumor development involves interaction with varied molecules, but the mechanisms remain largely unknown, which still require further investigation.
4.Construction of lentivirus RNAi vector pLenti6/V5-DEST-IP_3R1 and identification of its silencing effects

Objective To construct lentiviral expression vector of rat IP3R1 gene,and identify its silencing effect by using PC12 cell lines. Methods Oligo DNA sequences of 4 pairs of miRNA,named as miRNA1,miRNA2,miRNA3 and miRNA4,were designed according to IP3R1 gene sequence (GenBank:NM_001007235). The single strand of oligo DNA was annealed to form double strand DNA,and then connected with the empty plasmid pcDNA TM 6.2-GW/EmGFP-miR. By using gateway technology,the expression vector pcDNA TM 6.2-GW/EmGFP-miR-IP3R1 was linked into lentiviral destination vector pLenti6/V5-DEST to form the lentiviral expression vector pLenti6/V5-DEST-IP3R1,then it was transformed into infectious lentiviral particles and to infect PC12 cell lines. Silencing effect of gene IP3R1 was detected by Real-time PCR and Western blot. Results The sequence of expression vector pcDNA TM 6.2-GW/EmGFP-miR-IP3R1 was proved correct using sequencing method. After the transfection of letivirus vector pLenti6/V5-DEST-IP3R1 into PC12 cell lines,the IP3R1 mRNA and protein level were down-regulated 48h later,of which miRNA2 and miRNA3 sequence showed the best silencing effect,and the expression of IP3R1 in the blank control and negative control showed no significant changes. Conclusions Lentiviral expression vector pLenti6/V5-DEST-IP3R1 was constructed successfully. pLenti6/V5-DEST-IP3R1 may render the IP3R1 expression in PC12 cell lines down-regulated,and it provides a foundation for studying the function of calcium release channel IP3R1.
5.Preparation, Characteristics and Cell Imaging of Fluorescent Nanoparticles Based on Grafted Poly(acrylic acid)

Polyacrylic acid was firstly grafted by N-amino-4-N-methylpiperazine-1,8-naphthlimide (AMN) to prepare a amphiphilic polymer, which was self-assembled in water producing nanoparticles called as PAAMN.Then the morphology, structure and fluorescence properties of PAAMN were investigated by various methods including transmission electron microscopy, dynamic light scattering, ultraviolet-visible spectroscopy (UV-Vis), nuclear magnetic resonance spectroscopy (HNMR) and fluorescence spectroscopy.MTT assay was carried out to assess the cell compatibility of PAAMN.Finally, the fluorescence from PAAMN self and HeLa cells incubated with PAAMN was observed by fluorescence microscope.The results revealed that PAAMN had spherical structure, in which naphthlimide fluorphores were immobilized in the polyacrylic acid matrix with the degree of substitution of 4.1%.Under the physiological pH condition, PAAMN excited at 390 nm could emit strong and stable fluorescence at 534 nm.In the range of pH 4.0-10.0, its excitation and emission wavelengths had no obvious change.The fluorescence intensity of PAAMN increased with the decrease of pH values, but the pH sensitivity of PAAMN was much lower than that of AMN.PAAMN had good cell compatibility.From the pictures of fluorescence imaging, it was found that both PAAMN self and cells-engulfed PAAMN could emit green fluorescence upon excited at 390 nm, indicating the potential of the developed nanoparticle for cell imaging.

7.Spectrum and survival of cancer in cohort of HIV-infected population

Objection To investigate the spectrum and survival status of HIV positive cancer in HuBei province, China. Methods HIV positive cancer patients were added up and followed up who had registered in Zhongnan Hospital between January 2004 and June 2009. The prognostic features were determined for HIV patients with cancer. Results The average age of HIV positive and HIV negative group who suffered with malignant neoplasm were 42.5±8.8 years and 55.1±13.7 years respectively (P <0.05), mean CD4 counts were (220.9±142.3)/μl and (554.4±174.3)/μl, respectively (P <0.05), the types of common cancer were NHL, cervical cancer, liver cancer and lung cancer, colon cancer, breast cancer, respectively. Anticancer and/or ART treatment were the important positive prognostic factors. Additional factors such as age and CD4 count were associated with survival of cancer patients with HIV infection. Conclusion Mean age of HIV positive cancer patients is about 42 years old. NHL, cervical cancer and liver cancer are HIV associated malignant tumor. ART and anticancer can effectively prolong the survival of HIV infected patients with cancer.
8.Expression of inhibitor of growth 2 and mutant P53 and their clinicopathological significances in gastric cancer

Objective To investigate ING2 expression in normal gastric tissue,gastric carcinoma and precancerous lesions, and to explore correlation between ING2 expression and the carcinogenesis and progression of gastric carcinoma and the clinicopathological signficance as well as the correlation between ING2 and raP53 protein. Methods The expression of ING2 was measured by PV9000 two-step immunohistochemical staining. In total, 188 gastric cancer(GC), 128 matched normal gastric mucosa,35 chronic atrophic gastritis (CAG), 87 intestinal metaplasia (IM) and 36 dysplasia (DYS) samples were analyzed. Expression of mP53 was measured in 40 samples from the 188 gastric carcinomas mentioned above. Results Positive ING2 expression was significantly more frequent in CAG (74.29%) ,IM (91.95%) ,DYS (75.0%) and GCs (70.21%) than in normal gastric mucosa (36.72% ,P <0.05). Among GCs, ING2 expression varied by Lanren's classification. A significantly higher rate of positive ING2 expression was observed in intestinal type GCs (80. 56%) than in diffuse (64.49%) and mixed (55.56%, P < 0.05) type GCs. Furthermore, positive ING2 expression was significantly more frequent in well-to-moderately differentiated adenocarcinoma(80. 60%) than in poorly differentiated adenocarcinoma (62. 62%, P < 0. 05). No correlation was found between ING2 expression and mP53 expression in GC (P > 0. 05). Over-expression and mislocalization of ING2 may be involved in the development of CC, especially intestinal-type GC. Further investigations are needed to explore the relevant molecular mechanism.
9.The Projection from the Long Descending Propriospinal Tract to the Lumbar Enlargement in the Rabbit

ObjectiveTo explore the projections of the long descending propriospinal tracts to lumbar enlargement. Method Anterograde tracing with cupric-silver staining. 10 animals were injected b biotinylated dextran in the upper cervical cord. Following survival times of 14 days, projections of the long descending propriospinal tracts were immunohistochemically demonstrated in the lumbar enlargement.Results The degenerated terminals and labeled terminals were found in the bilateral gray matter of the lumbar enlargement, but predominantly ipsilaterally. The terminals were widely distributed in laminal Ⅴ-Ⅸ, heavily Ⅶ and Ⅷ. Conclusion The long descending propriospinal tract from the upper cervical cord projects to the lumbar enlargement.
10.Flashlamp-pumped pulsed dye laser (595 nm) for the treatment of port-wine stains: a clinical review of 1560 patients

Objective To evaluate the efficacy and safety of a 595-nm flashlamp-pumped pulsed dye laser in the treatment of patients at different ages with port-wine stains (PWS).Methods A retrospective review was performed in 1560 patients with PWS who had been treated with a 595-nm flashlamp-pumped pulsed dye laser.Treatment parameters were selected according to the age of and types of lesions in patients.Results The total response rate was 76.73% (1197/1560) in all of the patients.Clinical efficacy of the flashlamppumped pulsed dye laser was closely correlated with patients'age (x2 =83.47,P < 0.01) and types of lesions (x2 =46.30,P < 0.01 ).There was a low incidence of adverse reactions which were well tolerable.Conclusion The 595-nm flashlamp-pumped pulsed dye laser is safe and effective for the treatment of PWS.
