Objective To investigate the association between polymorphism of cytochrome P450 subfamily Ⅺ A polypeptide 1 (CYP11A1) gene and polycystic ovarian syndrome (PCOS) in Chinese population. Methods From May 2005 to Dec.2008,290 PCOS cases treated in the First affiliated hospital of Anhui Medical University matched with 344 reproductive women as controls were enrolled in this study. Genotypes of 7 tagging single nucleotide polymorphisms(tSNP,rs12438594,rs4077582,rs9806234,rsl6968477,rs4887139,rs1843090,rsl 1632698)covering CYP11A1 gene (r~2≥0.8) and 3 microsatellite markers (D15S1547,D16S520,D15S1546) were chosed from the phase II database of Han population in HapMap data.Genotype and frequency of allele were detected by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) and haplotype of gene polymorphism were analyzed in 290 PCOS cases and 344 controls. Results Among 7 tSNPs and 3 microsatellite markers, the frequency of rs4077582,D15S1547,D15S1546 and rsl 1632698 between two groups reached statistical difference (P =0.010,0.044,0.018 and 0.026).The allele frequency of rs4O77582,rs4887139,rsl843090,D15S1547 and Dl 6S520 showed significant difference between two groups(P=0.002,0.048,0.030,0.001 and 0.024).Among 5 haplotype of CYP11A1(ACGCA13/6/9AG,ACGTA16/6/11AA,GCACG12/8/8AA,GTACA14/4/7GG,GTGCA13/6/7AG),the frequency of GTACA14/4/7GG and ACGCA13/6/9AG were 7.8% (45/580) and 25.3% (147/580) in PCOS group and 11.9% and 19.6% in control group, which showed statistical difference (P< 0.05 ).There was no significant difference in the level of serum androgen at difference genotype from rs4077582 between two groups (P>0.05).Conclusion The polymorphism of CYP11A1 gene was associated with PCOS,however, the relationship between gene sequence covered by tSNP/microsatellite markers and hyperandrogenism of PCOS should be further investigated.

Objective To explore changes in circulating endothelial cells(CECs)numbers in neonatal hypoxemia and its clinical significance. Methods Hladovec method was used to measure the quantity of CECs in 40 cases of neonatus with different degree of hypoxemia(hypoxemia group),and 20 matched healthy newborns were enrolled as control group. Results The numbers of CECs in hypoxemia group were much more than those in control group, and the difference was statistically significant.Meanwhile, there were significant differences in CECs numbers between moderate group and mild group(t =13.43,P <0.05),as well as moderate group and severe group(t=7.698,P <0.05).Conclusion The quantity of circulating ECEs in hypoxic neonatus is correlated with the hy-poxic severity, which may be used as early diagnostic indicator for hypoxemia, and could provide new scientific evidences for early diagnosis, illness judgment and curative effect evaluation.

Journal of Southern Medical University 2012;32(4):539-543
Pituitary carcinoma is extremely rare. We report two cases of non-functioning pituitary carcinoma treated in our hospital. The two female patients, aged 39 and 40 years, were diagnosed by CT and MRI and postoperative pathological examination. The tumor was surgically resected followed by stereotactic radiotherapy. Despite of the treatments, both of the patients showed tumor recurrence and died at 1 and 2 months after discharge. The diagnosis of this disease relies on a combined evaluation of the clinical, radiological, cytological, immunohistochemical features. Due to the highly malignant nature and poor prognosis of this disease, early surgical intervention and systemic evaluation of the patients' condition are imperative. Currently more effective therapeutic approaches are needed to control the tumor progression.

Protein & Cell 2015;6(8):599-609
The mechanisms that specify and maintain the characteristics of germ cells during animal development are poorly understood. In this study, we demonstrated that loss of function of the zinc-finger gene lsy-2 results in various somatic cells adopting germ cells characteristics, including expression of germline-specific P granules, enhanced RNAi activity and transgene silencing. The soma to germ transformation in lsy-2 mutants requires the activities of multiple chromatin remodeling complexes, including the MES-4 complex and the ISW-1 complex. The distinct germline-specific features in somatic cells and the gene expression profile indicate that LSY-2 acts in the Mec complex in this process. Our study demonstrated that lsy-2 functions in the maintenance of the soma-germ distinction.

Chinese Journal of Hepatology 2018;26(9):660-665
Objective:
To investigate the action and antioxidant effects of CB2 agonist AM-1241 on rat hepatic stellate cell line (HSC-T6).
Methods:
HSC-T6 was randomly divided into four groups: control group, oxidative stress group, AM-1241 intervention group and AM-1241+AM-630 antagonist group. Survival rate of HSC-T6 was detected by thiazolyl blue assay under 24 h interventions with 0, 20, 50, 80 μmol/L AM-1241 and 0, 10, 20, 30, 40 μmol/L AM-630, respectively. Besides control group, the remaining groups were well cultured in low-glucose DMEM containing 100 mU/L glucose oxidase (GO) for 12 h to prepare the oxidative stress model. Then, AM-1241 intervention group was treated with 50 μmol/L low-glucose DMEM medium. After incubation for 12 h, the AM-1241+AM-630 antagonist group was treated with CB2 antagonist AM-630 (20 μmol/L) for 2 h, and cultured with 50 μmol/L AM-1241 in complete low-glucose medium for 12 h. The optimal drug concentration was selected according to the cell viability considered by the experiment results. Type III collagen (C III) content in the HSC-T6 supernatant was detected by enzyme-linked immunosorbent assay. Glutathione (GSH) content in HSC-T6 was detected by spectrophotometry. CB2 and heme oxygenase-1(HO-1) in each group of HSC-T6 were detected by western blotting.
Results:
HSC-T6 proliferation was inhibited in each group of AM-1241 in a concentration-dependent manner (

Objective To explore the efficacy of blunt separation method in midline catheter intubation in elderly patients with coagulation dysfunction. Methods A total of 80 elderly patients with coagulation dysfunction were selected in the convenience sampling method from October 2022 to April 2023 in our hospital, and they were randomly divided into an experimental(blunt)group and a control(routine)group, with 40 patients in each group. The differences in the degree of bleeding and exudation at the puncture site, the pain score and the incidence of complications(including bleeding and exudation, phlebitis, symptomatic catheter-related thrombus, catheter blockage, catheter pulling-off)were compared between 2 groups. Results In the experimental group, the degree of bleeding and exudation at the puncture point immediately after the operation, degree of bleeding and exudation at the puncture point 24 hours after the operation, pain score 1 day after the catheterization, pain score 3 days after the catheterization, incidence of bleeding and exudation, total incidence of complications and maintenance times were significantly lower than these in the control group(P<0.05).In terms of the pain score immediately after the operation, pain score 5 days after the operation, incidence of phlebitis, incidence of symptomatic catheter-related thrombosis, incidence of catheter blockage, incidence of catheter pulling-off,incidence of catheter related skin injury, incidence of unplanned extubation, success rate of one-time sheath delivery and the indwelling time, the differences between the experimental group and control group were not significant(P<0.05).Conclusion The application of blunt separation method in midline catheter indwelling can significantly reduce the incidence and degree of bleeding at the puncture point, decrease the maintenance times and relieve the pain in elderly patients with coagulation dysfunction.
