1.Effects of combination of tolterodine and tamsulosin on BPH induced LUTS in rats

Objective To investigate the efficacy of tolterodine plus tamsulosin in rats with benign prostatic hyperplasia(BPH)induced low urinary tract syndrome(LUTS).Methods Male rats were castrated,then testosterone propionate were given by subcutaneous injection to induce BPH/LUTS model.Model rats were intragastrically administrated with 0.4mg/kg of tolterodine,0.04mg/kg of tamsulosin or(0.4+0.04)mg/kg of tolterodine plus tamsulosin for 14 days and urodynamics,residual urine volume and volume of bladder were mearsured.Results Tolterodine plus tamsulosin could simultaneously shorten the micturition duration,prolong the interval duration between micturition,decrease the threshold intravesical pressure at micturition and the max intravesical Pressure at micturition significantly.The combined-therapy method could also decrease the residual urine volume to a certein extent and had no obvious influence on enlarging the volume of bladder(P<0.05 or P<0.01).Conclusion Combination treatment with tolterodine and tamsulosin may efficiently ameliorate bladder outlet obstrution and overactive bladder simultaneously,with no extra danger of uroschesis.
2.Experimental studies on microwave-assisted extraction of baicalin from root of Scutellariae baicalensis

Object To study the microwave assisted procedure for the extraction of baicalin from root of Scutellariae baicalensis Georgi Methods An MSP 100D domestically made microwave sample preparation system with a maximum power of 850 watts was used. The effects of solvents, pressure/temperature of solvents and microwave radiation time on the yield of baicalin were studied by orthogonal experimental design. Results The optimal experimental conditions were extraction at 70% of microwave power with 30 times of 35% ethanol at a constant heating pressure/temperature of 0.15 MPa for 30 s. Conclusion The microwave assisted extraction not only takes a shorter time with better parallel results, but also gave an increased yield of about 10% as compared with ultrasonic extraction.
3.Contents Determination of Five Components in Qinghou Liyan Granule by UPLC

OBJECTIVE:To establish a method for the contents determination of gallic acid,naringin,hesperidin,neohesperi-din and baicalin in Qinghou liyan granule. METHODS:UPLC was performed on the column of ACQUITY UPLC BEH C18 with mobile phase of acetonitrile- 0.1% phosphoric acid(gradient elution)at a flow rate of 0.40 ml/min,detection wavelength was 280 nm,column temperature was 30 ℃,and injection volume was 1 μl. RESULTS:The linear range was 0.048-0.480 μg for gallic ac-id(r=0.999 3),0.012-0.120 μg for naringin(r=0.999 9),0.016-0.160 μg for hesperidin(r=0.999 8),0.022-0.220 μg for neo-hesperidin(r=0.999 9)and 0.072-0.720 μg for baicalin(r=0.999 2);limit of quantitation was 2.4,1.6,1.8,1.8,1.6 ng,limit of detection was 7.5,5.0,5.6,5.6,5.0 ng;RSDs of precision,stability and reproducibility tests were no lower than 3.0%;recover-ies were 96.64%-102.02%(RSD=2.00%,n=6),95.86%-102.56%(RSD=2.86%,n=6),97.24%-102.54%(RSD=2.10%,n=6),97.44%-102.60%(RSD=2.40%,n=6)and 96.91%-103.13%(RSD=2.62%,n=6). CONCLUSIONS:The method is rapid and accurate,and can be used for the simultaneous determination of gallic acid,naringin,hesperidin,neohesperidin and baicalin in Qinghou liyan granule.
4.Synthesis and anti-platelet aggregation activities of ferulic acid esters

Aim: To study the synthesis and anti-platelet aggregation activities of ferulic acidic esters so as to search for novel antithrombus agents. Methods: Based on prodrug strategy and combination principles, monoesters were first synthesized by reaction of ferulic acid with alcohols, and then the monoesters were coupled with aspirin to afford bis-esters. The target compounds were assayed for anti-platelet aggregation activities in vitro. Results: Sixteen target compounds and eight sideproducts including 15 new compounds were synthesized and their struc-tures were determined by IR, MS and ~1H NMR. The results demonstrated that some tested compounds exhibited potential anti-platelet aggregation activities. Conclusion: The bis-esters of ferulic esters coupling with aspirin with 4 to 5 carbons in side chain might be used as lead compounds for further study in searching for novel antithrom-bus agents.
5. Treatment of avascular necrosis of femoral head in patients with human immunodeficiency virus infection by cementless total hip arthroplasty

Objective: To investigate the effectiveness of cementless total hip arthroplasty (THA) in treatment of avascular necrosis of the femoral head (ANFH) in patients with human immunodeficiency virus (HIV) infection. Methods: The clinical data of 22 patients (35 hips) with HIV infection and ANFH (positive group), who were treated with cementless THA between February 2014 and January 2017, was retrospectively analyzed. Tirty-three cases (43 hips) without HIV infection, who were treated with cementless THA during the same period, were collected as control (negative group). There were significant differences in the age, gender, incidence of bilateral ANFH, and disease duration between 2 groups ( P<0.05). There was no significant difference in the Ficat stage of ANFH between 2 groups ( P>0.05). The operation time, intraoperative blood loss, rate of blood transfusion, postoperative complications, and hospitalization time were recorded. The hip joint function was evaluated by Harris score and X-ray film was performed to observe the position of the prosthesis. Results: The follow-up time was 13-24 months (mean, 20.2 months) in positive group and 14-25 months (mean, 21.6 months) in negative group. The operation time of negative group was significantly shorter than that of positive group ( t=3.677, P=0.001). However, there was no significant difference in intraoperative blood loss, rate of blood transfusion, and hospitalization time between 2 groups ( P>0.05). There was no significant difference in Harris score between 2 groups before operation and at 1, 3, 6, and 12 months after operation ( P>0.05). There were 2 cases of superficial infection of incision and 1 case of pulmonary infection during hospitalization in positive group, and 1 case of superficial infection of incision and 1 case of venous thrombosis of lower extremity in negative group. No death or loosening and sinking of the prosthesis occurred in both groups during follow-up. Conclusion: Cementless THA is a safe and effective method for ANFH in patients with HIV infection.
6.Interaction of nourishing and tonifying blood effects of the combination of Angelicae sinensis Radix and Astragali Radix studied by response surface method.

The combination of Angelicae sinensis Radix (Danggui, DG) and Astragali Radix (Huangqi, HQ) is a popular herb pair commonly used in clinic for the treatment of blood deficiency syndrome in China. The aim of this paper is to study the interaction of DG and HQ nourishing and tonifying blood effects by response surface method. The blood deficiency mice were induced by injecting N-acetylphenylhydrazine (sc) and cyclophosphamide (ip). The blood deficiency mice were administrated intragastrically with DG-HQ extracts (0:1, 1: 5, 2:5, 2:3, 1:1, 3:2, 5:2, 5:1, 1:0). The changes of the peripheral blood indexes and organ indexes were observed. The indexes were integrated by comprehensive index method; the interactions of DG and HQ were analyzed by the response surface diagram established with Matlab software. The results showed that DG and HQ at most of their combination ratios had synergic effect. Within the range of 1:5 - 5:1, all of the extracts of DG-HQ showed synergic effect, and among which, high-doses had better effects than low-doses. The highest value (-1) of the synergic effect was showed when DG was 10 - 40 g at the same time of HQ as 90 -180 g, and DG was 50 - 100 g at the same time of HQ as 20 - 100 g. DG-HQ at all combination dosages within Chinese Pharmacopeia (DG: 6 - 12 g, HQ: 9 - 30 g) had certain synergic effect, and Danggui Buxue Decoction (DG: 6 g, HQ: 30 g) also was at this range. The results provided scientific basis to the clinical application of DG and HQ. And the response surface method was firstly applied to quantitatively evaluate the bio-activity change of herb combination, which provided a novel way for modern basic research on the interaction of herbs.

The heterogeneity of high-density lipoprotein-cholesterol (HDL-C) in hyperuricemia was analyzed in Uygur,Han,and Kazak ethnic groups in Xinjiang.A total of 2 396 subjects were surveyed.The HDL-C levels were significantly decreased in hyperuricemia group compared with control group in Han and Uygur,while the reverse occurred in Kazak.The serum uric acid (SUA) level in Kazak was the lowest.The SUA was negatively correlated with HDL C in Kazak (P < 0.05).HDL-C was a recognized protective factor for cardiovascular disease.With the lowered SUA level in the Kazak group,HDL-C level was increased.When SUA level was higher than a certain level (such as the SUA-4),there existed a negative impact on cardiovascular disease
8.Comparing tonifying blood effects of Danggui-Honghua with different proportions on blood deficiency mice

Aim To observe the differences of tonify-ing blood effect for the combination of Danggui and Honghua ( GH) with different proportions on blood de-ficiency mice, and choose the proportion of GH which has the optimal tonifying blood effect. Methods The blood deficient mice model was induced by injection of phenylhydrazine and cyclophosphamide. On the basis of multi-attribute comprehensive index method, the op-timal dose was determined through three doses of GH 1: 1 , which was the highest frequency proportion of GH used in the “Dictionary of Chinese Medicine Prescrip-tions”. According to the optimal dose, the change reg-ulation of tonifying blood effect of GH with different proportions was observed. Results Among three do-
ses (1, 3 and 5 times of clinical dose), the tonifying blood effect of GH was best when the dosing concentra-tion of GH was clinical dose. Among nine proportions (1 : 0, 4 : 1, 2 : 1, 3 : 2, 1 : 1, 2 : 3, 1 : 2, 1 : 4 and 0 : 1), GH 1 : 1 had the best effect. Conclusion The results are consistent with regulations of medi-cine usage that GH 1 : 1 has the highest frequency used in Herbal Formulae, which could provide scientif-ic basis for more effective application of Danggui and Honghua in modern clinic medicine.

10.The expression and significance of MAPK8 gene in oral squamous cell carcinoma

Objective: To study the the expression and significance of MAPK8 gene in oral squamous cell carcinoma (OSCC) .Methods: 17 cases of oral normal tissues and 42 of OSCC tissues were collected. The relative expression of MAPK8 mRNA was detected by Real-Time PCR, the expression of MAPK8 protein was detected by immunohistochemical SP method. Results: Over expression of MAPK8 mRNA and protein was observed in OSCC (P < 0. 05), the expression was correlated with the differentiation (P < 0. 05) and clinical stage (P < 0. 05) of the lesion, but not with the age (P> 0. 05) and gender (P> 0. 05) . Conclusion: MAPK8 gene may play an important role in the development of OSCC.
