1.The prediction value of cone margin status for risk of residual focal in patients with cervical adenocarcinoma in situ

Objective To observe the clinical value of the cervical conization operation margin in predicting residual focal in patients with adenocarcinoma in situ (ACIS).Methods 60 ACIS patients were reviewed,and postoperative specimens were analyzed.Negative resection margin was defined as:no residual tumor epithelial cell was seen in operation margin around,and normal cervical epithelial cells can be visible.Relations between the margin status and incidence of residual were analyzed.Results The margin positive rate in LEEP was 56.8%,which was significantly higher than that in CKC(26.1 %,χ2 =5.390,P =0.02).The corrected odds was 4.31 (OD =4.31 , 95%CI 1 .1 3 -1 6.43,P =0.03).None had no residual tumor in 26 cases with margin negative,and 1 7 cases (65.4%)were detected with residual tumor in 26 positive margins (χ2 =25.257,P <0.01 ).Conclusion The ACIS patients with negative margins have a lower risk for residual focal.The standard sampling and judging methods in evaluating margin criteria can enhance the predictive value of margins.
2.Treatment opportunity for early childhood caries

Objective:To estimate the effect of treatment opportunity for severe caries in primary molars on child dental fear and dent-al behavior.Methods:288 children aged 3 -6 years with primary molars caries were included on the first visit.The children in the ex-perimental group(n =144),with obvious clinical symptom,were initiative to see a dentist.Those in control group(n =144)without evident symptom were found with caries in conventional dental examination.The change of the children fear before and after the first treatment was assessed by MCDASf within the first week,the change of children's clinical behavior in their first and return visit was esti-mated by Venham Clinical Anxiety and Cooperation Behavior Measurement Scale.Data was statistically analyzed.Results:After the first treatment and during the return visit,the MCDASf average score of the experimental group was decreased(P <0.05),that of the control group was not(P >0.05).On their return visit,the behavior of the 2 groups was similar (P >0.05),and which was ameliora-ted in the experimental group during their own first and return visit,compared with the control group(P <0.05).Conclusion:Treat-ment of severe caries before acute symptom arising in young children may alleviatie the child dental fear.
3.Protective effects of fenofibrate on insulin resistance in mice of nonalcoholic fatty liver disease and its partly mechanisms of action

Objective To study whether the protective effects of fenofibrate on insulin resistance in mice of nonal-coholic fatty liver disease(NAFLD) are related to endoplasmic reticulum stress (ERS). Methods Male C57BL/6 mice were fed with high-calorie and high-cholesterol diet ( HCD) to induce a model of NAFLD, then one of those two groups from HCD was treated with fenofibrate 40 mg/( kg · d ) for 2 weeks ( HCF ) . Glucose tolerance test (GTT) and insulin tolerance test (ITT) were used to analyze the improvement on insulin resistance. The serum levels of triglyceride ( TG) , total cholesterol ( TC) , high density lipoprotein-cholesterol ( HDL-C) , low density lip-oprotein-cholesterol ( LDL-C) , alanine aminotransferase ( ALT) and aspartic transaminase ( AST) were detected. The pathological changes of livers were detected with HE and Oil Red O staining, the mRNA and protein expression of peroxisome proliferator-activated receptorα( PPARα) , glucose regulated protein 78 ( GRP78 ) and transcription factors GADD153 ( CHOP) were detected with real-time quantification PCR and Western blot analysis respectively. Results Compared with the SCD mice, the HCD mice showed significant insulin resistance, higher serum levels of TG, TC and LDL-C (P<0. 01, P<0. 05), lower serum level of HDL-C (P<0. 01), micro-and macrovesicular steatosis, ballooned hepatocytes and infiltration of inflammatory cells were observed in the liver, the expressions of PPARα and GRP78 at mRNA and protein levels were decreased (P<0. 05), however, the expressions of CHOP at mRNA and protein levels were increased ( P<0 . 05 ) . Fenofibrate intervention significantly improved insulin resist-ance and decreased the serum level of TG ( P<0. 05 ) . Fenofibrate also alleviated hepatic steatosis and inflammato-ry infiltration, and increased the mRNA and protein expressions of PPARα and GRP78, decreased the mRNA and protein expression of CHOP ( P<0 . 05 ) . Conclusion Fenofibrate significantly improves insulin resistance and de-creases the serum level of TG in NAFLD mice, which may be related to activating PPARα and relieving ERS.
4.Clinical observation on treatment of acute exacerbation of chronic respiratory failure with conventional western medicine therapy combined with Danhong injection

ObjectiveTo study the clinical effect of treating acute exacerbation of chronic respiratory failure with Danhong injection.MethodsAccording to random number table,64 cases of acute exacerbation of chronic respiratory failure were divided randomly into 2 groups,with 32 cases in each group.Both groups were given correspondingly conventional symptomatic treatment(including oxygen,anti-inflammatory,antispasmodic,sputum,etc.).On this basis,the treatment group was added Danhong injection 30 ml+5%glucose infusion 250 ml,once a day,continuous therapy for 14 days.ResultsThe total effective rate was 87.50% and 65.63% in Danhong injection treatment group and conventional treatment group respectively,showing obvious difference between 2 groups (x2=4.267,P<0.05).The blood low shear viscosity (7.11 ± 1.60) mPa ? s,high shear viscosity of whole blood (3.30 ± 0.83 ) mPa ? s,plasma viscosity ( 1.28 ± 0.32) mPa ? s,whole blood reduced viscosity ( 16.81 ± 3.32 ) mPa ? s,erythrocyte aggregation index ( 1.26±0.31 ),erythrocyte deformation index (0.64±0.27) in the Danhong injection treatment group all had statistical significance compared to the conventional treatment group(P<0.05).ConclusionClinical efficacy and safety of Danhong injection in treating acute exacerbation of chronic respiratory failure were good.

Objective To investigate the clinical effect of oxytocin combined with misoprostol in preventing postpartum hemorrhage of cesarean section.Methods 86 patients with cesarean section who were treated and diagnosed in our hospital from March 2016 to October 2016 were randomly divided into experimental group and control group according to the time of hospitalization.The patients were randomly divided into experimental group and control group.43 cases.Preoperative two groups of patients with different treatment methods for science introduction, so that pregnant women mental stress to relax, ease the heart anxiety and nervous mood.On the basis of conventional treatment, the experimental group received the uterine injection of oxytocin, while the patients under the tongue to take drugs;the control group of patients with oxytocin injection for preventive treatment.Results In the two groups, 32 cases were effective in the experimental group, 1 case was ineffective, the effective rate was 97.67%, compared with 10 cases in the control group, 16 cases were ineffective, the total effective rate was 62.79 % was statistically significant(P<0.05).Conclusion Oxytocin combined with misoprostol has a good therapeutic effect on the postpartum hemorrhage in the prevention and treatment of uterine bleeding.Compared with the original conventional treatment, there is a high efficiency, and the patient's comfort is Higher to improve, to a certain extent, is the more excellent clinical treatment.
6.Expression of IL-29 cDNA in cos-7 cells and its anti-HBV activity in vitro

Objective: To clone cDNA of human interleukin-29(IL-29) and express it in cos-7 cells, and to study its anti-HBV activity in vitro. Methods: Total RNA was extracted from PBMCs which had been infected with vesicular stomatitis virus(VSV) in vitro.IL-29 cDNA was amplified using one-step RT-PCR technique. The recombinant expressing plasimd of psectagB/His-myc-IL29 was constructed by inserting IL-29 cDNA into the vector and was then transfected into cos-7 cells. Stable expression stains were screened by Hygromycin B and limited dilution method. The target protein was purified through Ni2+-chelating Sepharose Fast Flow. Anti-viral bioactivity of the recombinant IL-29 fusion protein was analyzed through an in vitro model of production of HBV by the HepG2.2.2.15 cell line.ELISA was used for detection of the viral titers in the cell cultural supernants. Results: IL-29 was cloned and stably expressed in cos-7 cells successfully. SDS-PAGE and Western blot analysis showed multiple bands of the target protein with the molecular weights between 20 000 and 33 000, and the major band was located at about 33 000, indicating the fused IL-29 modified by additional glycosylation. The rhIL-29 was shown to dose-dependently inhibit secretion of HBsAg and HBeAg accompanied by the reduction of HBV genomic DNA in the cells tested. The inhibition ratio of HBsAg and HBeAg production was attained 85% at a concentration of 160 ?g/L of rhIL-29. Conclusion: The rhIL-29 with anti-HBV activity has been obtained.
7.Anti-inflammatory Effect and Underlying Mechanism of Ethanol Extracts of Ageratum Conyzoides. L. in Guangxi

Objective:To explore the anti-inflammatory effects and the underlying mechanisms of ethanol extract of Ageratum cony-zoides. L. from Guangxi. Methods:The auricle edema model was induced by dimethylbenzene in the mice and the paw edema model was induced by carrageenan respectively in the mice and rats to study the anti-inflammatory effects of ethanol extract of Ageratum cony-zoides. L. from Guangxi. The content of malondiadehyde (MDA) and proateglandin E2 (PGE2), and the activity of superoxide dis-mutase( SOD) in the mouse edema paw was measured. The contents of tumour necrosisfactor-α ( TNF-α) , interleukin-1β ( IL-1β) and interleukin-6(IL-6) in the rat serum were detected as well. Results:Compared with the model control group, the ethanol extracts of Ageratum conyzoides. L. from Guangxi could remarkably inhibit auricle edema in the mice and paw edema in the mice and rats( P<0. 05 or P<0. 01), the inhibition ratio for high, medium and low dosage group(6. 0, 3. 0, 1. 5 g·kg-1)was 29. 24%,16. 42% and 11. 21% in the auricle edema mice and 28. 66%,18. 79% and 13. 13% in the paw edema mice , respectively. It could remarkably re-duce MDA and PGE2 content and enhance the activity of SOD in the mouse inflammatory tissue(P<0. 05 or P<0. 01). In the paw e-dema rats, the inhibition ratio for high, medium and low dosage group(4. 5,2. 3, 1. 2 g·kg-1)was 43. 69%, 36. 01% and 23. 29%at the 3rd h, respectively , and it also could remarkably reduce serum TNF-α, IL-1βand IL-6 content(P<0. 05 or P<0. 01). Con-clusion:The ethanol extracts of Ageratum conyzoides. L. from Guangxi show significantly anti-inflammatory effects, and the mecha-nisms may be related to the ability of scavenging oxygen free radicals and reducing the release of inflammatory cytokines and proinflam-matory cytokines.
8.Transcriptional expression and promoter hypermethylation of RAS association domain family 1A gene in glioma

It was not found the promoter methylation in normal brain tissues and U251 cell line. In 11 patients with the aberrant promoter methylation, five showed the gene inactivation (P = 0. 022). Conclusion Aberrant promoter methylation was found in glioma. Although the gene inactivation of RASSF1 A was not so common in glioma, its expression was lower in glioma than in normal brain tissues. Promoter methylation may contribute to the low level or loss of RASSFT A mRNA expression.
9.Simvastatin improves high-fat diet-induced kidney damages by increasing expression of adiponectin and its receptors

Objective To probe the new mechanism of simvastatin on high-fat diet-induced kidney dam-age. Methods Female SD rats were subjected to a standard control diet(SCD)or high-fat diet(HFD)for 20 weeks,then the HFD group was randomly divided into HFD group and HFD group with simvastatin treatment (HFD+ST,10mg·kg-1·d-1 )for another 8 weeks. The expression of adiponectin,adiponectin receptors R1 and R2, Adenosine 5′-monophosphate(AMP)-activated protein kinase(AMPK),peroxisome proliferator-activated receptorα(PPARα),glucose regulated protein 78(GRP78)and GADD153(CHOP)in kidney were assessed respective-ly. Results Body weight and serum lipid levels in HFD group significantly increased ,expression of adiponectin and its receptors significantly down-regulated. Phosphorylation of AMPKα and PPARα expression decreased,and expression of GRP78 and CHOP up-regulated significantly. Above indexes in simvastatin treatment groups improved significantly. Conclusion Simvastatin can improve high-fat induced kidney damages ,probably by increasing expression of adiponectin and its receptors ,decreasing endoplasmic reticulum stress.
10.Influences of fenofibrate on oxidative stress and endoplasmic reticulum stress in livers of hyperlipidemic rats

Objective To study the influences of fenofibrate on oxidative stress and endoplasmic reticulum stress in livers of hyperlipidemic rats. Methods Male SD rats were fed with high-fat diet for 12 weeks to induce a model of hyperlipidemia, then divided into control group, high-fat diet group with another four-week high-fat diet and fenofibrate group, in which rats were treated with fenofibrate [100 mg / (kg·d)] for 4 weeks. Then improvement of insulin resistance was detected in rats with GTT and ITT experiment. The serum levels of glucose (GLU), fasting insulin (FINS),triglyceride (TG) and total cholesterol (TC) were detected. The pathological changes of livers were detected with Oil Red O staining. The oxidative stress indices such as T-SOD, Mn-SOD, GSH and T-AOC were detected with liver homogenate. The expression of GRP78 was detected with real-time quantification RT-PCR and Western blot analysis respectively. Results Compared with rats with high-fat-diet, rats after fenofibrate treatment showed obviously improved insulin resistance, lower serum level of TG, TC and FINS (P < 0.05), decreased number and size of lipid droplets in liver tissue sections. T-SOD level in liver homogenate was significantly increased (P < 0.05), while GSH and T-AOC levels increased but had no obvious differences when compared with control group (P > 0.05). The expression of GRP78 at mRNA and protein levels were increased significantly after fenofibrate treatment (P < 0.05). Conclusions Fenofibrate has significant effects on improving insulin resistance and lipid regulation, which might be related to decreased oxidative stress and subsequent endoplasmic reticulum stress.
