World Science and Technology-Modernization of Traditional Chinese Medicine 2014;(12):2731-2734
This study was aimed to establish a method for the determination of octylacetate in Jing-Fu-Kang (JFK) granules. The content was determined by GC with Agilent HP-5 (30 m × 0.32 mm × 0.25μm) capillary column, with hydrogen flame ionization (FID) as detector. The injector temperature was 220℃. The detector temperature was 260℃. The split ratio was 20:1. At first, the temperature should be maintained to 80℃for 2 min, and then heating up to 200℃with the speed of 10℃·min-1. This temperature was maintained for 10 min. The results showed that the good linear relationship of octylacetate was within the range of 0.156 5-7.824 2 mg·mL-1, r=0.999 9. The average recovery rate was 100.00%, and RSD was 1.91%(n=9). The lowest detection limit of octylacetate was 1.633 6 ng. It was concluded that the established method was simple and reproducible with no interference. It can be applied to the quality control of JFK granules.

BACKGROUND:The regulatory role of extracellular signal regulated kinase 1/2 (ERK1/2) and nuclear factor kappa B (NF-κB) signal pathways in the osteogenic differentiation of MC3T3-E1 cells subjected to mechanical strain remains unclear.
OBJECTIVE:To investigate the effects of ERK1/2 and NF-kB signal pathway on alkaline phosphatase, type Ⅰcol agen, osteocalcin and interleukin-6 expression in osteoblasts in response to mechanical strain, and to explore the regulatory effects of ERK1/2 and NF-kB signal pathway on osteoblast differentiation.
METHODS:MC3T3-E1 cells cultured in vitro were separately treated with ERK1/2 pathway specific inhibitor PD098059 and NF-kB pathway inhibitor PDTC for 30 minutes, and subjected to12%elongation for 24 hours. Normal cells and cells along loading 12%mechanical strain for 24 hours were considered as controls. Enzyme linked immunosorbent assay and real-time PCR were utilized to detect alkaline phosphatase activities, type Ⅰcol agen, osteocalcin and interleukin-6 mRNA expression before and after cellloading.
RESULTS AND CONCLUSION:Under 12%mechanical strain, alkaline phosphatase, type I col agen, and interleukin-6 expression was regulated by ERK1/2 signal pathway in MC3T3-E1 cells, but osteocalcin gene expression was not affected by ERK1/2 pathway. NF-kB signal pathway inhibitor PDTC significantly suppressed alkaline phosphatase activities in MC3T3-E1 cells under mechanical strain, and inhibited interleukin-6 gene expression. However, type I col agen and osteocalcin gene expression was not affected by NF-kB signal pathway. Results suggested that mechanical strain affected osteogenic differentiation and relevant gene expression in MC3T3-E1 cells by ERK1/2 and NF-kB signal pathway.
5. Advances of "help-me" signals from neurons after focal brain injuries
Chinese Journal of Cerebrovascular Diseases 2019;16(9):495-499
Injured neurons will send "help-me" signals to nearby cells after focal injuries. These neurons will be protected by proliferation of neural progenitors, transformation of microglia, and proliferation of endothelial cells. This article provides an in-depth review of the latest research on endogenous " helpme" signals after neuronal injury, especially after cerebral ischemia injury was made, suggesting findings of endogenous "help-me" signals after neuronal injury and its usage as a novel target for intervention may provide new ideas and methods for neuroprotection researches.

In recent years, for patients with acute ischemic stroke due to large vessel occlusion, intravascular mechanical thrombectomy has become the most recommended treatment method. Determining whether there is ischemic penumbra or not is very important for the selection of patients who are suitable for endovascular mechanical thrombectomy. In addition, the ischemic penumbra also provides guidance for extending the time window of traditional intravenous thrombolytic therapy. This article reviews the imaging evaluation methods of ischemic penumbra.

Objective:
Many physicians and patients still have concerns about the safety of breastfeeding in mothers infected with hepatitis B virus; we evaluate the safety of the newborn and the women with HBeAg positive and high viral load, who received nucleoside analogues to block maternal to child transmisssion and selected postpartum breastfeeding after drug discontinuance.
Methods:
This prospective, observational study enrolled 60 HBeAg positive patients and HBV-DNA >2*10E+ 5 IU/ml, all patients started antiviral treatment for blocking maternal to child transmission at 24-28 weeks of pregnancy. All the newborns received the active-passive immunization therapy with hepatitis B immunoglobulin (HBIG) and HBVac. After the delivery, patients with normal liver function discontinued the antiviral drug and selected breastfeeding voluntarily. The safety of breastfeeding were compared with patients selected artificial feedings, they were followed up for 7 months. Primary measurements were the proportion of mothers with abormal liver function after stopping the drug and the level of newborn’s anti-HBs at 6 months of age; secondary measurements were the positive rate of neonatal HBsAg and the HBV-DNA value of the patients at 6 weeks postpartum.
Results:
From December 1, 2015 to May 1, 2017, 415 patients were enrolled in Beijing You’an Hospital Affiliated to the Capital Medical University and all these patients were born following full-term single-child pregnancy. After the delivery and drug withdrawal, there was no significant difference in the incidence of ALT elevation between the breast fed group and the artificially fed group: 29 /220 versus 30/195, (χ2=0.411,

Objective To investigate the neuroprotective effect and its mechanism of local hypothermia combined with nicotinamide phosphoribosyltransferase (NAMPT) on cerebral ischemia-reperfusion injury in rats. Methods Fifty-four Sprague-Dawley rats were randomly divided into sham operation group, model group,NAMPT group, local hypothermia group, and combined treatment group (NAMPT + local hypothermia).A rat model of local cerebral ischemia-reperfusion was induced by suture method.Infarct volume and cerebral edema volume were assessed by 2,3,5-triphenyltetrazolium chloride staining after 2 h cerebral ischemia and 24 h reperfusion in rats.Evans blue staining was used to assess the extent of blood-brain barrier damage, and a 12-point scale was used to assess neurological deficits. Results The infarct volume in the local hypothermia group,NAMPT group, and combination treatment group was significantly lower than that in the model group (all P <0.05).The infarct volume in the combination treatment group was significantly lower than that in the NAMPT group (P <0.05).The infarct volume in the combination treatment group was lower than that in the local hypothermia group, but it did not reach statistical significance. The neurological function scores of the local hypothermia group,NAMPT group, and combination treatment group were significantly lower than those of the model group (all P <0.05).The score of the combined treatment group was significantly lower than that of the NAMPT group and the local hypothermia group (all P<0.05).Evans blue leakage in the local hypothermia group,NAMPT group, and combination treatment group was lower than that in the model group (all P <0.05),but the differences between each treatment group were not statistically significant. Conclusion NAMPT and local hypothermia combination therapy showed better neuroprotective effects on cerebral ischemia-reperfusion injury, suggesting that the combination therapy had clinical transformation prospects.
