1.Effect of transcutaneous electrical acupoint stimulation on stress reaction in elderly patients with intravenous patient-controlled analgesia after total hip replacement operation

Objective To observe the effect of transcutaneous electrical acupoint stimulation on stress reaction in elderly pa-tients with intravenous patient-controlled analgesia after total hip replacement operation.Methods A total of 60 cases of patients with patient-controlled intravenous analgesia spinal anesthesia after total hip replacement surgery were randomly divided into exper-imental group (n=30)and the control group (n=30).The experimental group respectively at the completion of surgery,postopera-tive 4,24,48 h was treated by transcutaneous electrical acupoint stimulation.The serum cortisol and blood glucose was detected in two groups at preoperative 30 min (T1 ),30 min after operation (T2 ),1 d after operation in the morning (T3 ),2 d after operation (T4 ).The pain scores,sedation scores,adverse reactions and analgesic dosage were investigated in two groups after operation.Re-sults In the experimental group,cortisol and blood glucose levels increased compared T2 and T1 (P <0.01 ).Compared with the control group,cortisol and blood glucose levels in the experimental group decreased at T3 and T4 (P <0.05 ).Compared with the control group,the VAS scores was significantly different in the experimental group(P < 0.01 ).The side effects in experimental group were significantly lower than those in the control group(P <0.05).After TEAS treatment,the physic liquor consumption and press the number decreased significantly (P < 0.05 ).Conclusion TEAS technology for elderly total hip replacement operation PCIA could effectively inhibit stress,enhance the analgesic effect,reduce or avoid the occurrence of adverse reactions.
2.Comparison of the results between two radiotherapeutic methods for cervical cancer patients

Purpose:To evaluate two different methods of radiotherapy, which affect the results of cervical cancers treated and complications of the rectum or bladder. Methods:From July 1991 to December 1996, 124 cases of cervical cancer were randomly allocated into two groups. Group A (59 cases:stage Ⅰ 1, Ⅱa 12, Ⅱb 30, Ⅲa 11, Ⅲb 5) received 45—55 Gy of external radiation(ER) to the parametrium and 50—65 Gy of intracavitary irradiation (4—5 Gy per fraction, 2 fractions per week). Group B(65 cases: stage Ⅰ 2, Ⅱa 15, Ⅱb 29, Ⅲa 10, Ⅲb 9) received 26—40 Gy ER to the whole pelvic and 30—40 Gy of intracavitary irradiation (4—5 Gy per fraction, 2 fractions per week), an additional 15—29 Gy ER to the parametrium. Results:In group A, 3 year and 5 year survival rates were 81.4% and 71.7%. In group B, they were 84.6% and 70.8%. 3.Application of refining management in sterilization and supply center

Objective To strengthen the management of contaminated area of sterilization and supply center, increase the cleaning quality of instruments and control nosocomial infection. Methods We executed the following measures: paying attention to the decontamination work, changing concepts, refining regulations, establishing programmed decontamination process, specification of work, thinning pest responsibility, strengthening infection mangement of contaminated area, effect examination and training and checking. Results The bacteria removing rate reached 95.3%. Conclusions Refining management could result in perfect cleaning effect which was applicable in the management of contaminated area of sterilization and supply center.
4.Design of a Detection System of Leaky Surface Acoustic Wave Biosensor in Liquid Phase and Test of HPV

OBJECTIVE To design the detection system of the leaky surface acoustic wave(LSAW) biosensor in(liquid) phase and detect human papilloma virus(HPV) target sequence with the LSAW biosensor.METHODS A detection delay line and a reference delay line were constructed,the phase of the two delay line changes was(evaluated) in liquid phase,and real time monitoring of HPV reaction was performed by using the LSAW biosensor system.RESULTS The phase of the two delay lines had different changes in liquid phase;HPV could lead to a great change of the phase in the detection delay line,and reference delay line had no change.CONCLUSIONS The(detection) system of the LSAW biosensor in liquid phase is successfully constructed and real time monitoring of(detection) of HPV is realized.
5.Effects of Aconite Alkaloids on Apoptosis and Cell Cycle of Human Gastric Cancer Cell Line SGC-7901

This study was aimed to investigate effect of aconite alkaloids on proliferation and apoptosis of hu-man gastric cancer cell line SGC-7901 . Effects of different concentrations of aconite alkaloids on proliferation of human gastric cancer cell line SGC-7901 were investigated with MTT assay; induced apoptosis and cell cy-cle blocking were detected with flow cytometer ( FCM ) . The results showed that the IC50 of aconite alkaloids on human gastric cancer cell line SGC-7901 was 0 . 2318 ± 0 . 0022 , 0 . 1601 ± 0 . 0227 , 0 . 1031 ± 0 . 0231 mg/ml at 24 h , 48 h and 72 h , respectively , compared to the control group with significant difference ( P <0.01). When the aconite alkaloids concentration was 0.8 mg/ml, it appeared with obvious apoptosis. The apop-tosis rate was ( 59 . 38 ± 5 . 05 )%. The FCM detection showed that compared with control group , the percentage of S-phase cells increased in the treatment group . And a typical sub-diploid peak appeared before G0 / G1 phase . It was concluded that aconite alkaloids can inhibit the proliferation of human gastric cancer cell line SGC-7901 in vitro, induce the apoptosis of cells and make SGC-7901 cells remain in S phase.
6.Desferroxamine preconditioning protects against hypoxia in neurons

Objective To investigate the possible effects and underlying mechanisms of desferroxamine (DFO) preconditioning against hypoxia in neurons. Methods Cortical neurons were cultured in DFO under ischemia condition of oxygen-glucose deprivation (OGD). Cell viability was determined by cell counting kit-8 (CCK-8) method; apoptotic cell ratio was examined with Hoechst 33342 staining; the morphological change was observed. Middle cerebral artery was occluded with or without DFO administration to establish the cerebral ischemia rat model. Infarct sizes were examined by TIC staining, and the neurological severity score was evaluated. Meanwhile immunofluorescent staining was employed to detect the protein synthesis of hypoxia inducible factor-1 (HIF-1) and erythropoietin (EPO), RT-PCR was performed to detect the mRNA expression of HIF-1 and EPO as well Results Neuronal viability kept in 49% (OGD group was 25%, t =8. 544, P<0. 05), the rate of apoptosis was 38% (OGD group was 30%, t = 4. 409, P <0.05 ) after administration of DFO (post-DFO) , the morphology of neurons improved. In the model of focal cerebral ischenfia of 30 mg/kg group, neurological severity score was reduced, the percentage of brain infarct decreased 8.5% (t=4.649, P<0.05) 3 days post-DFO(vs control). In the 100 mg/kg group, neurological severity score was 7.44 ±0.39 (t=2.903, P<0.05 ) ,5.60±0.47 (t=10.143, P < 0.01 ) ,6.97 ±0.73 (t=3.142, P<0.05 ), the percentage of brain infarct decreased 12. 0% (t=5.056, P<0.05), 32.3% (t =10.993, P<0.01), 10.6% (t =4.385, P<0.05)2,3 and7 days post-DFO(vs control), respectively. Immunofluorescent staining found synthesis of HIF-1α and EPO in cultured cortex neurons after DFO pretreated; HIF-1α and EPO were upregulated in the neurons of rat brain after DFO pretreated. The mRNA of HIF-1α and EPO upregulated in vivo and in vitro. Conclusion DFO preconditioning can protect the brain against ischemic damage, which is related to the protective effect on neurons. The mechanism of DFO preconditioning may be involved in the expression of HIF-1α and EPO in vivo and in vitro.
7.Study on Anti-hyperuricemia Effect of Geniposide

This article was aimed to study the effect of geniposide in hyperuricemia mice. Different doses (50, 100, 200 mg·kg-1) of geniposide were administrated to hyperuricemia mice induced by potassium oxonate by gavage for seven consecutive days. Then, serum uric acid (SUA), urinary uric acid (UUA) and the hepatic xanthine oxidase ac-tivities (XOD) of mice were detected. The results showed that the middle- and high-dose groups of geniposide re-duced the level of SUA significantly (P<0.01). It was concluded that geniposide can reduce the level of SUA in hy-peruricemia mice by promoting UUA excretion.
8.Primary results of the 3-in-1 technique of Disc-FX system for the discogenic low back pain

ObjectiveTo investigate the clinical efficacy of the 3-in-1 technique of Disc-FX system,namely:discectomy,radiofrequency ablation and annuloplasty for discogenic low back pain in the early stage.MethodsFrom February 2010 to February 2011,40 patients with the discogenic low back pain underwent discectomy,radiofrequency ablation and annuloplasty using Disc-FX system were retrospectively analyzed,including 22 males and 18 females with an average age of 38.7 years(range,32-58 years).The visual analogue scale (VAS) scoring was applied to evaluate the back and low limb pain at preoperative,postoperative 1week,3 months,6 months,and last follow-up.The Macnab score was also evaluated at last follow-up.Results All the patients were followed up for average 13.8 months (range,6-18 months).The operative time averaged 26 min(range,20-40 min).No surgical complication was found during the follow-up.The VAS of preoperative low back pain was 6.60±1.47,VAS score of low back pain at postoperative 1 week,3 months,6 months,and final follow-up were 1.05±0.68,1.15±0.70,1.00±0.62,0.95±0.63,respectively.The postoperative VAS of low back pain decreased significantly compared with that preoperative.The VAS of preoperative limb pain was 3.05±1.23,VAS score of limb pain at postoperative 1 week,3 months,6 months,and final follow-up after operation were 1.10±0.74,1.15±0.70,1.10±0.72,0.95±0.54,respectively.The postoperative VAS of limb pain decreased significantly compared with that preoperative.The evaluation of Macnab score were excellent in 20 patients,good in 17,fair in 2,and poor in 1,suggesting an effective rate of 92.5% (37/40).Conclusion Disc-FX system,which combined three surgical technique in one procedure,provides a new choice for orthopedics to treat discogenic low back pain.It can give satisfactory clinical outcomes during a short-term followup,but the long term clinical outcomes remains unknown.
9.Recombinant expression of Schistosoma japonicum fructose-1,6-bisphos-phate aldolase and its expression in different developmental stages of S. ja-ponicum

Objective To clone express and purify Schistosoma japonicum fructose?1 6?bisphosphate aldolase SjFBPA in E. coli and observe its expression in different developmental stages of S. japonicum. Methods FBPA gene was amplified from S. japonicum adult worm cDNA by using PCR. The amplified product was recombined into pET28a plasmid and inducibly expressed with IPTG in E. coli BL21. SDS?PAGE and Western blotting were employed to analyze and identify the recombinant protein SjFBPA rSjFBPA . Then rSjFBPA was purified by chromatographic purification and its purity was analyzed by SDS?PAGE. The protein concentration of rSjFBPA purified was measured by the BCA method. Furthermore SjFBPA mRNA was ana?lyzed in different developmental stages of S. japonicum by RT?PCR. Results SjFBPA was successfully amplified by using PCR and identified by restriction enzyme digestion and sequencing. The Western blotting analysis confirmed that the recombinant pro?tein could specifically reactive to the anti?His?tag monoclonal antibody. The concentration of the purified recombinant protein was about 4 mg/ml. The result of RT?PCR showed that SjFBPA mRNA was expressed in cercaria schistosomulum adult worm and egg of S. japonicum. Conclusion SjFBPA is successfully recombined and expressed in a prokaryotic system and SjFBPA mRNA is expressed in cercaria schistosomulum adult worm and egg of S. japonicum.
10.Protective effect of deferroxamine on glutamate induced neurotoxicity in cultured rat hippocampal neurons

Objective To investigate the protectve effects and underlying mechanisms of deferroxamine on glutamate-induced injury in cultured hippocampal neurons.Methods Primarily cultured hippocampal neurons from fetal rat were used in a model of glutamate induced neurotoxicity.There were two experimental groups.Neurons were pretreated with deferroxamine before glutamate in the deferroxamine group, and neurons were treated with glutamate only in the control group.The morphological change was examined under microscope.Hoechst 33342 DNA staining method was used to study the ratio of condensed nuclei.The levels of lactate dehydrogenase (LDH), malonaldehyde (MDA) and hydroxyl radical were determined using biochemistry.The change in calcium signal was detected using microfluorescent technique.Results The neurons pretreated by deferroxamine had intact morphology with the ratio of condensed nuclei at 14% ± 6% compared to 58% ± 6% (t= 8.98, P <0.01 ) in the control group.LDH level was (36.42 ± 8.99) U/L in the deferroxamine group and was (68.06 ± 11.26) U/L in the control group ( t =3.25,P<0.05).The respective levels of hydroxyl radical were (34.21 ±4.23) U/L and (47.06 ±8.79) U/L (t = 3.11, P <0.05 ).The respective levels of MDA were (12.26 ± 2.78 ) nmol/mg and (28.86±5.19) nmol/mg(t =4.88,P<0.01).Conclusion Deferroxamine can protect neurons from glutamate induced damage.The mechanisms include an inhibition of Ca2+ overload and reduction in the levels of MDA and hydroxyl radicals.
