The normal thyroid function is an important basis for maintaining cognitive function, and the different thyroid functional status may cause different cognitive function damages. This article expounds the correlation and its mechanism of thyroid function and cognitive function.
2.Effect of Gekko on Guinea pigs of Experimental Autoimmune Myositis

Objective To investigate the effect of Gekko on experimental autoimmune myositis(EAM)animal model.Methods From Nov.2003 to May.2004,in the first hospital of Ji Lin university,Guinea pigs were immunized with rabbit muscle homogenate in Freund's adjuvant and a generalized myositis fundamentally similar to humanpolymyositis was established.These animals were divided at random into A,B,C,D groups.A group were treated with small dose Gekko [1 250 mg/(kg?d)],B group were treated with large dose Gekko [3 750 mg/(kg?d)],C group were treated with preinisone [3 mg/(kg?d)],D group received no treatment.Sign,serum muscle enzyme,electromyography(EMG)and muscle biopsy changes were compared.Results A,B,C groups became better.D group showed no significant change.Conclusion Gekko is effective in therapy of EAM guinea pigs.The result of this study provides an evidence that Gekko mightbe used for the treatment of PM.
3.Effects of High Density Lipoprotein on Expression of Alpha-Granulemembrane Protein (CD62P) and Lysosome Intact Membrane Protein (CD63) on Platelet

Objective To investigate the effects of high density lipoprotein (HDL) on thrombin-activated platelet al-pha-granulemembrane protein (CD62P) and lysosome intact membrane protein (CD63) expressions in vitro. Methods The equivalent volume of washed platelets prepared by hand was preincubated with HDL (1 g/L) in 37℃water for 15 minutes, which was then stimulated with different concentrations of thrombin (0.5 U/mL, 1 U/mL and 10 U/mL) for 10 minutes in wa-ter of 37℃. Meanwhile another three groups of washed platelets were incubated with thrombin (0.5 U/mL, 1 U/mL and 10 U/mL) for 10 minutes, respectively. The CD62P and CD63 from each sample were analyzed by flow cytometry (FCM). Results The CD62P positive rates of HDL-preincubated groups were significantly lower than those of different concentrations of thrombin groups (0.5 U/mL,1 U/mL and 10 U/mL) in the absence of HDL (11.55%± 1.34% vs 18.14%± 1.50%, 17.19%± 0.17% vs 26.24%± 0.77% and 19.79%± 0.32% vs 80.38%± 5.66%,P < 0.01). Meanwhile, The CD63 positive rates of HDL-preincubated groups were also significantly lower than those of thrombin-treated (0.5 U/mL, 1 U/mL and 10 U/mL) groups without HDL, namely,2.92%±0.22%vs 8.09%±0.48%(P<0.001), 4.20%±0.98%vs 14.15%±1.39%(P<0.001) and 5.12%± 0.09% vs 24.48%± 1.71%(P < 0.01). Conclusion HDL inhibits the expression of CD62P and CD63 on throm-bin-stimulated platelets in vitro.
4.Clinical Study on Acupuncture Combined with Back Shu Acupoint Catgut Embedding Therapy in Treating Allergic Rhinitis

Objective To investigate the clinical efficacy of acupuncture combined with back Shu acupoint catgut embedding therapy in treating allergic rhinitis. Methods Totally 66 allergic rhinitis patients were included. Random number table method was used to divide the 66 patients into observation group and control group, with 33 cases in each group. The control group was given Ketotifen Fumarate Nasal Drops nose drops 2 drops each time, 3 times a day, Loratadine Tablets 10 mg orally, once a day for 4 weeks. The experimental group was treated with acupuncture, once a day, 10 d as a treatment course, three courses in total; at the same time, back Shu acupoint catgut embedding therapy was given, 15 d each time, twice treatment in total. After the end of treatment, clinical efficacy, clinical symptom score, life quality score, serum levels of inflammatory factors, serum immunoglobulin, peripheral blood eosinophil count and the incidence of adverse reactionsof the two groups were observed. Results The total effective rate was 93.94% (31/33) in observation group and 75.76% (25/33) in the control group, with statistical significance in the two groups (P<0.05). Compared with before treatment, the TNSS score, TNNSS score and RQLQ score of the two groups were significantly lower than after treatment (P<0.05); the IgE and EOS levels significantly decreased (P<0.05); the serum IL-17 and IL-22 levels were significantly lower (P<0.05). Compared with the control group, the TNSS, TNNSS and RQLQ score and the levels of IgE, EOS, IL-17 and IL-22 in the treatment group were lower after treatment (P<0.05). There was no significant difference in adverse reactions between the two groups (P>0.05). Conclusion Acupuncture combined with catgut embedding therapy in the treatment of allergic rhinitis has a significant clinical efficacy, which can improve the level of inflammatory factors in patients with high safety.

6.Cloning and expression of Schistosoma japonicum VCP gene and its mRNA expression levels in different stages

Objective To prokaryotically express the valosin-containing protein(VCP)of Schistosoma japonicum,and ana-lyze its VCP mRNA expressions in the cercaria,schistosomulum,adult worm(female and male worms)and egg. Methods RNA of S. japonicum eggs were extracted,and reversely transcribed into cDNA. The VCP gene of S. japonicum was amplified by using polymerase chain reaction(PCR),and subcloned into the prokaryotically expressed vector pET15b. The recombined plasmid was transformed into BL21 cells,and the expression of the target gene was induced with isopropyl-beta-D-thiogalactopyranoside (IPTG). The recombinant protein was yielded through the purification of inclusion body,and identified by using sodium dodecyl sulfate polyacrylamide gel electrophoresis(SDS-PAGE). The RNA(s)of cercaria,schistosomulum,female adult worm,male adult worm,and egg of S. japonicum were extracted,digested with DNase,purified,and reversely transcribed into cDNA. The mRNA expressions of the VCP gene in various developmental stages of S. japonicum were determined by using fluorescence-based quantitative real-time PCR. Results The VCP gene of S. japonicum was yielded by PCR amplification,and the recombinant pro-tein was obtained through recombinant plasmid expression and purification of inclusion body. The highest VCP mRNA expression in S. japonicum cercaria was detected by the fluorescence-based quantitative real-time PCR,while low expressions were found in the schistosomulum,egg,female and male adult worms. Conclusion The recombinant protein encoded by the VCP gene of S. ja-ponicum is successfully obtained,and the VCP mRNA expression is determined in various developmental stages of S. japonicum.

Medical insurance fund is the foundation for the healthy operation of the medical insurance system, which is related to the well-being of the people and social stability. Medical institutions, as important entities in the implementation of medical insurance policies and the use of medical insurance funds, should continuously improve and optimize their self-management, and strengthen the prevention and control of clean risks in the use of medical insurance funds. This is not only a necessary response measure in the context of strict supervision of national medical insurance funds, but also an inevitable choice for hospitals under the trend of high-quality development. This article took a certain hospital as an example to illustrate the practice of sorting out the relevant authority directory for the use of medical insurance funds, analyze the integrity risk points of doctors, patients, and medical insurance reimbursement auditors; And introduced the practice of using a clean risk prevention and control grid to strengthen the hospital′s medical insurance fund, including improving the medical insurance management system, cultivating a sustainable culture of integrity, optimizing process design, improving information system construction, and improving medical insurance service modes, so as to provide reference for other medical institutions.
8.Application of hazard analysis and critical control point in prevention and control of multi drug-resistant bacteria infection in Department of Neurosurgery

Objective To investigate the application of hazard analysis and critical control point (HACCP) in the prevention and control of multi drug-resistant bacteria infection in the Department of Neurosurgery.Methods Following the principle and method of HACCP,we conducted a hazard analysis on the medical environment or medical care in the Department of Neurosurgery;the critical control points and critical limits were determined,and therefore,corresponding measures of monitoring and rectifying,and procedures of verification and record system were established.Results After the implementation of HACCP management system,the discovery rate of multi drug-resistant bacteria infection in the Department of Neurosurgery decreased from 3.12% (January 2012 to June 2013) to 2.06% (July 2013 to December 2014) (x2=3.921,P=0.048),the average hospitalization days shortened from (62.02±68.59) d to (48.49±38.09) d (t=1.105,P=0.272),and the average cost of hospitalization reducedfrom (104571.41±71 721.62) Yuanto (77257.20±45 810.14) Yuan (t=2.182,P=0.032).Conclusion HACCP is worth promoting in the prevention and control of multi drug-resistant bacteria infection in the Department of Neurosurgery.
9.Expression of Bcl-2 and Bax proteins in hippocampus of rats with abnormal thyroid function and its relationship with cognitive Impairment

Objective To investigate the relationship between apoptotic protein Bcl-2 and Bax in rat hippocampal neurons and cognitive dysfunction in rats with abnormal thyroid function.Methods Thirty healthy Wistar rats of 8-week-old SPF grade were randomly divided into three groups:(1) Normal control group (n=10);(2)hypothyroidism group (n=10);(3) hyperthyroidism group (n=10).All rats were sacrificed at the 4th week by heart blood sampling.The serum T3 and T4,TSH were measured.Morris water maze was used to train rats in each group for 6 days.At the end of the experiment,the hippocampus was taken from the rats,and HE staining was performed.The expression of apoptotic protein bcl-2 and Bax in rat hippocampal neurons was detected by immunohistochemistry.Results ①The escape latency of hyperthyroidism and hypothyroidism group was higher than that of the normal group at different time points (P<0.05).In the test of the target area dwell time,the difference between the hyperthyroid group,the hypothyroid group and the normal group was statistically significant (P<0.05).In the distance test of the target quadrant,The differences between the three groups were statistically significant (P<0.05).In the number of passes through the target quadrant,the difference between hyperthyroidism group,hypothyroidism group and normal group was statistically significant (P<0.05),but there is no significant difference between hyperthyroidism group and hypothyroid group (P>0.05).②Hippocampus tissue HE staining:normal control group hippocampal neurons neatly arranged,the shape of the structure was complete and uniform,the nucleus was round or oval,nucleolus obvious,chromatin uniform level and more clear,nucleus round or oval,obviously,hyperthyroidism group,hypothyroidism group of neuronal structure loose,the number decreased,arranged disorder,deep nuclear staining,shrinkage,nucleolus disappeared,cytoplasm around the empty halo,cell spacing larger.③The positive cells expressing Bcl-2 and Bax protein in the hyperthyroidism group and the hypothyroidism group were increased compared with the normal control group (P<0.05).Compareds with hyperthyroidism group,the expression of Bcl-2 positive cells was increased in hypothyroidism (P<0.05).Conclusions ①The spatial learning and memory abilities of the rats with hypothyroidism and the hyperthyroid are lower than those in the normal control group.②The number of apoptotic protein positive cells in Bcl-2 and Bax neurons of hippocampus in rats with hypothyroidism and hyperthyroid increased,and the proportion of Bcl-2 and Bax was impaired,which indicates that apoptosis occurred in hippocampal neurons.This process may be one of the pathogenesis of cognitive impairment.
