1.EFFECTS OF CHROMIUM ON BLOOD GLUCOSE AND LIPID METABOLISM AND GENE EXPRESSION OF SKELETAL MUSCLES IN DIABETIC RATS

Objective: To study the effects of chromium on glucose and lipid metabolism and gene expression in diabetic rats.Methods: Male Wistar rats were assigned to three groups:normal control(NC), alloxan-induced diabetic control group(DM), and DM with chromium supplementation group(DM+Cr). Cr 200 ?g/(kg bw?d) was supplemented orally for 60 days. At the end of the treatment, the blood glucose, lipid and serum insulin were measured, and the changes in gene expression among three groups were studied by mRNA differential display technique.Results: Blood glucose in DM+Cr group decreased significantly than that before experiment. The levels of serum TG, TC, LDL-C and AI in DM+Cr group were lower than those of DM group, while the serum HDL-C levels were higher. Serum insulin was not improved obviously in DM+Cr group. 11 cDNA fragments larger than 400 bp expressed differences in skeletal muscles between DM+Cr group and DM group and were isolated, 4 of which expressed higher in DM+Cr group, while the rest expressed higher in DM group.Conclusion: Chromium supplementation could partially improve the disorders of glucose and lipid metabolism, and have an impact on some gene expression in diabetic rats, which may contribute to the regulating effects on its disorders of metabolism.

Objective:To observe the effect of different iodine intake on the thyroid function in euthyroid adult persons.Methods:One hundred and sixty-one euthyroid healthy volunteers aged 18-24 years were randomly divided into 7 groups.Each group was assigned to receive 500 μg,750 μg,1 000 μg,1 250 μg,1 500 μg,and 2 000 μg iodide/day for four weeks.Serum concentrations of free triiodothyronine(FT3),free thyroxin(FT4)and sensitive thyroid-stimulating hormone(sTSH)were measured by chemolumineseenee assays.Results:Serum FT3 concentration was found a small decline within the normal range in all the iodide supplemented groups(P < 0.05).The level of FT4 was significantly lower,when the dose was up to 1 500 μg (P < 0.05).The level of serum sTSH was increased after 2 weeks iodide supplement in all groups,and after 4 weeks in 500 μg and 750 μg groups(P < 0.05).No significant changes were observed in FT3,FT4 and sTSH between groups(P> 0.05).Conclusion:The thyroid function of normal people showed a rise in serum sTSH at a short time and a high-dose of iodine intake.
4.IDENTIFICATION AND TISSUE EXPRESSION OF NOVEL GENES IN DIABETIC RATS SUPPLEMENTED WITH SELENIUM

Objective: To clone novel gene fragments differentially expressed from diabetic rats supplemented with selenium and detect their expression distribution in various tissues. Method: cDNA fragments from former research project were cloned, sequenced and BLASTn analysed. The RT-PCR of the five novel genes were made using the primers designed according to the sequence of cDNA to observe the expression changes in liver of various groups and their expression distributions in various tissues. Results: Se-2, Se-6, Se-10, Se-14 and Se-18 cDNA were shown to be the novel gene fragments for no matched gene with them in GenBank. The expression levels of four cDNAs, including Se-2, Se-10, Se-14, Se-18, in DM group and DM+Se group were obviously lower than those in NC group. Objective: To study the regulation of selenium on gene expression of phospholipase D (PLD) gene in relation to liver metabolism of diabetic (DM) rats. Method: Differential display cDNA fragments were isolated from former research project in diabetic rats which were supplemented orally with 50 g/kg bw d selenium for 60d, and were cloned, sequenced, and the homology was analyzed. RT-PCR was made using the primers designed according to the sequences of cDNA fragments. 6.RESEARCH OF THE SAFE DIETARY IODINE INTAKE IN ADULTS

Objective To explore the safety of iodine intake in adults.Method Euthyroid healthy volunteers aged 18 to 24 years(22.54?2.65y)were adopted and divided into 7 groups randomly.Each group was assigned to receive 500,750,1000,1250,1500,and 2000?g iodide oral doses daily for 4 w.Blood and urine samples during morning fast were obtained on weeks 0,2 and 4.We used chemiluminescence immunoassay(CLIA) to measure sTSH,FT4 and immunoradiometric assay(IRMA) to measure TPOAb and TGAb.Urinary iodine was evaluated.The dietary survey was done by recording method to collect food consumption data for 7 d.We also measured the iodine amount in drinking water,food and iodinized salt.Results All the volunteers were in adequate iodine levels.The median urinary iodine was 272.25?g/L,and the dietary iodine intake including iodinized salt was 346.24?g/d.We found all the iodide supplemented groups responded in the same way with a significant rise in urinary iodide excretion and in serum sTSH levels,with a small decline in serum FT4 concentration at normal range.After 2 w the serum sTSH levels in the iodide supplemented groups increased nearly more than double,and nearly two fold after 4 w.The subclinical hypothyroidism appeared at 500~2000 ?g/d after 2 w.After 4 w the prevalence of subclinical hypothyroidism kept the same level at 500?g/d(15%),and increased at other iodine supplemented groups. The subclinical hypothyroidism prevalence at the 750-2000?g/d doses was at the range of 28.00%~47.37%.No clinical hypothyroidism was observed among the people during iodide administration.Conclusion The dysfunction of thyroid in normal population was mainly subclinical hypothyroidism with an exposure to a high dose iodine intake(500?g~2000?g) in a short time.It can lead to subclinical hypothyroidism in the subjects at 500?g /d doses.In iodine-sufficient area iodine supplemented dose was not appropriate to exceed 500?g/d.We suggested the UL of iodine below 900 ?g/d.
7.Clinicopathological features of unicystic ameloblastoma with special reference to its recurrence.

OBJECTIVETo clarify the clinicopathological spectrum and biologic behavior of unicystic ameloblastomas (UA).
METHODSRetrospective analysis of 33 UA was undertaken to relate clinicopathological variables to recurrence.
RESULTSThe clinical features of this series were in general agreement with that of the previous reports. UA tended to occur at an earlier age (mean 25.3 years), with a male predilection and predominant mandibular involvement (90.9%). Microscopically, the tumors demonstrated a generally monocystic growth pattern, with 8 being simple cystic, 10 comprising intraluminal nodules, and 15 exhibiting a conspicuous component of infiltrative tumor islands in the cyst capsule. Follow-up of 29 patients revealed 6 recurrences with an average interval of 7 years. Recurrence also related to histologic subtypes of UA, with those invading the fibrous wall having a rate of 35.7% but others 6.7%.
CONCLUSIONSDespite the fact that unicystic ameloblastoma may, in general, compare favorably with its solid or multicystic counterpart in terms of clinical behavior and response to treatment, the subsets of the maxillary lesions or tumors exhibiting mural invasion could have a high risk of recurrence.

Objective To study the dietary nourishment of adult patients with leukemia and compare acute leukemic patients with chronic leukemic patients. Methods Adopting dietary review of 24 hours and seven consecutive days of dietary records method to obtain the food category and quantity of 122 patients with acute leukemia and 10 patients with chronic leukemia. Using statistic software SPSS11.0 to calculate the patients'intake of various kinds of nutfiments. and the difiences between acute and chronic leukemic patients were analyzed. Results The rate of most ontrients of patients'intake reaches RNI/AI is lower,especially vitamin A,vitamin C and caleium.There's a tendency that intake diet,energy and nourishments of acute leukemic patients is lower than that of those chronic leukemic patients. Conclusion There is a tendency of unbalanced dietary intakes in leukemic patients.including the low intakes.There is the tendency that nutritional status of acute leukemic patients iS poorer than that of chronic leukemic patients.
