1.Comparison of auditory evoked potential index, bispectral index and spectral edge frequency for monitoring depth of anesthesia during emergence from anesthesia

Objective Auditory evoked potential index (AAI) has been proposed for monitoring anesthetic depth. The aim of this study was to compare tins new technique with hispectral index (BIS) and 95% spectral edge frequency (SKF) for assessing anesthetic depth during emergency from propofol-isoflurane anesthesia. The ability of these techniques in distinguishing consciousness from unconsciousness was also evaluated. Methods Thirty six ASA I - II patients ( 15 male, 21 female) undergoing elective surgery under propofol-isoflurane anesthesia were enrolled in the study. Age ranged from 18 to 75 years and body weight from 35 to 80 kg. Patients with psychoneural diseases or hearing disturbances were excluded. The patients were premedicaled with phenobarbitai sodium 0.1g and airopine 0.5mg. Anesthesia was induced with rnidazolam 0 .05-0.1 mg.kg1,fentanyl 5- 10ug.kg1 and vecuronium 0. 1-0.2mg.kg-1 and maintained with propofol infusion (8-16ml. h 1) and isoflurane inhalation (0.5% -1.0%). Intermittent IV boluses of vecuroniuni were given when needed. The patients were transported to recovery room after surgery. AAI, HIS, SKF and hemodynamic parameters were monitored and recorded on entering the recovery room, before extubation, during extubalion, 5, 10, 20 min after exlubation and before release from recovery room. Results AAI, BIS and SEF were 40.9?11.7,73.64?10.8 and 17.5?2.8 respectively on entering recovery room and increased to 72.6 ?11.0, 88.2?7.3 and 22.5?2.6 during exlubation. The increase in AAI was significantly greater. The mean values of AAI before and after responding to light glabellar tap or loud auditory stimulus were 36. 1?11.5 and 52.4?12.3 respectively, the mean values of BIS were 71.9?11.5 and 78 . 6?11.9 and of SKF 16.7?3.0 and 18. 6?3.2 .Only AAI demonstrated a significant difference detween consciousness and unconsciousness. Conclusion AAI, HIS and SKF all increase gradually during emergence from anesthesia. AAI is most sensitive among the three techniques and is most useful in detecting the transition from unconsciousness to consciousness.
2.Comparison between pharmacokinetics of propofol in the elderly and young patients during induction

Objective To compare the pharmacokinetic profile of propofol after a single intravenous dose during induction in the elderly and young patients. Methods Eighteen ASA I-II patients undergoing elective gastro-intestinal and intracranial surgery were studied. Patients with abnormal liver and/or kidney function were excluded. The patients were premedicated with intramuscular phenobarbital 100mg and scopolamine 0.3mg 1h before operation. The patients were divided into two groups according to their age; the young and middle-aged group, aged between 31-57, on average 46. 5yr (A, n=6); the elderly group, aged between 67-81 yr, on average 74.6 yr(E, n = 12) . Group E was further divided into two subgroups: E1 aged between 67-73 yr, on average 69 .3 yr (n = 6); E2 aged between 76-81 yr,on average 78. 7 yr ( n=6 ) . In group A anesthesia was induced with propofol 1 . 5rng kg-1 , midazolam 0.03-0.06mg kg-1 , fentanyl 3-5ugkg-1 and vecuronium 0. 1 mgkg-1 . In group E propofol 1.0 mgkg-1 was given but the doses of the other three drugs for induction were the same as in group A. Anesthesia was maintained with isoflurane 0.5%-2.0% supplemented with intermittent iv boluses of fentanyl, vecuronium and midazolam. ECG, BP, SpO2 , PET CO2 , CVP and urine output were continuously monitored during anesthesia. Propofol was given in bolus through the vein in the forearm slowly over 30-45 s, and blood samples were taken from internal jugular vein before propofol injection and 1 ,2,4 ,6,10,15,30,45,60,75, 90,120 ,150 ,180,240 ,300,360min after the end of propofol injection for measurement of plasma propofolconcentration by HPLC with fluorescence detection. Results The pharmacokinetics of propofol in the 18 patients were best described by a three compartment pharmacokinetic model. 3.RHD Gene Analysis of A Blood Donor with Del Phenotype.

OBJECTIVE:
To analyze the RHD genotype of a blood donor with Del phenotype in Yunnan.
METHODS:
Rh serological phenotype was identified. RHD gene was detected by PCR-SSP typing, and its 10 exons were sequenced. Exon 9 was amplified for sequencing and analysis. RHD zygosity was detected.
RESULTS:
The Rh phenotype of this specimen was CcDelee. Genomic DNA exhibited a 1 003 bp deletion spanning from intron 8, across exon 9 into intron 9. The deletion breakpoints occurred between two 7-bp short tandem repeat sequences. There was no variation in the sequences of the remaining exons. The Rh hybridization box test showed that there was one RHD negative allele.
CONCLUSION
This specimen is Del type caused by deletion of RHD exon 9.

4.Precision gene editing technologies based on CRISPR/Cas9: a review.

Gene editing technology is a genetic operation technology that can modify the DNA sequence at the genomic level. The precision gene editing technology based on CRISPR/Cas9 system is a gene editing technology that is easy to operate and widely used. Unlike the traditional CRISPR/Cas9 system, the precision gene editing technology can perform site-directed mutation of genes without DNA template. This review summarizes the recent development of precision gene editing technology based on CRISPR/Cas9, and prospects the challenges and opportunities of this technology.

5.Retrospective analysis of strabismus surgery in adolescents and children in Yunnan province

AIM: To analyze the strabismus surgery situation of adolescents and children in Yunnan province.METHODS: A retrospective analysis was conducted on medical records data of 3 068 adolescents and children who received strabismus surgery at Affiliated Hospital of Yunnan University from January 2017 to December 2021. The analysis included gender, constituent ratio of age, distribution of strabismus types and combination with other ocular diseases, etc.RESULTS: Among the included patients, 52.12% were males, and 47.88% were females. Preschool patients(1 to 6 years old)accounted for 32.89%, primary pupils(7 to 12 years old)accounted for 45.89% and high school students(13 to 18 years old)accounted for 21.22%. Exotropia accounted for 63.17% of the total strabismus, of which intermittent exotropia was the most common type. Esotropia accounted for 19.69%, and concomitant esotropia was the most common type. The special type of strabismus accounted for 17.14%, and A-V syndrome and dissociative vertical deviation(DVD)were the most common types. Strabismus combined with ametropia accounted for 61.02% and amblyopia accounted for 10.89%. A few patients also combined with other eye diseases.CONCLUSION: In Yunnan province, intermittent exotropia was the most common type of strabismus among adolescents and children. Some patients combined with other ocular diseases.
6.Capability building and professional staff of schistosomiasis control institutions in Yunnan Province: a cross-sectional survey

To understand the current status of professional staff and capacity building among schistosomiasis control institutions in Yunnan Province, the data pertaining to the current status of professional staff and capacity building were captured from the schistosomiasis control institutions at all levels in Yunnan Province. There were 20 schistosomiasis control institutions at a level of above county in Yunnan Province, with 411 employees, and there were 11 institutions with an independent legal personality, with a mean building area of 69.83 m2 per capita. Among all institutions, the number of posts was 382, and the actual number of employees was 360, with 22 job vacancies. The mean age of professional staff was 41.6 years at a province level, 44.1 years at a city level and 42.4 years at a county level, and there were no professionals at ages of < 30 years in city-level institutions. Of all professionals, there were 45.76% with a college degree or below, and 54.26% with a bachelor degree or higher, and 16.55%, 24.57% and 39.90% had senior, moderate and primary professional titles, with mean ages of 49.10, 44.37 years and 38.36 years, respectively. The results of the cross-sectional study demonstrate that there are problems of job vacancy, population aging and low education levels in schistosomiasis control institutions in Yunnan Province. Therefore, the capability building and professional quality should be further improved in schistosomiasis control institutions to meet the requirements of schistosomiasis control activities in Yunnan Province.
7.Research progress on effect of IL-17 in ocular diseases

Interleukin-17(IL-17)is an important pro-inflammatory factor in autoimmune diseases and many inflammatory diseases. Studies have shown IL-17 plays a vital role in the pathogenesis of eye disease. This paper reviews the function and mechanism of IL-17 in keratitis, dry eye, uveitis, diabetic retinopathy, and age-related macular degeneration, providing new ideas and targets for the treatment of these diseases.
8.Drug resistance characteristics and epidemic trend of Escherichia coli in Anning, Yunnan

Objective To investigate the drug resistance characteristics and epidemic trend of Escherichia coli in Anning, Yunnan, and to provide a reference for clinical rational use of antibiotics. Methods A total of 376 strains of Escherichia coli were isolated from June 2018 to June 2019 from Kungang Hospital of Yunnan Province. The ESBLs producing strains were screened by double disk method and the sensitivity of ESBLs to antibiotics was detected by K-B method. The genotyping of ESBLs strains and the class I, II and III integrase genes in integron 3' and 5' conservative regions was conducted using polymerase chain reaction (PCR). Results The detection rate of ESBLs producing Escherichia coli was 53.19% (200/376). ESBLs-producing Escherichia coli had a low resistance rate to carbapenem antibacterial drugs with a relatively high sensitivity. The resistance rates to amikacin and piperacillin were 7.50% and 12.50% respectively, which had good antibacterial activity. The resistance rate to other antibiotics was high. There were 74.00% (148/ 200) of ESBLs-producing Escherichia coli carrying CTX-M gene, 34.50% (69/200) carrying TEM genes, and 1.00% (2/200) of strains carrying SHV genes. In addition, there were 9.00% (18/200) of strains carrying both CTX-M and TEM genes, and 0.50% (1/200) of strains carrying both CTX-M and SHV genes. The detection rate of integrons accounted for 37.00% (74/200), all of which were class I integrons. Conclusion The prevalence of ESBLs-producing Escherichia coli was relatively high in Anning, Yunnan. ESBLs-producing Escherichia coli showed a trend of multi-drug resistance. The genotype was mainly CTX-M, and the integron was classified as class I.
9.Advances in research of peripheral blood miRNAs in early diagnosis of tumors

[Abstract] MicroRNA (miRNA) is non-coding RNA molecule consisting of 20-25 nucleotides. It plays an important role in regulation of tumorigenesis and progression, including proliferation, differentiation and apoptosis of cancer cells, which directly affect the progress of tumors. Peripheral blood miRNA is relatively more stable, and easier to acquired and detected than tissue miRNA. It is a new generation biomarker for early detection and early diagnosis of tumors. It is also one of the main development directions of research and application in precision medicine. Methods commonly used in peripheral blood miRNA detection are RT-PCR, electrochemical detection, NanoString Technologies, genechip and high-throughput sequencing etc. Multiple miRNAs in peripheral blood are the early diagnostic markers for non-small cell lung cancer, esophageal squamous cell carcinoma, pancreatic cancer, squamous cell carcinoma of the head and neck, ovarian cancer, colorectal cancer, breast cancer, prostate cancer and hematological malignancies. Combined detection of multiple peripheral blood miRNAs, as well as combined detection of tumor-specific miRNAand serological, imaging and other auxiliary methods, can improve the sensitivity and specificity of tumor diagnosis at early stage.
10.Progress of interruption of schistosomiasis transmission and prospects in Yunnan Province

Schistosomiasis was once hyper-endemic in Yunnan Province. Following concerted efforts for over 70 years, remarkable achievements have been made for schistosomiasis control in the province. In 2004, the Mid- and Long-term Plan for Schistosomiasis Prevention and Control in Yunnan Province was initiated in Yunnan Province, and the target for transmission control of schistosomiasis was achieved in the province in 2009. Following the subsequent implementation of the Outline for Key Projects in Integrated Schistosomiasis Control Program (2009—2015) and the 13th Five - year Plan for Schistosomiasis Control in Yunnan Province, no acute schistosomiasis had been identified in Yunnan Province for successive 12 years, and no local Schistosoma japonicum infections had been detected in humans, animals or Oncomelania hupensis snails for successive 6 years in the province by the end of 2020. The transmission of schistosomiasis was interrupted in Yunnan Province in 2020. This review summarizes the history of schistosomiasis, changes in schistosomiasis prevalence and progress of schistosomiasis control in Yunnan Province, and proposes the future priorities for schistosomiasis control in the province.
