1.Determinination of Taurine in Taurine Soft Capsules by Pre-column Derivatization HPLC

Objective:To develop a method for the determination of taurine in Taurine Soft Capsules by RP HPLC.Methods: Taurine was derivated by phthalaldehyde and ethanethiol. The analytical column was C 18 . The UV detective wavelength was set at 360nm. The mobile phase was methanol water phosphate buffer (1∶1∶1). The external standard method was used. Results: The recovery was 95.8%~107.2%. RSD was 2.1%. The linear range of concentration was 6.7 ~40.4 mg/mL. The minimum detectability was 2.47mg/mL. Conclusions: The method is simple, quick and accurate.
2.Therapeutic Effect of Modified Simiao Powder for Drug-induced Hyperuricemia

[Objective] To observe the therapeutic effect of heat-clearing and diuresis-promoting herbal medicine for hyperuricemia induced by pyrazinamide. [Methods] Eighty-six patients with pyrazinamide-induced hyperuricemia were randomized into 2 groups: group A ( n = 43) received antiphthisic drugs (including isoniazid, rifampin, pyrazinamide and ethambutol) , allopurinol tablets and Modified Simiao Powder (one dose per day); group B ( n = 43) was treated with antiphthisic drugs and allopurinol tablets. Fourteen days constituted one treatment course and the two groups were treated for 2 courses. After treatment, the therapeutic effect was assessed and the changes of blood uric acid level were observed. [Results] In group A, 28 (65.12%) were cured, 12 (27.91%) effective, 3 (6.97%) ineffective and the total effective rate was 93.02%; in group B, 16 (37.21%) were cured, 17 (39.53%) effective, 10 (23.26%) ineffective and the total effective rate was 76.74% . 3.Optical coherence tomography of choroidal neovascularization of exudative age-related macular degeneration and central exudative chorioretinopathy

Objective To investigate the characteristics and analyze the differences of choroidal neovascularization (CNV) image between exudative age-related macular degeneration (AMD) and central exudative chorioretinopathy (CEC) using optical coherence tomography (OCT). Methods Twenty-three eyes of 22 patients with exudative AMD and 20 eyes of 19 patients with CEC, which were diagnosed as CNV using fundus fluorescein angiograph (FFA) or indocyanine green angiograph (ICGA), were exami- ned by OCT. The size of CNV and the thickness of retinal neurosensory layer in foveola were measured by OCT software. Results On OCT image, CNV had three main types of morphological features including simple CNV, CNV with serous retinal neurosensory layer detachment and CNV with choroidoretinal exudation. Exudative AMD mainly showed CNV with choroidoretinal exudation (56.52%) and CEC showed simple CNV (90 0%). The thickness of retinal neurosensory layer in foveola of patients with exudative AMD was thicker than that of patients with CEC and the size of CNV of patients with exudative AMD was larger than that of patients with CEC. Negative correlation was found between retinal neurosensory layer thickness in foveola and vision in both groups (?=-0.521, P =0.001). Conclusions There were certain discrepancy in morphology and area involved of CNV between exudative AMD and CEC on OCT images.
4.Clinical observation of three dimensional conformal radiation therapy in combination with chemotherapy in stage Ⅲ patients with non-small-cell lung cancer

Objective To observe the toxicity and effection of three dimensional conformal radiation therapy(3DCRT)based on conventional fractionation radiotherapy in combination with chemotherapy in non-small-cell lung cancer(NSCLC).Methods 62 NSCLC patients with stage Ⅲ were chosen as research objects.All the patients received chemotherapy two cycles with NP regimen firstly(NVB 40 mg d1,8,DDP 30 mg d2-5),then they accepted radiotherapy in twentith day after chemotherapy when hemocyte got normal.In the first half period of the radiotherapy,conventional radiotherapy were given with 2 Gy/f,after the dose got 40 Gy/20 f,then three dimensional conformal radiation therapy were given with 4~6 Gy/f every 1 to 2 days until the total dose got 24~30 Gy/4~6 f.The patients with supraclavicular lymphnode metastatic carcinoma received conventional fractionation radiotherapy with 60Co or 6 MV-X ray combined electronic ray until the dose(DT)up to 64~66 Gy.After radiotherapy,the patients accepted chemotherapy two cycles with NP regimen again.Results All the 62 patients completed treatment plan,and follow-up survey had lasted for 3 years.The short-term efficacy was evaluated in 3 months after radiotherapy. Among 62 patients, 9 cases gained a complete remission(CR),40 cases partial remission(PR),and the total remission was 79.0%.The acute toxicity showed acute radiation esophagitis,radiation pneumonia and acute gastrointestinal reaction.However,expectant treatment was effective.The 1,2,3 year survival rate was 7 1.0%,48.4%,30.6%,respectively.The 1,2,3 year local control rate was 80.6%,62.9%,40.3%,respectively.Conclusion 3DCRT based on conventional fractionation radiotherapy for NSCLC have good effection and mild toxicity.
5.Effect of central retinal artery occlusion on retinal microstructure of macula in vivo

AIM: To investigate the effect of ischemia induced by central retinal artery occlusion on retinal microstructure of macula using optical coherence tomography (OCT). METHODS: Fourteen eyes of 14 patients with unilateral central retinal artery occlusion (CRAO) in two to three days without fully recovery of retinal circulation underwent OCT examination with 4.5 mm length horizontal and vertical line scans through foveola to measure the retinal neurosensory layer (RNL) thickness on foveola, 175 ?m (fovea), 750 ?m (macula) to foveola, respectively. The other normal eyes of patients as control group underwent the same examination and measurement. RESULTS: The mean RNL thickness(?m) on foveola, fovea, macula were 169 91?10 96, 176 36?11 74 and 256 45?16 95 respectively in normal control eyes, and 235 64?47 02 , 241 84?49 36 and 401 57?54 53 respectively in CRAO eyes with retinal ischemia. 7.Effects of silencing RPB5-mediating protein (RMP) gene on cell proliferation and migration of liver cancer SMMC-7721 cells

Objective:To establish the RPB5-mediating protein (RMP)-silenced stable cell lines and study the inhibitory effects of small interfering RNA (siRNA) targeting RMP gene on the proliferation and migration of human hepatoma SMMC-7721 cells. Methods:Three RMPi siRNAs were designed and synthesized in vitro and transfected into SMMC-7721 cells. The inhibitory effect of siRNA on RMP gene expression was measured by RT-PCR to select the best siRNA. The expression vector pGPU6-Neo-RMP-484 was transfected into SMMC-7721 cells by the lipofectamine and the cells stably expressing the siRNA were selected by G418. RT-PCR was used to detect the interference efficacy against RMP gene. Cell proliferation and adhesion were measured by MTT assay. Wound healing test was used to observe the migration ability of cells. Results:The SMMC-7721 cell lines with down-regulated RMP expression were established by using RNA interference technology. Compared with the negative control cells, expression of RMP mRNA was down-regulated by(83.67±2.56)% .The proliferation of stable-transfected cells was inhibited by(74.33±0.58)% . The adhesion capability of stable-transfected cells was enhanced but the migration capacity was decreased compared with the negative control cells. Conclusion:The pGPU6-Neo-RMP-484 cell lines with stable transfection of RMP siRNA recombinant vector are successfully screened,which can be used as a cellular model for studying the molecular mechanism of RMP. Down-regulation of RMP gene expression can effectively inhibit the proliferation, enhance the adhesion, and decrease the migration of SMMC-7721 cells.
8.Pre-bend elastic stable intramedullary nail fixation for distal radial shaft fractures in children

Objective To investigate the functional and radiographic outcomes of pre-bent elastic stable intramedullary nail in treatment of distal radial shaft fractures in children. Methods From January 2006 to December 2008, 18 children with distal radial shaft fracture were treated by close reduction and internal fixation with a pre-bent elastic stable intramedullary nail. The age range was from 5 years to 15 years,with an average of 9 years and 8 months. All the cases were closed fractures, and followed up at least 12 months. The time interval from injury to surgery were 1 to 9 days. Fracture sites were located at the distal radial shaft. Among them, 15 cases suffered ipsilateral ulnar fracture, and the others for solitery radial fracture. Closed reductions were performed under C-arm fluoroscopy. All affected arms were immobilized in the functional position postoperatively by the plaster cast beyond elbows. Results The average time of fracture complete union was 5 months (ranged from 3 months to 7 months). The average preoperative angulation of the fractures: anteroposterior view 16.2±7.5 degrees, lateral view 26.2±13.1 degrees. The average postoperative angulation of the fractures: anteroposterior view 2.7±1.5 degrees, lateral view 1.4±1.2 degrees. All fractures maintained good alignment postoperatively, and 94.4% (17/18) of the patients regained a full range of rotation of the forearm. One patient suffered limitation of rotation in forearm less than 10°, and this had improved by final follow-up. Complications included soft tissue irritation at the site of nail insertion in one patient and transient scar hypersensitivity in another. Conclusion Fixation with a pre-bent elastic stable intramedullary nail is an effective, safe and convenient method for treating distal radial shaft fractures in children.
9.Binary cardiac rehabilitation in patients with acute myocardial infarction and the measurement of 6-minute Walk Test

Objective To study the effects of binary cardiac rehabilitation (CR) composed of hospital-based and home-based CR in patients after acute myocardial infarction (AMI) evaluated by six-minute walking distance (6MWD). Methods A total of 38 post-MI patients were divided into two groups, namely conventional CR group (n=20) and binary CR group (n=18). In the first seven days, CR training was carried out in all 38 patients in our hospital, and then 20 patients were kept at CR training in the hospital (hospital-based CR group), whereas the remaining 18 patients (binary CR group) got tele-monitored walking training at home for eight weeks (binary model). Before discharge from hospital based CR or after 8 weeks CR at home, all patients received Cardiopulmonary Exercise Testing and Six-minute Walk Test, and the data of patients' body mass index (BMI),thyroid function, serum lipid metabolism and echocardiography were collected before and after CR. Results After CR, BMI and low density lipoprotein (LDL) decrease in both groups compared with those before CR[hospital-based group: BMI, (25.99±3.36)kg/m2 vs. (23.04±3.72) kg/m2,P<0.05; LDL, (3.40±1.38) mg/dl vs. (2.04±0.73)mg/ dl,P<0.01; binary group: BMI, (24.84±2.70) kg/m2 vs. (22.88±2.56) kg/m2,P<0.05; LDL, (3.40±1.01) mg/dl vs. (92.11±0.37) mg/dl,P<0.01]. After 8 weeks CR at home or hospital based CR until discharge, the anaerobic threshold (AT), maximum rate of oxygen consumption (VO2max),metabolic equivalent (MET) and 6MWD improved significantly in both groups compared with those at 7 days after CR in hospital [Hospital-based group: AT, (12.37±1.53) mL/(kg. min) vs. (14.77±1.57) mL/(kg. min); VO2max (17.87± 1.66 mL/(kg. min) vs. (20.73±2.14) mL/(kg. min); MET (5.02±0.36) vs. (6.09±0.53); 6MWD (500±53.36) m vs. (582.5±57.6) m,P<0.01; Binary group: AT, (12.56±1.11) mL/(kg·min) vs. (14.30±1.23) mL/(kg. min); V02max, (17.28±1.38) mL/(kg. min) vs. (20.02±1.37) mL/(kg. min); MET, (5.07±0.47)vs (5.94±0.46); 6MWD,(511.4±50.96) m vs. (590.3±56.1)m,P<0.01]. There was no significant difference in CR effects observed between two groups (P>0.05). Conclusions In post-MI patients,a binary model of CR training improved physical capacity and was a similarly effective form of CR as a entirely hospital-based approach. A home-based tele-monitored program facilitated patients' adherence to CR. The 6 minute walk experiment is economical and good evaluation on the CR effect of binary cardiac rehabilitation.
10.Antiviral activity of nano-realgar against herpes simplex virus Type II in vitro.

To explore the antiviral activity of nano-realgar against herpes simplex virus Type II (HSV-2) in vitro.
Methods: Acyclovir (ACV) as a positive control, the cytotoxicity of nano-realgar at different concentrations (including 200.00, 150.00, 100.00, 50.00, 25.00, 12.50, 6.25, 3.13, 1.54, 0.78, 0.39 and 0 mg/L) on normal Vero cells were determined by 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2-H-tetrazolium bromide (MTT) assay. HSV-2 virus titer was determined by plaque assay, and the Vero cells model of HSV-2 infection was established. Subsequently, the antiviral effects of nano-realgar at different concentrations (including 20.00, 10.00, 5.00, 2.50, 1.25, 0.63, 0.31, 0.15, 0.08, 0.04 and 0 mg/L) on infected cells model were evaluated by the observation of cytopathic effect (CPE) and MTT method under the 3 modes including pre-treatment, treatment and direct inactivation.
Results: The 50% cytotoxic concentration (CC50) of nano-realgar on Vero cells was 37.15 mg/L. The titer of HSV-2 was 7.30 log PFUs/mL. In the 3 modes, the half-maximal effective concentration (EC50) of nano-realgar on HSV-2 infected Vero cells were 0.13, 1.80 and 0.52 mg/L, and the corresponding therapeutic index (TI) were 285.77, 20.64, 71.44, respectively. The TI value of nano-realgar on pre-treatment mode was higher than that of nano-realgar on treatment and direct inactivation modes.
Conclusion: Nano-realgar can play a good anti-HSV-2 activity in the 3 modes (pre-treatment, treatment and direct inactivation), and the anti-HSV-2 efficacy of nano-realgar on pre-treatment mode is better than that of nano-realagr on other 2 modes.
