1.The Correlation between Angiotensinogen Gene M235T Polymorphism and Hypertension Combined with Depression

Objective To investigate the correlation between angiotensinogen gene M235T polymorphism and gerontism hypertension combined with depression. Methods The polymerase chain reaction (PCR) and restriction fragment length polymorphism (RFLP) technique were used to detect the M235T genetic polymorphism in 117 patients with hypertension and 91 patients with hypertension and depression, and 101 healthy subjects without clinical evidence of hypertension and depression.Result The frequency of TT genetype and T allele was higher in group with hypertension and the group with hypertension and depression than that in the control group ( <0.05) . Conclusion Angiotensinogen gene M235T polymorphism is significantly correlated with the gerontism hypertension combined with depression.
2.Determination of cognition function and sleep status in goungsters with internet addiction disorder and correlation analysis

Objective To explore the changes of cognition and sleep status in the youngsters with internet addiction disorder (IAD), and to clarify the relation between cognition and sleep-related indicators of IAD youngsters.Methods Event related potential (ERP) and Pittsburgh Sleep Quality Index (PSQI)test were used for forty-two IAD (case group)and forty non-IAD (control group) in youngsters, and the indexes of period latency (PL)of N1 ,P2 ,N2 ,P3 and amplitude of P3 (P3 amp)of ERP were analyzed,the factor scores of sleep quality (SQ), sleep latency (SL), sleep time (ST), sleep efficiency (SE), sleep dysfunction (SD), sleeping pills (SP), daytime dysfunction (DD)and total score of PSQI were analyzed. The correlation between the P3 PL, P3 amp of ERP and all the indexes of PSQI in case group was analyzed.Results The PL of N2 ,P3 of ERP in case group was significanly longer and the P3 amp was significantly lower than those in control group (P<0.01);the factor scores of the SQ,SL,ST,SE,SD,DD,and the total score of PSQI test had significant differences between two groups (P<0.01).The correlation analysis results showed that there were remarkable positive correlations between the P3 PL and the factor seores of the SQ,SL,ST,SE,SD,DD and the total score of PSQI test in case group (r=0.46,0.34,0.51,0.40,0.48,and 0.54;P<0.01),and there were remarkable negative correlations betweeb P3 amp and the indexes of PSQI test in case group (r=-0.42,-0.45,-0.49,-0.38,-0.38,and -0.50;P<0.01).Conclusion The IAD youngsters have cognition and sleep dysfunction,and the both factors can affect each other.
3.Relationship on personality characteristics and event related potential P300 with internet addiction disorder of youngsters

Objective To explore the relation between personality characteristics and event related potential P300 with internet addiction disorder(IAD) of youngsters.Methods Eysenck Personality Questionnaire and event related potential test were used for forty-two IAD and forty non-IAD in youngsters,and the results were compared between the two groups.Results The latency of N2,P3 ((267.2±24.2) ms,(374.5 ±22.0) ms) of event related potential in IAD group were significantly longer and the amplitude of P3 ((3.9± 2.5) μV) was significantly lower than those in non-IAD group((235.7±21.8) ms,(332.3±23.5) ms,(7.8±2.4) μV) (P<0.01).And the indexes of P,N score (56.2±9.4,49.8±7.4) of Eysenck Personality Questionnaire were significant higher than those in non-IAD group(51.9±9.7,42.4±7.5)(P<0.01),but the indexes of E,L score (45.7±9.0,44.3±7.9)of Eysenck Personality Questionnaire were significant lower than those in non-IAD group(51.2±9.4,50.9±8.3) (P<0.01).Correlation analysis showed that there were remarkable positive correlation with latency of P300 and P,N score of Eysenck Personality Questionnaire respectively in IAD group(r=0.49,0.54,P<0.01),and a significant negative correlation with E,L score(r=-0.43,-0.46,P<0.01),there was a remarkable negative correlation with amplitude of P300 and P,N score of Eysenck Personality Questionnaire respectively in IAD group(r=-0.41,-0.50,P<0.01),and a significant positive correlation with E,L score(r=0.45,0.47,P<0.01).Conclusion IAD in youngsters has personality characteristics and cognitive dysfunction,and it has a relation with personality and event related potential P300.
4.Determination of Sulfadiazine Residues in Pork by Molecular Imprinted Column Coupling with High Performance Liquid Chromatography

A method for the quantitative monitoring of sulfadiazine (SD) residues in pork was established by molecular imprinted column coupling with high performance liquid chromatography (HPLC).The molecular imprinted column was selected as an extraction device.To obtain the optimum extraction efficiency,several parameters related to the molecular imprinted column,including column solvent,flow-rate,eluent of the sample matrix and eluent volume,were investigated.The sample solution was directly injected into the device for the extraction after simple extraction.Under the optimum conditions,the relative standard deviations (RSD) was ≤6.1% and the recoveries for SD were higher than 75.6%.In comparison with the AL-SPE column,the MIP-SPE column had good reusability and extraction efficiency.This method was successfully applied to the determination snlfadiazine residues in pork.
5.Lactoferrin improved radioactive lung damage by regulating HMGB1/TLR4 inflammation

Objective:To investigate the protective effect of lactoferrin(Lf) on lung injury in mice exposed to irradiation.Methods:C57BL/6 J mice were randomly divided into control group, 15 Gy irradiation group (IR group) and lactoferrin combined 15 Gy irradiation group (Lf+ IR group), with 5 mice in each group. The mice in the Lf+ 15 Gy group drank lactoferrin solution (10 mg/ml) from 3 days before irradiation and contained the whole experiments. Then, single chest 15 Gyirradiation was performed both in the IR and Lf+ IR groups. The body weight and other characteristics were monitored during the experiment. The mice were killed at day 14 after irradiation. The lung histopathology was observed by HE staining. Serum inflammatory cytokine such as HMGB1, TNF-α, IL-1β and IL-6 was determined by ELISA method . The expression of inflammatory related protein in lung tissue including HMGB1, TLR4, MyD88 and NF-κB were performed by immune histochemistry and Western blot method.Results:Compared with the control group, lung weight was significantly increased ( t=3.20, P<0.05), pulmonary hyperemia and inflammatory cell infiltration was observed in the IR group. Exposure also significantly increased serum level of TNF-α[(291.80±5.49) vs.(332.25±22.18)pg/ml]( t=3.07, P<0.05), up-regulated the expression of inflammatory related protein in lung tissue ( t=4.04, 4.78, 3.77, 6.14, P<0.05). Lactoferrin intervention (Lf+ IR group) significantly decreased lung weight ( t=2.18, P<0.05), alleviated histopathologic changes, decrease serum levels of HMGB1, TNF-α and IL-1β ( t=4.67, 2.97, 3.49, P<0.05). On the other hand, lactoferrin intervention decreased the positive cell number of HMGB1 and NF-κB, and down-regulated the protein expression of HMGB1, TLR4, MyD88 and NF-κB in lung tissues, with significant difference with the IR group ( t=8.06, 9.80, 3.07, 5.56, P<0.05). Conclusions:Lactoferrin plays the protective effect of radiation-induced lung injury through the downregulation of inflammatory response, such as HMGB1/TLR4/MyD88/NF-κB signaling pathway.
6.Expression analysis of neuregulin 1 gene in schizophrenia model rats

ObjectiveTo analyze the gene expression of neuregulin1 (Nrg1)mRNA and protein in encephalic region and peripheral blood,and to explore the consistency of the expression in central and peripheral in schizophrenia model rat induced by dizocilpine maleate ( MK801 ).Methods30 adult male SD rats were randomly divided into 3 groups ( 10 cases per group):model group injected with MK801 (0.6 mg/kg and 100μl/20 g)on the right rear of ventrolateral compartment,control group 1 ( no treatment) and control group 2 injected with equal volume normal saline.Nrg1 mRNA was measured in peripheral blood and in prefrontal lobe by using semiquantitative RT-PCR in 3 groups,and Nrg1 protein was measured in encephalic region (prefrontal lobe,dentate band and hippocamp) by using immunohistochemistry.ResultsThere was significant difference of the amount of Nrg1 mRNA among 3 groups (for center:F=9.141,P =0.001 ;for peripheral blood F =8.389,P =0.001 ),and it was higher in model group ( center:2.08 ± 0.64; peripheral blood:1.43 ± 0.46) than that in two control groups ( control group1,center:1.17 ± 0.42,peripheral blood:0.78 ± 0.39 ; control group 2,center:1.31 ± 0.44,peripheral blood:0.79 ± 0.37 ),but no statistical significant difference existed between two control groups.There was positive correlation of Nrg1 mRNA between center and periphery.There was significant difference of Nrg1 protein in prefrontal lobe,dentate band and hippocamp among 3 groups ( F value was 7.275,21.50 and 4.619,and P value was 0.003,0.000 and 0.019,respectively),and it was higher in model( 7.71 ± 2.55,11.67 ± 1.83and 10.18 ±2.08,respectively)than that in two control groups( control group 1:4.89 ± 1.06,7.53 ± 1.14 and 7.10 ± 2.52,respectively; control group 2:5.31 ± 1.39,8.10 ± 1.60 and 7.81 ± 2.50),but no statistical significant difference existed between two control groups.ConclusionMK801 can effect on the expression of Nrg1 gene,Nrg1 mRNA and protein increase in MK801 model rat,and the change is synchronous between center and periphery.
7.Comparative study between Charlison′s weighted index of comorbidities score,the diagnostic criteria for emergency sepsis score and combination of the two scoring systems to predict the emergency pulmonary infection prognosis

Objective To compare the predictive effect of Charlison's weighted index of comorbidities (WIC), the diagnostic criteria for emergency sepsis (MEDS) and combination of the two scoring systems to predict the emergency pulmonary infection prognosis. Methods A total of 327 patients with pulmonary infection admitted from January 2016 to January 2017 were enrolled in this study whose WIC score,MEDS score and risk stratification were recorded at admission.They were divided into survival group and death group according to the 28 d treatment outcome,the optimal cutoff of WIC score and MEDS score to predict the prognosis were found by ROC curve, and the prediction effect of WIC score, MEDS score, the combined use of both and APACHEⅡto predict the prognosis were compared. Results The mortality of low,middle and high risk of WIC score were 13.7%(29/212),48.7%(38/78)and 78.4%(29/37)with significant difference(χ2=82.097,P=0.000),mortality of low,middle and high risk of MEDS score were 11.3%(23/203),50.6%(40/77)and 73.3%(33/45)with significant difference(χ2=145.526,P=0.000).The WIC scores in survival group and death group were 1.3 ± 0.9 and 2.7 ± 1.1 with significant difference(t=11.030,P=0.000).The MEDS score of live group(6.1 ± 4.0) was significantly lower than death group(12.6 ± 4.9)(t=11.502,P=0.000).the optimal cutoff values of WIC and MEDS to predict prognosis were 1.7 points, 11.6 points, the ROC curve area between WIC, MEDS score and combined application to predict prognosis were 0.632, 0.798, 0.897, and the sensitivity and accuracy of the combined prediction[93.8%(212/226)/89.9%(294/327)] were significantly higher than those of the individual WIC[72.7%(168/231)/75.2%(246/327)] and MEDS[67.5%(156/232)/72.2%(236/327)] (χ2=0.562-42.594, P < 0.05). The sensitivity and accuracy of the combined application and APACHE Ⅱto predict of prognosis had no statistical significant difference(P>0.05).Conclusions The sensitivity and accuracy of WIC score combined with MEDS score to predict the prognosis of patients with acute lung infection is higher than the individual WIC score and MEDS score,and its prediction effect is more better.
8.Efficacy observation of P-Gemox chemotherapy combined with intensity modulated radiotherapy in treatment of extranodal NK/T cell lymphoma, nasal type

Objective:To evaluate the therapeutic efficacy and side effects of P-Gemox regimen combined with intensity modulated radiotherapy in the treatment of extranodal NK/T cell lymphoma, nasal type (ENKTL).Methods:The data of 60 patients with ENKTL confirmed by pathomorphology and immunohistochemistry in Guizhou Cancer Hospital from July 2014 to October 2019 were retrospectively analyzed. All patients received P-Gemox chemotherapy combined with intensity modulated radiotherapy (at least 2 cycles), and the efficacy and adverse reactions were evaluated.Results:The complete remission rate of 60 patients was 65.0% (39/60), the partial remission rate was 25.0% (15/60), and the total effective rate was 90.0% (54/60). The main side reactions were myelosuppression, transaminase elevation and radiation mucositis; most of them were mild to moderate, which were relieved after treatment or the withdrawal of radiotherapy and chemotherapy. No treatment-related death cases were found. The overall survival rate of 1-year, 2-year, 3-year was 91%, 75% and 69%; the progression-free survival rate of 1-year, 2-year, 3-year was 86%, 68% and 62%. During the treatment, 3 cases died due to the progress of the disease and infection. Multivariate analysis showed that with and without hemophagocytic syndrome and radiotherapy dose were related to prognosis (all P < 0.05). Conclusion:P-Gemox, as the first-line induction chemotherapy regimen combined with intensity modulated radiotherapy has good short-term efficacy and safety for patients with ENKTL.
9.Advances in the biosynthesis of cyclodipeptide type natural products derived from actinomycetes.

Cyclodipeptide (CDP) composed of two amino acids is the simplest cyclic peptide. These two amino acids form a typical diketopiperazine (DKP) ring by linking each other with peptide bonds. This characteristic stable ring skeleton is the foundation of CDP to display extensive and excellent bioactivities, which is beneficial for CDPs' pharmaceutical research and development. The natural CDP products are well isolated from actinomycetes. These bacteria can synthesize DKP backbones with nonribosomal peptide synthetase (NRPS) or cyclodipeptide synthase (CDPS). Moreover, actinomycetes could produce a variety of CDPs through different enzymatic modification. The presence of these abundant and diversified catalysis indicates that actinomycetes are promising microbial resource for exploring CDPs. This review summarized the pathways for DKP backbones biosynthesis and their post-modification mechanism in actinomycetes. The aim of this review was to accelerate the genome mining of CDPs and their isolation, purification and structure identification, and to facilitate revealing the biosynthesis mechanism of novel CDPs as well as their synthetic biology design.
