3.Enzyme histochemical study of germanium dioxide-induced mitochondrial myopathy in rats.

The purpose of this study were 1) to determine the earliest pathological changes of germanium dioxide (GeO2)-induced myopathy; 2) to determine the pathomechanism of GeO2-induced myopathy; and 3) to determine the minimal dose of GeO2 to induce myopathy in rats. One hundred and twenty five male and female Sprague-Dawley rats, each weighing about 150 gm, were divided into seven groups according to daily doses of GeO2. Within each group, histopathological studies were done at 4, 8, 16, and 24 weeks of GeO2 administration. Characteristic mitochondrial myopathy was induced in the groups treated daily with 10 mg/kg of GeO2 or more. In conclusion, the results were as follows: 1) The earliest pathological change on electron microscope was the abnormalities of mitochondrial shape, size and increased number of mitochondria; 2) The earliest pathological change on light microscope was the presence of ragged red fibers which showed enhanced subsarcolemmal succinate dehydrogenase and cytochrome c oxidase reactivity; 3) GeO2 seemed to affect the mitochondrial oxidative metabolism of muscle fibers; 4) GeO2 could induce mitochondrial myopathy with 10 mg/kg of GeO2 for 4 weeks or less duration in rats.

4.Sex Determination with Sex Chromatin of Epithelial Cells from the Oral Mucosa in Koreans.

The authors studied 100 Koreans for sex determination using the sex chromatin of the epithelial cells from the oral mucosa according to Marberger's method. Turbid suspensions obtained by scraping the buccal mucosa were used; these were stained with a modified Feulgen technic for DNA. Summarizing our observations, the percentage of the sex chromatin in the nuclei of the epithelial cells showed a remarkable difference between the male and feamle of 23.6%. The authors confirmed the fact that the method of oral smear which had already been introudced by Marberger and his associates (1955) might be applicable to the Korean, also.

5.Histological Studies on the Degranulation of Mesenteric Mast Cells of the Rat by Water Extracts of Ginseng.

Ever since Fujidani made his report in 1905, many workers have studied the chemical components of Panax Ginseng and their effects on depression of blood pressure. Ri1ey (1952) and other workers have demonstrated the degranulation of mast cells in experimental animaIs treated with some histamine liberators, and the existence of a histamine liberator in the water extract of ginseng has been demonstrated by pharmacological assay by Lee et aI (1960). This present study was intended observe the disruption and degranulation of mesenteric mast cells of rats administered the water extract of ginseng, which might contain the histamine liberator. Variab1e doses of the water extract were injected intraperitoneally, and the degranulation of mesenteric mast cells was histologically demonstrated by means of toluidin blue, Giemsa, May-Gr nwa1d and Wright's stains. Degranulation began in the experimental group given 4ml of the extract mixed with 16cc. of Tyrode solution; the severity of degranulation increased probably with the dose of the extract, and extreme degranulation took place in the groups injected with dose of 6 or 8ml of the extract.

6.The Morphologic Changes of Parvalbumin- Immunoreactive Interneurons of the Dentate Gyrus in Kainate-Treated Mouse Hippocampal Slice Culture Epilepsy Model.

PURPOSE: Loss of hippocampal interneurons in dentate gyrus has been reported in patients with severe temporal lobe epilepsy and in animals treated with kainic acid(KA). Interneurons contain Ca2+- binding protein parvalbumin(PV). The effects of kainic acid on parvalbumin-immunoreactive (PV-IR) interneurons in dentate gyrus were investigated in organotypic hippocampal slice cultures. METHODS: Cultured hippocampal slices from postnatal day nine C57/BL6 mice were exposed to 10 muM KA, and were observed at 0, 8, 24, 48, 72 hours after a one hour KA exposure. Neuronal injury was determined by morphologic changes of PV-IR interneuron in dentate gyrus. RESULTS: Transient(1 hour) exposure of hippocampal explant cultures to KA produced marked varicosities in dendrites of PV-IR interneuron in dentate gyrus and the shaft of interbeaded dendrite is often much thinner than those in control. The presence of varicosities in dendrites was reversible with KA washout. The dendrites of KA treated explants were no longer beaded at 8, 24, 48 and 72 hours after KA exposure. The number of cells in PV-IR interneurons in dentate gyrus was decreased at 0, 8 hours after exposure. But there was no significant difference in 24, 48 and 72 hours recovery group compared with control group. CONCLUSIONS: The results suggested that loss of PV-IR interneurons in dentate gyrus is transient, and is not accompanied by PV-IR interneuronal cell death.

7.Two cases of human infection by Echinostoma cinetorchis.

Eggs of Echinostoma sp. were found from two patients who were hospitalized between Oct. and Dec. 1985. Single dose of 10 mg/kg of praziquantel was prescribed to each patient and after purgation with magnesium salt seven and two adult worms of Echinostoma cinetorchis in respect were recovered from patients. The first case was a 54-year-old male residing at Chungju city in Chungbuk province. He was hospitalized because of severe weight loss and frequent urination. The second case was a 71-year-old male residing at Chungsun-kun in Kangwon province, and hospitalized due to suspected lung cancer. The average size of the eggs found in the stools was 98 x 63 micrometer (the first case) and 100 x 67 micrometer (the second case) respectively, and total nine worms collected were identified as E. cinetorchis.

8.Development of Korean version of World Health Organization Disability Assessment Schedule II (WHODAS II-K) in Community Dwelling Elders.

OBJECTIVES: This study aimed to develop the Korean version of World Health Organization Disability Assessment Schedule II (WHODAS II-K) with community dwelling elderly population. METHODS: The WHODAS II-K was administered to 1204 community residents aged 65 or over in two areas of Kwangju, South Korea, in 2001. For assessing 'health condition', data on physical illness, depression (Korean version of Geriatric Mental State Schedule B3), and cognitive dysfunction (Korean version of Mini-Mental State Examination) were collected. For evaluating 'contextual factors', informations on demographic characteristics (age, gender, living area, marital state, and religiou), socioeconomic status (education, type of accommodation, number of room, previous occupation, and current employment), and social network were obtained. RESULTS: WHODAS II-K showed high levels of internal consistency, split-half reliability, and inter-rater and test-retest reliabilities. In the correlation analyses, scores on the WHODAS II-K were significantly correlated with the unfavorable conditions in the all variables on health condition and contextual factors. Partial correlations of scores on the WHODAS II-K with health condition were significant even after controlling for contextual factors. CONCLUSION: The WHODAS II-K is a reliable and valid instrument for assessing disability in elderly population since it reflects physical illness, depression, and cognitive impairment, which are common in elderly.

Background:
For acute ischemic stroke (AIS) patients with history of prior stroke (PS) and diabetes mellitus (DM), intravenous recombinant tissue plasminogen activator (IV-tPA) therapy in the 3- to 4.5-hour window is off-label in Korea. This study aimed to assess the safety and efficacy of IV-tPA in these patients.
Methods:
Using data from a prospective multicenter stroke registry between January 2009 and March 2021, we identified AIS patients who received IV-tPA in the 3- to 4.5-hour window, and compared the outcomes of symptomatic intracranial hemorrhage (SICH), 3-month mortality, 3-month modified Rankin Scale (mRS) score 0-1 and 3-month mRS distribution between patients with both PS and DM (PS/DM, n=56) versus those with neither PS nor DM, or with only one (non-PS/DM, n=927).
Results:
The PS/DM group versus the non-PS/DM group was more likely to have a prior disability, hypertension, hyperlipidemia, coronary heart disease and less likely to have atrial fibrillation. The PS/DM and the non-PS/DM groups had comparable rates of SICH (0% vs. 1.7%; p>0.999) and 3-month mortality (10.7% vs. 10.2%; p=0.9112). The rate of 3-month mRS 0-1 was non-significantly lower in the PS/DM group than in the non-PS/DM group (30.4% vs. 40.7%; adjusted odds ratio [95% confidence interval], 0.81 [0.41-1.59]).
Conclusions
In the 3- to 4.5-hour window, AIS patients with PS/DM, as compared to those with non-PS/DM, might benefit less from IV-tPA. However, given the similar risks of SICH and mortality, IV-tPA in the late time window could be considered in patients with both PS and DM.
