1.Effects of Tonifying Kidney-Yin on the Activities of Embryonic Stem Cell

Objective To study the effects of tonifying kidney-yin on activities of embryo stem cell. Methods Embryonic stem cell line CRL-1825 was choosed as model cell in present study, and tonifying kidney-yin herb serum was added to CRL-1825 cell. The influence of tonifying kidney-yin on stem cell differentiation, cell cycle, apotosis and express of key genes were observed. Results Tonifying kidney-yin can inhibit stem cell differentiation and apotosis, promote stem cell proliferation and progression of cell cycle. In addition, tonifying kidney-yin can up-regulate Wnt, Oct4 gene expression, and down-regulation P16INK4a expression. Conclusions Tonifying kidney-yin can promote stem cell proliferation and maintain the state of stem cell.
2.Pharmacokinetics Study of Danshensu in Danqi Injection in Dogs

Objective To study the pharmacokinetics of Danshensu (tanshinol) in Danqi Injection(DQI) in dogs. Methods HPLC-UV detection was used to determine the Danshensu level in biological samples. The Lichrospher C18 column(4.6?250 mm,5?m) was used as an analytical column with a mobile phase consisted of acetonitrile-1 %acetic acid (8∶92),the flow rate being 1.0mL?min-1 and the wave-length being 280 nm. Results After intravenous injection of DQI in dogs,the plasma concentration-time curve of DQI in dogs fitted well to a two-compartment model,with the characteristics of fast absorption and slow elimination.Its pharmacokinetics parameters are as follows:?=1.533 h-1,?=0.5111 h-1,T(peak)=0.25 h,T1/2?=0.573 h. Conclusion The pharmacokinetics of DQI in dogs fits the two-compartment model.In the pharmacokinetics of Danshensu,elimination course is the main course with the slow and lasting characteristics.
3.The correlation between chronic infection of microorganism and the severity of coronary artery disease

Objective To study the correlation between chronic infection of three microorganism and coronary artery lesion by detecting their antibodies and high-sensitivity C-reactive protein(hs-CRP) .Methods According to their severity of coronary artery lesion ,patients enrolled in the study were divided into 4 groups ,including normal control group(n=37) ,single-branch lesion group (n=36) ,double-branch lesion group(n= 30) and multi-branch lesion group(n= 29) .Serum Chlamydia pneumonia IgG(CPN-IgG) ,Helicobacter pylori IgG(HP-IgG) ,human cytomegalo virus IgG (HCMV-IgG) and hs-CRP tests were performed .Results CPN-IgG ,HP-IgG ,HCMV-IgG positive rates and the mixed infection positive rates increased sequentially from normal group to multi-branch group .There were significant differences of CPN-IgG ,HP-IgG ,HCMV-IgG and hs-CRP contents among the 4 groups (P<0 .05) .Logistic analysis showed that CPN-IgG ,HP-IgG and HCMV-IgG were dangerous factors to coronary artery lesion . Conclusion Chronic infections of three microorganism might affect the development of coronary artery lesion .
4.IGF-Ⅱ Promotes the Proliferation and Apoptosis of Human Colorectal Adenocarcinomas and Its Clinical Significance

Objective To investigate the relationship among IGF-II expression and cell proliferation and apoptosis in human colorectal adenocarcinoma. Methods 60 paraffin embedded samples of human colorectal adenocarcinomas were used in this study. IGF-ⅡmRNA expression was detected by in situ hybridization, proliferation cell nuclear antigen(PCNA) protein was measured by immunohistochemical staining and apoptosis was determined by TUNEL technology. Fifteen normal colorectal tissues were used as controls. Computerized image analysis system qualitatively analyzed the positive ratio of above three indices in the cancer and normal tissues. 5.Protective effect of sodium selenite on human keratinocytes from ultravio-let-B irradiation

AIM: To investigate the protective effect of sodium selenite ( Na2 SeO3 ) on human keratinocytes under ultraviolet-B (UVB) irradiation.METHODS: The cultured HaCaT cells were divided into 4 groups: (1) normal control group;(2) Na2 SeO3 group:pretreated with Na2 SeO3 at doses of 10 nmol/L, 50 nmol/L, 100 nmol/L, 200 nmol/L and 1 μmol/L for 24 h;(3) UVB group: irradiated with UVB at doses of 300, 600 and 900 J/m2; (4) Na2SeO3 +UVB group:after pretreated with Na2SeO3 for 24 h, irradiated with UVB at doses of 300, 600 and 900 J/m2.The cell pro-liferation was detected by MTT assay .The apoptotic rates of HaCaT cells treated with UVB at dose of 300 J/m2 were as-sessed by flow cytometry .RESULTS:Compared with normal control group , the cell proliferation activity in UVB group de-creased significantly ( P<0.05 ) .The cell activity was inversely correlated with the irradiation intensity .No significant difference of the cell activity between Na 2 SeO3 group and normal control group was observed .The cell proliferation in Na2SeO3 +UVB group was higher than that in UVB group significantly (P<0.05).Na2SeO3 at concentration of 100 nmol/L showed the strongest activity to promote cell proliferation .After 300 J/m2 UVB irradiation, the apoptotic rate in Na2SeO3+UVB group decreased significantly ( P<0.05) compared with UVB group .The inhibitory effect of Na 2 SeO3 at concentra-tion of 100 nmol/L on apoptosis was the strongest .CONCLUSION: The damage of human keratinocytes by UVB irradia-tion is in a dose-dependent manner .The photoprotection performance of Na 2 SeO3 reduces the damage of human keratino-cytes induced by UVB irradiation .
6.Effects of estrogen versus letrozole on chicken embryo frontal bone osteoblast

BACKGROUND: There are plenty of studies of estrogen effects on mammalian osteoblast, but the studies of estrogen effects on bird osteoblast cannot be found. There are many reports about the side effects of letrozole on bone metabolism, but there are no reports about the effect of letrozole on osteoblast.OBJECTIVE: The effects of estrogen and letrozole on the proliferation, cell cycle, estrogen receptor mRNA expression and alkaline phosphatase (ALP) activity of chicken osteoblast in vitro were studied in order to illustrate the mechanism of medullary bone osteogenesis.METHODS: The osteoblasts were harvested from the frontal bone of 15-day SPF chicken embryos by the enzyme digestion, and treated with various mass concentrations of estrogen (0, 5, 10, 20, 100. 200, 400,800, 2 000, 20 000 ng/L) and letrozole (0, 5.10, 25, 50, 100, 250, 500, 1 000, 5 000 μg/L). The proliferation rates of the osteoblast treated with estrogen or letrozole were measured through the MTT method, The ALP activities of osteoblast were measured by the pNPP method. The cell cycle was measured by flow cytometry. The expression of estrogen receptor mRNA was detected using real-time fluorescent quantitative polymerase chain reaction (PCR).RESULTS AND CONCLUSION: The estrogen could promote proliferation of osteoblast in concentration- and time-dependent fashion. Estrogen could increase the expression of estrogen receptor mRNA, impulse cell cycle, and elevate ALP activities.Letrozole could increase the cell population, impulse cell cycle, inhibit estrogen receptor mRNA expression, but letrozole has no effects on ALP synthesis and secretion.
7.CW bio-radar vital sign detector and experiment study.

Non-contact vital sign detection technique provides an effective usage in health monitoring applications. A vital sign detector was designed based on microwave bio-radar technique. Using Doppler principle, continuous wave bioradar was designed for tiny body movement detection, short-time Fourier transform and interpolation algorithm were adopted for heart and respiration rate extraction, embedded system was used for system integration, real-time signal processing software was designed on it. Experiments were done by using simulation device and human body for research and performance evaluation. The result shows that the proposed prototype can be used for single target vital signs detection at the distance of 90 cm, and the heart rate result shows a 96% recognition rate.

8.Effect of Panax Notoginseng Saponins in Jiqi Injection on Pharmacokinetics of Silibinin in the Beagle Dogs

Objective To study the pharmacokinetic features of silibinin from Jiqi Injection(JI) in Beagle dogs,and to observe the effect of Panax notoginseng saponins,another active component in JI,on pharmacokinetics of silibinin. Methods The Beagle dogs received intravenous injection of JI and silibinin,and then its plasma sample was collected in different time. The plasma samples of Beagle were prepared by hydrolysis with sulfatase-? glucuronidase complex enzyme and liquid-liquid extraction with aether. A high-performance liquid chromatographic method was developed to determine the plasma concentration of silibinin,and the pharmacokinetic parameters were performed by BAPP2.3 program. Results The pharmacokinetics of two tested preparations met with two-compartment model. There were not significant differences between pharmacokinetic parameters of JI and that of Silibinin Injection. Conclusion The silibinin in Jiqi Injection has a fast in-vivo clearance rate after intravenous injection,and Panax notoginseng saponins have no effect on its pharmacokinetic parameters.
9.Study on the Relationship between Severity of Coronary Artery Lesion and the Level Cystatin C,Nitric Oxide and Superoxide Dismutase of Serum

Objective To study the correlation between coronary lesion and Cystatin C (CyC),Nitric Oxide (NO)and Super oxide dismutase,accumulate the clinical experience in prevention and diagnose of Coronary Heart Disease (CHD)by analy-zing the contents of serum Cystatin C (CyC),Nitric Oxide and Superoxide Dismutase in patients with coronary lesion.Meth-ods According to their severity of coronary artery lesion,all cases were divided into four groups:Normal compared group (40 cases);Simple branch pathological group (44 cases);Double branch pathological group (43 cases):Multi-branch patho-logical group (35 cases).All cases were tested CyC,NO,SOD and hs-CRP in serum.Results The level of CyC and hs-CRP were increased gradually from normal group to multi-branch group.The level of NO and SOD were decreased gradually from normal group to multi-branch group.There were significant differences the levels of CyC,NO,SOD and hs-CRP among the four groups.Coronary lesion Gensini in the CHD patients had significant positive relationship to the level of CyC or hs-CRP respectively (r=0.473,0.429),had significant negative relationship to the level of NO or SOD respectively (r=-0.356, 0.384)(P<0.05).CyC in the CHD patients had significant positive relationship to the level of hs-CRP (r=0.458),had sig-nificant negative relationship to the level of NO or SOD respectively (r=-0.426,0.484,P<0.05).Conclusion By aggra-vation of coronary artery lesion,the level of CyC increased,reduced the contents of NO,SOD,aroused endothelial dysfunc-tion,induced occurrence and aggravation of coronary artery lesion.CyC,NO and SOD play important mark in the assistant diagnosis of the disease.
