1.The expression of ASC and Caspase-1 in liver cancer and its clinic significance

Objective To analyze the apoptosis-related spots like protein (ASC) and caspase -1 in HCC and adjacent noncancerous tissues and its clinical significance. Methods ASC and Caspase 1 were determined with immunohistochemistry LSAB method and the various indexes were analyzed in 30 cases of liver cancer and adjacent tissues which did not receive preoperative radiotherapy and chemotherapy. Results The positive rate of ASC in HCC and adjacent noncancerous tissues was 10.00% (3/30) and 73.33% (22/30). Positive rate between the groups had significant difference (P<0.01). Caspase-1 positive rate had significantly difference between the groups (23.33% (7/30) vs. 66.67% (20/30), P<0.01). ASC and the two indicators of Caspase-1 expression in cancer tissues were low, but no significant correlations between the two groups (P>0.05). ASC and the two indicators of Caspase-1 expression in adjacent tissue were significantly increased, and it showed a very significant correlation (r=0.722,P<0.01). Conclusions ASC, Caspase-1 expression in liver cancer were low expressed, and significantly lower than the adjacent tissues, which suggested that ASC and Caspase-1 were closely related to the occurrence and development of the apoptosis of liver cancer, and it might be used as diagnosis and treatment of targets in liver cancer.
2.Effect of uncoupling protein 2 on activation of hepatic stellate cell in liver fibrosis of rats

Objective To explore the role of uncoupling protein 2 ( UCP2) in the development of hepatic fibrosis and its molecular mechanism .Methods The CCl4-induced liver fibrosis rat model in vivo was established to observe the pathological changes of rat livers .The expression levels of UCP2 and p38 mitogen activated protein kinase (p38 MAPK) were detected by using the techniques of Western blotting , Real-time PCR and immunohistochemistry .The hepatic stellate cells ( HSC) were stimulated by CCl 4 and UCP2-specific inhibitor Genipin to mimic liver fibrosis in vitro.The expression levels of UCP2 and p38MAPK were determined by using Western blotting .Results We found that UCP2 and α-SMA expression levels increased significantly (P <0.05, n =10) in the liver of rats with CCl4-induced liver fibrosis when compared with that of the normal control rats in vivo.Similarly, the expression levels of UCP2 and p38 MAPK were up regulated (P <0.05, n=6) in CCl4-treated HSC cells in vitro.However, the expressions of UCP2 and p38 MAPK were down regulated (P <0.05, n=6) in genipin-treated HSC cells in vitro.Conclusion UCP2 is involved in liver fibrosis, and probably contributed to the activation and proliferation of hepatic stellate cells .

5.The clinical value of measurement of urine nuclear matrix protein 22 and urinary bladder cancer antigen content in patients of bladder transitional cell carcinoma

Objective To assess the feasibility of nuclear matrix protein 22(NMP22)and urinary bladder cancer,antigen (UBC) for the early diagnosis of bladder transitional cell carcinoma and its influencing factors.Methotis 105 subjects,including 60 patients of bladder cancer,25 patients of urological benign disease and 20 normal (healthy)individuals were enrolled in this study.Urine NMP22 and UBC wag assessed by enzyme-linked immunosorbent assay(ELISA).Urine NMP22 and UBC as well as exfoliocytology were conducted for the purpose to compare the sensitivity,specificity,positive and negative predictive value of these three ways.Results The sensitivity of NMP22(88.3%)and UBC(86.7%)were significantly better than exfolioeytology(40.0%,P<0.01).The specificity of NMP22,UBC and exfoliocytology were 80.0%,84.0%and 92.0%,respectively, the positive predictive values were 91.4%,92.9%and 92.3%,and the negative predictive values were 74.1%.72.4%and 38.9%.Conclusions NMP22 and UBC are sensitive,specific,simple,feasible and noninvasive diagnostic markers for the early detection of urinary bladder transitional cell cancer.
6.Effect of ligustrazine on Th17 cell and balance of Th17/Treg in Asthma model mouse

Objective:To explore the effect of ligustrazine on Th17 cell and the expression of Th17/Treg characteristic cytokines in a model of asthma.Methods: Male BALB/c mice were randomly devided into four groups , including the normal control group,the model group,the ligustrazine treatment group and the DXM treatment group ,with 10 mice in each group.In the model group, mice were sensitized with the OVA respectively.In the treatment group ,mice were also sensitized and challenged as model group and in addition were intraperitoneally injected with the ligustrazine or DXM.The mice of control group were intraperitoneally injected normal sodium instead of OVA and challenged by inhaling normal sodium instead of OVA in challenge phase respectively .Animals were sacrificed to detect the percentage of CD 4+CD25+T regulatory cells and Th17 cells in the blood were measured by flow cytometry ,and the cytokines,including IL-17 and IL-10,in the blood serum by ELISA.Results:The mice of model group showed the performances of a typical asthma.In treatment group ,the symptoms above mentioned became lighter.The mice of control group were generally in good condition;the lung tissue from the mice of model group showed widen alveolar septum , narrowed bronchial lumen , thicken bronchial wall,and numerous inflammatory cells infiltrated around the small vessels and bronchioles.In ligustrazine treatment group, the pulmonary inflammation was obviously relieved compared with the model group;the percentages of CD 4+CD25+T regulatory cell both in the peripheral blood and in the total CD 4+T cells were significantly lower in the mice of model group than in the mice of control group , and were higher in the treatment group than in the model group;the level of IL-10 in serum was significantly lower in the model group than in the control group ,and was higher in the treatment group than in the model group.The level of IL-17 of serum was significantly higher in the model group than in the control group , and was significantly lower in the treatment group than in the model group.Conclusion:Ligustrazine can adjust the Th 17/Treg balance and reduce the inflammation of lung tissue in asthma mice by up-regulating CD4+CD25+T regulatory cell and then restraining the Th 17 response , and play a preventive/control asthma attack role in asthma model mouse.

Hypoxic-ischemic brain damage is a serious complication of perinatal asphyxia. Enriched environment is the living condi-tions for the promotion of feeling, cognition and behavior. Enriched environment may promote the repairing of hypoxic-ischemic brain dam-age in term of praxiology, morphology and molecular biology, that enriched environment may regulate the secretion and release of various factors, and enhance synaptic plasticity, induce neuron regeneration, inhibit its apoptosis, and result in ethological and morphological recov-ery.
8.Biomechanical characteristics of bipolar femoral head prosthesis replacement and internal fixation for femoral neck fractures in the elderly

BACKGROUND:Bipolar femoral head prosthesis has achieved the desired repair effect in patients with femoral neck fracture, but there is stil much controversy on the biomechanical properties of bipolar artificial femoral head prosthesis. OBJECTIVE:To compare biomechanical properties of the bipolar femoral head prosthesis replacement and internal fixation in the repair of femoral neck fracture in the elderly. METHODS:Twenty-three (46 sides) senile femoral neck fracture specimens were analyzed and randomized into internal fixation group(n=23)and bipolar femoral head prosthesis replacement group (n=23). Specimens in the internal fixation group underwent conventional metal implant fixation. Specimens in the bipolar femoral head prosthesis replacement group underwent bipolar femoral head prosthesis replacement. Mechanical properties of femoral prosthesis stem were tested on universal testing machine. Biomechanical properties after bipolar femoral head prosthesis replacement and internal fixation were compared. RESULTS AND CONCLUSION:(1) The maximum loadwassignificantly higher in the bipolar femoral head prosthesis replacement group than in the internal fixation group (P< 0.05). The maximum load trend images showed that the maximum load was significantly higher in the bipolar femoral head prosthesis replacement group than in the internal fixation group (P< 0.05). (2) The displacement in axial pul test was significantly shorter in the bipolar femoral head prosthesis replacement group than in theinternal fixation group (P< 0.05). The displacement trend images exhibited that the displacement in axial pul test was significantly shorter in the bipolar femoral head prosthesis replacement group than in the internal fixation group (P< 0.05). (3) These findings suggest that compared with the implant, bipolar femoral head prosthesis replacement in the repair of femoral neck fracture in the elderly is more stable, has better repair effects, can elevate hip function and obtain good initial stability.
9.Comparison of Ku protein Expression in CNE1 and CNE2 nasopharyngeal carcinoma cell lines

Objective Radiosensitivity is highly correlated with DNA doub le stra nd breaks(DSBs). The repair of human DSB is possible mainly through nonhomologou s DNA end joining(NHEJ)pathway. This study was designed to determine the relat ionship between expression of 70Ku/ 80Ku prot ein (a modulating subunit of DNA-PK, which is an important component in NHEJ pathway) and radiosensitivity of nasoph aryngeal carcinoma cell lines. Methods Radiosensitivity parameters of the nasoph aryngeal carcinoma cell lines were obtained by the colony forming assay and radi ation dose-survival curve was done by linear quadratic model. Western blot was p erformed to determine the expression of 70Ku and 80 Ku in total extract of CNE1 a nd CNE2 at baseline level、different time after 4?Gy irradiation、12 h after di ff erent doses of irradiation. To perform a semi-quantitated assay of protein expr e ssion, the optic density (OD) value of each band was estimated by automatic imag e analysis system. Results Surviving fraction of CNE1 was higher than those of CNE2 at each dose point. Mean inactivation dose was higher in the CNE1(2.35)t han that in the CNE2(1.11), but the quantity of 70Ku/ 80Ku in either cell line was similar at both baseline and postirradiation levels. Conclusions CNE2 is mo re radiaosensitive than CNE1; as there was no marked correlation observed betwee n the quantity of Ku protein and radiosensitivity of the nasopharyngeal carcinom a cell lines. X-irradiation cannot result in any change in total quantity of Ku protein.
