1.Separative Purification of and Characteristic Research on Human Blood Type Substance P1

A Human blood type antigen P1 with very high activity was separated from sheep hydatid cyst fluid. Determination of the antigen by ultraviolet spectrum, polyacrylamide electrophoresis, gel filtration, and determination of its carbohydrate content and aminoacid residue had indicated that the substance was a glycoprotein, its total carbohydrate content was 80% about one-fifth of which was hexosamine, and other carbohydrate components were glucqse, galactose and glucosamine. Its protein consisted of 14 aminoacids, and its molecular weight was about 30000 Daltons. By immunizing the animals with this substance, the anti-P1 serum of high titer could be obtained. The successful extraction of this substance P1 with high activity would provide the abundant resource for reseach on human blood type antigen and its immunochemistry, and create the good conditions for studies in preparation and application of anti-P1 serum or monoclonal antibody toward P1.
2.Therapeutic Observation of Acupuncture plus Auricular Point Electroacupuncture for Post-stroke Depression and Its Effect on the Quality of Life

Objective To evaluate the clinical efficacy of acupuncture plus auricular point electroacupuncture and its effect on the quality of life (QOL).Method Ninety-eight patients with post-stroke depression were randomized into a treatment group and a control group, 49 cases in each group. The treatment group was intervened by acupuncture plus auricular point electroacupuncture, while the control group was by oral administration of Fluoxetine hydrochloride. The Hamilton Depression Scale (HAMD), Side-effect Rating Scale (SERS), and the World Health Organization Quality of Life Questionnaire-Brief Version (WHOQOL-BREF) were observed before and after intervention, and the clinical efficacies were also compared.Result The total effective rate was 89.1% in the treatment group versus 87.0% in the control group, and the between-group difference was statistically insignificant (P>0.05). Respectively after 2-week, 4-week, and 8-week treatment, the HAMD scores were significantly different from that before intervention in the treatment group (P<0.05). Respectively after 2-week, 4-week, and 8-week treatment, the HAMD scores were significantly different from that before intervention in the control group (P<0.05). There were no significant differences in comparing the HAMD scores between the two groups after 2-week, 4-week, and 8-week treatment (P>0.05). The SERS scores in the treatment group were significantly different from that in the control group after 4-week and 8-week treatment (P<0.05). In the treatment group, the SERS score after 8-week treatment was significantly different from that after 4-week treatment (P<0.05). After treatment, the scores of general health domain 1 and 4, physical health domain, psychological health domain, and environment domain from WHOQOL-BREF were significantly changed in both groups (P<0.05). After intervention, the physical health domain score of the treatment group was significantly different from that of the control group (P<0.05).Conclusion Acupuncture plus auricular point electroacupuncture is an effective approach in treating post-stroke depression, and it can improve the quality of life of the patients.

4.Pharmacokinetic Study on Luteolin and Cynaroside in Rats

OBJECTIVE:To establish the method for pharmacokinetic study of luteolin and cynaroside in rats and to determine pharmacokinetic parameters. METHODS:16 SD rats were randomly divided into luteolin group (sublingual iv,1.34 mg/kg) and cynaroside group(sublingual iv,0.64 mg/kg). 0.5 ml blood were collected before administration and 0,15,30 min and 1,2,3,4,6, 8,12,24,48 h after administration respectively to prepare plasma. UPLC-TQ-MS was adopted to determine plasma concentration, and pharmacokinetic parameters were calculated. A CORTECSTM UPLC? C18(100 mm×2.1 mm,1.6 μm)column was used with mobile phase consisted of acetonitrile-water (containing 0.1% formic acid) at a flow rate of 0.4 ml/min,the column temperature was set at 40 ℃,and quercetin was used as internal standard. RESULTS:The linear range of luteolin and cynaroside were 2.5-500 ng/ml (r=0.998 2) and 10-2 500 ng/ml (r=0.993 5). The lowest quantitation limits were 1 and 2.5 ng/ml,and extraction were 70.75%-87.72% and 75.40%-91.18%(n=6);RSD of inter-day and intra-day were all lower than 10%(n=3). Pharmacokinetic parameters as t1/2 were (1.88 ± 0.32) and (1.57 ± 0.08) h;CL were (0.77 ± 0.18) and (0.06 ± 0.01) L/(h·kg);AUC0-6 h were (189.60±40.04)and(1 093.14±187.36)ng·h/ml;AUC0-∞ were(195.18±38.37)and(1 097.11±188.07)ng·h/ml. CONCLU-SIONS:The method can be used for pharmacokinetic study of luteolin and cynaroside in rats,and the pharmacokinetics of them in rats are in line with two-compartment model.
5.Expression of adenovirus-mediated CTLA4Ig-IRES-CTLA4 gene in bone marrow mesenchymal stem cells

Objective To construct adenovirus vector harboring CTLA4Ig-IRES-CTLA4 gene, and to express the gene in bone marrow mesenchymal stem cells (BMMSCs). Methods The cDNA fragments of CTLA4, and the extracellular domains of CTLA4 and IgGFc were cloned to pT-Easy vectors by RT-PCR from the activated splenocytes of Wistar rats. Recombinant adenovirus vector harboring CTLA4Ig-IRES-CTLA4 was produced from AdeasyTM Adenoviral Vector System by homologous recombination between the Adtrack vector (pAdtrack-GFP-CTLA4Ig-IRES-CTLA4) and the plasmid pAdEasy-1 containing most of the human adenovirus serotype 5 (Ad5) genome in E. coli BJ5183 strain, and then packaged and propagated in 293 cells. BMMSCs were infected with the recombinant adenovirus, and the co-expression of both CTLA4Ig and CTLA4 was then detected by RT-PCR and Western blotting. The immunosuppression function of the transfected BMMSCs was investigated by mixed lymphocyte response (MLR). Results Fusion gene CTLA4Ig was constructed successfully, and the recombinant CTLA4Ig-IRES-CTLA4 adenovirus was then generated by homologous recombination and packaged in 293 cells. The transduction efficiency of recombinant adenovirus in BMMSCs was elevated up to 98.67% determined by flow cytometry using a GFP as marker. CTLA4Ig and CTLA4 were expressed at the same time in BMMSCs detected by RT-PCR and Western blot, and the transfected stem cells showed stronger immunosuppression function than that of BMMSCs or BMMSCs transduced by Ad-CTLA4Ig. Conclusion The BMMSCs transfected by the recombinant adenovirus can co-express CTLA4Ig and CTLA4, which improved the immunosuppression function of BMMSCs in the way of costimulatory pathway blockade.
6.Influence of Sijunzi Decoction in various dosages on digestive and immune function of mice with Pi-deficiency syndrome

Objective To study the influence and the optimal dosages of Sijunzi Decoction (SJZD) on treating digestive and immune function of mice with Pi-deficiency syndrome (PDS). Methods The experimental model of mice with PDS was induced by rhubarb decoction. Then the mice were treated with three dosages of SJZD (12.5, 25, and 37.5 g/kg), respectively. The body weight, body temperature, mean retention time of chyme in digestive tract, gastric emptying velocity, gastrin and motilin levels, and visceral index of mice were measured. Results Being compared with the normal group, the PDS mice showed a slower growth of body weight and a lower body temperature, a shorter mean retention time of chyme in digestive tract and faster gastric emptying velocity, a lower gastrin content, and a higher motilin content. Also splenic and thymus indexes were found to follow a downtrend in the PDS mice. Three dosages of SJZD showed a restoration effect on the above all indexes, and the high dosage was the most effective one, especially. Conclusion The high dosage (37.5 g/kg) of SJZD possibly is the optimal dosage on treating the digestive and immune diseases of PDS mice.
7.Determination of Warfarin and Its Metabolite in Human Plasma by Ultra Performance Liquid Chromatog-raphy Tandem Quadrupole Mass Spectrometry (UPLC-MS/MS)

Objective:To establish an ultra performance liquid chromatography-tandem quadrupole mass spectrometry method for the determination of warfarin and its metabolite 7-hydroxywarfarin in human plasma. Methods: An ACQUITY UPLC? BEH C18 (50 mm × 2. 1 mm, 1. 7 μm) column was used as the stationary phase at 40℃. The mobile phase consisted of acetonitrile and water (con-taining 0. 1% formic acid) with gradient elution at a flow rate of 0. 4 ml·min-1 . Warfarin-d5 was used as the internal standard. The analytes were detected on a triple-quadrupole mass spectrometer equipped with an ESI interface in a positive mode. Results:The reten-tion time of warfarin and 7-hydroxywarfarin was 1. 8 min and 1. 5 min, respectively. Excellent linear calibration curve of warfarin and 7-hydroxywarfarin was obtained within the concentration range of 25-2 000 ng · ml-1 ( r =0. 999 3 ) and 5-500 ng · ml-1 ( r =0. 999 6), respectively. The lower limit of quantification of warfarin and 7-hydroxywarfarin was 5 ng·ml-1 and 2. 5 ng·ml-1 with the average recovery of 96. 9%-105. 3% and 97. 1% -103. 3%, respectively. The intra-and inter-day standard deviations were both less than 10%. Conclusion: The method is accurate and simple, and suitable for the determination of warfarin and its metabolite 7-hydroxywarfarin in human plasma.
8.The investigation of doctor-patient communication skills of medical students and the exploration of training measures

ObjectiveTo investigate and analyze the doctor-patient communication skills of medical students and hereby to explore the efficient training system.MethodsUsing the questionnaires and statistic analysis,we focused on the doctor-patient communication skills of the observed 914 medical students in our college,their awareness of doctor-patient communication and the existing problems in the field.Results96.6% of medical students realized the importance and necessity of doctor-patient communication,while 62.7% of them lacked such kind of communication skills.Additionally,some other problems are exposed in medical history taking.ConclusionDoctor-patient communication skills might be the most important prerequisite for establishing harmonious relationship between doctors and patients,improving the quality of medical service and reducing the number of medical disputes.Consequentially,it's time to redesign relevant curriculum,to improve the old-fashioned training methods,to enhance the practice of doctor-patient communication training.Furthermore,improvements of teaching quality and a scientific evaluation system are also effective approaches to resolving the issue.
9.Clinical characteristics and treatment of infective endocarditis in children

Objective To study the clinical characteristics, treatment and prognosis of infective endocarditis in children. Methords Clinical data from 83 patients of infective endocarditis admitted from 1998 to 2012 were retrospectively analyzed. Results In a total of 83 patients, there were 53 males and 30 females, and the average age was 6.8±4.6 years. The main clinical characteristics were fever (77.1%) and mild to moderate anemia (71.1%). The C-reaction protein (67.5%), erythrocyte sedimen-tation rate (60.2%), and white blood cell (47.0%) were elevated. Twenty (24.1%) patients had embolism. Blood culture was pos-itive in 56 (67.5%) cases with bacteria mainly being Gram-positive and Streptococcus and Staphylococcus accounted for 89.3%. Vancomycin and other sensitive antibiotics were effective. Neoplasm was detected in 68 cases (82%) by transthoracic echocar-diograerphy. Fifty-ifve (66.2%) patients underwent cardio surgery. Seven patients (8.4%) died. Conclusion In recent years, the distribution of pathogenic bacteria in infective endocarditis had changed. Streptococcus mitis and Staphylococcus aureus has become a major pathogens and need to be treated by vancomycin and other sensitive antibiotics. The detection rate of neoplasm is higher by echocardiography.
10.Determination of Equilibrium Solubility of Albendazole and Its Apparent Oil-Water Partition Coefficient

OBJECTIVE:To determine the equilibrium solubility of albendazole(ABZ)and its apparent oil-water partition coef-ficient. METHODS:The equilibrium solubility of ABZ in various solutions was determined at 37 ℃ by HPLC and saturation solu-bility method,including water,7 kinds of common organic solvents with different polarities(methanol,ethanol,etc.),organic ac-id(oleic acid,glacial acetic acid,lactic acid,formic acid),hydrochloric acid of pH 1.2,phosphate buffer solution(PBS)of dif-ferent pHs(2.0-7.8)and 6 kinds of common surfactants with mass concentrations of 10,50 and 100 mg/ml(polysorbate 80,polox-amer,etc.). The apparent oil-water partition coefficient(P)was calculated with the concentration ratio of ABZ in oil phase(N-oc-tyl alcohol)and water phase(water and PBS of different pHs)after partition equilibrium. RESULTS:The equilibrium solubility of ABZ reached(0.26±0.02)μg/ml in water,with the lgP of 3.66±0.01. The equilibrium solubility of ABZ in common organic sol-vents and organic acids was higher than in water. The higher the polarity of the organic solvent was and the weaker of organic acid was,the weaker of its ability to solubilize ABZ would be. The equilibrium solubility of ABZ was higher in the medium of pH 1.2-2.5 than in that of pH 5.0-7.8. The ability of the surfactant to solubilize ABZ was related to its type,and the higher of the mass concentration of the surfactant was,the stronger of its ability to solubilize ABZ became. lgP was less than 1.6 at pH 1.2-2.0 and changed little at pH 5.0-7.8 [(3.71 ± 0.26)-(3.68 ± 0.26)]. CONCLUSIONS:ABZ is insoluble in water. Its equilibrium solubility demonstrates a negative correlation with the polarity of the organic solvent and a positive correlation with the acidity of the organic acid and with the mass concentration of the surfactant. It has higher water solubility and lower lipid solubility in a strong acidic en-vironment,and higher lipid solubility and weaker water solubility in weak acidic,weak basic and neutral environments.
