1.The regulatory effect of endothelial cells of pulmonary arteriole on the tone of pulmonary artery

The regulatory effect of endothelial cells of the rabbit pulmonary arteriole(PAECs) on the tone of pulmonary artery was studied. The conditioned medium (CM) ofPAECs, cultivated under normal and hypoxic condition for 24 hours, did not induce con-striction of pulmonary artery ring. The aqueous phase was able to induce the constriction.The constriction of pulmonary artery rings was reduced by the heated aqueous phase, but it was disappeared when the heated aqueous phase was treated with trypsin. The constric-tion caused by heated aqueous phase was stronger in hypoxic endothelial cells than innormal cells. Treatment of PAECs with indomethacin and captoril, and administration ofFPL 55712, BN 52021, chlorpheniramine and prazozin to contraction pool did not affectthe constriction of pulmonary artery rings. It was concluded that PAECs might producelong half-life lipid substances which could induce the dilatation of pulmonary artery.Hypoxia inhibted the release of long half-life aqueous heat-unstable vasoconstrictive sub-stances. but enhanced that of the heat-stable fraction which was probably a peptide.
2.Influence of Package on Weight and Appearance and Active Ingredient Content of Unguent

OBJECTIVE:To study the influence package on weight and appearance and active ingredient content of unguent.METHODS:Unguent samples from6hospitals were examined and their appearance observed by adopting Ch.P.2000“Determination of least volume”and active ingredient content of analytic methods for each sample and observe appearance.RESULTS:No remarkable change of airtight packing samples and differ change of general packing samples.CONCLUSION:Suggested to improve the packing and original validity can be suitable for unguent of airtight packing.
3.Gene transfection mediated by ultrasound and Pluronic P85 in HepG2 cells.

In order to assess whether gene transfection could be mediated by ultrasound in association with P85 and find the appropriate parameters of ultrasound irradiation, the effects of ultrasound with or without P85 on gene transfection of HepG2 cells were examined. The HepG2 cells were irradiated by ultrasound at 1 MHz, 0.4-2.0 W/cm(2) and 50% duty cycle with plasmid encoding enhanced green fluorescent protein (EGFP) as a report gene. Forty-eight h later, the expression of EGFP was detected under the fluorescence microscopy. Transfection efficacy was quantitatively assessed by flow cytometry, and cell viability was evaluated by trypan blue exclusion. The results showed that the transfection efficacy was increased with the increases in ultrasound output power and the ideal transfection efficacy was achieved in HepG2 cells irradiated by ultrasound at 0.8 W/cm(2) for 30 s. The transfection efficacy in ulstrasound+P85 group was three times higher than in single ultrasound group [(17.63+/-1.07)% vs (5.57+/-0.56)%, P<0.05]. The cell viability was about 81% and 62% in ultrasound group and ultrasound+P85 group respectively. It was concluded that ultrasound in combination with P85 could mediate the gene transfection of HepG2 cells, ideal transfection efficacy was achieved by ultrasound irradiation at 0.8 W/cm(2) for 30 s, and P85 could somewhat increase the damage to cells caused by ultrasound.

4.Mechanism and abnormal expression of sox4 gene in tumor

Recent studies have revealed that sox4 gene expresses abnormally in many kinds of tumor tissues and it probably involves in tumorigenesis,development and metastasis of cancer.Regulating proliferation,differentiation,apoptosis and other process may participate in the work mechanisms of sox4 gene.Therefore,further studies about the relationship between sox4 gene and tumor would provide new ideas of exploring special diagnosis markers and novel targets for tumor therapy.
5.Application of combined therapy based on the principle of machanical ventilation in acute respiratory failure induced by aspiratory pneumonia

6.Application of multi-slice spiral CT angiography in diagnosis of cerebral vascular diseases

Objective To evaluate the application of multi-slice CT angiography(MSCTA) in cerebral vascular diseases.Methods 16-slice spiral CT angiography of 32 patients which clinically suspected cerebrovascular diseases were analyzed retrospectively.Source images were processed by GE Lightspeed CT Scanner;all axial images were transferred to an external workstation.The reconstructed images were processed VR,MPR,MIP in working-station.Results 10 of the 32 patients were normal, 17 were aneurysms,could be clearly displayed with the source, size,and neck width.3 were AVM,the location and scope could be observed.2 were vaso-occlusive.Conclusion Multislice spiral CT angiography is a safe ,quick and noninvasive method in diagnosing the cerebrovascular disease,it may be proposed as a first choice technique for screening and preoperative assessment for cerebrovascular disease.CTA is a valuable diagnostic method for various intracranial cerebral disease.
7.Clinical assessment of pelvic external fixation in hemodynamically unstable patients with pelvic fracture

Objective To evaluate the clinical effect of emergency pelvic external fixation in hemodynamically unstable patients with pelvic fracture.Methods The clinical data of 56 hemodynamically unstable patients with pelvic fracture from January 2008 to December 2013 were selected.The changes of hemoglobin,mean arterial pressure,heart rate,serum lactate,urine volume,norepinephrine dosage before and after surgery for 24 h were compared and analyzed.Results Compared with those before surgery,the levels of hemoglobin,mean arterial pressure,serum lactate,urine volume,heart rate after surgery for 24 h were significantly improved:(109 ± 12) g/L vs.(69 ±22) g/L,(87.8 ±6.5) mmHg (1 mmHg =0.133 kPa) vs.(55.8 ± 7.7) mmHg,(6.3 ± 5.1) mmol/L vs.(10.8 ± 6.6) mmol/L,(35.9 ± 9.9) ml/h vs.(17.6 ± 8.4) ml/h,(84.2 ± 15.4) times/min vs.(146.4 ± 12.1) times/min,and the norepinephrine dosage was significantly reduced:(0.8 ± 0.2) μ g/ (kg· min) vs.(2.2 ± 0.8) μ g/ (kg· min).The differences had statistical significance (P < 0.05 or < 0.01).Conclusion Bedside pelvic external fixation can rapidly improve shock symptom in hemodynamically unstable patients with pelvic fracture,which is safe and efficient.
8.Sonographic characteristics of high-frequency ultrasound and elastography of thyroid microcarcinoma and the analysis of misdiagnosis

Objective To summarize the sonographic characteristics of high frequency ultrasound and elastography of thyroid microcarcinoma(TMC),and to analyze the causes of misdiagnosis.Methods The preoperative ultrasonic data of 245 suspicious TMCs in 202 patients,as confirmed by operation pathology,were retrospectively analyzed.Results Preoperative ultrasonography accurately diagnosed 221 TMCs,the diagnosis rate was 90.2%,and the misdiagnosis rate was 9.8%.Among the missed diagnosed lesions,18 lesions were nodular goiter,others were 3 nodular goiter with focal papillary hyperplasia of follicular epithelium,2 nodular goiter with adenomatous hyperplasia,1 focal lymphocytic thyroiditis,respectively.The thyroid lesions≤0.5 cm in diameter were more easily to misdiagnose.According to the importance of the ultrasonographic features of TMC,the order were aspect ratio (A/T) ≥ 1,irregular-shape,microcalcifications,low or very-low echo.Based on the above corresponding characteristics and considering other features together,the diagnostic accuracy rate were 94.1 %,93.9%,92.4% and 90.5%,respectively.Less blood supply and ill-defined boundary were the secondary sonographic signs of TMC.The elastographic scores of TMC were most showed 4 to 5 points.Diagnosis of TMC relied on elastography alone is less effective,but when elastograph diagnosis based on high frequency ultrasound,the diagnostic accuracy is much higher,especially when there is no calcification in the lesions.Conclusions High-frequency ultrasound has a very important value in the diagnosis of TMC,while elastography has certain assistant value on the basis of high-frequency ultrasonic diagnosis.
9.Relationship between albumin supply and prognosis in mechanically ventilated patients: a retrospective analysis.

OBJECTIVE:
To investigate the relationship between enteral nutrient albumin supply and prognosis in patients with mechanical ventilation.
METHODS:
The clinical data of 418 mechanically ventilated patients receiving enteral nutrition support treatment in intensive care unit (ICU) of the First Hospital of Jiaxing from January 2016 to June 2019 were retrospectively analyzed. According to whether the daily albumin supply was up to standard, the patients were divided into the standard group (albumin supply was ≥ 1.2 g×kg-1×d-1) and the non-standard group (albumin supply was < 1.2 g×kg-1×d-1). Prealbumin, transferrin levels before and after treatment, 28-day mortality, mechanical ventilation time and the length of ICU stay were compared between the two groups.
RESULTS:
A total of 418 patients were included, including 225 in the standard group and 193 in the non-standard group. There were no significant differences in gender, age and disease composition between the two groups, and the baseline data were comparable. There was no significant difference in daily calories between the standard group and the non-standard group (kJ/d: 119.73±31.55 vs. 110.05±28.98, P > 0.05), but the daily albumin supply of the standard group was significantly higher than that of the non-qualified group (g×kg-1×d-1: 1.38±0.83 vs. 0.95±0.75, P < 0.05). There was no significant difference in the levels of prealbumin, transferrin between the two groups before treatment. The levels of prealbumin, transferrin in standard group and non-standard group were significantly higher after treatment than before [prealbumin (mg/L): 188.53±69.25 vs. 119.44±57.62, 145.18±56.92 vs. 108.81±69.50; transferrin (g/L): 2.99±0.87 vs. 1.85±0.76, 2.09±0.81 vs. 1.52±0.76, all P < 0.05]. Moreover, prealbumin and transferring in the standard group were further improved than the non-standard group [prealbumin (mg/L): 188.53±69.25 vs. 145.18±56.92, transferrin (g/L): 2.99±0.87 vs. 2.09±0.81, both P < 0.05]. In addition, mechanical ventilation time, the length of ICU stay of the standard group were significantly shorter than those of the non-standard group (hours: 147.2±7.5 vs. 216.6±8.2, 198.8±9.5 vs. 295.4±8.9, both P < 0.05), but there was no statistically significant difference in 28-day mortality [11.56% (26/225) vs. 15.03% (29/193), P > 0.05].
CONCLUSIONS
Under the condition of standard enteral nutritional calories, increased of the albumin supply can improve the clinical nutritional status of patients with mechanical ventilation, shorten mechanical ventilation time and hospital stay.

10.Ultrasound and microbubbles: their functions in gene transfer in vitro.

To examine the role of ultrasound in gene delivery in vitro, three cells lines were exposed to the low-frequency ultrasound of varying intensities and for different durations to evaluate their effect on gene transfection and cell viability of the cells. Microbubble (MB), Optison (10%), was also used to observe the role of the microbubbles in gene transfection. The results demonstrated that as the ultrasound intensity and the exposure time increased, the gene transfer rate increased and the cell viability decreased, but at high energy intensities, the cell viability decreased dramatically, which caused the transfer rate to decrease. The most efficient ultrasound intensity for inducing gene transfer was 1 W/cm(2) with duration being 20 s. At the same energy intensity, higher ultrasound intensity could achieve maximal gene transfer rate earlier. Microbubbles could increase ultrasound-induced cell gene transfer rate by about 2 to 3 times mainly at lower energy intensities. Moreover, microbubbles could raise the maximum gene transfer rate mediated by ultrasound. It is concluded that the low-frequency ultrasound can induce cell gene transfer and the cell gene transfer rate and viability are correlated with not only the ultrasound energy intensity but also the ultrasound intensity, the higher ultrasound intensity achieves its maximal transfer rate more quickly and the ultrasound intensity that can induce optimal gene transfer is 1 W/cm(2) with duration being 20 s, and microbubbles can significantly increase the maximal gene transfer rate in vitro.
