1.Effects of VALD-3,a derivative of o-vanilla Schiff base ligand,on proliferation,migration and apoptosis of colorectal cancer cells and its mechanism

Objective To the effects of VALD-3,a derivative of o-vanilla Schiff base ligand,on proliferation,migration and apoptosis of colorectal cancer cells and evaluate its mechanism.Methods HT-29 and HCT116 cells were cultured in vitro,and the inhibitory effects of VALD-3(5,10,20 and 40 mg/L) on proliferation of the two kinds of cells were detected by MTT assay;The effects of VALD-3(10,20 and 40 mg/L) on the morphological changes of the cells were observed by inverted microscope,while the effects on the migration ability of HT-29 cells were detected by cell scratch test,and the effects on the apoptosis of HT-29 cells were detected by flow cytometry and Hoechst 33258 fluorescence staining;The effects of VALD-3(5,10,20 and 40 mg/L) on the expression of apoptosis-related proteins in HT-29 cells were detected by Western blot.Negative control groups were set up(with no VALD-3).Results Compared with the negative control group,the survival rates of HT-29 and HCT116 cells in 10,20 and 40 mg/L VALD-3 treated groups significantly decreased(t=7.717~2 006.148,each P <0.05);the number of HT-29 and HCT116 cells in 10,20 and 40 mg/L VALD-3groups decreased significantly with the increase of VALD-3 concentration,the cells appeared irregular morphology and gradually became round and smaller,and cell fragments increased;In 10,20 and 40 mg/L VALD-3 treated groups,the migration rate of HT-29 cell scratches decreased significantly(t=7.596~73.780,each P <0.01),the apoptosis rate increased significantly(t=7.092~8.057,each P <0.01),and the number of apoptotic cells increased significantly with strong bright blue fluorescence,chromatin concentration and nuclear fragmentation;The levels of cleaved caspase-3 and Bax protein in HT-29 cells treated with 5,10,20 and 40 mg/L VALD-3 significantly increased(t=2.998~24.901,each P <0.05),the level of Bcl-2 protein in 40 mg/L VALD-3 group decreased significantly(t=10.035,P <0.05),and the levels of cleaved caspase-8 in 20 and 40 mg/L VALD-3 group significantly increased(t=12.630 and 8.064 respectively,each P <0.01).Conclusion VALD-3 inhibited the proliferation and migration of colorectal cancer cells and induced apoptosis by regulating the expression of cleaved caspase-3,cleaved caspase-8,Bax and Bcl-2 proteins.
2.Clinical value of Antibody D5F3 and Survivin in patients with non- small cell lung cancer

Objective To investigate the clinical value of D5F3 and Survivin in patients with non-small cell lung cancer. Methods 100 patients with non - small cell lung cancer admitted in our hospital from January 2016 to June 2017 were examined by immunohistochemical method, to analyze the clinical value of D5F3 and Survivin antibody in non - small cell lung cancer (NSCLC). Results The coincidence rate of D5F3 was (20%), (++) was 33.33%, and the coincidence rate was (87.50%), and the difference was statistically significant (P<0.05). The positive rate of Survivin protein expression was correlated with the differentiation degree of non-small cell lung cancer, TNM stage and lymph node metastasis (P<0.05). Conclusion D5F3 has high accuracy, sensitivity and specificity, and has important clinical diagnostic value; The detection of Survivin expression is helpful for clinical differentiation, clinical staging and lymph node metastasis in patients with non-small cell lung cancer.
3.Oral azithromycin,oral and intravenous erythromycin treatment of respiratory tract mycoplasma infection curative effect and adverse reaction of contrast

Objective To summarize analysis of respiratory tract mycoplasma infection in patients with oral azithromycin,oral and intravenous erythromycin treatment the clinical effect and safety.Methods From April 2014 to September 2016 received during the period of 120 patients with respiratory tract mycoplasma infection as the research object,were randomly divided into oral azithromycin, oral erythromycin group and intravenous erythromycin group,all patients were retrospectively analyzed the clinical therapeutic effects and adverse reactions occur.Results The total effective rate in treatment,oral azithromycin is higher than oral and intravenous treatment of erythromycin,Signs and symptoms in the incidence of adverse reactions and improve the time aspect,oral azithromycin is lower than the oral and intravenous treatment of erythromycin,diff there is statistical significance(P<0.05).Conclusion For respiratory tract mycoplasma infection adopt oral azithromycin compared with oral and intravenous erythromycin treatment,the effect is better,shorter symptom improvement,higher security,worthy of clinical popularization and application.
4.The clinical effect of local application of domestic fluconazole in treatment of chronic obstruction pulmonary disease complicated with oral candidiasis

Objective To study the safety, efficiency and efficacy economics of local application of domestic fluconazole in treatment of chronic obstruction pulmonary disease (COPD) complicated with oral candidiasis. Methods Selected 74 patients with chronic obstruction pulmonary disease (COPD) complicated with oral candidiasis from May, 2013 to Oct. 2015 in our hospital were divided into the observation group in 35 cases and the control group in 39 cases according to table of random number. The observation group was given local application of domestic fluconazole, and the control group was given systemic application of imported fluconazole. The clinical effects and side effects of both groups were observed and compared. Results The total effect rate in observation group (100%) was higher than that in the control group (97.4%) without significant difference (P > 0.05). The cute rate in observation group (94.3%) was higher than that in control group (87.2.4%) without significant difference (P > 0.05). The average course of treatment was shorter in the observation group (2.9 d) than that in control group(5.2 d), with significant difference (P < 0.01). The observation group had no adverse reaction and the control group had 5.1% (P > 0.05). The cost of antifungal drugs in the observation group was lower than that in the control group, (t = -23.398, P = 0.000). The cost-effectiveness ratio in the observation group was lower than that in the control group. Conclusion Local application of domestic fluconazole has similar effect and side effect to systemic application of imported fluconazole in treatment of COPD complicated with oral candidiasis,but has short course of treatment and low cost .
5.Analyzing Complications of 73 Cases of Laparoscopic Gynecologic Surgery

Objective:The causes, management, and prevention of complications in laparoscopic gyneco-logic surgery were investigated. Methods:A retrospective analysis was done based on clinical data of 8279 cases of laparoscopic gynecologic surgery from January 1996 to December 2008 in this hospital. Results:The complication rate was 0.88% (73/8279). These included five cases intraperitoneal bleeding, five cases injury of ureter, 20 cases abdominal vessel damage, 10 cases vulva emphysema, 25 cases wound healing delay and each one case respectively for stomach injury, bladder injury, secondary abdominal cavity preg-nancy,infection and incision hernia. The occurrence rates of complications between hysteroscopy combined with laparoscopic surgery and pure adnexal surgery were no difference( P >0.05). Compared to adnexal op-erations, there was a significantly higher complication rate in myomectomy and laparoscopic assistant vaginal hysterectomy (P < 0.01 ). Conclusions:Complications of laparoscopic gynecologic surgery are correlated with the difficulty of operation and the skillfulness of operators. In order to reduce complications, careful oper-ation and avoiding over self -confidence are essential.
6.Clinical efficacy of naloxone plus aminophylline in the treatment of children with acute respiratory failure

Objective To observe the clinical efficacy of naloxone plus aminophylline in the treatment of children with acute respiratory failure.Methods 72 patients with acute respiratory failure were randomly divided into the study group and the control group.36 cases in the study group were given naloxone plus aminophylline therapy, 36 cases of the control group received conventional therapy.And the clinical efficacy was compared.Results The total effective rate of the study group was 91.67%,which was significantly higher than 66.67% of the control group (χ2 =6.82,P <0.05).In the comparison of the effect of improving blood indicators,arterial blood pressure of the study group was (67.51 ±4.11)mmHg,which was significantly higher than (61.03 ±4.08)mmHg in the control group (t =2.64,P <0.05).The oxygen saturation of the study group was (93.55 ±8.05)%,which was significantly higher than (79.62 ±10.22)% of the control group (t =2.29,P <0.05).The arterial carbon dioxide partial pres-sure of the study group was (69.03 ±5.71)mmHg,which was also higher than (61.52 ±4.09)mmHg of the control group (t =2.22,P <0.05).The incidence rate of adverse reactions of study group was 2.78%,which was lower than 22.22% in the control group (χ2 =6.22,P <0.05).Conclusion Naloxone plus aminophy -lline used in children with acute respiratory failure obtain the desired therapeutic effect,not only can effectively improve blood indicators of children,and without significant adverse reactions,drug safety is high,it is worthy of promoting.
7.Effect of Different Penetration Enhancers on Percutaneous Absorption of Xiaoyan Runma Mucilage

Objective To investigate the effects of different penetration enhancers on percutaneous absorption of Xiaoyan Runma mucilage in vitro, and to improve the curative efficacy of this mucilages through selection of the effective penetration enhancers. Methods Xiaoyan Runma mucilage was prepared with different penetration enhancers. An intelligent permeability instrument was used for in vitro percutaneous absorption test of rats , with isolated mice abdomen skin serving as in vitro transdermal barrier and saline isotonic solution as receptor fluid. Then the contents of lidocaine hydrochloride in receptors were determined by HPLC.The accumulative transit dose (Q) and percutaneous permeability (J) within 12 h were calculated and compared with those of mucilage without any enhancer. Results With Q value serving as an index, different enhancers had different promote permeation effects on Xiaoyan Runma mucilage, and the effects in descending order were as follows:4% azone [(222.75±3.4) μg?(cm2)-1]>2% azone[(207.42±5.1) μg?(cm2)-1]>3% menthol [(183.38±4.9) μg?(cm2)-1]>5%menthol [(160.82±5.4) μg?(cm2)-1]>2% azone+3% menthol [(151.25±5.5) μg?(cm2)-1]>2% azone+5% isopropyl myristate [(127.26±7.1) μg?(cm2)-1]>2% oleic acid [(125.16±6.5) μg?(cm2)-1]>no enhancer [(109.82±8.2)μg?(cm2)-1].4% azone was the best penetration enhancer for the mucilage delivery in vitro, with Q and J value as [(222.75± 3.4)μg?( cm2 )-1 ] and 19. 896 μg?( cm2 )-1?h-1 , respectively, which was 2. 08 times those of mucilages without any enhancer. Conclusion Being as a transdermal absorption enhancer of Xiaoyan Runma mucilage, 4% azone has the best effect. This study can provide the optimal formulation for transdermal delivery system of Xiaoyan Runma mucilage.
8.The value of early electronic bronchoscopy on patients with nonresponding pneumonia

Objective To investigate the effect and safety of early electric bronchoscope on patients with nonresponding pneumonia. Method Seventy non responding pneumonia patients were selected from our hospital , who received treatments from January 2015 to April 2016.According to the random table ,patients were randomly divided into the observation group(34 cases)and the control group(36 cases). According to the experience and sputum culture results,patients in the control group were given the conventional therapy,and patients in the observation group were given bronchoscope and transbronchil lung biopsy(TBLB). The effect of bronchoscopy,the treatment curative effect,duration of fever,the period of the elevated CRP dropped to baseline,the length of hospital stay and medical costs were compared between the two groups. Results The diagnostic rate of bronchoscopy and the positive rate of airway suction bacterial culture in the observation group were similar with those in the control group,with no significant differences. The positive rate of airway suction bacterial culture was significantly higher than that of sputum culture in the observation group. The positive rate of airway suction bacterial culture was higher than that of sputum culture in the control group ,but with no significant difference.The total effective rate in the observation group(87.1%)was significantly higher than that in the control group(58.8%). The duration of fever and the time of the elevated CRP dropped to baseline in the observation group were significantly shorter than those in the control group. The hospital stay and hospital costs in the observation group were also significantly lower than those in the control group. Conclusion Early bronchoscopy can obtain diagnosis evidence of etiology and histology and improve the diagnostic rate,meanwhile,it also has a therapeutic effect.It can improve the clinical treatment effect and shorten the healing time and the cost of hospitalization. In addition , early electronic bronchoscopy has fewer adverse reactions and it may play an important role in diagnosis and treatment on nonresponding pneumonia patients.
9.CT-guided percutaneous puncture and chemical mutilation of gasserian gangtion in the treatment of trigeminal neuralgia:comparison of the effects of ethyl alcohol and adriamycfal

Objective Percutaneous puncture of gasserian ganglion was guided by CT scan to improve the accuracy and reduce complications and to compare the efficacy of chemical mutilation with ethyl alcohol and adriamycin.Methods Ninety-two patients with primary trigeminal neuralgia (39 males, 53 females) aged 37-84 yrs were divided into two groups : A ethyl alcohol group ( n = 47) and B adriamycin group ( n = 45) . Secondary trigeminal neuralgia was excluded by preoperative CT scan. Percutaneous puncture of oval foramen was guided by CT and correct location of needle tip was confirmed by injection of contrast medium. 100% ethyl alcohol 0.5ml (group A) or 2% adriamycin 0.5ml was injected.Results The patients were followed up for 12 months after treatment. Neuralgia disappeared in 31 out of 47 patients in group A (alcohol group) and in 36 out of 45 patients in group B (adriomycin group) . Treatment failed in 13 patients in group A and 6 patients in group B, and was partly effective in the other patients. There were no severe side effects or complications. Conclusion Percutaneous puncture of gasserian ganglion guided by CT is effective and reduces complications. Adriamycin is more effective than ethyl alcohol for chemical mutilation.
10.Determination of Deltamethrin Residue in Water by Rephase High Performance Liquid Chromatogrophy

Objective An analytic method with rephase high pressure liquid chromatogrophy(RP-HPLC) was developed to determine the residue of deltamethrin in water.Methods Water sample was filtrated and determined by RP-HPLC.Chromatographic column was YMC C_(18)(250 mm?4.6 mm,5?m),mobile phase was methanol:water=90:10(V/V),the flowing rate was 0.8 ml/min, DAD detecting wavelength was 205 nm.Results The regression equation was y=655.6 x-1.8,r=0.999 99.The lowest detectable concentration was 2.9?g/L.The recovery rate was 98.0%-102.0%and the relative standard deviation was 0.29%-0.48%. Conclusion The method is proved to be satisfactory in precision,accuracy and sensitivity and can be used for the rapid determination of deltamethrin residue in water.
