1.Method of Extract Genomic DNA of Yeast for AFLP

The high molecular weight genomic DNA of yeast was extracted using three methods.Products were separated on agarose gel electrophoresis,quantified by spectrophotometer ND-1000 and restricted by EcoRⅠand MesⅠ.The result was shown that the genomic DNA extracted by modified benzyl chloride method was the best.The products of wild isolates supported it,too.This method was suitable for restriction of genomic DNA from yeast.
2.Protective effect of Danxuetong injection against testicular injury after testis torsion/detorsion in rats.

OBJECTIVETo investigate the protective effect of Danxuetong injection (DXT, a combination of Danshen and Xueshuantong injections) against testicular ischemia-reperfusion injury following testis torsion/detorsion in rats.
METHODSThirty-two 4-week-old healthy male SD rats were randomly divided into four groups of equal number: sham operation, normal saline, single DXT injection, and successive DXT injection. The rat models of testicular ischemia-reperfusion injury were established by 2-hour 720-degree torsion/detorsion of the unilateral testis. At 6 weeks after modeling, the rats were killed and their testes were harvested for measure- ment of testicular coefficients, sperm counts, sperm motility, and the levels of total anti-oxidative capacity (T-AOC) , superoxide dismutase (SOD) , nitric oxide synthase (NOS) , and malondialdehyde ( MDA) in the testis tissue.
RESULTSCompared with the rats of the normal saline group, those of the single DXT injection and successive DXT injection groups showed significant increases in the testicular coefficient (0.11 ± 0.03 vs 0.35 ± 0.04 and 0.40 ± 0.06, P < 0.05), sperm count ([0.46 ± 0.10] vs [1.44 ± 0.50] and [3.00 ± 1.28] x10(9)/ml, P < 0.05), sperm motility ([13.63 ± 14.04] vs [39.63 ± 5.04] and [76.31 ± 3.67]%, P < 0.05), the activity of SOD (72.76 ± 5.58 vs 116.25 ± 8.83 and 133.20 ± 13.84, P < 0.05), and the level of T-AOC (5.58 ± 1.07 vs 13.34 ± 5.81 and 19.21 ± 5.69, P < 0.05), but a remarkable decrease in the content of MDA (42.38 ± 8.94 vs 20.94 ± 5.65 and 15.02 ± 1.03, P < 0. 05) in the injured testes.
CONCLUSIONDXT can effectively rid the testis tissue of oxygen free radicals, improve sperm count and motility by antioxidation, and protect the testis tissue of prepubertal rats against testicular ischemia-reperfusion injury after testis torsion/detorsion. It also has a protective effect on the contralateral testis, and successive injection has a better effect than single injection of DXT.

3.Complete Genomic Sequence of a Chinese Isolate of Duck Hepatitis Virus

The complete genomic sequence of Duck hepatitis virus 1 (DHV-1) ZJ-V isolate was sequenced and determined to be 7 691 nucleotides (nt) in length with a 5'-terminal un-translated region (UTR) of 626 nt and a 3'-terminal UTR of 315 nt (not including the poly(A) tail). One large open reading frame (ORF) was found within the genome (nt 627 to 7 373) coding for a polypeptide of 2 249amino acids. Our data also showed that the poly (A) tail of DHV-1 has at least 22 A's. Sequence comparison revealed significant homology (from 91.9% to 95.7%) between the protein sequences of the virus in the Picornaviridae family, its genome showed some unique characteristics. DHV-1 contains 3copies of the 2A gene and only 1 copy of the 3B gene, and its 3'-NCR is longer than those of other picornaviruses. Phylogenetic analysis to do sequence homology based on the VP1 protein sequences showed that the ZJ-V isolate shares high sequence homology with the reported DHV-1 isolates (from 92.9% to 99.2%), indicating that DHV-1 is genetically stable.
4.Animal model of facial neuritis induced by herpes simplex virus.

OBJECTIVETo study the role of herpes simplex virus type 1 ( HSV-1 ) in facial paralysis by developing an experimental animal model of viral facial paralysis.
METHODSBoth sides of posterior auricular branch of facial nerve were anatomies and incised in 66 mice. The HSV-1 was inoculated into right ear branch and fetal bovine serum was inoculated into left ear branch as control. The symmetry of mouse face was observed and scored. The temporal bones were serially sectioned and stained with hematoxylin and eosin. The extratemporal facial nerves were stained with osmium tetroxide. HSV-1 DNA in bilateral facial nerve, brain stem, trigeminal ganglion and spinal cord was detected by the polymerase chain reaction.
RESULTSTwenty-eight (42. 42%) mice developed right facial paralysis between 2 and 5 days after inoculation. Continuing 3-6 days, the facial paralysis recovered spontaneously. Thirty-eight mice had no signs of facial paralysis. Compared with the left, nerve swelling, inflammatory cell infiltration were manifested in right temporal facial nerve of paralyzed mice. The ratio of the cross-sectional area of the facial nerve to the facial canal ( FN/FC ) was significantly higher than that on the control side (P < 0.01). Demyelinated nerve fibers were seen in the right extratemporal facial nerve. Not only in paralyzed mice, but also in non-paralyzed mice, HSV DNA was detected in some nerve tissues.
CONCLUSIONSInoculating HSV-1 into posterior auricular branch of facial nerve can produce an acute and transient facial paralysis in mice. The possible pathophysiologic mechanism of the facial paralysis is viral invasion and transportation from distal branch to main trunk. Then the viral facial neuritis causes facial paralysis.

5.Application of endobutton in the treatment of acute acromioclavicular joint dislocation.

OBJECTIVETo study the clinical efficacy of the endobutton in the treatment of acute acromioclavicular joint dislocation by reconstructing coracoclavicular ligaments.
METHODSFrom October 2008 to January 2010,12 patients with acute acromioclavicular joint dislocation were immobilized with the endobutton. All the patients had the dislocations of or above type III according to Rockwood classification. Among the patients, 9 patients were male and 3 patients were female, with an average age of 55 years (ranged from 31 to 83 years). Eight patients had injuries in the left, and 4 patients in the right. Four patients had accompanied injuries of rib fractures, 2 patients had brain injuries,and 1 patient had femoral fracture. Seven patients were injured by traffic accident, 4 patients were injured by falling down,and 1 patient was sports injuries. All the patients had pain and tenderness at the shoulder, positive piano sign, and shoulder confined activity. The duration from injury to operation ranged from 2 days to 10 days (averaged 6 days). The therapeutic effects were evaluated by Karlsson criteria based on range of motion of acromioclavicular joint, subjective feeling,and postoperative X-ray.
RESULTSAll the patients were followed up, and the duration ranged from 4 months to 19 months (averaged 11 months). The motion of the shoulder joint recovered to normal about 15 to 35 days after operation. There were no displacement, dislocation and redislocation occurred. All the patients got A degree results according to Karlsson criteria.
CONCLUSIONReconstruction of coracoclavicular ligament by using the endobutton to treat acute acromioclavicular dislocation of or above type III is a perfect method with advantage of rigid fixation, micro-injury, and early functional exercise.

6.1,8-cineol based influenza vaccine can improve vaccine efficacy on mice infected influenza virus

Objective:To evaluate 1,8-cineol as vaccine adjuvants to enhance influenza vaccine immune efficacy.Methods:80 BALB/c mice were divided into blank control group,1,8-cineol group,HA group and HA+ 1,8-cineol group,each group of 20.Blank control group normal breeding,HA group accepted HA protein 0.2 μg,the HA+1,8-cineol group was given 0.2 μg HA+ 100 mg/kg 1,8-cineol,1,8-cineol group was given 100 mg/kg 1,8-cineol.Except the blank group,other groups of mice intranasal immunized for three times interval of one week.At the fourth weeks each mice was infected with 10LD50 influenza virus FM1.On the 6 days after the infected,10 mice in each group were collected serum,using enzyme-linked immunoassay(ELISA)to detect serum IgG,IgG1b,IgG2a, IgG2b in 450 nm absorbance values(OD).Removed the lung tissue and measured the lung index and pathological.Remaining mice continue to observe 15 days and record the deaths,calculating the infected mice survival rate.Results:Compared with the HA group, HA + 1,8-cineol group can reduce the lung index,alleviate the pulmonary lesions,improve the serum IgG,IgG1b,IgG2a,and increases the survival rate of infected mice.Conclusion:1,8-cineol as vaccine adjuvants,can strengthen the immune effect of influenza vaccine.
7.Development and Psychometric Evaluation of the Career Growth Scale for Nurses

Purpose:
The purpose of this study was to develop the career growth scale for nurses (CGSN) and evaluate its psychometric properties.
Methods:
This study was conducted in four phases: (1) creating a pool of potential items through a qualitative design; (2) developing a preliminary scale using a modified two-round Delphi process; (3) refining the preliminary scale to finalize the scale using a cross-sectional survey; and (4) evaluating the psychometric properties of the final scale using another cross-sectional survey. A sample of 858 registered nurses from 12 general hospitals was recruited for this study.
Results:
The final scale consisted of 17 items categorized into three factors: career goal progress, professional ability and attribute improvement, and career promotion and prestige increase. The three factors accounted for 75.4% of the observed variance in career growth. The overall Cronbach's α was .96, and the intraclass correlation coefficient was .92. The content validity index was .97. Confirmatory factor analysis showed acceptable model fitness.
Conclusions
These results showed that the CGSN has good psychometric properties and can be used to evaluate specific career growth among nurses. This new instrument can further help nurse managers and clinical nurses themselves assess career growth and identify unsatisfactory aspects of growth, thereby designing tailored training programs and evaluating the effectiveness of such interventions.
8.Inhibition effects of Yiai capsules on tumor of xenografts in nude mice transplanied hunman lung cancer cell NCI-H460

Objective To investigate the effects of Yiai capsules on human lung cancer NCI-H460 cells of xenografts in nude mice.Methods Subcutaneous transplantation tumor model of xenografts in nude mice was established.The nude mouse model were randomly into 5 groups:model group,positive control group,and low,medium,high does experimental groups,8 nude mice in each group.The model group was given equal volume of saline with stomach perfusion.The positive control group was injected intraperitoneally(ip) with cyclophosphamide 20 mg · kg-1,three times a week.The mouse in low,medium,high does experimental groups were received Yiai capsules 0.9,1.8,3.6 g · kg-1,pid,respectively,stomach perfusion.Duration of drugs groups were given drugs for two weeks.At the end of the treatment,the nude mice were sacrificed and the tumor weight of nude mice were weighed and the volume of the transplanted tumor was measured.Morphological changes were observed by hematoxylin eosin (HE) method.The apoptosis rate in each group was detected by TdT-mediated dUTP nick-end labeling (TUNEL) method.Results The inhibitory rates in the model group,positive control group and experimental-L,experimental-M,experimental-H groups were 0,60.30%,18.00%,33.70%,36.30%,respectively.The tumor volume of nude mice in the5 groups were (2879.40 ±546.64),(1124.10 ±256.21),(2048.69 ±747.35),(2017.54 ±719.33),(1642.65 ±546.55)mm3,respectively.Comparison between drugs group and the model group,the difference on the tumor volume was significant (all P < 0.01).The tumor weight in the 5 groups were (2.67 ± 0.59),(1.06 ± 0.31),(2.19 ± 0.88),(1.77 ± 0.82),(1.70 ± 0.89) g,respectively.Comparison between experimental-M,experimental-H groups,positive control group and the model group,the difference on the tumor weight was significant (all P < 0.05).Under the optical microscope,the cells morphology of the model group had no obvious change.The cell morphology of experimental groups and control group changed significantly,and there were different degrees of cell necrosis.After administration,the apoptotic rate of tumor cells in the 5 groups were (5.40 ± 0.02)%,(29.00 ±0.01)%,(11.00 ± 0.03) %,(16.20 ± 0.01) %,(22.20 ± 0.02) %,respectively.Comparison between drugs groups and the model group,the difference on the apoptotic rate had significantly (all P < 0.01).Conclusion The Yiai capsule can inhibit NCI-H460 cells in nude mice transplanted tumor growth.
9.Effect of different methods cleaning equipment by machine washing on Ruhof lubricating oil pollution

Objective To observe the polluted Ruhof lubricating oil with two kinds of cleaning ml, only then tested them after the examination aseptic situation. The 400 recycling of surgical instruments were randomly divided into hand washing group and machine washing group (each group was 200 ), The recycling of surgical instruments were respectively soaked into the oil for 30s after hand washing and machine washing to oil.Adapting Ruhof lubricating oil made bacterial culture by setting people and time to check the sample, and compared the results. Results The hand washing group:percent of pass was 78.5 %, no bacterial growth was 23 times;The machine washing group: percent of pass was 50%, no bacterial growth was 9 times. There was significant difference between the two groups (P < 0. 01 ). Conclusions Hand washing is better than machine washing, it can extend the life cycle of Ruhof lubricating oil, save consumption and reduce costs.
10.Impact of gender on in-hospital death in hospitalized patients with acute myocardial infarction

Objective To investigate whether gender is an independent predictor of in-hospital death in hospitalized patients with acute myocardial infarction (AMI).Methods This retrospective study compared baseline characteristics, therapeutic approaches and the occurrence rate of angina pectoris, reinfarction, heart failure and death during hospitalization between 1501 male and 635 female hospitalized patients with AMI.Multivariate logistic regression analysis was performed to identify risk factors predicting in-hospital death.Results In-hospital death rate was significantly higher in female than male patients with AMI ( 11.7% vs.6.3% ,P <0.01 ).Female patients with AMI were significantly older than male patients [ (67.8±9.2)years vs.(61.1±11.9)years, P <0.01] had a higher incidence of hypertension (52.1% vs.41.1%, P <0.01), diabetes mellitus (35.4% vs.17.3%, P <0.01), cardiac function ≥ Killip class Ⅲ(11.7% vs.5.1%, P<0.01) and TC >4.68 mmol/L (71.3% vs.55.0%, P<0.01). Cigarette smoking, however, was more common in males than in females (69.4% vs.15.7%, P <0.01 ). Reperfusion therapy within the first 24 hours after symptom onset, β-blockers and statins use during hospitalization were significantly fewer in females compared with males (22.2% vs.31.5%, P < 0.01; 64.6% vs.71.2%, P=0.003; 43.1% vs.48.0%, P =0.041, respectively).An increased mortality was demonstrated in females during the hospitalization phase of AMI( 11.7% vs.6.3%, P <0.01 ).The results of logistic regression demonstrated that age, diabetes mellitus, hypertension, Killip classification of cardiac function, administration of reperfusion therapy and beta receptor blockers use were significant predictors of in-hospital death in patients with AMI, with odds ratios being 1.06 (95% CI: 1.04-1.08), 1.96(95% CI:1.32-2.90), 1.80(95% CI: 1.25-2.58), 2.86(95% CI:2.35-3.48), 0.44(95% CI:0.30-0.66) and 0.51 (95% CI: 0.36-0.74), respectively.conclusions The in-hospital mortality of females is significantly higher than that of males in this patient cohort.Older age, higher risk factor rates, less reperfusion therapy and β-blockers use contributed to the higher in-hospital mortality in female patients with AMI compared to males.
