1.Effect of Eliminating Pyrogens with Ultrafiltration on Quality of Chuanshentong Injection

Objective: To study the effect of eliminating pyrogen with ultrafiltration on the quatity of Chuanshentong Injection. Methods: The sample solution was filtered with the ultrafiltration membranes of molecular weight 6000 and 20000. Then,the results of eliminating pyrogens and the examination results of quality of sample solution before and after ultrafiltration were compared.Results: The results of elminating pyrogens is satisfactory. But there are larger changes in many items of quality markers after ultrafiltration. Conclusion: Therefore, the ultrafiltration should be used with caution in eliminating pyrogens of injection of Chinese medicinals.

Objective:To study inhibited tecomerase activity of HL-60 cells in human cancer cell by Icarrin(ICA) and its mechanism.Methods:MTT assay,NBT assay, TRAP-PCR,RT-PCR assay,Flow Cytometry.Results:ICA inhibited telomerase activity in HL-60 cells significantly.It was negative correlation between telomerase activity and the expression ratio of CD11b antigen in HL-60 cells.It can induced HL-60 cells to differentiate into mature granulocytes.It can changed cell cycle distribution of HL-60 cells,which was reflected by the accumulation of the vast majority of cells in the G0/G1phase and the loss of cells in the S phase. It can downregulated cell proliferation related gene c-myc;upregulated p21 gene protein and mRNA expression.Conclusion:This study proved mechanism of ICA on inhibited telomerase activity from gene-protein-effect of cell level in HL-60 cells.

5.Expression of MT-1 and MT-2 Genes in Liver of Chronic Exposure to Inorganic Arsenate Mice

Objective To investigate the effect of MT-1 and MT-2 in liver damage in chronic inorganic arsenate exposure mice and to explore the mechanism of arsenic-induced liver damage. Methods The male Kunming mice were randomly divided into control and exposed groups. The control group was given ordinary feed and tap water. The exposed group was given ordinary feed and 300 mg/L of sodium arsenite solution by drinking water. After 10 months of treatment, the activity of serum alanine aminotransferase (ALT), aspartate aminotransferase(AST) and globulin (Glb) content were determined. The total RNA was extracted by the TRIzol-phenol-chlorofor-method from the liver tissue. The quantity of the RNA was determined by spectrophotometry and its purity was judged at a ratio of A260/A280. Then real-time PCR(RT-PCR) was used to measure the mRNA expression of MT-1 and MT-2. 6.Effects of fluoride on the expression of c-fos in mouse fibroblast and osteoblast

Objective To observe the expression of c-fos mRNA and protein in fluoride treated mouse fibroblast (FB) and osteoblast (OB) and to further explore the effects of c-fos in the osteogenic action of FB. Methods Mouse FB and OB were divided into control group and six fluoride groups (0, 0.0001, 0.0010, 0.1000, 1.0000, 10.0000,20.0000 mg/L F-), and the levels of c-fos protein at 2,4,24,48,72 h and c-fos mRNA at 48 h were measured by using ELISA and RT-PCR methods. Results Compared with the control group, fluoride increased the content of c-fos protein obviously in all FB group(P<0.01); and it is increased in 0.0001,0.0010 mg/L groups at 48 h (0.73±0.04, 0.64±0.14) and 0.0001 mg/L group at 72 h(0.70±0.17) in OB compared with the control group (0.32±0.04,0.27±0.05, P<0.01). Compared with the control group (0.95±0.11), RT-PCR revealed an increasing tendency of the expression of c-fos mRNA at 48 h in FB (1.06±0.16, 1.06±0.12,1.12±0.16,1.04±0.15,1.04±0.10,1.15±0.29), but the difference was not statistically significant(P>0.05); however, a statistically significant difference(P<0.01) of c-fos mRNA in 20.0000 mg/L group(1.40±0.17) in O B was found compared with the control group (1.06±0.06). Conclusion The higher expression of c-fos mRNA and protein in FB induced by fluoride may play an important role in the transformation of osteoblastic phenotype as well as increase the osteogenesis ability in FB.

8.Release of hepatocyte growth factor mediated by heparin

Objective: To observe the effects of sodium heparin and low molecular weight heparin on the release of plasma hepatocyte growth factor (HGF) in senior coronary heart disease patients.Methods: Fifty-four senior patients with coronary heart disease were divided into three groups: intravenous sodium heparin, subcutaneous sodium heparin, and subcutaneous low molecular weight heparin (LMWH). Plasma HGF and vascular endothelial growth factor (VEGF) were measured before and after injection.Results: Plasma HGF was increased rapidly and significantly after intravenous injection of sodium heparin, reaching its peak level (about 48 fold) after approximately 10 minutes. Plasma HGF was also increased rapidly and significantly after subcutaneous injection of sodium heparin and LMWH, reaching its peak level (about 4 and 5 fold in sodium heparin and LMWH respectively) after approximately 2-3 hours. Conclusion: The rise of plasma HGF after heparin treatment suggests that heparin has some other biological effects in addition to its anticoagulant property through HGF. By this mechanism, the administration of heparin may be of some importance in the reparation of cardio-vascular diseases.
9.Improved technique for preparation of deacetylated chitin

BACKGROUND: The application of deacetylated chitin (chitosan, DC) prepared by traditional technique was limited because of its lower degree of deacetylation, impurity and relative lower molecular mass.OBJECTIVE: To improve the traditional technique for extraction and preparation of DC from the rustae f oratosquila oratoria.DESIGN: Contrasting observation.SETTING: Shandong Institute of Biopharmaceutics.MATERIALS: The experiment was carried out in the Shandong Key Laboratory for iopharmaceutics, Shandong Institute of Biopharmaceutics from February 2003 to January 2004. Experimental nstrument: Mettler AE240 electronic analytical balance and ZK-40AB electrothermal vacuum drying oven were rovided by Mettler-Toledo Instruments Co., Ltd. And ShangHai Shuli Instruments Co.,Ltd., respectively. Xperimental materials: oratosquila oratoria was purchased from Jinan aquatic products market.METHODS: ①C as traditionally prepared from chitin by decoloring with strong oxidant and deacetylating with concentrated alkaline liquor under high temperature. Chitin was extracted from crustae of shrimp and crab fter decalcification with diluted acid and deproteinization with diluted base. ②The improved preparation echnique: The crustae of oratosquila oratoria was decalcificated and decolorized with diluted acid, and as deproteinized and decolorized with diluted base at room temperature, and then the chitin was obtained fter the second decalcification and decolorization with diluted acid. The crude DC was obtained from hitin after deacetylation and decolorization with concentrated ase at 55-65 ℃. The crude DC was issolved n diluted acid and filtered. The filtrate was collected, precipitated with dilute base. After filtering, he sediments were retained, washed with water and dried. Then pure DC was obtained.MAIN OUTCOME MEASURES: he comparison between improved technique and traditional technique; Color, dissolvability in diluted cid, ield, degree of deacetylation and dynamic viscosity of DC prepared by different techniques.RESULTS: ①echnique comparison: Compared with traditional technique, the reaction of improved preparation technique as milder, and the deproteinization was at room temperature; it did not need an isolated decolorization;deacetylation was at low temperature, and had a refine process. ②Detection indexes of DC: he yield of DC prepared by improved technique was little lower than that by traditional technique (15%, 7% respectively). The degree of deacetylation and dynamic viscosity were both higher than those by raditional technique (> 95%,< 70%; > 120mPa·s,< 80 mPa·s , respectively). The color was white or lmost white, which was better than that by traditional technique (yellow or gray). The DC could ompletely issolve in dilute acid, and the dissolvability was better than that by traditional technique (dissolved artly in dilute acid).CONCLUSION: Compared with the traditional technique, the improved technique was mpler and the reaction conditions were milder. And also the quality of obtained DC was obviously improved.
10.Analysis of monitoring results of coal-burning borne endemic fluorosis in Fuyuan county Qujing city in 2009

ObjectiveTo explore the prevalence state of children in coal-burning borne endemic fluorosis in Fuyuan county Qujing city,to know the implementation of preventing measures,and to provide a scientific basis for prevention of endemic fluorosis.MethodsIn 2009,105 families were sampled to check the service condition of defluoridation stove and the effect of health education (the awareness of use defluoridation stove reasons) in Xiaoyuwang,Tuoga and Shaliang villages,Fucun town of Fuyuan county.Food fluorine of indoor stored peppers and corns was monitored of 20 families in Xiaoyuwang village,5 household were randomly selected from east,west,south,north and middle places of the village.Air fluorine was monitored from indoor air samples of 5families,dental and urinary fluorine was monitored of the native-born children aged 8 to 12,and dental fluorosis of these children was checked.Clinical endemic fluorosis and X-ray endemic fluorosis was monitored of local adult residents over the age of 30 that had been lived in the area for more than 5 years.ResultsThe rate of correct use of the defluoridation stove was 100%,and adult awareness rate of use defluoridation stove reasons was 82.86%(87/105).Of the 19 pepper samples,fluorine content were 9.98 - 538.40 mg/kg and median was 32.38 mg/kg,all exceeded the standard(1.5 mg/kg).Of the 25 corn samples,fluorine content were 0.42 - 101.54 mg/kg and median was 6.32 mg/kg,with 18 samples exceeded the standard(1.0 mg/kg).Of the 6 air samples,fluorine content were 0.000 98 - 0.003 33 mg/kg and median was 0.001 45 mg/kg.Detection rate of dental fluorosis of children aged 8 - 12 was 42.3%,urinary fluorine content were 0.13 - 1.80 mg/L,geometric mean was 0.58 mg/L,4 adults were diagnosed clinical skeletal fluorosis,detection rate was 2.46%(4/162),5 adults were found by X-ray with endemic fluorosis,detection rate was 9.61%(5/52).ConclusionPeppers and cons fluorine pollution is serious,general public should be guided to change their lifestyles to minimize the fluorine pollution.
