1.Study on the activation effects to T lymphocytes by gastric cancer cell-dendritic cell fusion vaccines

Objective To study the activation effects to cytotoxic T lymphocytes by gastric cancer cell-dendritic cell(DC) fusion vaccines and to provide theoretical data for biological therapy of gastric cancer based on(anti-tumor) fusion vaccines.Methods(1)Peripheral blood mononuclear cells from gastric cancer patients were separated and co-cultured with granulocyte macrophage colony stimulating factors,interleukin-4 and tumor necrosis factor-? to generate mature dendritic cells.(2)The dendritic cells and SGC7901 cells were fusioned by use of polyethylene glycol,and the pure fusion cells were screened out and cultured with HAT and HT(selective) culture systems.(3)The ability of fusion cells to activate the cytotoxic T lymphocytes was(investigated) by using MLA method.Results(1)Mature dendritic cells were gained from gastric cancer(patients)′ peripheral blood mononuclear cells by co-culturing with granulocyte-macrophage colony stimulating factors,interleukin-4 and tumor necrosis factor-?.(2)Dendritic cells and SGC7901 cells could be fusioned by PEG and could get pure fusion cells by culture with HAT/HT selective culture systems.(3)The fusion vaccines could strongly activate cytotoxic T lymphocytes,and there were significent differences between the(fusion) vaccine group and the control group.(P
2.Study on the induction culture and phenotypes of dendritic cells originating from peripheral blood of gastric cancer patients

Objective To analyse the morphological and phenotypic characteristics of dendritic cells obtained by induction of peripheral blood monocytes of gastric cancer patients and provide an experimental basis of (dendritic) cells for the biological treatment of gastric cancer. Methods Peripheral blood monocytes of gastric cancer patients were co-cultured with GM-CSF, IL-4 and TNF-? to obtain the dendritic cells. The (morphological) characteristics of the dendritic cells were observed by light and electron microscopes and the (phenotypes) were studied by flow cytometry. Results Mature dendritic cells were oblained seven days after (induction) culture. These cells showed numerous typical dendritic protuberances on their surface.The (phenotypes) of the cells increased gradually with their maturation and reached stable levels seven days later. Conclusions Mature dendritic cells could be obtained from peripheral blood monocytes of gastric cancer (patients) after induction culture with GM-CSF,IL-4 and TNF-?. The obtained cells had typical morphological and phenotypic characteristics. These cells could be used for further research in the biological treatment of (gastric) cancer.
3.A study on the biological characters of SGC7901 gastric cancer cell-dendritic cell fusion vaccine

Objective To provide experimental basis for the study of SGC7901 gastric cancer cell-dendritic cell fusion vaccine. Methods The fusion of SGC7901 gastric cancer cells and dendritic cells was induced by PEG. We obtained pure fusion cells by selective culture with HAT/HT culture systems. The growth curve characters, clone abilities, abilities to irritate T lymphocytes and the ability to grow were detected. Results The fusion cells had weaker proliferative abilities and clone abilities with increased abilities to irritate T lymphocytes. The CPM value of T lymphocytes were 15083?231、 9608?83、 4214?135、 3020?28 respectively when they were co-cultured with the fusion cells on different proportions. Conclusion SGC7901 gastric cancer cell-dendritic cell fusion cells keep strong abilities to irritate T lymphocytes while inable to grow into new planted tumors in immunodeficiency animals.
4.Effect of EGCG-Zn on the Learning,Memory and Antioxidant Abilities in Vascular Dementia Rats

OBJECTIVE:To study the effect of EGCG-Zn chelated by epigallocatechin gallate (EGCG) and Zn ion on learn-ing,memory and antioxidant abilities in vascular dementia(VD)rats. METHODS:Rats were randomly divided into sham group, model group,zinc gluconate group (positive control,70 mg/kg),EGCG group (5 mg/kg),and EGCG-Zn low-dose,mid-dle-dose,high-dose groups (2,5,10 mg/kg),10 in each group. The VD models were induced by middle cerebral artery occlu-sion. The rats were intragastrically given relevant medicines after 7 d of modeling,once a day,for 28 d. Morris water maze were used to evaluate learning and memory abilities of rats. Spectrophotometric method was adopted to detect the superoxide dismutase (SOD),catalase(CAT),glutathione peroxidase(GSH-Px)activities and malondialdehyde(MDA)content in brain tissue of rats. RESULTS:Compared with sham group,escape latency was extended in model group,percentage of platform quadrant swimming distance(dP/dT)was reduced(P<0.01);SOD,CAT,GSH-Px activities in brain tissue were reduced,and MDA content was in-creased (P<0.01). Compared with model group,escape latency on 4th d of recording was obviously shortened in EGCG group and EGCG-Zn dose groups,dP/dT was obviously increased and dose-depended(P<0.05 or P<0.01);SOD,CAT,GSH-Px activi-ties in brain tissue were increased,MDA content was decreased(P<0.05 or P<0.01)and dose-depended. Compared with EGCG group and zinc gluconate group,escape latency was obviously shortened in EGCG-Zn dose groups,dP/dT was increased;SOD, CAT,GSH-Px activities in brain tissue were increased,MDA content was decreased;and the effects in high-dose,medium-dose groups were more obvious(P<0.05 or P<0.01). CONCLUSIONS:EGCG-Zn can improve the learning,memory and antioxidant abilities in VD rats,and the effect is superior to EGCG.

ObjectiveTo study the effect of α7 ( α7 AChR) agonist nicotine on regulating sensitivity of regular chemotherapeutic agent in cholangiocarcinoma cells,and explore the possible target.MethodsThe effect of nicotine and α-BTX pretreatment on the survival ability of cholangiocarcinoma cells was investigated when applied with 5-FU by using MTT and Flat cloning formation experiment.ResultsApplied with 5-FU,in various con centrations nicotine stimulating group( 10-3 g/L,10-4 g/L,10-5 g/L ),the survive rate of QBC939 was 128%,124%,118%,while that in α-BTX stimulating group and combined stimulation group was 92%,94%,93%,92%,respectively.The cloning formation ability of nicotine- stimulating group (6.2 ± 0.40) was significantly higher than α- BTX stimulating group (3.2 ± 0.20 ),combined stimulation group ( 3.2 ± 0.20 ) and control group ( 3.4 ±0.33).ConclusionNicotine can prevent chemotherapy-induced apoptosis,and improve cholangiocarcinoma cell survival via α7 nicotine acetylcholine receptor in vitro.
6.Pathological studies on lachrymal dilated drainage tube implantation in rabbits

AIM:To observe the pathological changes of the lower segment of nasolacrimal duct mucosa in rabbits at different stages after retrograde lachrymal dilated drainage tube implantation.
METHODS:Totally 14 New Zealand rabbits were used in the present study. One side of nasolacrimal duct was obstructed to produce an experimental model and operated the reverse implantation of nasolacrimal duct intubation. Histological changes of the lower segment of nasolacrimal duct mucosa were observed by routine light microscope at 2, 4, 6, 8, 10, 12 and 14wk after the operation.
RESULTS: Compared with the control side, the group of 2 and 4wk after surgery presented the inflammatory cytokine. The group of 12wk after the operation presented isolated granuloma. Group 12 and 14wk presented scattered granuloma. The size of the granulomas was smaller and the density of epithelioid cell and fibroblast were lower in group 12wk than those in group 14wk by HE and Masson trichrome stain.
CONCLUSION:Recurrent Silicone Tube is used to treat nasolacrymal duct obstruction. Nasolacrimal duct can be narrowed and blocked again by granuloma, progressive fibrosis and adhesion of surrounding tissues when tube is in the duct more than 12wk.
7.Minimally invasive percutaneous osteosynthesis for the treatment of 53 patients with complex ankle fractures.

OBJECTIVETo explore the surgical method and its clinical effects of minimally invasive osteosynthesis on the treatment of complex ankle fractures.
METHODSFrom January 2007 to December 2011, 53 patients with complex ankle fractures were treated with minimally invasive osteosynthesis. There were 31 males and 22 females, with an average age of 38.2 years old (ranged, 18 to 65). According to the system of Lauge-Hansen, 32 fractures were supination external rotation injury (grade WV), 13 fractures were pronation external rotation (grade III or IV), 5 fractures were pronation abduction (grade III); and 3 fractures can not be classified due to serious comminution fracture of fibula. According to the system of Denis-Weber, there were 4 cases with type A, 34 cases with type B and 15 cases with type C. Seven cases were open fractures. The duration from injuries to operation ranged from 2 hours to 14 days with an average of 5 days. The sequence of reduction and fixation of ankle fractures was firstly posterior malleolus, then medial malleolus and lateral malleolus, and inferior tibiofibular syndesmosis lastly. The fractures of posterior malleolus were reduced and fixed through anterior ankle approaches; the fractures of medial and lateral malleolus were percutaneously fixed with bolts or blade plate or tensile force band; and inferior tibiofibular syndesmosis were firmly fixed if necessary. Baird-Jackson scoring system was used to evaluate clinical effects.
RESULTSForty-eight patients were followed up from 10 to 36 months with an average of 13 months. The fractures got healing with an average time of 12 weeks (ranged, 10 to 18). According to the Baird-Jackson scoring system, the mean score of ankle function was 94.7 +/- 4.2, and 28 cases obtained excellent results, 15 good, 3 fair and 2 poor. One case experienced superficial infections and was cured by changing dressings, 2 cases experienced fixed syndesmosis screw breakage.
CONCLUSIONThe surgical method of minimally invasive osteosynthesis can ensure the anatomical join restoration, protect the blood supply of fracture end, rebuild the function of ankle joint, obtain satisfactory clinical results in treating complex ankle fractures.

In order to standardize the management of fixed assets in basic medical research, and to solve the problem ofone equipment with more than one code , we discussed the fixed assets coding of instrument and equipment in this paper.The existing equipment classification of the Institute of Basic Medical Sciences of the Chinese Academy of Medical Sciences was analyzed.Depending on application of the experiment and the principle of equipment, the novel fixed assets encoding dictionary of instrument and equipment is generated, which fits in the application and development of basic medical research.The managers may find the corresponding code quickly with the standardized equipment name.The new encoding dictionary may facilitate the classification of basic medical experimental instruments, prevent multiple coding situations of equipment and improve the management.

10.Effects of horse-riding squat exercise plus Governor Vessel-regulating Tuina therapy on static balance function in patients with stroke

Objective:To observe the effect of horse-riding squat exercise plus Governor Vessel-regulating Tuina(Chinese therapeutic massage)therapy on static balance function in patients with stroke.Methods:A total of 176 stroke patients were enrolled as the study subjects and divided into four groups by the random number table method,namely horse-riding squat+Tuina group,horse-riding squat group,Tuina group,and control group,with 44 cases in each group.The control group was treated with rehabilitation balance training.The horse-riding squat+Tuina group,horse-riding squat group,and Tuina group were treated with additional horse-riding squat exercise plus Tuina,horse-riding squat exercise,and Tuina treatment,respectively.Four weeks of treatment was regarded as one treatment course.After 1 treatment course,the balance function of the four groups was compared.Results:After treatment,the ellipse area of motion and the length of motion in all four groups were reduced,and the intra-group differences were all statistically significant(P<0.05).The difference in the ellipse area of motion before and after treatment in the horse-riding squat+Tuina group and the horse-riding squat group was larger than that in the control group,and the differences were statistically significant(P<0.05).The difference in the ellipse area of motion before and after treatment in the horse-riding squat+Tuina group was larger than that in the Tuina group,and the difference was statistically significant(P<0.05).The difference in the length of motion before and after treatment in the horse-riding squat+Tuina group,the horse-riding squat group,and the Tuina group was greater than that in the control group,and the differences were statistically significant(P<0.05).There was no statistically significant difference in the pairwise comparison between the horse-riding squat+Tuina group,the horse-riding squat group,and the Tuina group(P>0.05).Conclusion:Horse-riding squat exercise plus Governor Vessel-regulating Tuina therapy can effectively improve the static balance function in patients with stroke.Training the affected lower limb weight-bearing exercise,adjusting the center of gravity distribution,and promoting the mutual balance and coordination between muscle groups may be the mechanism of improving the static balance function.
