1.Construction and primary characterization of cDNA expression library of Humulus pollen

Objective To construct a cDNA expression library of Humulus pollen and provide the basis for screening the major allergenic components and producing recombinant allergen of Humulus pollen.Methods Verdure Humulus pollen were collected and preserved in liquid nitrogen after being sift out.Total RNA was extracted from the Humulus pollen with trizol reagent,and cDNA was synthesized by RT-PCR with purified total RNA.Then the cDNA was digested by SfiⅠ and the fragments smaller than 400bp were removed by chroma spin-400 column,and the fragments longer than 400bp were ligated with ?TripIEx2 Vector.The library was packaged in vitro and a small portion of packaged phage was used to infect E.coli XL1-Blue MRF′ for titration.The diversity of the library and the length of the inserted fragments were analyzed by PCR.Results The cDNA expression library contained 5?10~(5) recombinants and the percentage of recombination was 90%.The average length of inserted cDNA fragments was about 1.02kb.Conclusion The constructed cDNA expression library contains appropriate contents and sizes of cDNA fragments and is qualified for screening target cDNA clone.
2.The clinical significance of central electric fetal monitoringfor ante-partum observation

Objective:To study the clinical significance of central electric-fetal monitoring system for ante-partum observation and measurement.Methods:The experimental group (253 pregnant women who delivered in our hospital from January,2001 to August,2003) was monitored with the central electric fetal monitoring system.The control group (250 pregnant women who delivered the children in our hospital from July,1998 to September,2000) was not monitered with this system.Then,the results of fetal monitoring and the outcome of prenatal fetuses of the two groups were compared and analyzed.Results:The rate of abnormal FHR was markedly higher in experiment group(30.8%) than in the control group(17.2%),and the rate of variable deceleration in abnormal graph was markedly higher in the experimental group (28.8%) than in the control group (12.4%).There was a significant difference is significant between the two groups.The incidence of asphyxia among newborns was lower in the experimental group (4%) than in the control group (8.4%),and the difference is significant between the two groups.However,there was no significant difference in the rate of cesarean delivery and Simpson forceps delivery between the two groups.Conclusion:Ante-partum monitoring assisted by central electric-fetal monitoring system can improve the prenatal prognosis and increase the rate of cesarean delivery.
3.Investigation and analysis of sexual concept and sex status of pregnant women

Objective:To investigate the sexual concept and sex status of pregnant women so as to instruct pregnant health care and improve living quality.Methods:112 random sample candidates who have mature fetus for delivery in hospital were chosen to investigate and analyze by means of self-edited questionnaires and Eysenck protracted sexual latitude scale test.Results:Both sex frequency and sex continuing time of pregnant women reduced remarkably;their knowledge on sex related to pregnancy was generally poor;average value of Eysenck protracted sexual latitude scale test was 5.25,lying below the average points;professionals and individual professionals have statistics differences in occupation team;the single-mode of sexual intercourse in the sex of pregnant women has the highest percentage.Conclusion:Pregnant women lack the related health care knowledge on sex,and their concept of sex is comparatively conservative,so they can not enjoy full sex and need further improvement.
4.Exploration of the mode for measurement and evaluation of nurses'occupational diathesis in employment systemed

Objective:Under the background that the army hospitals need to employ civilian nurses in a large quantity,we try to grope a scientific,objective method to choose the nurse who matches post.Methods:Referring to related data and the ritual of talented person evaluation,we chose the evaluation methods including paper test,computer test,personal interview and data integration;arranged the steps of planning,assigning the weight,performing evaluation,integrating the test results,and tracking for feedback;in order to form the mode to test the employed nurses' occupational diathesis.Results:We fulfilled a series of scales for mood and hobby,weight score,quality structure,evaluation method comparison and nurse's professional quality evaluation,etc,and determined the evaluation methods and procedures.Conclusion:Evaluating more than 100 nurses who are newly employed using this new mode,the result demonstrates that this evaluation mode can be easily operated,and is a guide in employing quantities of professional nurses for a hospital.
5.Effect of dietary and physical activity interventions on reducing excessive weight gain during pregnancy, cesarean section and macrosomia :A Meta-analysis

Objective To systematically review the effect of dietary and physical activity interventions on weight management during pregnancy.Methods The PubMed (from Jan.1990 to Sep.2013),Springer (from Jan.1960 to Sep.2013),Willy (from Jan.1990 to Sep.2013),Web of Science (from Jan.1990 to Sep.2013),Wanfang data (from Jan.1995 to Sep.2013),China National Knowledge Infrastructure (CNKI) (from Jan.1995 to Sep.2013)were searched for relevant articles.All published randomized controlled trails (RCT) on dietary and physical activity interventions during pregnancy were considered.The quality of RCTs was appraised and data were extracted by 3 reviewers independently.Meta-analysis were conducted by RevMan 5.2 software.Results In total,eleven RCTs were included in this review.Meta-analysis revealed that dietary and physical activity intervention significantly reduced incidence of cesarean section and macrosomia;However,there were no statistically significant differences in the total gestational weight gain.Conclusions Interventions based on dietary and physical activity during pregnancy appears effective in reducing incidence of cesarean section and macrosomia.However,there are many limitation of the Meta analysis,therefore more multi-center,large sample with high quality RCTs are required for future verification.
6.Expression of nuclear factor kappa B and inducible nitric oxide synthase in the respiratory syncytial virus infection on human lung epithelial cells

AIM: After human lung epithelial cells (A549) infected with respiratory syncytial virus (RSV), inducer cells over-express active transmitter. The study investigated the expression changes in nuclear factor ?B (NF-?B), inducible nitric oxide synthase (iNOS) mRNA and protein, nitric oxide (NO) and malonaldehyde (MDA) on human lung epithelial cells (A549) infected with RSV. METHODS: Experiments were performed at the Laboratory of Etiology and Immunology, Anhui Medical University between December 2006 and May 2007. ① Gyopreserved virus broliferated in Hep-2 cells after recovery. A549 cells infected with RSV in vitro were used to collect cells and cellular culture supernatant at hours 4, 8, 16 and 24. Cells non-infected with RSV were served as controls. ② Reverse transcription-polymerase chain reaction (RT-PCR) was used to evaluate the expression of iNOS mRNA. The expression of iNOS protein and NF-?B was detected by Western-Blot. The concentrations of NO and MDA were measured by nitrate reductase method and thiobarbituric acid method. RESULTS: ①The basal expression of NF-?B was increased with the prolongation of RSV infection to A549 cells. ②RSV infection to A549 increased the amounts of mRNA and protein of iNOS in a time-dependent manner. The expression of iNOS mRNA was about 12 times as many as basal expression. RSV infection promoted the expression of iNOS protein, which was higher than the control group. The activities of NF-?B significantly were positively correlated with the mRNA and protein expression of iNOS. ③RSV could improve A549 cells secreting NO and MDA, but the NO levels rose slowly during the 24 hours of infection. CONCLUSION: The inflammatory response of RSV can increase the activity of NF-?B and it is positive correlated with the increase in iNOS mRNA and protein levels.

8.Laparoscopic Sphincter Preserving Surgery for Ultra Low Rectal Cancer:Report of 108 Cases

Objective To explore advantages of laparoscopic sphincter preserving surgery for ultra low rectal cancer . Methods From April 2006 to January 2015, we performed laparoscopic sphincter preserving surgery in 108 cases of ultra-low rectal cancer.After laparoscopic mesorectal resection and lymph node dissection were completed , transection of the rectum was performed with the Endo-GIA at 2 cm from the lower margin of the tumor .The right lower abdomen main operation port was expanded to remove the lesion and introduce the stapler base .The proximal colon was returned and the pneumoperitoneum was re-established .A colorectal end-to-end anastomosis was conducted through the anus by using a carliber 29 circular stapler . Results Laparoscopic operation was successfully accomplished in all the 108 cases.No conversion to open surgery was required .No operative mortality was encountered . The operation time was 68 -145 min ( mean, 104 min ) , the intraoperative blood loss was 10 -100 ml ( mean, 40 ml ) , the postoperative flatus time was 1-3.5 d (mean, 2.3 d), and the number of removed lymph nodes was 8-37 (mean, 12.5).There were 98 patients followed up for 6-62 months (mean, 24.6 months).No tumor metastasis or recurrence was found . Conclusion Laparoscopic total mesorectal excision of ultra low sphincter preserving operation is feasible .

MicroRNAs (miRNAs) are a class of small endogenous RNAs, which handle post-transcriptional gene regulation activity through targeting messenger RNAs.As the deep understanding of them, miRNAs had been found to participate in numerous basic biological processes, including the normal growth process of tissues and organs ,as well as the pathologic course of diseases.Different miRNAs may be involved in different tissues and organs, the same miRNA may have different expression in various physiological process or stages of pathological conditions.Now, the recent studies about the miRNAs in the lung development and pulmonary diseases were reviewed.
10.Association study between clinical pharmacokinetic of paclitaxel and chinical effect in patients with gynecological malignant tumor

Objective To observe the association between clinical pharmacokinetic of paclitaxel and adverse reactions in patients with gynecological malignant tumor.Methods 78 patients with female malignant tumor selected from Jan.2009 to Dec.2013 in Kailuan General Hospital were treated with paclitaxel (intravenous drip 175 mg/m2 3 h).The blood drug level of paclitaxel was measured at different time after treating.The DAS software was used to calculate the parameter of pharmacokinetic of paclitaxel,and the SPSS software was used to analyze the association between clinical pharmacokinetic of paclitaxel and adverse reaction in patients with female malignant tumor.Results The peak plasma concentrations (Cmax)was (3.99 ± 1.07)mg/L,the areas under the plasma concentration-time curve (AUC)was (9.74 ±2.45)mg/(h·L),the plasma elimination half-life (t1/2z)was (13.7 ±3.2)h,the apparent volumes of distribution (Vd)was (162.9 ±49.5)L/m2,the plasma clearance rates (CL)was (8.2 ±1.0)L/(h·kg).The kinetics of paclitaxel was fitted to two-compartment model.The time of drug concentration >0.1 μmol/L (t>0.1 )in responder patients (16.8 ± 4.8)h was higher than that in nonresponder patients (13.5 ±2.9)h,and the difference was significant (P<0.05).The Cmax in patients with severity adverse reactions (4.61 ±0.86)mg/L were higher than those in patients without severity adverse reaction (3.65 ±1.13)mg/L,and the difference was significant (P<0.01).Conclusion The Cmax of paclitaxel is associated with efficacy after treatment and severity adverse reactions.
