1.Healing effect analysis of rhegmatogenous retinal detachment in 288 myopic cases

AlM: To understand the prognostic factors of myopic rhegmatogenous retinal detachment ( RRD) surgery, and provide guidance for clinical treatment.
METHODS: This retrospective study included 288 myopic RRD eyes ( excluding trauma, aphakia, congenital diseases, tractional and exudative retinal detachment ) from April 2007 to April 2014 in our department. The surgical anatomic reduction situation was statistically recorded, the effects of hole's size, retinal detachment duration on sight restoration and surgery success rate were analyzed
RESULTS:The statistical results showed that 245 eyes (85. 1%) healed after their first operation, about 95. 1%cases finally healed after their second or third operation. The degree of myopia and aging factor influenced the RRD operation:the higher degree and the older age were, the lower success rates were (P<0. 05). Through surgery, postoperative visual acuity was significantly better than the preoperative ones ( P<0. 01 ). A higher degree of myopia RRD led to poor visual acuity ( P < 0. 01 ). Postoperative visual acuity of 152 eyes ( 52. 9%) were improved, of which 106 eyes' visual acuity was 0. 05 or more.
CONCLUSlON:Surgery is an effective means of treating myopic RRD. The older patients have lower success rates. A higher degree of myopia led lower reset rate after retinal detachment surgery and poor postoperative visual acuity. When treat myopic RRD, we should pay attention to the recovery of visual function.

Objective To explore the relationship of traditional Chinese medical syndrome patterns with renal pathological changes and clinical laboratory parameters in patients with lupus nephritis.Methods Syndrome patterns were differentiated according the clinical manifestations.The data of laboratory examination and renal pathology were collected and analyzed in 40 patients with lupus nephritis.Results(1) Of the fundamental syndrome patterns,pattern of spleen and kidney Qi deficiency had higher activated partial thromboplastin time(APTT),erythrocyte sedimentation rate(ESR) and 24-hour urine protein amount than the pattern without spleen and kidney Qi deficiency.(2) Of the incidental syndrome patterns,water-damp pattern had higher APTT than damp-turbidity pattern;hemoglobin(HGB) level in water-damp pattern and damp-turbidity pattern was lower than the normal,and HGB level in water-damp pattern differed from that in blood-staiss pattern and damp-heat pattern(P

Objective:To investigate the expression of CXCR4/CXCL12 and effect on the expression of CXCR4 by hypoxia in trophoblasts of human first trimester gestation.Methods:To detect the expression of CXCR4 and CXCL12 in placental tissue and primary cytotrophoblasts by immunohistochemistry and to assess the level of CXCR4mRNA under the condition of hypoxia in the trophoblasts of first trimester gestation by reas-time PCR.Results:The expression of CXCR4/CXCL12 were detected in the villous column,blood vessels and trophoblasts in the early pregnancy;CXCR4 mRNA was significantly higher through 12,24,48 and 72 h incubation periods in the hypoxic condition than that under normal oxygen condition respectively.Conclusion:Human first trimester trophoblasts produced CXCR4/CXCL12,which may play important biological role on the normal human pregnancy;Hypoxia could regulate the expression of CXCR4,which may contribute to the physiological pregnancy process.
6.Study on the synergistic proliferation-inhibiting and apoptosis-inducing effects of recombinant human-derived interleukin-24 and recombinant human-derived decorin on human hepatocellular carcinoma cells HepG2

Objective To investigate the synergistic proliferation-inhibiting and apoptosis-inducing effects of recombinant human-derived interleukin-24 (rhIL-24) and recombinant human-derived decorin (rhDCN) on human hepatocellular carcinoma cells HepG2.Methods Cellular growth and morphological changes of HepG2 cells were observed under the inverted microscope at 48 h after being transiently transfected with pcDNA3.1 (+)-IL-24 and pcDNA3.1 (+)-DCN by Lipofectamine.The proliferation-inhibiting effects of IL-24 and DCN on HepG2 cells,respectively and jointly,were observed with MTT assay at 24 h,48 h and 72 h post-transfection.Apoptosis and cell-cycle of HepG2 cells were analyzed by flow cytometry at 48 h post-transfection.Results Compared to control groups,the cells of target gene groups presented typically changes of proliferation inhibition and apoptotic morphology,which occurred obviously in co-transfection group.The results of MTT assay showed that at 48 h and 72 h post-transfection,the profiferation-inhibiting rates in the group of cells co-transfected with IL-24 and DCN were (31.88±6.57) % and (36.83±3.76) %,respectively,displaying significant difference with those of other groups (P < 0.01).The results of flow cytometry showed that IL-24 and DCN can induce HepG2 cells apoptosis to some extent.Compared to the early apoptosis rate of cells of control groups,plasmid (2.98±0.72) %,blank cell (3.50±0.92) %,IL-24 (20.01±1.08) % and DCN (22.20±0.91) %,a statistically remarkable apoptosis rate,(32.56±0.90) %,can be seen in the group of cells treated with IL-24 and DCN jointly (P < 0.01).The result of cell cycle analysis revealed that,compared to control groups,the proportion of cells was higher in the phase of G2/M in the IL-24 group (11.24±0.35) % and in the phase of G0/G1 in the DCN group (77.93±0.67) %.The proportions of cells in the phases of G2/M increased to (71.36±0.60) % and that of G0/G1 statistically increased to (10.39±0.67) % in the group of cells co-transfected with IL-24 and DCN (P < 0.01).Conclusions Combinatorial treatment of HepG2 cells with IL-24 and DCN can exert stronger synergistic proliferation-inhibiting effect and apoptosisinducing activity-in comparison to single therapies.IL-24 and DCN can induce cell cycle arrest on HepG2 cells,occurred in the phase of G2/M and G0/G1,respectively.Promoting effect of cell cycle arrest in the phase of both G2/M and G0/G1 can be seen on HepG2 cells co-transfected with IL-24 and DCN,which maybe the possible mechanism of the synergistic proliferation-inhibiting and apoptosis-inducing effect.

Because of lots of ethical problems,military medical triage has been unaccepted by general doctors.But it's adopted by American Army in front of facts of the war.The main principles of the military medical triage are justice and efficiency.They practice the military medical triage through tow steps.First,all patients are assigned into urgent,immediate ,delayed,minimal or ambulatory and expectant status in decreasing order of medical urgency.Second,choosing the model of triage depends on nonaustere,austere or extreme conditions.The ethical issues of American military medical triage focus on four aspects - how to deal with minimally injured patient treatment first,how to treat expectants,how to care for noncombatant casualties and who decide the model of triage.

Aim To study the role of ubiquitin carboxyl terminal hydrolase L1 (UCH-L1)involved in the pro-tective effect of fluoxetine against monocrotaline-in-duced pulmonary arterial hypertension in rats.Meth-ods Monocrotaline (60 mg·kg -1 )was used to es-tablish pulmonary arterial hypertension in rats and low-dose (2 mg·kg -1 ·d -1 )or high-dose (10 mg·kg -1 ·d -1 )fluoxetine was applied to inhibit pulmonary ar-terial hypertension.The hemodynamics,morphology of pulmonary arterioles and lungs,UCH-L1 protein ex-pression and nuclear factor-κB (NF-κB)nuclear trans-location were observed.Results Monocrotaline not only increased pulmonary arterial pressure and promo-ted pulmonary arterial remodelling and lung inflamma-tion,but also down-regulated UCH-L1 protein expres-sion and increased NF-κB activity in lungs.Fluoxetine inhibited these changes in a dose-dependent manner. However,UCH-L1 protein expression of pulmonary ar-teries did not significantly change among different groups.Conclusion Fluoxetine inhibits monocrotal-ine-induced lung inflammation in rats,involved in NF-κB activity inhibited by up-regulated UCH-L1 protein expression.
10.Mesenchymal to epithelial transition in malignant mesenchymal tu-mors

Mesenchymal to epithelial transition (MET), whereby mesenchymal cells become more epithelial like in phenotype, was observed to occur during normal development and in cancers. Numerous investigations have been conducted on MET in carcino-mas. In addition, accumulating evidence also suggests the critical function of MET in sarcomas. Integrated analyses reveal that MET may be an important biological and clinical process in sarcomas, and transcription factors such as Slug may also perform central func-tions in epithelial differentiation in several sarcomas such as leiomyo-sarcoma and synovial sarcoma. Given the scarcity of investiga-tions and evidence, several important issues about MET, such as its molecular markers, signaling mechanisms, micro RNA regulations, and clinical significance, need to be clarified. In this article, we review several important questions about MET in sarcomas, including molecular markers, signaling mechanisms, regulation by miRNAs, and therapeutic implications.
