1.Increased apoptosis of A549 cells by overexpression of helicase-like transcription factor induced by radiation exposure

Objective To study the effects of overexpression of helicase-like transcription factor (HLTF) on radiation-induced apoptosis of human lung carcinoma cells.Methods Human lung carcinoma cellline A549 were cultured,transfected with FLAG-tagged HLTF expression plasmid pcDNA3.1/HLTF-FLAG (HLTF gene transfection group) ,empty plasmid vector pcDNA3.1 (empty vector group) ,and mock transfection group,respectively,and then exposed to 60Co γ-ray irradiation.Cell apoptosis was observed by flow cytometry 48 h post-irradiation.Western blotting was used to detect the levels of HLTF in the nucleus and cytoplasm and the release of cytochrome C from mitochondria into cytoplasm.Results The apoptosis rate of the HLTF gene group was (32.2 ± 2.1) %,significantly higher than those of the empty vector group [(11.4±2.3)%] and mock transfection group [(11.1 ±1.8)%] (F=101.85,P<0.05).The level of cytochrome C in the cytoplasm of the HLTF gene group was significantly higher than that of the empty plasmid group.Conclusions HLTF protein sensitizes human lung carcinoma cells to radiation-induced apoptosis,partly by promoting the nuclear-cytoplasma transition of HLTF protein and the increased release of cytochrome C.
2.Laparoscopic diagnosis and treatment of non-palpable undescended testis: Experience of 29 cases (34 sides)

Objective To discuss the laparoscopic diagnosis and treatment for non-palpable undescended testis.Methods Laparoscopy was utilized in the diagnosis and treatment of 29 cases of non-palpable undescended testis(34 sides) from July 2002 to March 2005.Results The diagnosis was clarified in all the 29 cases.Intraabdominal testis was found in 21 sides(primary orchidopexy in 16 sides and two-staged Fowler-Stephens orchidopexy in 5 sides) and absent testis,13 sides.Follow-up observations for 0.5~1 year in 29 cases showed no testicular retraction or atrophy.Conclusions Laparoscopic intervention,characterized with accurate diagnosis,little invasion,and quick recovery,can be the first choice for the diagnosis and treatment of(non-palpable) undescended testis.The primary orchidopexy of testis may have satisfactory results if possible.
3. The effect of DLX4 on the apoptosis of choriocarcinoma cells and its related mechanism

Objective: To explore the effect of homeodomain protein DLX4 on apoptosis of choriocarcinoma cells and its related mechanism. Methods: RNA interference (RNAi) was used to knockdown DLX4 expression in JEG-3 cells. Then the inhibitory effect of DLX4 on apoptosis was determined by flow cytometry and the expression of apoptosis-related protein cleaved caspase-3 and caspase-8 and bax was detected by western blotting. Results: RNA interference targeted for DLX4 effectively and specifically inhibited DLX4 mRNA expression and decreased DLX4 protein level in JEG-3 cells. The apoptotic rate of JEG-3 cells was increased, the expression levels of cleaved both caspase-3 and caspase-8 were elevated but the bax protein expression did not change after knocking down DLX4. Conclusion: RNA interference silences DLX4 expression and induces apoptosis of JEG-3 cells. The mechanism for apoptosis was related with transcriptional regulation on caspase-3 and caspase-8.
4.Changes of Serum Nitric Oxide and Trace Element Zinc in Children with Pneumonia and Their Correlation

Objective To explore the changes of serum nitric oxide(NO) and trace element Zinc in children with pneumonia and their clinical significance.Methods The observing group contained 48 patients with pneumonia in our hospital from Oct.2005 to May 2006,who were collected 3 mL of blood sample on empty stomach on the second day.Twenty-six of them had been collected serum during their convalescence stage.The control group contained 20 children,who were healthy in the same stage.The levels of NO of 48 pneumonia,26 convalescence stage(recovery group) and 20 healthy patients were determined by UV-2100 spectrophotometer.The Zinc in serum was determined by P-E503-mode atomic absorption spectrophotometric analysis antigenic in those patients.Blood viscosity was measured and analyzed with the statistic analysis SPSS 10.0 software.Results The levels of NO in pneumonia children[(57.76?19.41) ?mol/L] were significantly higher than that in control group [(25.09?5.51) ?mol/L] and recovery group[(30.08?8.05) ?mol/L](P_a
5.Design of competitive teaching model for medical literature retrieval practice course

6.Theoretical model for compatibility of medicinal property combination of traditional Chinese medicines.

Medicinal properties are specific attributes of traditional Chinese medicines(TCM). The medicinal property theory is an important principle for the compatibility of traditional Chinese medicines. In this paper, medicinal properties, flavors and meridian tropism were combined to represent TCM medicinal properties; and multiple medicinal properties were further combined into medicinal property combination modes. TCMs and medicinal property combination modes were divided according to their efficacies, which were regarded as the concept of inductive logic programming and finally got medicinal property combination and compatibility rules with different efficacies. These medicinal property combination and compatibility rules were used to form the theoretical model through the entity grammar system, realize the automatic reasoning process from the medicinal property combination and compatibility to the efficacies, verify the reasoning result and analyze their rationality and limitations, in order to provide new ideas for revealing the relations between the TCM compatibility rules and efficacies.

7.DNA repair protein degradation mediated by helicase-like transcription factor in radiation-induced apoptotic cells

Objective To study the effects of helicase-like transcription factor (HLTF)transfection on DNA repair protein level in radiation-induced apoptotic cells.Methods Human lung carcinoma A549 cells were cultured and transfected with FLAG-tagged wild type HLTF (wild type HLTF transfection group),RING structure domain (ubiquitin conjugating region) mutatation HLTF expressing plasmid (mutant transfection group),empty plasmid (congtrol group) respectively.And the other cells were used as mock transfection group.All cells were irradiated with 15 Gy of 60Co γ-rays to induce apoptosis.Western blotting was used to detect the protein levels of the DNA repair proteins HRAD17 and HRAD52 in the transfected cells.Results The levels of HRAD17 and HRAD52 in the wild type HLTF transfection group was significantly lower than that of the control group.There was no significant difference in HRAD17 and HRAD52 levels between the mock transfection group and ubiquity in conjugating region mutation group.complexes of HLTF and HRAD17 and HRAD52 could be found in the irradiation-induced cells.Conclusions HLTF mediates the degradation of HRAD17 and HRAD52 in the irradiation-induced apoptotic cells possibly by the interaction of the protein complex causing ubiquitination of the repair proteins.
8.Intervention of Effective Parts of Acanthopanax Senticosus on the Expression of LRRK 2 in Injured PC12 Cells Induced by MPP+

Objective:To observe the effects of effective parts of Acanthopanax senticosus on the expression of LRRK 2 in injured PC12 cells induced by MPP+ to explore the mechanism of neuroprotection of the effective parts of Acanthopanax senticosus.Methods:The PC12 cell model of Parkinson's disease (PD) was constructed by MPP+,and intervened by the effective parts of Acanthopanax senticosus.The cell survival rate was detected by MTT,the mRNA expression level of LRRK 2 was studied by PCR,and the protein expression of LRRK 2 was determined by immunohistochemistry and Western Blot.Results:Compared with that of the model group,the survival rate of the drug group increased with significant difference (P<0.01).Compared with those in the model group,the LRRK 2 gene and protein expression all decreased with significant differences (P<0.01).Conclusion:The effective parts of Acanthopanax senticosus show protective effects on the PC12 cell model of PD,and one of the mechanisms may be related with the decreased expression level of LRRK2 caused by the effective parts of Acanthopanax senticosus.

10.CdTe quantum dot inhibits cell survival and induces mitochondrial dysfunction in primary human umbilical vein endothelial cells

OBJECTIVE To elucidate the toxicological properties of CdTe quantum dots (CdTe QD) by investigating their effect on mitophagy in human umbilical vein endothelial cells (HUVECs).METHODS The purity of primarily cultured HUVECs was detected by immunofluorescence.Then,HUVECs were incubated with CdTe QD 0.1-100 mg· L-1 for 24 h.After treatment,the cell viability of HUVECs was detected with MT-T assay.The mitochondrial morphology was observed under a laser scanning confocal microscope after labeling with Mitotracker.The treated HUVECs were also labeled with JC-1 probe,and the mitochondrial membrane potential (MMP) was then examined by flow cytometry.The expression of mitophagy-related proteins including microtubule-associated protein 1 light chain 3 Ⅰ /Ⅱ (LC3 Ⅰ /Ⅱ),moesin-like BCL2-induced protein1(Beclin1),phosphatase and tensin (PTEN) homologinduced putative kinase 1(PINK1) and dynamin-related protein Ⅰ (DRP1) was determined by Westem blotting.RESULTS More than 95% of the cultured cells expressed vascular endothelial cadherin and herein were vascular endothelial cells.The MTT result showed that the cell survival of HUVECs was significantly decreased after incubation with CdTe QD (0.1-100 mg,L-1) for 24 h (P<0.05,P<0.01).CdTe QD also induced extensive fragmentation of the mitochondrial network.The results of JC-1 assay showed that CdTe QD (0.1-100 mg· L-1) caused the disruption of MMP.The percentage of HUVECs with higher MMP was reduced from (91.8±0.6)% in cell control group to (90.2±1.1)%,(84.4±0.9)% (P<0.05) and (78.1 ±1.3)% (P<0.01),respectively.The Western blotting data suggested that CdTe QD 10 mg·L-1 significantly increased the expression of autophagy-related protein beclin 1 and the ratio of LC3 Ⅱ/LC3 Ⅰ (P<0.05,P<0.01),CdTe QD 1 mg· L-1 also raised the level of mitophagy-related proteins like PINK1 and DRP1.CONCULSION CdTe QD can induce mitochondrial dysfunction as well as mitophagy in HUVECs.
