1.Cultural Conditions for Production of Glutathione by Mutant Saccharomyces J-X25

The production conditions of glutathione with shaking flask fermentation by the mutant Saccharomyces J-X25, a methionine-defected strain were studied, and the optimum culture conditions are as follows: initial pH6. 0, temperature 30℃, 100ml in 500ml flask, the inoculum size 10% and agitation rate 220r/min. The emphasis was on the stimulating effect on the cells by dioxogen and the sodium lactate as surfactant. Both of which were added at the logarithmic phase of fermentation, and the GSH production was up to 0. 253g/L , 52% higher than the control without the additions. Compared with the production of GSH initial strain that by the mutant in optimum conditions was increased by 79%.
2.Diagnostic value of MRI on Creutzfeldt-Jakob disease

Objective To investigate the diagnostic value of MRI on Creutzfeldt-Jakob disease(CJD).Methods The imaging data of 15 patients with CJD were analyzed retrospectively.Results In the 15 cases,the diagnosis was clinical probable CJD 10 cases,maybe CJD 5 cases.There was no abnormal signal found by routine T 1WI and T 2WI scanning in the 15 cases.DWI showed that 10 cases had hyperintense abnormalities in both the cerebral cortex and the striatum,5 cases had hyperintense abnormalities only in the cerebral cortex.The hyperintense abnormalities were symmetric in 8 cases,but were asymmetric in the other 7 cases.1H-MRS were applied in the 4 cases and showed that N-acetylas partate(NAA),NAA/creatine(Cr) were decreased to some extent.Choline(Cho),Cho/Cr were decreased in 2 cases and increased in 2 cases.mI peak did not appear in all 4 cases.Conclusion MRI,especially DWI and 1H-MRS give can valuable information in detecting and diagnosing CJD early.
3.Dynamic Changes of Th1/Th2 Cell Factor in Induced Sputum and Clinical Significance in Children with My-coplasma Pneumoniae Pneumonia

Objective To detect levels of IL - 4 and INF - ? in induced sputum dynamically in children with mycnplasma pneamo-niae pneumonia( MPP), and to analyze the function of Th1 /Th2 cell factor immune response in the genesis and development of MPP, so as to evaluate the clinical value of induced sputum method in MPP research. Methods There were 38 cases who were diagnoses as MPP using 3% high osmotic pressure of saline water to ultrasonic atomizing inhalation for inducing sputum. ELISA was used to detect IL-4 and INF-?. Results The content of IL-4 in acute stage was higher than that in convalescence stage in induced sputum of MPP children. Severe stage was higher than mild stage. However, the comparison between acute and convalescence stage didn't have statistics difference in the content of INF-?, neither did the comparison between severe and mild stage. IL- 4/INF- ? in acute stage was higher than that in convalescence stage. Severe stage was higher than mild stage. In convalescence stage, the comparison of INF - ?, IL - 4, IL - 4/INF - ? between the severe and the mild didn' t have statistic significance. Conclusions IL-4 and INF - ? have participated in the monogenesis of MPP. The disequilibria of Th1 /Fh2 is existed in MPP and Th2 reaction predominates. So induced sputum analysis can be a better way to judge the light or heavy press degree of MPP practically, conveniently and sensitively.

5.Fingerprints of saline semen Trigonellae by HPLC

AIM: To establish the HPLC fingerprints of semen Trigonellae processed with salt-water.(METHODS:)HPLC analysis was performed on a phenomenex Hydro-RP 80A(4.6 mm?250 mm,5 ?m) with mobile phase acetonitrile-water under isocratic and gradient elutions,and detection wavelength was 211nm,the flow rate was 1 mL/min,column temperature was 40 ℃. RESULTS: HPLC fingerprint of 17 main peaks was separated.It was found that the preparative HPLC of standardized processing saline semen Trigonellae from different places have no significant differences. CONCLUSION: The method is reliable,accurate and can be used for quality control of saline semen Trigonellae.
7.Experimental research on antibacterial effect of Belamcanda Ch inensis DC and portulaca oleracece L on P. aeruginosa in vitro

Objective:To explore the antibacterial effect of Belamcanda Chinensis DC and Por tulaca oleracece L on P.aeruginosa (PA) in vitro.Methods:Fourty six strains PA were tested for minimum inhibition concentrati on ( MIC) by water decoct agents of the two drugs.MIC50 and MIC90 were st atistically studied.Results:For Belamcanda Chinensis DC,MIC was 31.25~3.90 g/L;MIC50 was 7 .81 g/L,and MIC90 was 15.62 g/L;whereas, for portulaca oleracece L,MIC was 31 .25~7.81 g/L, MIC50 was 15.62 g/L,and MIC90 was 31.25 g/L.Conclusion:Both of the two drugs have stronger antibacterial effects on P.ae ruginsosa in vitro.
9.Effect of Shuwel Decoction on Enteric Nervous System-Interstitial Cells of Cajal-Smooth Muscle Network Structure Injury in Deep Muscle Nerve Plexus of Functional Dyspepsia Rats.

OBJECTIVETo observe morphological changes of enteric nervous system (ENS)-interstitial cells of Cajal (ICC)-smooth muscle cell (SMC) structure injury in deep muscle nerve plexus offunctional dyspepsia (FD) rats, and the repair of Shuwei Decoction (SD) on it, and to explore its effecton FD.
METHODSTotally 72 rats were randomly divided into the control group, the model group, the lowdose SD group, the medium dose SD group, and the high dose SD group, the Mosapride group, 12 ineach group. Rats in the low dose SD group, the medium dose SD group, and the high dose SD group were intragastrically fed with SD at 0.767, 1.534, 3.068 g/mL, respectively. Rats in the Mosapride group were intragastrically fed with Mosapride (1.37 mg/kg). FD rat model with Gan depression Pi deficiency syndrome (GDPDS) was established using complex pathogenic factors. Corresponding liquors were respectively administered to rats in corresponding groups from the 3rd day after modeling. Distilled water(10 mL/kg) was administered to rats in the control group and the model group, once per day for 14 successive days. Rats were sacrificed and small intestine tissues collected for observing ENS-ICC-SMC structure injury using immunofluorescence double labeling, laser scanning confocal microscope, and transmission electron microscope at day 15. Repair of SD on it was also observed.
RESULTSENS-ICC SMC structure was incomplete, with obvious injury in mutual link of ICC, ICC, SMC, and connecting structure. ENS-ICC-SMC structure was more complete in high, medium, and low dose SD groups, with close link of ICC and SMO. Their connecting structures were in good conditions.
CONCLUSIONSD could keep the integrity of ENS-ICC-SMC structure by promoting regeneration and morphology of ICC, thereby, improving gastrointestinal movement disorder and showing therapeutic effect on FD.

10.Effects of 1.8mm coaxial micro incision phacoemulsification on corneal endothelial injury and postoperative visual acuity

AIM:To investigate the effects of 1.8mm coaxial micro incision phacoemulsification on corneal endothelial injury and postoperative visual acuity.METHODS: Totally 145 eyes in 120 patients underwent phacoemulsification from July 2013 to July 2015 were randomly divided into observation group 60 cases (73 eyes) and control group 60 cases (72 eyes).The observation group 60 cases were given 1.8mm coaxial micro incision cataract phacoemulsification operation,while the control group were given traditional 3.2mm coaxial micro incision cataract surgery.The uncorrected visual acuity (UCVA),best corrected visual acuity (BCVA),corneal thickness of incision area,incision width,incision length,macular retinal thickness,surgically induced astigmatism,corneal endothelial cell counts and complications of the two groups were compared.RESULTS: The UCVA and BCVA on 1wk after surgery of the observation group were significantly higher than the control group (t=3.604,7.109;P<0.05);the width of incision on 1wk and 1mo after surgery of the observation group were significantly less than the control group (t=205.3,225.2;P<0.05).The length of incision in observation group was significantly greater than the control group (t=3.926,5.009;P<0.05).Macular retinal thickness 1wk after surgery of the observation group was significantly less than the control group (t=2.817,P<0.05).The surgically induced astigmatism was significantly less than the control group (t=19.43,22.16;P<0.01);the difference of corneal edema between the two groups was not significant (8.22% vs 11.11%) (x2=0.348,P>0.05).CONCLUSION: The 1.8mm micro incision phacoemulsification is helpful to improve the visual acuity of patients with cataract phacoemulsification,which may be related to the reduction of corneal cell injury,enhancement of corneal closure and decrease post-operation corneal original astigmatism.
