2.Construction of automatic elucidation platform for mechanism of traditional Chinese medicine.

Aim at the two problems in the field of traditional Chinese medicine (TCM) mechanism elucidation, one is the lack of detailed biological processes information, next is the low efficient in constructing network models, we constructed an auxiliary elucidation system for the TCM mechanism and realize the automatic establishment of biological network model. This study used the Entity Grammar Systems (EGS) as the theoretical framework, integrated the data of formulae, herbs, chemical components, targets of component, biological reactions, signaling pathways and disease related proteins, established the formal models, wrote the reasoning engine, constructed the auxiliary elucidation system for the TCM mechanism elucidation. The platform provides an automatic modeling method for biological network model of TCM mechanism. It would be benefit to perform the in-depth research on TCM theory of natures and combination and provides the scientific references for R&D of TCM.

5.Effects of Heat Needle Treatment on Inflammatory Reaction in Damaged Tissues of Rats with Third Lumbar Transverse Process Syndrome

Objective To observe the effect of heat needle treatment on the inflammatory reaction in the damaged tissues of rats with the third lumbar transverse process syndrome. Methods Ninety Sprague-Dawley rats were randomly divided into 6 groups, namely normal group, model group, drug group, heat needle group, conventional acupuncture group and warming acup-moxibustion group, 15 rats in each group. The rat model of the third lumbar syndrome was established by embedding gelatin sponge. Two weeks later, the rats in various groups were given corresponding treatment respectively. Before treatment and on the 7th, 14th and 28th day of treatment, the blood samples were collected from jugular vein, and then the serum inflammatory cytokines of tumor necrosis factor alpha (TNF-α), interleukin (IL)-1β and IL-6 were detected by enzyme-linked immunosorbent assay (ELISA). On the 28th day of treatment, all of the rats were sacrificed, the expression levels of TNF-α, IL-1βand IL-6 mRNA in the muscle tissue were detected by reverse transcription quantitative real-time polymerase chain reaction (RT-qPCR), and the pathological changes in local muscle tissue were observed by HE staining method. Results The serum contents of the inflammatory cytokines of TNF-α, IL-1β and IL-6 in the model group and treatment groups after modeling were significantly higher than those in the normal group (P < 0.05). After treatment, the contents of TNF-α, IL-1β and IL-6 in various treatment groups were decreased significantly as compared with those in the model group, and the decrease was most obvious in the heat needle group. The expression levels of TNF-α, IL-1β and IL-6 mRNA in muscle tissues of drug group, heat needle group, conventional acupuncture group and warming acup-moxibustion group were lower than those of the model group (P < 0.05), and the heat needle group had the lowest levels. HE staining results showed that heat needle group had milder inflammatory reaction and better muscle fiber structure than the model group. Conclusion Heat needle treatment can improve the local inflammatory response and promote the repair of tissue damage through decreasing the expression levels of inflammatory cytokines of TNF-α, IL-1βand IL-6.
6.Outcomes and prognostic factors of 25G minimally invasive vitrectomy for intraocular foreign body

AIM:To explore clinical characteristics, outcomes and prognostic factors for cases with intraocular foreign bodies(IFB)and treated with 25G minimally invasive vitrectomy(PPV).
METHODS: Patients traumatized with retained IFB and treated with PPV were retrospectively collected from the Department of Ophthalmology, Affiliated Hospital of Southwest Medical University from 2016-1-1 to 2019-1-1. The clinical records including general condition, time and cause of injury, locations andsize of IFBs, comorbidities, and best corrected visual acuity(BCVA)were reviewed and statistical analyzed.
RESULTS: Among the 105 patients, most of them are middle-aged and young men, mostly from townships. The nature of IFB was mainly metals(62 cases, 59.0%). The entrance of IFB of 78 cases(74.3%)located at Zone I of the eye. There were 17 patients with BCVA≥0.1 before surgery, 88 patients with BCVA<0.1. And 43 patients gained BCVA≥0.1 after operation, and 62 patients with BCVA<0.1. Postoperative BCVA was significantly improved compared with preoperative BCVA(P<0.05). Through multivariate Logistic regression analysis, poor preoperative BCVA, retinal detachment, and endophthalmitis are risk factors for poor visual prognosis.
CONCLUSION: PPV for treatment of IFB can achieve a better outcome. The poor prognosis of visual acuity is related to retinal detachment, endophthalmitis, and the position and size of IFB.
8.Leptin receptor of the hind brain nuclei is involved in the conditioned taste preference of rats.

OBJECTIVEConditioned taste preference (CTP) is a taste learning reflex by which an animal learns to prefer a substance which tastes not well and has been studied with much interest in recent years. However, the neural substrates of CTP are less known. This study aimed to determine the possible neural path- ways of CTP and whether serum leptin level and the leptin receptor (OB-Rb) in the hind brain are involved following CTP formation.
METHODSWe established CTP of quinine in rats with a 2-bottle preference test. The serum leptin concentrations were detected, the expression of c-fos in the rat brain was tested to determine the nuclei in relation with establishment of CTR Finally, the OB-Rb mRNA expression was examined by RT-qPCR assay in parabrachial nucleus (PBN) and the nucleus of the solitary tract (NST) of the hind brain.
RESULTSCompared with control group, the level of serum leptin was higher in the CTP group (4.58 ± 0.52 vs 1.67 ± 0.25 µg/L, P < 0.01); increased c-fos positive cells were found in the anterior hypothalamus (AH, 221.75 ± 4.96 vs. 178.50 ± 6.63 cells/mm², P < 0.05), the basal lateral amygdala (BLA, 70.75 ± 6.17 vs 56.50 ± 3.62 cells/ mm², P < 0.05) and the nucleus of the solitary tract (NST, 41.25 ± 1.32 vs 32.50 ± 1.02 cells/mm², P < 0.05). But in ventromedial nucleus of the hypothalamus (VMH, 20.75 ± 2.73 vs 38.5 ± 1.54 per 1 mm², P < 005), PBN (21.50 ± 2.24 vs 36.25 ± 1.49 cells/mm², P < 0.05) and the central nucleus of the amygdala (CeA, 22.25 ± 1.53 vs 35.50 ± 2.11 cells/mm², P < 0.05), the number of c-fos positive cells was decreased in the CTP group. In addition, we found OB-Rb mRNA expression in PBN of CTP group rats was higher than that of control group (0.95 ± 0.055 vs 0.57 ± 0.034, P < 0.05), while there was no significant difference of OB-Rb mRNA expression in NST between the two groups.
CONCLUSIONNuclei AH, BLA, NST, VMH, PBN and CeA participate in the formation of CTP. Leptin and its receptor in PBN may be involved in the formation and maintenance of CTP.

9.Morphology and Molecular Characterization of Alternaria argyranthemi on Chrysanthemum coronarium in China.

Chrysanthemum coronarium is an economically important plant in Asia, and used medicinally, ornamentally and as a vegetable. In April 2017, leaf spot disease on C. coronarium was observed in Shiyan, Hubei, China. A single-spore isolate was obtained and identified based on morphology and sequence analysis using four regions (rDNA ITS, GAPDH, EF-1α, and RPB2). The results indicated that the fungus is Alternaria argyranthemi. The pathogenicity tests revealed that the species could cause severe leaf spot and blight disease on the host. This is the first report of leaf spot disease on C. coronarium caused by A. argyranthemi in the world, which is also a new record of Alternaria species in China.

10.Genotyping of Epstein-Barr virus in Epstein-Barr virus associated gastric carcinoma.

Tumor samples from 236 gastric carcinoma patients and throat washings (TWs) from 135 healthy adults were collected and screened for EBV genome. 17 Epstein-Barr virus associated gastric carcinoma (EBVaGC) and 33 EBV positive TWs were further examined for type 1/2 EBV and polymorphism at Bam HI F, Bam HI W1/I1 boundary regions and Xho I restriction site in LMP1 gene. No type f of Bam HI F polymorphism was found in all the cases, nor type 2 in EBVaGC. 25 of 33 TWs (75.8%) analyzed detected type 1 virus, while 8 TWs (24.2%) detected type 2 virus. Type I and i of Bam HI W1/I1 polymorphism accounted for 1(5.9%) and 16 (94.1%) in EBVaGC and 11 (33.3%) and 19 (57.6%) in TWs, respectively. LMP1 Xho I(+) and (-) polymorphism accounted for 0(0) and 15 (88.2%) in EBVaGCs and 12 (36.4%) and 18 (54.5%) in TWs, respectively. Among those specimens with determinable genotypes through all the polymorphism, the distribution rate of type 1/i/Xho I(-) in EBVaGC (15/15, 100%) was significantly higher than that in TWs (4/28, 14.3%) (chi2 = 29.098, P < 0.0001), suggesting that the distinctive EBV strain (1/i/Xho I(-)) could be associated with Chinese EBVaGC.
