1.Clinical study on coronary artery calcification score combined with carotid intima-media thickness measure in elderly patients with coronary artery disease

Objective To evaluate the clinical diagnostic value of coronary artery calcification score(CACS)of multi-slice spiral computed tomography(MSCT)combined with carotid intima-media thickness(IMT)measure in elderly patients with coronary artery disease(CAD). Methods CACS of MSCT,carotid IMT measure,and coronary angiography were performed in 68 patients,including 36 cases with CAD(CAD group)diagnosed by coronary angiography and 32 cases(control group)with coronary arterial stenosis(<50% stenosis).CACS and carotid IMT were compared between two groups. Results The coronary artery calcification score was significantly increased in CAD group compared with the control group[(349.5±86.3)vs.(74.7±25.2),t=13.670,P<0.01],and it was increased with the severity of coronary arterial stenosis.The carotid intima-media thickness in CAD group showed significant difference with that in control group[(1.11±0.05)mm vs(0.69±0.13)mm,t=13.587,P<0.01].In CAD group,CACS exhibited a significant positive correlation with carotid IMT(r=0.950,P<0.01).The positive rates of CACS and carotid IMT were both 77.8% (28 cases)in CAD group and both 12.5%(4 cases)in control group,which showed significant difference between two groups(X2=28.976,P<0.01). Conclusions CACS of MSCT combined with carotid IMT have high sensitivity and specificity in evaluating coronary arteriaI stenosis.It can be used as a non-invasive examination to diagnose CAD in the elderly.

4.Effect of low intensity white light irradiation on the retinas of mice

AIM: To investigate the effect of low intensity white light irradiation on the retinas of mice.
METHODS: Thirty C57BL/6J mice were randomly divided into two groups. The number of the mice in each group was 15. The mice in experimental group received dark adaptation from 5:00p. m. to 6:00p. m. , and then exposed to LED white light from 6:00p. m. to 7:00p. m. everyday for a month. At 1, 3, 7, 14 and 30d after the beginning, we examed the histology of mice retinas, calculated the thickness of outer nuclear layer ( ONL ) , inner nuclear layer ( INL ) and analyzed electrophysiology of mice.
RESULTS:One month after experiment, compared to the control group, the latency of Rod-R a wave of the mice in experimental group significantly prolonged, the amplitude of Cone-R b wave of the mice in experimental group significantly decreased and the latency of b wave of the mice in experimental group significantly prolonged ( P<0. 05). There are no significant difference in the histology of retina, ONL and INL thicknesses.
CONCLUSION: 100lux low intensity white light could give rise to the impairment of the retinal functions in dark-adapted mice.
5.In silico cloning and bioinformatics analysis of an AP2/EFR family gene from Arnebia euchroma.

A cDNA sequence of Arnebia euchroma AP2/ERF named AeAP2/ERF was cloned by in silico cloning in this study, using ACX71873 sequence from Lithospermum erythrorhizon as the probe sequence. Some characters of the AP2/ERF gene and encoded protein sequences were predicted and analyzed by the bioinformatics methods, including general physical and chemical properties, hydrophobieity, signal peptide, secondary structure, localization sites in cells. Results showed that the 876 bp long gene included a 1 077 bp ORF and encoding 205 amino acid. The AeAP2/ERF protein had no signal peptide, it was a hydrophilic proteins located in nucleus. The function of the AP2/ERF protein was mainly involved with metabolism controlling and signal transduction.

6.Blood supply features and interventional therapy of pedunculated hepatocellular carcinoma

Objective To evaluate the blood supply features and effectiveness of arterial chemoembolization for pedunculated hepatocellular carcinoma.Methods Angiography and chemoembolization via supplying blood arteries of tumor were performed in five patients with pedunculated hepatocellular carcinoma.Interventional procedure was carried out with tumor vascular infusion of 350 mg hot elemene emulsion and tumor embolization by cisplantin-lipidol emulsion(cisplantin 60-80 mg+lipidol 8-15 ml)and glutin.Results Ten interventional procedures(TACE)were undertaken in 5 patients.Angiography showed that tumor blood supply mainly coming from collateral circulation adjacent to the tumors,but partially from hepatic artery.Tumor sizes decreased from 30% to 50% in 5 cases,and AFP declined in 4 cases after the treatment. Conclusion Pedunculated hepatocellular carcinoma possessing different blood supply features from intrahepatocellular carcinomas.But transarterial ehemoembolization is still an effective method of choice for this treatment.
7.Effect of rosiglitazone on glucose tolerance in newly-diagnosed type 2 diabetes patients

One hundred and thirteen newly-diagnosed type 2 diabetic patients were treated with rosiglitazone for 16 weeks. Glucose tolerance was restored in 46 cases ( group A), but not restored in 67 cases (group B). Compared with group B,the patients of group A were younger and had shorter course of diabetes and lower body mass index (BMI,P<0.05); Homeostasis model assessment for β-cell function (HOMA-β) and earlyphase insulin secretion in group A were better than those of group B before and after treatment respectively (P<0. 05 ) ,while homeostasis model assessment for insulin resistance ( HOMA-IR )in group A was lower than that of group B before treatment (P<0. 05 ). In conclusions, rosiglitazone improved insulin resistance of type 2 diabetes patients, thus facilitated restoration of β-cells function and glucose tolerance. The related factors of glucose tolerance restoration included age ,diabetes duration, BMI, HbA1 c, β-cells function, and insulin resistance.
8.Effect of Chinese medicine intestine adjusting therapy on patients with respiratory failure caused by acute exacerbation of chronic obstructive pulmonary disease and undergoing noninvasive ventilation.

OBJECTIVETo observe the effect of Chinese medicine intestine adjusting therapy (IAT) on patients with respiratory failure caused by acute exacerbation of chronic obstructive pulmonary disease (AECOPD) and undergoing noninvasive ventilation, their immune function, ventilation indices and incidence of complication.
METHODSPatients matched with the inclusive criteria were randomized into two groups, 30 in each group. All received bi-level positive airway pressure ventilation and conventional drug therapy, but to patients in the treatment group, IAT was applied additionally by electro-acupuncturing (EA) acupoints Zusanli (ST36), Shangjuxu (ST37), Fenglong (ST40), and Quchi (LI11), also the retention enema with Xuanbai Dachengqi Decoction. The nutritional indicators, including serum total protein (TP), serum albumin (ALB) and hemoglobin (HGB); immune indices, including immuno-globulins (IgG, IgA, IgM), complements, and T-lymphocyte subsets; and the incidence of ventilation complications in the two groups were dynamically observed and compared.
RESULTSAfter treatment, the nutritional indicators went down in both groups (P < 0.05, P < 0.01), but the lowering in the treatment group were lesser. Moreover, the treatment group showed a higher TP level (P < 0.05) and lower depressive amplitude of ALB (P < 0.01) than those in the control group. Immune indices, excepting IgM, increased significantly in both groups (P < 0.05 or P < 0. 01), but the increments in the treatment group were higher, so significant difference was shown between groups (P < 0.05 or P < 0.01). As for comparison in ventilation complication, the incidence of abdominal distension (which was extensively occurred in the control group), belching and error aspiration in the treatment were significantly fewer (P < 0.05, P < 0.01). Besides, the maximum PS and PEEP, and the mechanical ventilation time were significantly reduced in the treatment group (P < 0.05).
CONCLUSIONIAT of Chinese medicine is facilitated to improve the nutritional status of AECOPD patients with respiratory failure undergoing noninvasive ventilation, enhance their immune function, improve the ventilatory efficiency, reduce the duration of mechanical ventilation and the occurrence of complications.

9.Killing effect improved by fusion gene HRE1.Egr-1. yCDglyTK on gene-radio therapy of nasopharyngeal cancer in vitro.

OBJECTIVE:
To construct hypoxia/radiation inducible promotor HRE1.Egr-1, and to observe its promotive effect on the expression of yCDglyTK gene in nasopharyngeal cancer HNE-1 cells and the anti-tumor effect of yCDglyTK and to lay an experimental foundation for further exploration of new gene-radio therapy of nasopharyngeal cancer.
METHODS:
pcDNA3.1(-)HRE1.Egr-1.yCDglyTK was constructed by gene recombination technique. Stable yCDglyTK-expressing HNE-1 cells were generated by transfecting the recombinant plasmid into the target cells with liposome. The expression of yCDglyTK was detected by Western blot in 4 groups: a normoxia group, a radiation group, a hypoxia group, and a hypoxia and radiation group. The killing effect of 5-FC in different circumstances was determined by MTT.
RESULTS:
The expression of yCDglyTK/5-FC gene in all the groups was significantly different(P<0.01),especially in the hypoxia and radiation group. The killing effect of 5-FC on HNE1 cells varied under different conditions, especially in the hypoxia and radiation group.
CONCLUSION
Hypoxia and radiation can induce the activity of fusion promoter HRE1.Egr-1, and obviously promote the anti-tumor effect of yCDglyTK/5-FC system, suggesting that yCDglyTK may be a candidate suicide gene for gene-radio therapy of NPC.

10.Establishment and clinical application of TapMan real-time RT-PCR method for detection of HHV-6.

OBJECTIVETo establish a rapid, sensitive and specific real-time PCR method for detection of Human Herpesvirus-6 (HHV-6).
METHODSAccording to the reference, a pair of primers and a probe were designed located in U65-66 gene and to set up the standards. We established a real-time RT-PCR method for detection of HHV-6, and to verify the specificity, sensitivity, reproducibility.
RESULTSThe correlation coefficient was 0.999, E = 97.9%, the coefficient of variation values of Ct were 0.61% and 3.13% in real-time PCR assay for inter and intra assay, respectively. The results of all viruses were negative except of HHV-6 for the assay. The quantitative detection limit of the assay was 3 x 10(0) copies/microl.
CONCLUSIONThe real-time PCR assay is highly specific, sensitive and reproducible, which can be used to quatitative detecting clinical samples.
