China Journal of Chinese Materia Medica 2014;39(3):536-539
Triptolide (TPT), an active compound extracted from Chinese herb Tripterygium wilfordii , has been used in therapy of rheumatoid arthritis (RA). In this study, after synovial fibroblasts from rheumatoid arthritis (RASFs) were treated with TPT, we investigated its effect on the differentiation of Th17 cells. Firstly, the mRNA level of cyclooxygenase (COX) wad detected by qRT-PCR and the protein level of prostaglandin E2 (PGE2) was tested by ELISA in RASFs treated with different concentrations (0, 10, 50, 100 nmol L-1 ) of TPT. Then after TPT pre-treated RASFs and RA CD4 + T cells wer e co-cultured for 3 days in the presence or absence of PGE2, IL-17 and IFN-gamma production in CD4 T cell subsets were detected by flow cytometry. The results showed TPT decreased the mRNA experssion of COX2 and the secretion of PGE2 in RASFs in a dose-dependent manner(P <0. 05). We further found that differentiation of Thl7 cells was downregulated in a dose-dependent manner, and exogenous PGE2 could reverse the inhibition of Th17 cell differentiation(P <0. 05). Taken together, our results demonstrated that TPT inhibited the mRNA level of COX2 and the secretion of PGE2 in RASFs, which partly led to impaired Th17 cell differentiation in vitro.

ObjectiveTo study possible differences in cognitive dysfunction between left hemisphere injury(LHI) patients and right hemisphere injury(RHI) ones.MethodsCognitive function of 55 LHI patients and 43 RHI patients were examined by aphasia screening test and the Loewenstein Occupational Therapy Cognitive Assessment Battery(LOTCA).ResultsThe scores of RHI group were significant higher than that of LHI group in orientation in place, overlapping figures, spacial perception,praxis(P<0.001-0.005),but significant lower than that of LHI group in visual identification of shapes and visuomotor organization(P<0.001-0.02).The possibilities of RHI group were significant more than that of LHI group in disorientation in time, shape agnosia, disorder in figure-ground identification, unilateral neglect, disordered attention(P<0.005-0.025),but significant fewer than that of LHI group in aphasia, ideomotor apraxia(P<0.005-0.01).The possibilities were no significant differences in disorientation in place, objects agnosia, colour agnosia, spacial agnosia, motor apraxia, constructional apraxia, thinking disorder between RHI group and LHI group(P>0.05).Conclusions There is characteristic cognitive dysfunction after right or left hemisphere injury.
10. Establishment of a cell model for high-throughput TRPV4 regulator screening based on CaCC
Medical Journal of Chinese People's Liberation Army 2020;45(10):1032-1039
Objective To construct a high-throughput screening model for transient receptor potential vanilloid 4 (TRPV4) channel modulators based on calcium-activated chloride channels (CaCC). Methods RT-PCR was used to detect the endogenous expression of TRPV4 in Fischer rat thyroid (FRT) cells. The PCR products obtained were subjected to nucleic acid sequencing using gel-recovery technology. Western blotting was employed to detect the expression of TRPV4 protein in FRT cells. The liposome transfection method was applied to construct the FRT cell model that co-expressed anoctamin 1 (ANO1) and YFP-H148Q/ I152L. The expressions of ANO1 and YFP-H148Q/I152L in cells were identified by the inverted fluorescence microscope and the fluorescence quenching kinetics test. After adding TRPV4 activators and inhibitors, the fluorescence quenching kinetics experiment was used to test whether the model could screen TRPV4 modulators. The Fura-2 fluorescent probe method was applied to detect the calcium concentration in cells after adding TRPV4 activators; The Z' factor was calculated to evaluate the sensitivity and specificity of the cell model. Results RT-PCR and Western blotting confirmed the endogenous expression of TRPV4 in FRT cells; ANO1 was clearly expressed on the FRT cell membrane and YFP-H148Q/I152L was clearly expressed in the cytoplasm of FRT cells under the inverted fluorescence microscope. The FRT cell model co-expressing ANO1 and YFP-H148Q/I152L was successfully constructed. Fluorescence quenching kinetics experiments confirmed that the model could screen TRPV4 regulators, and the slope value of fluorescence change and the concentration of TRPV4 regulator concentration were in a dose-dependent manner. The model could sensitively detect changes in intracellular calcium concentration, and the slope value could reflect intracellular calcium concentration. The Z' factor was 0.728, which demonstrates its capacity for high-throughput screening. Conclusions We successfully constructed a high-throughput model that could screen TRPV4 modulators sensitively and efficiently.
