1.Inhibition of visceral hypersensitivity in irritable bowel syndrome model rat by intrathecal administration of corticotrophin releasing factor

Objective To investigate whether intrathecal administration of corticotrophin releasing factor(CRF) has an efficacy on visceral hypersensitivity in irritable bowel syndrome model rat.Methods Thirty rats were randomly divided into three groups(n=10 ).After establishment of irritable bowel syndrome rat model were intrathecal injected with CRF or preemptive peritoneal injected with CP-154526,which is inhibitor of CRF-1 receptor,and the control group to give saline.After experiment all rat with the method of rectal balloon distention,the perception thresholds and the number of abdominal withdrawal reflexes (AWR) of different balloon volume were observed.Results The perception threshold of intrathecal administration of CRF group was(0.62±0.10)ml and higher than other two groups [(0.52±0.09)ml,(0.56±0.08)ml;F=3.25,P<0.05].At the same time,the number of AWR to the lower balloon content (1.0 ml) was(9.10±1.97)in intrathecal administration of CRF group slightly lower than other two groups[(14.4±1.71),(15.6±2.32);F=29.4,P<0.01],but no difference was found to the higher balloon content (1.5 ml and 2.0 ml).Besides,the areas and OD value of c-fos positive neurons in cornu posterius medullae spinalis in group with intrathecal administration of CRF were significantly lower than other two groups (P<0.05).Conclusion Intrathecal administration of CRF can degrade the visceral hyperensitivity in irritable bowel syndrome model rat,and lower expression of c-fos in CNS is probably involved in the process.
2.The curative effect of couple therapy for patients with postpartum depression

Objective To investigate curative effect thymoleptics with couple therapy for the patients with postpartum depression (PPD) Methods 27 patients first time diagnosed with PPD consistent with DSM-Ⅳwere randomly divided into study group and control group.The study group received regular thymoleptics with couple therapy,a control group were administered solo thymoleptics.Hamilton Depression Rating Scale(HAMD-17) and Edinburgh Postnatal Depression Scale (EPDS-10) and OLSON marriage quality questionnaire assessed the efficacy of each group before treatment and after 12 weeks treatment.Results (1) According to the total effective rate,there was no significantly differences in two groups after 12 weeks treatment from evaluation of HAMD-17>(P0.05),but complete remission of study group was higher than control group (78.6% ,46.2% ;P<0.05).(2) After 12 weeks treatment there were significantly differences in mean scores of HAMD-17 and EPDS-10 between two groups (P<0.05).The most decreased scores of HAMD-17 and EPDS-10 was found in the study group,and mean scores of each group was lower than pretherapy(P<0.05).(3) There was no significant difference on the scores of eight factors in OLSON marriage quality questionnaire between the two groups before treatment (P>0.05).After 12 weeks treatment the study group had higher score than control in seven factors in addition to sexual life factor (P<0.05).Conclusion Thymoleptics with couple therapy can improve the marriage quality and increase curative effects outstrip solo thymoleptics for the patients with postpartum depression.

4.Surveillance of Schistosomiasis in Yongsheng county of Lijiang city, Yunnan from 2002 to 2010

Objective To learn the effects of control measures,the screening situation and the epidemic situation of Schistosomiasis in Yongsheng county of Lijiang city of Yunnan province,and to provide a scientific basis for developing control strategies.Methods Annual surveillance data of Schistosomiasis of Yongsheng county from 2002 to 2010 were collected and analyzed by material retrospective method.Schistosomiasis screening situation,infection rate of people and cattle as well as snails area was statistically analyzed.Infection rate of people was from the screening results by indirect hemagglutination(IHA),and pathogenic examination of the positives(1 ∶ 10)was carried out through collection of eggs with nylon bag and hatching the stool.Infection rate of cattle was from etiology examination results by plastic cup pipe jacking method.Results From 2002 to 2010,257679 individuals were screened and 26540 cases (10.30%) were seropositive; with stool examination,451 (1.72%) were identified positive of 26233 persons; 603 (0.71%) were found positive of 85658 oxen.All of the positive detection rate of IHA method,the feces-positive detection rate in the population and the prevalence rate of oxen showed a declining trend(x2 =3615.53,785.68,434.50,all P< 0.05).The snails area was gradually reduced,the rate of snails environmental area was decline from 23.82% in 2004 to 1.98% in 2010.Conclusions The endemic situation of Schistosomiasis in Yongsheng county is controlled in the past eight years,but remains epidemic,and we will continue to strengthen monitoring,in order to achieve the goal of blocking the spread of Schistosomiasis.
5.Effect of five fungicides on growth of Glycyrrhiza uralensis and efficiency of mycorrhizal symbiosis.

In order to obtain the fungicides with minimal impact on efficiency of mycorrhizal symbiosis, the effect of five fungicides including polyoxins, jinggangmycins, thiophanate methylate, chlorothalonil and carbendazim on the growth of medicinal plant and efficiency of mycorrhizal symbiosis were studied. Pot cultured Glycyrrhiza uralensis was treated with different fungicides with the concentration that commonly used in the field. 60 d after treated with fungicides, infection rate, infection density, biomass indexes, photosyn- thetic index and the content of active component were measured. Experimental results showed that carbendazim had the strongest inhibition on mycorrhizal symbiosis effect. Carbendazim significantly inhibited the mycorrhizal infection rate, significantly suppressed the actual photosynthetic efficiency of G. uralensis and the most indicators of biomass. Polyoxins showed the lowest inhibiting affection. Polyoxins had no significant effect on mycorrhizal infection rate, the actual photosynthetic efficiency of G. uralensis and the most indicators of biomass. The other three fungicides also had an inhibitory effect on efficiency of mycorrhizal symbiosis, and the inhibition degrees were all between polyoxins's and carbendazim's. The author considered that fungicide's inhibition degree on mycorrhizal effect might be related with the species of fungicides, so the author suggested that the farmer should try to choose bio-fungicides like polyoxins.

6.Production of discrepant monoclonal antibody against HL60 and HL60/ADR by SI technique

Objective:To prepare and characterize specific and discrepant mouse hybridoma antibodies on membrane of HL60 and HL60/ADR cell lines.Methods:BALB/c mice were immunized by subtractive immunization induced Cp(Cyclophosphamide).McAbs were prepared by hybridoma technique,screened and detected by FACS and LSCM.Results:51 candidates and discrepant antibodies were found,and one of them (5F6) was purified and identified.Conclusion:Combination of SI with discrepant screening method should facilitate the preparing and identifying discrepant McAbs for identifying antibodies that can distinguish the differences in proteins expressed in HL60 and HL60/ADR,which is a significative and potential method in the research and target therapy associated drug-resistance.
7.Construction of a recombinant adenovirus vector for interferon-gamma-inducible protein 10 and the adenovirus preparation.

OBJECTIVETo construct a recombinant adenovirus vector for expressing interferon-gamma-inducible protein 10 (IP-10) by homogenous bacterial recombination.
METHODSIP-10 gene was cloned into the shuttle plasmid pAdTrack-CMV that contained the coding sequence of enhanced green fluorescent protein (EGFP). The shuttle plasmid was then transformed into E. coli BJ5183 with pAdEasy-1 vector by chemical transformation. The recombinant adenovirus vector pAd/IP-10 was identified by enzyme digestion with Pac I and the linearized plasmid was transfected into HEK293 cells.
RESULTSThe positive clones were identified with enzyme digestion and polymerase chain reaction (PCR) and were further verified by DNA sequencing. The recombinant adenovirus of high titration was obtained after transfection and packaging in HEK293 cells.
CONCLUSIONA recombinant adenovirus vector for expression of IP-10 has been constructed successfully and high-titer active adenovirus is obtained for functional study of IP-10 protein.

8.Transcriptome profiling and analysis of Panax japonicus var. major.

The rhizome of Panax japonicus var. major have been used as the natural medicinal agent by Chinese traditional doctors for more than thousand years. Most of the therapeutic effects of P. japonicus var. major had been reported due to the presence of tetracyclic or pentacyclic triterpene saponins. In this study, Illumina pair-end RNA-sequencing and de novo splicing were done in order to understand the pathway of triterpenoid saponins in this species. The valid reads data of 15. 6 Gb were obtained. The 62 240 unigenes were finally obtained by de novo splicing. After annotation, we discovered 19 unigenes involved in ginsenoside backbone biosynthesis. Additionally, 69 unigenes and 18 unigenes were predicted to have potential function of cytochrome P450 and UDP-glycosyltransferase based on the annotation results, which may encode enzymes responsible for ginsenoside backbone modification. This study provides global expressed datas for P. japonicus var. major, which will contribute significantly to further genome-wide research and analysis for this species.

9.Microwave radiation induces injury to GC-2spd cells.

OBJECTIVETo explore the impact of microwave radiation on GC-2spd cells.
METHODSWe exposed cultured GC-2spd cells to microwave radiation at the average power densities of 0, 10 and 30 mW/cm2 for 15 minutes and, from I to 24 hours after the exposure, we observed the changes in cell proliferation, histology and ultrastructure, cell apoptosis, and cAMP content by MTIT, light microscopy, electron microscopy, flow cytometry and ELISA.
RESULTSCompared with the control group, the GC-2spd cells showed a significant decrease in proliferation ability at 1 -24 hours after 10 and 30 mW/cm2 microwave radiation, except at 12 hours after 30 mW/cm2 radiation (P <0.05 or P <0.01), with reduced length and number of cell enation and increased intra cytoplasm vacuoles. The rate of cell apoptosis (%) was significantly increased in the 10 and 30 mW/cm2 groups at 6 hours (4.56 +/- 2.09 vs 14.59 +/- 1.09 and 8.48 +/- 1.73, P <0.05 or P <0.01) , with agglutination and margin translocation of chromatins and obvious dilation of endo cytoplasmic reticula. The cAMP content (nmol/g) in the GC-2spd cells was remarkably reduced in the 10 and 30 mW/cm2 groups at 6 and 24 hours (2.77 +/-0.24 vs 1.65+/- 0. 17 and 1.96+/-0.10, 3.02 +/-0.47 vs 2.13 +/-0.33 and 1.69 +/-0.27, P <0.05 or P <0.01).
CONCLUSIONMicrowave radiation at 10 and 30 mW/cm2 may cause injury to GC-2spd cells, which is manifested by decreased content of intracellular cAMP, reduced activity of cell proliferation, and increased rate of cell apoptosis.

Receptors play a crucial role in determining the host specificity and tissue tropism of virus. Foot-and-mouth disease virus(FMDV)has been showed to use four integrins, ?v?1, ?v?3, ?v?6 and ?v?8 as receptors to initiate infection and ?v?6 functions as the major receptor.The cDNA encoding bactrian camel integrin ?6 from the lung tissue was cloned and sequenced. The 2367bp cDNA of bactrian camel integrin ?6 encodes a polypeptide of 788 amino acids consisting of a 26-residue putative signal peptide, a 681-residue ectodomain with 8 potential N-linked glycosylation sites and 58 cysteine residues, a 29-residue transmembrane domain, and a 52-residue cytoplasmic domain with a NPLY motif and 1 potential N-linked glycosylation site. The nucleotide sequence similarity of integrin ?6 between bactrian camel and cattle, pig, sheep, human, mouse, Norway rat is 91.1%、91.8%、90.6%、90.5%、83.7%、84.1%, and the amino acid sequence similarity is 94.3%、93.4%、93.4%、93.7%、88.7%、88.6%, respectively. The bactrian camel ?6 gene exhibited the higher sequence homology with the ?6 gene of cattle, pig and sheep, indicating their close genetic relationships. It is possible that host tropism of FMDV may related to divergence in ?6 receptors among different species.
