2.The role of NgR - Rhoa - Rock signal pathway in retinal ganglion cell apoptosis of early diabetic rats
International Eye Science 2014;(9):1570-1573
To study the function and mechanism of the NgR-Rhoa- Rock signal pathways which exists in the retinal ganglion cells apoptosis in diabetes mellitus (DM) rats.
● METHODS: Some healthy SD rats were operated by means of single intraperitoneal injection of 1%streptozotocin based on the standard of 50mg/ kg wight, after that the blood sugar value was greater than 16. 7mmol/ L as DM model, then randomly divided into 3 groups, each group was 10 rats. ln addition to take 10 healthy SD rats as control group. Four groups of rats were bilaterally eyeball intravitreal injection in turn with NgR-siRNA virus 10μ L (siRNA group), NgR-siRNA virus diluted 10μ L ( DM group), NgR - siRNA virus - negative -control solution 10μ L (siRNA blank group), NgR- siRNA virus diluted 10μ L ( normal control group ), and fed normally. During that time, some life indexes like blood glucose, body mass, etc. were measured and recorded. After 12wk, the expression of NgR and Rhoa, HE staining, and TUNNEL staining were detected by Western blot analysis.
● RESULTS: Western blot analysis: compared with normal control group, the expression of NgR and Rhoa in DM group and siRNA blank group increased significantly (P<0. 01), while siRNA group was no significant change (P > 0. 05); compared with DM group and siRNA blank group, the expression of those proteins significantly lowered in siRNA group. HE staining: compared with normal control group, some extent ganglion cells arranged disorder, irregular shape, spacing not consistent were all found in three groups of model rats;compared with DM group and siRNA blank group, there was some improvement in siRNA group of ganglion cells about the order and shape size. TUNEL staining:compared with normal control group, there were retinal ganglion cells apoptosis in all of three groups of model rats. Compared with DM group and siRNA blank group, the number of retinal ganglion cells apoptotic cells was less, and the shape of cells had improved significantly in siRNA group.
●CONCLUSlON: ln the DM phase, the expression of NgR and Rhoa were up - regulation, the condition of diabetic retinal ganglion cell apoptosis was improved after that the NgR-Rhoa-Rock signal pathways had been inhibited.

Journal of Pharmaceutical Analysis 2001;13(1):30-32
Objective To investigate the urinary endothelin-1 (ET-1) excretion and urinary sodium excretion, microalbuminuria and ambulatory blood pressure(ABP) in salt-sensitive(SS) hypertension patients. Methods Twenty-one cases of normotensive subjects and 32 cases of uncomplicated hypertensive patients were recruited in this study. Salt sensitivity was determined by acute venous saline loading test. Before saline loading, 24-hour ABP measurements were performed. Urine samples were collected to assay ET-1 ,urinary sodium excretion and urinary albumin excretion(UAF). Results Compared to slat-resistant(SR) subgroup, SS showed low urinary ET-1 excretion in normotensive group (P<0.05) or hypertensive group (P<0.01) ,regardless of saline loading or not. The nighttime MAP of SS was higher than SR subgroup in normotensive or hypertensive group. Urinary sodium excretion during 4h of saline loading was significantly lower in SS than that in SR hypertensive patients (P<0. 05). Twenty-four-hour UAE of SS patients was higher than SR group (P<0.01). Results of further correlation analysis indicated that the urinary ET-1 excretion was positively related to urinary sodium content and negatively to ABP and UAE. Conclusion Urinary ET-1 is low in SS normotensives or hypertension patients, which may play a role in renal sodium retention and renal impairment of SS hypertension patients.

Objective:To explore the correlation between white matter lesions (WML) at different locations and cognitive function. Methods:The cognitive function of 68 patients with WML in Beijing Electric Power Hospital from January 2019 to May 2020 were assessed with Montreal Cognitive Assessment Scale (MoCA), and the extent of WML were assessed with age-related white matter changes rating scale(ARWMCRs). Correlation between the ARWMCRs scores of different locations and the scores of all locations and the scores of cognitive function, as well as the relationship between the ARWMCRs scores of different locations and each subitem of cognitive function were analyzed. Results:In 68 patients with WML, the degree of lesions in different parts from heavy to light was frontal area, parietooccipital area, basal ganglia area and temporal area, and no white matter lesions were found in infratentorial area. Except the basal ganglia region, the ARWMCRs scores in other regions and the total score were negatively correlated with MoCA score, among which, the ARWMCRs score in left frontal area, right frontal area, left parietal pillow and right parietal pillow region had the strongest correlation with MoCA score( rs=- 0.388, - 0.479, -0.418, -0.502, all P<0.01).In WML patients, the left frontal area was negatively correlated with language, abstraction and orientation ( rs=- 0.479, - 0.324, - 0.288, all P<0.01).The ARWMCRs score in right frontal area was negatively correlated with visual spatial and executive function, memory, language, abstraction, and orientation ( rs=- 0.324, -0.272, -0.459, -0.382, -0.352, all P<0.05). The ARWMCRs score in left parietal occipital region was negatively correlated with visual spatial and executive function and memory ( rs =-0.287、- 0.270, all P<0.05). The ARWMCRs score in right parietal occipital region was negatively correlated with memory, language, abstraction and orientation ( rs=-0.366, -0.289, -0.260, - 0.307, all P<0.05).The ARWMCRs score in left temporal region was negatively correlated with language, abstraction and orientation ( rs=- 0.248, -0.372, - 0.608, all P<0.05).The ARWMCRs score in right temporal region was negatively correlated with abstraction and orientation ( rs=- 0.525, -0.465, all P<0.01).The ARWMCRs score in right basal ganglia was negatively correlated with orientation and memory ( rs=- 0.455, - 0.275, all P<0.01), while The ARWMCRs score in left basal ganglia was not correlated with MoCA subitems. Conclusions:The frontal area and parietal occipital area are the most serious lesions in WML patients, and the lesions in these two areas also cause the widest range of cognitive impairment.
