1.Effect of body position intervention on sleep quality of patients undergoing video-assisted thoracic surgery

Objective To explore the body position intervention on the sleep quality of patients undergoing video-assisted thoracic surgery (VATS).Methods Totals of 148 patients were randomly divided into control group (n =75) and observation group (n =73),control group received the routine nursing,while observation group received special body position intervention nursing.Then,sleep quality of two groups in one week before the surgery and three days after the surgery were compared.Results No significant difference was found in the score of sleep quality of VATS patients between two groups before one week of thoracic surgery (t =0.8623,P>0.05).Three days after the thoracic surgery,the score of sleep quality of observation group was statistically higher than that of control group [(9.82 ± 2.81) vs (13.29 ± 3.32),t =6.8 591,P < 0.05)].Conclusions Body postural intervention can promote to improve sleep quality of VATS patients after thoracic surgery,also promote the recovery of the patients,and improve nursing quality.
2.Determination the concentration of antofloxacin in human plasma by LC-MS /MS and its application in the pharmacokinetic study of antofloxacin hydrochloride

Objective To develop a LC-MS/MS method for the determination of antofloxacin in human plasma and to study the pharmacokinetic characteristics of antofloxacin after oral administration of antofloxacin hydrochloride tablets.Methods The samples were precipitated by acetonitrile and ornidazole was selected as the internal standard.The separation was carried on a Agilent Poroshell 120 SB-C18(2.1 mm × 50 mm,2.7 μm) column with a mobile phase of acetonitrile:5 mmol · L-1 ammonium formate (containing 0.1% formic acid) =12∶88 at a flow of 0.4 mL · min-1 12 healthy subjects were given orally single dose of antofloxacin hydrochloride 400 mg dose,blood samples were collected at different time points,the concentration of antofloxacin in human plasma was measured,and the main pharmacokinetic parameters were calculated by DAS 2.1.1 and Excel 2003.Results All the analytes showed good linearity over their concentration ranges on 0.01-15.00 μg · mL-1 The accuracy,inter-run and intra-run precisions were all below 15%.Analytes were stable during the study.The main pharmacokinetic parameters after antofloxacin 400 mg single drug administration:Cmax was (3.57 ±0.54) μg · mL-1,tmaxwas (2.25 ±0.92) h,t1/2 was (11.76 ± 1.64) h,AUC0-1 was (60.38 ± 7.47)μg · mL-1 · h.Conclusion This method can be used to determine the concentration of antofloxacin in human plasma and to study the pharmacokinetics of antofloxacin hydrochloride tablets.
3.Dissection of seed development of Gastrodia elata at different temperatures.

The study is aimed to create seed materials and dissect the molecular mechanism of sexual propagation of Gastrodia elata. In this research, thirteen characteristics of flowers, flower stem, fruits, seeds and embryo of G.elata f. glauca and G.elata f. elata after bolting at room temperature(RT) and constant temperature(CT, 22 ℃) were determined. It was found that the constant temperature condition could prolong the bolting duration of G.elata and increased the number of flowers, while the variety of G.elata only affected the bolting duration, but had no effect on the number of flowers, and the G.elata f. elata was more likely to bolting than the G.elata f. glauca. The variety of G.elata was the main factor affecting the time of dehiscent fruit of G.elata, the temperature was the main factor affecting the fruits number and fruits diameter, and the constant temperature was more conducive to the fruits shape of G.elata than the room temperature. There was no significant difference in seed phenotype of G.elata varieties, but the seed embryo of G.elata seeds cultivated at constant temperature was fuller than that of G.elata cultivated at room temperature, and temperature had less influence on the seed phenotype of G.elata. But it was interesting to find that temperature and varieties had greater influence on the seed embryo of G.elata, constant temperature cultivation was more conducive to the formation of seed embryo of G.elata, and more the seed embryo of G.elata f. elata was easier to form than the seed embryo of G.elata f. glauca. However, the development of seeds and embryos of G.elata was significantly affected, and the development of seeds and embryos of G.elata f. glauca was more sensitive to temperature than G.elata f. elata. The research suggested that it is advisable for G.elata to produce seed materials by bolting at constant temperature(22 ℃).

4.Age estimation using content of sjTREC in human peripheral blood.

OBJECTIVE:
To determine and verify the correlation formula of age estimation using the content of signal joint T-cell receptor excision DNA circle (sjTREC) in human peripheral blood and to discuss its application value in forensic biological practice.
METHODS:
The samples of peripheral blood stains were collected from 30 healthy unrelated individuals whose ages were known. The DNAs were extracted from the samples stored at room temperature after 4 weeks. The content of sjTREC was measured by real-time fluorescent quantitative PCR technique, and the TATA box binding protein (TBP) was selected as reference genes. The age of each sample was predicted with the formula which was Age = -7.181 5 Y-42.458 +/- 9.42 (Y = dCtTBP-sjTREC), and the result was compared with the real age of each individual to determine the accuracy of the formula.
RESULTS:
sjTREC and TBP gene were detectable in all 30 samples of peripheral blood. The contents of sjTREC in human peripheral blood showed a decreasing tendency with aging. The accuracy rate for the age estimation by this method was 76.67%.
CONCLUSION
The method for the age estimation with the content of sjTREC was simple, fast, sensitive, and good species specific with important potential application prospect.
