Chinese Journal of Applied Physiology 2012;28(3):205-208
OBJECTIVETo study the influence of 14- day simulated weightlessness on emotional behaviour in rats.
METHODSSixteen from twenty male SD rats were selected and assigned to a 2-group design: the control group and the tail suspension (TS) group (n = 8). The essay deployed typical methods for assessing emotional activity in the current, including food-intake and body weight variation, open field testing, sucrose preference testing and the evaluation of emotional arousal level to test emotional behavior.
RESULTS1. After 14- day simulated weightlessness for rats, their food-intake and body weight increase were less than those in the control group. 2. The ability of movement reduced in rats, the number of locomotion was significantly lower than that in the control group (P < 0.05) and the frequency of self-grooming was significantly higher than that in the control group(P <0.01. 3. A higher level of emotionality in TS group was recorded. 4. Sucrose preference was not observed in TS group.
CONCLUSIONIt demonstrated depression, anxiety and nervous symptom occurred in the TS rats with a certain degree of nervous reaction but no anhedonia.

OBJECTIVETo observe the relation between Pi deficiency syndrome (PDS) and the configuration and functions of extensor digitorum longus (EDL)and soleus (SOL).
METHODSTotally 36 ICR mice were randomly divided into 3 groups according to weight matching principle, the control group, the exhausted group, and the rhubarb group, 12 in each group. Two PDS models were established by either purgation with rhubarb diarrhea (as Group A) or exhausted swimming plus sleep deprivation (as Group B).The cross sectional area (CSA) of type I and II fibers of extensor digitorum longus (EDL) and soleus (SOL), relative proportions of type I and II fibers were measured by m-ATPase histochemical method. The isotonic contraction and the maximum tetanus contraction of EDL and SOL were detected by PowerLab system.
RESULTSCompared with the control group, the body weight, body temperature, and the general health condition of PDS model rats obviously decreased; the spleen index and the thymus index were also lower; the maximal isotonic contraction and the maximum tetanus contraction obviously decreased; the cross section areas of EDL and SOL were reduced with loosely arranged cells. In EDL, the proportion of type I fibers was added and the proportion of type II fibers was lowered. In SOL, there was no change in the proportion of type I and type II fibers.
CONCLUSIONSEDL and SOL were obviously atrophied in the two PDS model mice. The type I fibers of SOL was more significantly atrophied in Group B.

Objective To observe the immediate hemodynamic effects of the intercoronary ischemic preconditioning on the coronary perfusion pressure. Methods The observational study recruited 17 patients who were hospitalized for stable coronary disease and had severe stenosis lesions (70% ~ 85% ) in one or two vessels per coronary angiography. They were randomized into ICPC ( n =8 ) group ( receiving two cycles of 1-min balloon inflation and 5-min reperfusion) and control group (n =9).Before interventional treatment, the ICPC group was given 2 cycles of intercomary ischemic preconditioning. The occlusive and non-occlusive pressures proximal and distal to the stenosis were collected before and after ICPC.Fractional flow reserve (FFR) was calculated upon the following equation:FFR =Pd/Pa ( Pa:hyperemic mean aortic pressure,Pd:hyperemic coronary pressure distal to the stenosis). Results Before and after ischemic preconditioning, coronary wedge pressure and FFR of ICPC group were significantly increased ( coronary wedge pressure was from [ 22.08 ± 19.14 ]mm Hg to [25.46 ±19.04]mm Hg,P=0.011;FFR.was from [70.30±16.05]% to [77.53 ±13.42]%,P=0.001).The collateral flow was increased significantly.Coronary wedge pressure and FFR of control group did not improved obviously. There were significant differences in FFRs and coronary wedge pressures between ICPC and control groups. Conclusion Coronary ischemic preconditioning can improve coronary perfusion pressure, and rapidly increase the coronary pressure distal to the severe stenosis lesions.

Aim To develop the in vitro and in vivo models induced by shrimp tropomyosin( ST) and mono-clonal tropomyosin-specific murine IgE antibody ( anti-ST-IgE mAb) . Methods ST was purified from Metap-enaeusensis by an isoelectric precipitation method. The anti-ST-IgE mAb was obtained from hybridomas. After RBL-2 H3 cells were sensitized with anti-ST-IgE mAb and challenged with ST,β-hexosaminidase release was determined. Passive systemic anaphylaxis ( PSA ) was induced in mice and the rectal temperature was recor-ded after ST challenge within 30 min by a thermal probe. Results A significant increase ofβ-hexosamin-idase was observed in sensitized cells after ST chal-lenge. The average temperature drop after ST challenge was 1. 44℃ in PSA mice within 30 min. Conclusion The in vitro and in vivo models induced by ST and anti-ST-IgE mAb are established as an improvement of pres-ent models of type Ⅰ allergy.

Objective:To develop murine models of Th2 response induced by shrimp tropomyosin (ST). Methods:Mice were sensitized with ST for 6 weeks. The serum antigen-special IgE (sIgE),total IgE and sIgG level,Th1/Th2 cytokines production were measured by ELISA. The basophil activation in mice was measured by flow cytometry. Results:The intraperitoneal sensitization with ST for 6 weeks induced significant increase of serum sIgE,total IgE and sIgG (sIgG1,sIgG2a and sIgG2b) level in mice. Th2 cell response was induced and cytokines (IL-4,IL-5,IL-10 and IL-13) production increased in splenocytes stimulated by ST,while Th1 cytokine (IFN-γ) production decreased. As the markers of basophil activation,CD200R and CD41 expression also increased in response to ST. Conclusion:The Th2 response is dominant in ST-induced anaphylaxis in mice.
