1.Surgical treatment of Helveston syndrome: two cases report

·AIM: To report two patients with Helveston syndrome who received surgical treatment.·METHODS: During last year in department of oph-thalmology in West China Hospital, the surgery was designed to correct A-exotropia by lateral rectus' weaken and vertical offsets, and dissociated vertical deviation (DVD) by superior rectus muscles recession simultaneously.·RESULTS: A-exotropia and DVD of the patients vanished six months after the surgery. Both patients were clinically cured.·CONCLUSION: As the degree of horizontal and vertical strabismus was in the allowed range, complete examination and full assessment of the function of the four muscles were performed before operation, single operation can be considered in the treatment of Helveston syndrome.
5.Analysis of characteristics and novelty of researches in clinical medicine based on scientific literature novelty assessment files

Objective To improve the scientific literature novelty assessment by studying the characteristics and novelty of researches in clinical medicine.Methods The objects, levels, locations, methods and novelty of 380 scientific literature novelty assessment files in Library of Guilin Medical College from 2012 to 2014 were analyzed by chi-square test.Results The novelty of researches was mainly manifested as an effect index in 2012-2013 and as an in-tervention factor in 2014(x2=110.12, P<0.01).The novelty of effect index was mainly manifested as a compre-hensive index in 2012-2013 and as a sensitivity index in 2014(x2=44.10, P<0.01).Conclusion Understanding and keeping abreast of the characteristics and novelty of researches in clinical medicine can serve them better and more effectively encourage their innovation .
6.Gene expression of AAV-ITR ssDNA mini vector in skeletal muscle of mice.

AAV-ITR single strand DNA mini vector (AAV-ITR ssDNA mini vector) is a novel gene expression vector based on AAV-ITR. We have shown efficient gene expression of AAV-ITR ssDNA mini vector in HEK 293T. Here, we studied the efficacy of gene expression of AAV-ITR ssDNA mini vector in vivo. We injected the skeletal muscle of ICR mice separately with equal molars of AAV-ITR ssDNA mini vector, ITR mutated AAV-ITR single strand DNA mini vector (AAV-ITRmm ssDNA mutant vector), AAV-ITR dsDNA and pUC57-minivector-GFP, combined with TurboFect. Florescence microscope analysis of skeletal muscle section shows that AAV-ITR ssDNA mini vector had higher expression efficiency and longer expression period. We extracted DNA from the muscle three months after injection and quantified three vectors by Real-time PCR. RT-PCR analysis shows that there were highest copy numbers of AAV-ITR ssDNA mini vector existing in muscle. Stable existing of AAV- TR ssDNA mini vector in muscle could be the molecular basis of long term gene expression of the vector. The results suggest that AAV-ITR ssDNA mini vector might be a promising vector for gene therapy.

7.Preliminary study on inhibitory effects of sub-MIC matrine alone or in combination with erythromycin On Staphylococcus epidermidis biofilm

Objective To observe the inhibitory effects of sub-MIC matrine alone and in combination with erythromycin on Staphylococcus epidermidis biofilms and their influences on morphological changes of the biofilms.Methods Minimum inhibitory concentrations (MIC) of matrine and erythromycin against Staphylococcus epidermidis were determined by the serial dilution method,antibacterial activity of matrine combined with erythromycin against planktonic S.epidermidis was evaluated by the checkerboard method.S.epidermidis biofilms were constructed in vitro,XTT reduction assay was used to evaluate influences of sub-MIC matrine alone and in combination with erythromycin on metabolism and adhesion of S.epidermidis biofilms,and scanning electronic microscope(SEM) was applied to observe the morphological and the structural changes of the biofilms.Results The MIC of erythromycin to S.epidermidis was 7.8125 μg/ml,while the MIC of matrine was greater than 1000 μg/ml,besides,a synergistic effect between erythronmycin and matrine on planktonic S.epidermidis was shown (FIC<0.5).The sub-MIC matrine had no significant inhibitory effect on adhesion of S.epidermidis,and also the combination of the two agents was better than was used alone.However,the sub-MIC matrine had inhibitory effects on metabolism and morphology of S.epidermidis biofilms,and the combination of the two agents was weaker than was used alone.Conclusion Both the sub-MIC matrine and erythromycin had a significant inhibitory effect on S.epidermidis biofilm formation.Combination of the two agents showed synergistic effects on plankton and adhesion of S.epidermidis,but showed no synergistic effect on metabolism and morphology of the biofilms.
8.Effects of aging on midgastric transverse band and gastric emptying

Objectives To explore the effects of aging on midgastric transverse band(MTB)and gastric emptying.Methods In our prospective study,57 healthy volunteer were divided into young, middle and old-age groups.After taking test meal labeled by 99mTc-iethylenetriaminepentaacetic acid(99mTc-DTPA), the pictures were collected using double probe single photon emission computed tomography(SPECT).Stomach in each frame of the pictures was divided into proximal, midgastric transverse band(MTB)and distal parts.And half gastric emptying time,gastric remnant rate at 90 min, areas of different gastric parts were tested and calculated respectively.Results Half gastric emptying time of whole stomach was(43.24± 11.87)min,(42.07 ± + 9.31)min and(45.81 ± 10.73)min in young,middle and old-age groups, respectively, with P>0.05.Among young, middle and oldage groups, half gastric emptying time was(38.09 ± 10.26)min,(37.33 ± 9.28)min and(26.74 ± 12.07)min in proximal stomach, and it was(38.35 ± 12.96)min,(37.73 ± 7.46)min and(46.41 ± 10.74)min in distal stomach,respectively.The half gastric emptying time was significantly reduced in proximal stomach and increased in distal stomach in old-age group(both P<0.05).The gastric nuclide remnant rate at 90min in total stomach was(30.38 ± 19.32)%,(29.03 ± 10.36)% and(31.92 ± 13.47)% ,in young, middle and old-age groups, respectively, with P>0.05.This rate in proximal stomach was(25.01 ± 12.35)%,(26.36± 15.29)% and(19.54±8.47)% among three group, respectively.The rate in proximal stomach was(42.25 ± 12.36)%,(41.56 ±± 9.33)% and(56.05 ± 11.72)% among three group,respectively.The gastric remnant rate at 90min was reduced in proximal stomach and was increased in distal stomach significantly in old-age group(both P<0.05).Compared with young and middle-age group,the old-age group showed no difference in areas of total stomach in all the time, while the areas were reduced in proximal stomach and increased in distal stomach significantly from 30 min to 90 min(all P<0.05).Total stomach versus proximal and distal stomach showed no difference in count/pixel ratio in all time,while a count/pixel ratio was reduced in proximal stomach and increased in distal stomach significantly from 30 min to 90 min(both P< 0.05).Both areas and count/pixel ratio of MTB at 60 min and 90 min were significantly increased in old-age group(both P<0.05).Conclusions The total gastric emptying is not delayed along with aging, while the gastric emptying is increased in proximal stomach and reduced in distal stomach in the elderly.This abnormity of intragastric distribution of food might be related with larger area of midgastric transverse band.
9.The HA Gene of H5N1 Avian Influenza Virus Expressed in Insect Cells and Its Biological Activity

The 1.7 kb full-length hemagglutinin (HA) gene fragment of H5N1 subtype avian influenza virus was amplified by RT-PCR and then cloned into the pFastBacHT donor plasmids. The recombinant plasmid pFastBac-H5 was identified by restriction enzyme digestion and sequencing. Following the transposition pFastBac-H5 into the bacmid in DH10Bac E.coli competent cells, the colonies were identified by blue and white selection. The recombinant bacmid (rBacmid-H5) was verified by PCR analysis. Transfection of rBacmid-H5 DNA into sf9 cells using Cellfectin reagent results in the production of recombinant viral stock. Cells were harvested 72h post infection and analyzed by SDS-PAGE, Western-blot, hemagglutination and hemagglutination inhibition test;the expressed HA protein (rH5)shows hemaggluting activity and can be inhibited by H5N1 virus immunized chicken sera. On the other hand, immunization of chickens with rH5 protein results in high titers of H5N1 virus specific hemagglutation inhibition antibodies, which proved its biological activity.
