1.Efficacy of mometasone furoate aqueous nasal spray combined with loratadine in pediatric allergic rhinitis and its effects on IL-10, IL-17

Objective To study the curative efficacy of mometasone furoate aqueous nasal spray combined with loratadine in the treatment of pediatric allergic rhinitis, and its effects on IL-10, IL-17 in serum.Methods 126 patients of pediatric allergic rhinitis who were admitted in our hospital from May 2011 to September 2013 were selected as research objects and randomly divided into treatment group and control group, 63 patients in each group.The control group were treated with loratadine, while the treatment group were treated with mometasone furoate aqueous nasal spray in combination with loratadine.The curative efficacy, scores of clinical symptoms and nasal examination, curing time, medication time and serum IL-10, IL-17 levels were compared between the two groups.Results The total therapeutic efficacy ratio in the treatment group was 98.41%, which was statistically higher than that of 80.95% in the control group (P<0.05).5 weeks after the treatment, scores of sneezing, runny nose, nasal congestion, nasal itching, eye itching and nasal examination in the treatment group were statistically lower than those in the control group (P<0.05).The curing time and medication time in the observation group were statistically shorter than those in the control group ( P <0.05 ) .The inflammatory factors, in comparison with the control group after treatment, the treatment group had statistically higher level of serum IL-10 and lower level of serum IL-17 ( P <0.05 ) . Conclusion Mometasone furoate aqueous nasal spray in combination with loratadine is effective for pediatric allergic rhinitis with a high therapeutic efficacy ratio.And it could significantly improve clinical symptoms, shorten treatment time, increase level of IL-10 and reduce level of IL-17.
2.Effects of NF-κB/p65 siRNA on Apoptosis and Bax Expression in Lung Tumour Cell Xenografts of Nude Mouse

Objective To investigate the effects of nuclear factor-kappaB(NF-κB)/p65 signaling transduction on the apoptosis and expressions of apoptosis-related genes Bax in nude mouse lung tumour cell xenografts. Methods The nude mice Lewis lung carcinoma cell xenograft model was established,and then the mice were intraperitoneally injected with NF-κB/p65 small interfering RNA(siRNA). The apoptosis of xenografted tumor cells in nude mice was detected by TUNEL assay. Expressions of Bax mRNA and protein were detected by RT-PCR and Western blotting. Results The result of TUNEL assay demonstrated that p65 siRNA evidently evoked cell apoptosis. Compared to the PBS treatment group or the normal control mice,both mRNA and protein expression levels of Bax in tumor xenografts were significantly up-regulated. There were significant differences among three groups(P<0.05). Conclusion NF-κB/p65 subunit may play an essential role in cell apoptosis of Lewis lung tumor.

4.The expression and clinical significance of MKK4 in osteosarcoma

Objective To explore the clinical significance of expression of MKK4 in osteosarcoma,and to discuss the relationship between MKK4 and osteosarcoma occurrence,development and prognosis.Methods The expression of MKK4 protein was determined with immunohistochemical SP method in 30 cases of osteosarcoma.Results The MKK4 was obviously expressed in both nucleus and cytoplasm in osteosarcoma.The expression of MKK4 in the cytoplasm of osteosarcoma was positively correlated with clinical stage (r =0.72,P =0.000),the expression of MKK4 in the nucleus of osteosarcoma had no correlation with clinical stage (r =0.44,P =0.080).MKK4 expression in the cytoplasm was higher in transfer osteosarcoma than no transfer osteosarcoma(x2 =9.69,P <0.01),while no obvious difference in the nucleus of osteosarcoma(x2 =1.00,P =1.000).Conclusion The MKK4 is highly expressed in the nucleus and cytoplasm of osteosarcoma,suggests that MKK4 may be related with the carcinogenesis development of osteosarcoma.The expression of MKK4 in the cytoplasm of osteosarcoma is related to the malignant,metastasis and prognosis of osteosarcoma,may be used as a potential prognostic indicator of judgment osteosarcoma.
5.Variability of Pyramidal Neurons in Hippocampal CA1 Area with Time after Heat Stroke in Rats

Objective To observe the pyramidal neurons in the hippocampal CA1 area varying with time after heat stroke in rats. Meth-ods Sprague-Dawley rats were divided into sham group (n=5) and heat stroke group (n=14), and the heat stroke group was divided into 7-day subgroup and 21-day subgroup (n=7 at each group) after heat stroke. Heat stroke group was established model of heat stroke. The brain tissues of rats were observed with Nissl staining to count the living pyramidal neurons in hippocampal CA1 area. Results The number of living pyramidal neurons in hippocampal CA1 area decreased in the heat stroke group (F=11.80, P<0.01), and decreased more in the 21-day subgroup than in the 7-day subgroup (P<0.05). Conclusion Pyramidal neurons in hippocampal CA1 area decrease with time after heat stroke, which may be associated with the learning and memory impairment.
6.Expression of MT-1 and MT-2 Genes in Liver of Chronic Exposure to Inorganic Arsenate Mice

Objective To investigate the effect of MT-1 and MT-2 in liver damage in chronic inorganic arsenate exposure mice and to explore the mechanism of arsenic-induced liver damage. Methods The male Kunming mice were randomly divided into control and exposed groups. The control group was given ordinary feed and tap water. The exposed group was given ordinary feed and 300 mg/L of sodium arsenite solution by drinking water. After 10 months of treatment, the activity of serum alanine aminotransferase (ALT), aspartate aminotransferase(AST) and globulin (Glb) content were determined. The total RNA was extracted by the TRIzol-phenol-chlorofor-method from the liver tissue. The quantity of the RNA was determined by spectrophotometry and its purity was judged at a ratio of A260/A280. Then real-time PCR(RT-PCR) was used to measure the mRNA expression of MT-1 and MT-2. Objective To discuss the value of high frequency ultrasound in assessment the changes of knee,ankle and elbow joints with haemophiliac arthropathy in children.Methods Eighty joints of 42 haemophiliac patients were enrolled in this study.Sixty two joints of 29 patients were examined by X-ray at the same time.The correlation of ultrasonography and X-ray on bone aspects were compared.Results Among 80 joints of 42 haemophilic patients,soft tissue swelling was showed in 12 joints,synovial hypertrophy was in 48 joints,the thickness range of synovial membrane was from 2.01 to 11.12 mm [(4.71± 2.26)mm],increased blood flow was in 40 joints,effusion in 53 joints,haemosiderin was in 33 joints,bone erosion was in 36 joints,subchondral cyst formation was in 20 joints,cartilage damage or disappear was in 31 joints.Ultrasonograthy showed bone erosion in 29 joints,subchondral cyst formation was in 16 joints,cartilage damage was in 24 joints.X-ray showed bone erosion in 23 joints,subchondral cyst was in 16 joints,narrowing joint space was in 20 joints.There was high correlation between the osteochondral domain of the ultrasonography and X ray:Kappa value were 0.803,0.832,0.790 respectively(P <0.05).Conclusions Ultrasonography is sensitive to detect the signs of enrolled joints in hemophiliac patients.It is an important method on diagnosis for hemophiliac arthrophv.
8.Role of PI3K/Akt signaling pathway in reduction of hypoxia-reoxygenation-induced injury to cardiomyocytes by propofol postconditioning in rats

Objective To evaluate the role of Phosphatidylinositol 3-kinase/protein-serine-threonine kinases (PI3K/Akt) signaling pathway in reduction of hypoxia-reoxygenation (H/R)-induced injury to cardiomyocytes by propofol postconditioning in rats.Methods Primary cardiomyocytes were obtained from neonatal rats aged 1-3 days and cultured in DMEM culture medium.The cells were seeded in 96-well plates (density 1 × 105/ml,200 μl/well) or 6-well plates (density 5 × 105/ml,2 ml/well) and randomly assigned into 4 groups (n =24 each):control group (C group),H/R group,H/R + propofol group (H/R + P group) and H/R + propofol + wortmannin (a specific PI3K inhibitor) group (H/R + P + W group).The cells were routinely cultured for 6 h in group C.The cells were exposed to 2 h hypoxia followed by 4 h reoxygenation.Propofol with the final concentration of 50 μmol/L was added to the culture medium at the end of hypoxia in group H/R + P.Wortmannin (final concentration 100 nmol/L) and propofol (final concentration 50 μmol/L) was added to the culture medium at the end of hypoxia in group H/R + P + W.At the end of reoxygenation,the cell viability was measured by MTT assay,the lactic dehydrogenase (LDH) activity was detected in the culture medium,the cell apoptosis was assessed by flow cytometry,and the expression of phosphorylated Akt (p-Akt),Bcl-2 and Bax in cardiomyocytes were determined by Western blot.The apoptotic rate and Bcl-2/Bax ratio were calculated.Results Compared with C group,the cellviability was significantly decreased,the LDH activity and apoptotic rate were increased,p-Akt and Bax expression was up-regulated and Bcl-2 expression was down-regulated in H/R group (P < 0.05).Compared with H/R group,the cell viability and Bcl-2/Bax ratio were significantly increased,the LDH activity and apoptotic rate were decreased,p-Akt and Bcl-2 expression was up-regulated and Bax expression was down-regulated in H/R + P group (P < 0.05).Compared with H/R + P group,the cell viability and Bcl-2/Bax ratio were significantly decreased,the LDH activity and apoptotic rate were increased and p-Akt and Bel-2 expression was down-regulated in H/R +P + W group (P < 0.05).Conclusion The mechanism by which propfol postconditioning attenuates H/R-induced injury to cardiomyocytes is related to the activation of PI3K/Akt signaling pathway.
9.Determination of Plasma Concentration of Magnesium Isoglycyrrhizinate in Patients Underwent Liver Re-section by HPLC

OBJECTIVE:To establish the method for the determination of plasma concentration of magnesium isoglycyrrhiz-inate in portal vein and peripheral venous blood of patients underwent liver resection,to further validate and evaluate pharmacoki-netic characteristics,rational and safe use of drugs in the clinic. METHODS:31 patients underwent liver resection in our hospital during Oct. 2014-Mar. 2015 were given magnesium isoglycyrrhizinate intravenously at the beginning of surgery. Portal vein and pe-ripheral venous blood of patients were drawn at 1 hour after drug use,and HPLC-UV detection method was used to determine the plasma concentration of drug. RESULTS:The retention time of isoglycyrrhizinate magnesium was 4.5 min,which showed a good peak shape,and was not interfered with the determination by plasma endogenous peak. The plasma concentration ranged from 0.55 to 55.00 mg/L. The minimum quantitative concentration was 0.55 mg/L. The extraction recoveries were 84.7%-87.1%,and method recoveries were 101.2%-105.4%,and RSDs of intra-day and inter-day were less than 6%. Plasma concentration of magnesium iso-glycyrrhizinate in portal vein blood was significantly higher than in peripheral vein blood of patients underwent liver resection (close to 2 times);and plasma concentration was not affected by primary liver diseases and underlying diseases such as cirrhosis. CONCLUSIONS:The method is simple and has high recovery rate of extraction,high accuracy and high sensitivity. It can meet the needs of pharmacokinetic study. After the application of magnesium isoglycyrrhizinate during liver resection,there is higher blood concentration of magnesium isoglycyrrhizinate in portal vein,which is beneficial to protect liver cells and improve liver func-tion. It is suitable during perioperative period of liver.
