1. Role of PTCH1 gene hypermethylation in tumorigenesis of gastric cancer

Objective: To study the effect of PTCH1 methylation on gastric carcinogenesis and the therapeutic effect of methylation inhibitor, 5-aza-2′-deoxyeytidine (5-aza-dC), for treatment of gastric cancer. Methods: The total RNAs were extracted from 10 gastric cancer tissues, their corresponding adjacent normal tissues, and gastric cancer cell line AGS. The PTCH1 mRNA expression was detected by Quantitative real-time PCR (QRT-PCR) and the methylation of the promoter was examined by methylation specific PCR (MSP). AGS cells were treated by 5-Aza-dC; the cell cycle and apoptosis were examined by flow cytometry, and the methylation level was also observed. Results: PTCH1 expression was negatively correlated with promoter methylation in gastric cancer tissues, their corresponding adjacent normal tissues, and gastric cancer cell line AGS (r = -0.591, P = 0.006). 5-Aza-dC treatment caused apoptosis and G 0/G1 phase arrest of AGS cells, and also induced demethylation of PTCH1 and increased its expression. Conclusion: Hypermethylation of PTCH1 gene promoter region is one of the main causes of low PTCH1 expression in AGS cells. Demethylation agent 5-Aza-dC can reverse this methylation status of PTCH1 and regulate the expression of PTCH1, suggesting a role for it in gastric cancer treatment.
2.The effects of sinter temperature on the crystalline phase and microstructural evolution of lithium disilicate glass-ceramic

Objective:To investigate the influence of sinter temperature alteration on the crystalline phase and microstructure of lithium disilicate glass-ceramic.Methods:Samples of lithium disilicate glass-ceramic were heated using the standard two stage heating-schedule at different final temperatures.All samples were observed by X-ray diffractometer (XRD),field emission scanning electron microscope(FESEM) and energy-dispersive X-ray spectroscopy(FE-SEM-EDS) for the evaluation of crystalline phase and microstructural evolution.Results:Crystalline phase and microstructure of ceramic samples showed significant changes during processing at different temperatures,especially at 800 ℃.Samples sintered at 840 ℃ displayed higher crystallinity(P<0.05) and denser microstructure.Conclusion:Temperature control is important in ceramic sintering.800 ℃ and 840 ℃ are the critical temperature points in the crystalline phase and microstructural evolution of lithium disilicate glass-ceramic.
3.Comparison of efficacy and adverse reactions of oxaliplatin combined with gemcitabine and S-1 combined with cisplatin for advanced three negative breast cancer

Objective To investigate the efficacy and adverse reactions of SOX and GP combined with two chemotherapy methods.Methods From January 2014 to September 2016 came to our hospital for treatment of three advanced breast cancer patients with negative 84 cases as the research object of observation,were randomly divided into SOX group and GP group were given chemotherapy measures and clinical observation on the treatment of.Results Two groups of patients with CR,PR,SD,PD,the number of cases and the total effective rate comparison,P values were 0.693,0.637,0649,0.801,0.823; two groups of patients with diarrhea,liver damage,thrombocytopenia,nausea and vomiting,neurotoxicity,anemia,leukopenia,rash,renal toxicity,appetite the incidence rate of decline,P values were 0.374,0.629,0.643,0.374,0.645,0.434,0.450,0.693,0.645,0.815.Conclusion Standard chemotherapy regimens for advanced three negative breast cancer was not uniform,but the SOX and GP regimens are safe and effective,can be used for clinical treatment.
4.Clinical observation of raltitrexed combined with irinotecan as first-line chemotherapy for recurrent or metastatic gastric cancer

Objective To observe the efficacy and adverse reaction of raltitrexed combined with irino-tecan as first-line chemotherapy for recurrent or metastatic gastric cancer.Methods From January 201 4 to March 201 5,39 patients of recurrent or metastatic gastric cancer who received treatment in the First People′s Hospital of Zhangjiagang of Jiangsu Province were collected.All patients received raltitrexed (3.0 mg/m2 , 1 5 min intravenous drip)on the first day and irinotecan (1 80.0 mg/m2 ,90 min intravenous drip)on the first day.One cycle lasted 21 days.The efficacies were evaluated every 2-cycle.Adverse reactions were evaluated every cycle.Results The efficacies and adverse reactions could be evaluated in 39 patients.The study received 0 complete remission,1 6 partial remission,1 1 stable disease,1 2 progression disease.The objective response rate was 41 .03% (1 6 /39).The disease control rate was 69.23% (27 /39).The median overall sur-vival time was 9.3 months (95%CI:8.8-1 1 .1 months).The median progression-free survival time was 6.0 months (95%CI:5.1 -6.8 months).The adverse reactions were mainly neutropenia,anemia,liver dysfunc-tion,the incidence of them were 35.90%,33.33% and 28.21 % respectively.Conlusion Raltitrexed com-bined with irinotecan as first-line chemotherapy for recurrent or metastatic gastric cancer acquires an definite efficacy,and the adverse reactions can be tolerated,which is worthy of further clinical research.
5.Comparative Study of Environmental Risk Factors for Han and Uyghur Children with Dyslexia

Objective To investigate differences between Han and Uyghur children in dyslexia prevalence and potential environmental risk factors as well as to provide diagnosis and treatment evidence for dyslexia children . Methods We used cluster sampling to recruit 2 854 students in grades 3~6 from five Uyghur -Chinese bilingual primary schools in Xinjiang province .The children with dyslexia were selected step by step according to the defini‐tion of ICD-10 and DSM -IV .The children with DD and children without DD were selected and compared by 1∶1 of the same class ,ages and genders .Then single factor analysis and logistic regression analysis were used to as‐sess children'environmental risk factors .Results In total ,2 438 effective quostionnaires have been got .The difference between Han (3 .89% ) and Uyghur (7 .05% ) dyslexia prevalence was statistically significant .The factor analysis revealed that educational grades ,family income ,father's and mother's occupations ,and their education levels as well as some home literacy environmental factors were significantly different for the two groups of children with dyslexia (P<0 .05) .Conclusion The prevalence of dyslexia was high in both groups ,and especially for Uyghur children . Some environmental factors may be responsible for the differences noted ,especially for the occupation of mother .
6.Efficacy observation of Guan's quadruple therapy for kidney disease on the treatment of chronic renal failure.

OBJECTIVETo compare the differences in the clinical efficacy between Guan's quadruple therapy for kidney disease and the conventional western medication in the treatment of chronic renal failure (CRF).
METHODSEighty patients were randomized into an observation group and a control group, 40 cases in each one. In the observation group, besides the physician instruction of low protein and low phosphorus diet in the two groups, Guan's quadruple therapy for kidney disease was applied, including (1) the isolated herbal moxibustion at Shenshu (BL 23) and Pishu (BL 21), or Ganshu (BL 18), Zusanli (ST 36) and Guanyuan (CV 4) alternatively; (2) acupuncture at the auricular points such as shen (CO10), jiaogan (AHR6), Shenshangxian (TG2), fei (CO14) and pizhixia (AT4); (3) injection of mixture of astragalus injection and lidocaine injection at Feishu (BL 13), Pishu (BL 21), Shenshu (BL 23) and the others; (4) modified Shenshuai Yihao decoction according to syndrome differentiation. In the control group, the conventional western medication was used. After 6 months of treatment, the differences were observed between the two groups in the clinical efficacy, serum creatinine (Scr), blood urea nitrogen (BUN), endogenous creatinine (Ccr) and 24 h urine protein quantitation (UPQ).
RESULTSIn the observation group, after treatment, the remarkably effective rate was 50.0% (20/40) and the total effective rate was 82.5% (33/40), which were superior to 25.0% (10/40) and 45.0% (18/40) in the control group (both P < 0.05). After treatment, the levels of Scr, BUN and 24 h UPQ were reduced apparently (all P < 0.05), and the level of Ccr was increased apparently in the two groups (both P < 0.05). After treatment, the levels of Scr, BUN and 24 h UPQ were reduced much more apparently in the observation group as compared with those in the control group (all P < 0.05).
CONCLUSIONGuan's quadruple therapy for kidney disease achieves a better efficacy on CRF compared with the conventional western medication. This therapy improves renal functions, relieves clinical symptoms and physical signs and benefits the life quality of patients.

7.Chemical constituents of Aconitum brachypodum from Dong-Chuan area.

Aconitum brachypodum is traditionally known to be toxic chinese medicie, but its chemical constituents is not enough studied to date. To further elucidate the chemical constituents of A. brachypodum, 80% ethanol extract of A. brachypodum collected from Dong-Chuan area was investigated, which led to isolation of seventeen compounds. By spectroscopic methods, their structures were determined as hypaconitine (1), mesaconitine (2), talatisamine (3), neoline (4), fuziline (5), aconine (6), bullatine A (7), lepeine (8), songrine (9), isocorydine (10), beta-sitosterol (11), daucosterol (12), stearic acid (13), triacontanol (14), palmitic acid (15), benzoic acid (16), and inosine (17), respectively. All compounds except for compounds 1 and 7 were isolated from A. brachypodum for the first time.

Objective To study the effects of found in inflammatory zone 1(FIZZ1) on the migration of cul- tured mouse vascular smooth muscle cell(VSMC) and the potential mechanism.Methods Using Boyden chamber, immunofluorescence and Western blot,the migratory effects,F-actin content and Akt/Akt1 expression were deter- mined in VSMC after stimulation by FIZZ1.Results FIZZ1 markedly increases the migratory ratio,F-actin con- tent and the Akt1 expression of VSMC.Ly294002,a PI3K inhibitor,attenuated migratory ratio,F-actin content and the Akt1 expression of VSMC promoted by FIZZ1 in a dose-dependent manner (10-30 ?mol/L).Conclusion Our data demonstrated FIZZ1 by activating PI3K/Akt1 pathway induced the expression of F-actin of VSMC,and promoted the migration of VSMC.

10.Experience on the diagnosis and treatment of intrathoracic gastro-airway fistulae after esophagectomy for esophageal carcinoma

Objective To summarize our results and experience in dealing with the postoperative intrathoracic gastro-air-way fistulae after esophagectomy for esophageal carcinoma.Methods From January 2010 through February 2012,1490 patients with esophageal carcinoma underwent esophagectomy in our department.The postoperative intrathoracic gastro-airway fistulae were documented in 10 patients,with a frequency of 0.67%.Five of them died.The possible etiology,clinical characters,treatment and prevention of this complication were reviewed.Results The location of the fistulate were 7 at left main bronchus,1 at right main bronchus,and 2 at distal trachea.After 2-3 weeks conservative treatment,1 patient underwent primary surgical repair and cured,1 refused any further intervention and sacrified,8 patients underwent endoscopic insertion of covered stent and only 3 healed.For the remaining 5 cases with failed stent therapy,2 died of severe aspiration and lung infection,3 had surgical repair,one of them successed and 2 died of aspiration and aortic rupture,respecively.Conclusion The development of intrathoracic gastro-airway fistulae was associated with the iatrogenic injuries and suturing material irritation of the gastric tube to the tracheal/bronchial wall.Therefore,a meticulous closure and wapping of gastroplasty and appropriate isolation using artifical patch or great omentum between airway and esophageal substitution could effectively reduce the fistulae.The stent therapy usually fails in treating this entity and surgical repair remains the final and ratical therapeutic option.Primary repaire is suggested and careful preoperative assessment is crucial.
