The S100 proteins are a multi-gene calcium-binding family,which are differently expressed in a variety of tumors.It is found to be associated with tumor invasion and metastasis.As a tumor-associated biomarkers,unraveling the relationship between S100 and prostate cancer progression as well as molecular mechanisms,would provide an important basis for the clinical diagnosis and therapeutic monitoring.
2.Determination of Doxorubicin Hydrochloride Concentration in Human Plasma by RP-HPLC

OBJECTIVE:To establish an RP-HPLC method for the determination of doxorubicin hydrochloride concentration in human plasma.METHODS:Tianhe Kromasil C18 was used as column,A solution of methenol-0.01 mol?L-1ammonium dihydrogen phosphate-acetic acid(30200.1) was used as the mobile phase at a flow rate of 1.0 mL?min-1.The detection of wavelength was set at 233 nm.The sample size was 20?L and the column temperature was 35℃.The internal standard was daunorubicin.RESULTS:The calibration curve was linear in the range from 0.25~20.16 ?g?mL-1(r=0.999 6);the average methodological recovery was 96.45%(RSD=2.16%).CONCLUSIONS:The method is rapid,accurate,reproducible and easy to use in clinical detection of blood concentration.
3.Determination of Plasma Concentration of Paclitaxel in Tumor Patients by RP-HPLC

OBJECTIVE:To establish a RP-HPLC method to determinate the plasma concentration of paclitaxel in tumor patients.METHODS:Paclitaxel was extracted from plasma with organic phase(ethyl-acetate)by two-step extraction on Tianhe Kromasil C18 column(250 mm?4.6 mm,5 ?m)with a mobile phase consisted of acetonitrile-methanol-water(40∶25∶40)at a flow rate of 1 mL?min-1.The detective wavelength was set at 227 nm and the column temperature was maintained at 35 ℃.RESULTS:The linear range of paclitaxel was 0.05~5.00 mg?L-1(r=0.999 7)with average recovery rate at 98.75%~100.44%.Both intra-day RSD and inter-day RSD were less than 5%(n=5).The plasma concentration-time profile in 11 patients after iv infusion of paclitaxel was in line with a two-compartment model.CONCLUSION:This established method is simple,accurate,reproducible and applicable for clinical determination of blood drug concentration and pharmacokinetic studies.
4.Baizhu suppresses S180 sarcoma growth and inhibits Bcl-2 expression in tumor-transfected mice

Objective To study Baizhu's inhibitory effect on S180 sarcoma growth and its effect on Bcl-2 expression in tumor-transfected mice. Methods S180 cells were subcutaneously injected into 60 healthy Kunming mice. Meanwhile the mice were dealt with MTX or Baizhu, separately; the weight of tumor was measured in the following two weeks; the expression of Bcl-2 was detected by RT-PCR. Results The tumor's weight in Baizhu group was lower than that in model group, but higher than in MTX group (P<0.05), but Baizhu's inhibition was not associated with its dose (P>0.05), and Baizhu's rate of tumor growth inhibition was lower than that of MTX. Compared with that in control group, Bcl-2 expression was lower in Baizhu group obviously. Conclusion Baizhu can inhibit tumor growth and serve as an adjuvant drug in tumor therapy.
5.Design and manufacture of hypobaric cabin simulator for experimental animals

Objective To design and manufacture a hypobaric cabin simulator for experimental animals.Methods The simulator was composed of a fully transparent working chamber,an electrical chamber,a pressure control component and a program control component,which had its body made of polymethyl methacrylate plastics,the hatch made of metal plate and support by reinforced metal bar.The air inside chamber was exhausted by the diaphragm vacuum pump,the air inflow was controlled by proportional valve,and the inner pressure and the speed of rise and decline were exactly controlled by the balance of inflow and exhaust.Results The new type of hypobaric cabin simulator had the characteristics of accurate pressure control,low fluctuation range and controllable up and down time,low noise as well as adaptable air exchange rate.Conclusion The new type of hypobaric cabin simulator matches the national standard for experimental animals,and can be used in making the animal model of high altitude diseases.
7.Determination of paeoniflorin in Fufang Biejia Ruangan Tablet by TLC scanning

OBJECTIVE To establish a method for the determination of paeoniflorin in Fufang Biejia Ruangan tablet.METHODS The tablets were extracted with 50% ethanol.The sample solution was applied on a silica gel G-CMC plate and developed with chloroform-methanol-glacial acetic acid (85∶13∶2) and was sprayed with sulfuric acid containing 5% vanillin.The separated spot was determined by scanning with CS-930 at λS=575 nm,λR=700 nm.RESULTS The average recovery was 97.60% and the RSD was 3.67%.CONCLUSION The results showed that this method is sensitive,specific,accurate and reproducible.
8.Applications of global left ventricular myocardial strain in detection of anthracycline-induced cardiotoxicity in breast cancer survivors with postoperative chemotherapy

Objective To assess the clinical significance of three-dimensional speckle tracking imaging (3D-STI) in the evaluation of left ventricular myocardial function in breast cancer survivors with postoperative anthracycline-based chemotherapy.Methods A total of 51 breast cancer survivors with postoperative anthracycline-based chemotherapy were recruited and 31 female healthy volunteers as the controls.Conventional echocardiography and 3D-STI derived parameters [including global longitudinal strain (GLS) and global area strain (GAS)] were measured before and at 3 and 6 anthracycline-based chemotherapeutic cycles.The receiver operating characteristics (ROC) curve was constructed to determine optimal sensitivity and specificity of 3ID-STI derived parameters for the prediction of future cardiotoxicity.Results In comparison with the controls and baseline cases,GLS and GAS deteriorated significantly (P < 0.05 for both).No statistical difference in GCS and conventional parameters showed before and after the initiation of chemotherapy (P <0.05,respectively).The ROC curves showed that GAS as the best 3D-STI predictor of patients who develop cardiotoxicity during the follow-up.The area under ROC(AUC) of GAS was 0.894,and its optimal cut-off value was-28.4%,with a specificity of 88.0% and a sensitivity of 82.9%.AUC of GLS was 0.802,and its optimal cut-off value was-14.3%,with a specificity of 62.0% and a sensitivity of 85.4%.Conclusions Early decreases in GAS and GLS based on 3D-STI may allow the prediction of subsequent cardiotoxic development accurately in breast cancer survivors with postoperative anthracycline-based chemotherapy.
9.Distribution and antimicrobial resistance of clinically isolated pathogens in hematology department from 2011 to 2013

Objective To analyze the distribution and antimicrobial resistance of pathogens isolated from department of hematology during the past three years.Methods Pathogenic strains isolated from patients hospitalized in a hematology de-partment between January 2011 and December 2013 were collected,antimicrobial susceptibility testing was performed by Kirby-Bauer disk diffusion method or automatic system,antimicrobial susceptibility testing results were judged according to American Clinical and Laboratory Standards Institute 2011, data were analyzed by WHONET 5.6 software. Results A total of 462 clinical isolates were collected in 2011—2013,including 161 gram-positive cocci isolates,279 gram-negative bacilli,and 22 fungi.Of Staphylococcus spp ,detection rate of methicillin-resistant coagulase negative Staphylo-coccus(MRCNS)and methicillin-resistant Staphylococcus aureus (MRSA)was 81.37% and 62.50%respectively.The re-sistant rate of Staphylococcus spp .and Enterococcus spp .to linezolid was 1.69% and 3.57% respectively,resistant rate of Staphylococcus spp .to teicoplanin was 3.39%,vancomycin-resistant gram-positive coccus was not found.Enterobacte-riaceae strains Escherichia coli and Klebsiella pneumoniae were highly susceptible to carbapenems,the sensitivity rates were 97.56%—98.88%;while nonfermentative gram-negative bacilli Pseudomonas aeruginosa and Acinetobacter bauman-nii strains were obviously resistant to carbapenems,the resistance rates were 38.71%—64.00%.Conclusion Antimicrobial resistance of major pathogenic strains from hematology department is high,antimicrobial agents should be used according to pathogenic distribution characteristics and antimicrobial susceptibility testing results,healthcare-associated infection control should be strengthened to reduce antimicrobial resistant rate.
10.Identification study of Compound Biejiaruangan Tablets

Objective: To establish the identificational method of Compound Biejiaruangan Tablets. Methods: The microscopic identification and TLC were used. Results: Carapax Trionycis, Radix Paeoniae Rubra, Radix Notoginseng, Placenta Hominis, and Cordyceps in Compound Biejiaruangan Tablets can be distinguished by microscopic identification and Cordyceps, Fructus Forsythiae and Radix Notoginseng can be identified by TLC, respectively. Conclusion: This method is simple, rapid and with a good reappearance, and available for quality control of the preparation.
