1.The influence of Lactobacillus salivarius on IgE,eosinophil cationic protein and transforming growth factor-βin serum and lung tissue homogenates of asthmic Balb/c mouse

Objective To explore the influence of Lactobacillus salivarius on IgE,eosinophil cationic protein(ECP)and transforming growth factor(TGF)-βof serum and lung tissue homogenates of asthmic Balb/c mouse.Methods Thirty female Balb/c mice weighing 16-18 g(4 weeks old),SPF grade,were ran-domly divided into three groups:normal control group(group A),asthma group(group B),asthma combined Lactobacillus salivarius group(group C).Acute asthma model was established applying ovalbumin,behavioral changes were observed in mice,the changes in airway inflammation and the number changes of tracheal cilia were observed by pathological HE staining and the electron microscope respectively.The levels of IgE,ECP and TGF-βin the serum and lung tissue homogenate were measured by ELISA method.Results (1 )The mice in group B presented obvious restlessness,head and facial itching,scratching,shortness of breath,peri-oral cyanosis with abdominal breathing,sneezing,stooping,forelimbs shrinkage,urine and feces incontinence and other asthma acute phase performances in 5-1 0 minuts after 4% ovalbumin saline solution atomization excitation,the above reaction alleviated in group C compared to group B.(2)Pathology of HE staining in group B showed the thickening bronchial wall,narrow lumen,peribronchial and perivascular infiltration with a large number of eosinophil-based inflammatory cells,lumen with many inflammatory secretions,pathological changes in lung tissue of mice in group C significantly alleviated compared with group B.Under the electron microscope,the number of cilia in group B significantly decreased compared with group A,while group C significantly increased compared with group B.(3)Expression levels of serum IgE and ECP in group C were sig-nificantly lower than those in group B[(0.86 ±0.14)μg/ml vs.(1.30 ±0.12)μg/ml;(2.30 ±0.15)ng/ml vs.(3.41 ±0.15)ng/ml,P <0.05,respectively].The contents of lung tissue homogenate IgE[(0.97 ± 0.06)μg/ml]and ECP[(5.88 ±0.13)ng/ml]in group C significantly decreased,respectively,compared with the IgE[(1.13 ±0.05)μg/ml]and ECP[(8.08 ±0.15)ng/ml]in group B(P<0.05).The levels of serum TGF-β[(0.17 ±0.01 )μg/ml]and lung tissue homogenates TGF-β[(0.18 ±0.01 )μg/ml]in group C were higher than the serum TGF-β[(0.13 ±0.01 )μg/ml]and lung tissue homogenates TGF-β[(0.15 ±0.01 )μg/ml]in group B,respectively,the differences were statistically significant(P<0.05 ). Conclusion Lavage with Lactobacillus salivarius before sensitization could alleviate clinical symptoms of asthmatic mice,as well as reduce airway inflammation in some degree,TGF-βmay be involved in the mecha-nism of Lactobacillus salivarius playing a protective role.
2.Effect of lactobacillus salivarius on asthma Balb/c mouse in IL-4,IFN-γ and IgE of serum and lung tissue homogenates

Objective To invstigate the effect of Lactobacillus salivarius on asthma Balb/c mouse in IgE,IL-4 and IFN-γ of serum and lung tissue homogenates.Methods Thirty female Balb/c mice weighing 16-18 g (4 weeks old), SPF grade, were randomly divided into three groups: normal control group (N), asthma group(A), asthma combined Lactobacillus salivarius group (AH).Acute asthma model was established by ovalbumin challenge method, mice were observed in behavioral changes, non-invasive method for the determination of airway responsiveness in mice, the changes in airway inflammation and the number changes of tracheal cilia were observed by pathological HE staining and the electron microscope.Respectively, the levels of IgE and IL-4/IFN-γin the serum and lung tissue homogenate were measured by ELISA.Results five ~ ten min after 4% OVA saline solution atomization excitation mice in A group appeared obvious restlessness, head and facial itching, scratching, shortness of breath, perioral cyanosis with abdominal breathing, sneezing, stooping, forelimbs shrinkage, urine and feces incontinence and other asthma acute phase performances, The above reaction alleviated in AH group compared to A group;Pathology of HE staining in asthma group showed the thickening bronchial wall, narrow lumen, peribronchial and perivascular infiltration with a large number of eosinophil-based inflammatory cells,lumen with many inflannatory secretions,Pathological changes in lung tissue of mice in AH group significantly alleviated compared with asthma group;Under the electron microscope, the number of cilia in A group significantly decreased compared with N group, while AH group significantly increased compared with A group;Expression level of IgE,IL-4 in the serum and lung homogenate of A group was significantly higher than that of N group (P < 0.05), the level of IFN-γexpression was significantly lower than N group (P < 0.05), however the expression level of IgE, IL-4 in AH group significantly decreased compared with A group (P < 0.05), and IFN-γexpression level elevated compared with A group(P < 0.05).Conclusion Lavage with Lactobacillus salivarius before sensitization could alleviate clinical symptoms of asthmatic mice, partially correct Th1/Th2 imbalance as well as reduce airway infammafion.

4.Clinical comparison of polymethylmethacrylate and bone cement in the treatment of osteoporotic vertebral compression fractures: a retrospective study

Objective:To analyze the outcomes of patients with osteoporotic vertebral compression fractures treated with polymethylmethacrylate (PMMA) and GeneX(R) bone cement in percutaneous kyphoplasty and to provide guidance in selecting the vertebral filling material for this procedure.Methods: In this study,86 patients with osteoporotic vertebral compression fractures treated at Peking University Third Hospital between June 2012 and June 2013 were retrospectively analyzed.The patients were followed-up with questionnaires and X-ray and CT examinations after 1 month,3 months,6 months,and 1 year.Postoperative conditions,including recovery rate of vertebral height,recovery rate of vertebral Cobb angle,CT data,reductions in visual analogue scale (VAS) and Oswestry scores,and postoperative complications,were evaluated.The results were analyzed using SPSS 18.0.Results: According to the rank sum test,there were significant differences in the recovery rate of vertebral height,vertebral Cobb angle and vertebral volume between the two groups after 3 months,6 months,and 1 year (P<0.05).The GeneX(R) group had greater losses in vertebral height,Cobb angle and volume than did the PMMA group.The PMMA group had 3 cases of cement leakage and 1 case of an adjacent vertebral fracture after percutaneous kyphoplasty.The GeneX(R) group had 2 cases of cement leakage.Conclusion: GeneX(R) bone cement is similar to PMMA in terms of postoperative pain relief.As the filling material in percuta-neous kyphoplasty,it is effective at maintaining vertebral height,quickly improves strength and stiffness of the vertebral body and has fewer complications.However,it is not satisfactory in maintaining long-term postoperative vertebral height.GeneX(R) bone cement can be used as the filling material for patients with osteoporotic vertebral compression fractures in percutaneous kyphoplasty.Patients with severe osteoporosis and vertebral compression should be treated with standardized osteoporosis treatment and should try to avoid early postoperative walking exercises.Sustained vertebral filling materials,such as PMMA,are more suitable for such patients.Research into better biodegradable materials is still needed.
5.Effect of Lactobacillussalivariuson the Number of CD4+CD25+Foxp3+Treg Cells and the Expression of TGF-β1 in Asthma Balb/c Mice

Objective To explore the effect of Lactobacillussalivariuson the number of CD4+CD25+Foxp3+Treg cells and expression of transform?ing growth factorβ1(TGF?β1)in asthma Balb/c mice. Methods Thirty?two female Balb/c mice were randomly divided into four groups:the nor?mal control group,the asthma group,the Lactobacillus salivarius group,and the asthma combined Lactobacillussalivariusgroup. Acute asthma mod?el was established by the ovalbumin challenge method. After extraction of primary spleen cells,flow cytometry was used to test CD4+CD25+Foxp3+Treg/CD4+T ratio in spleen lymphocytes. The levels of IL?4,IFN?γand TGF?β1 in the spleen cell culture supernatant were measured by ELISA method. Results The level of Th2 cytokine(IL?4)in the spleen cell culture supernatant of the asthma group was significantly higher than that of the control group(P<0.05),however,the level of Th1 cytokine(IFN?γ)was significantly lower than that of the control group(P<0.05). The ex?pression level of Th2 cytokine(IL?4)in the Lactobacillussalivariusintervention group was significantly decreased compared with the asthma group, and the Th1 cytokine(IFN?γ)expression level was elevated compared with the asthma group(P<0.05). The level of TGF?β1 in the Lactobacillus salivarius intervention group was higher than in the asthma group(P<0.05). The proportion of CD4+CD25+Foxp3+Treg/CD4+T in spleen lympho?cytes in the asthma group was lower than that in the control group(P<0.05),and was higher in the Lactobacillus salivarius intervention group than in the asthma group(P<0.05). Conclusion CD4+CD25+Foxp3+Treg was associated with the pathogenesis of asthma. Lactobacillus salivarius could adjust Th1/Th2 imbalance and reduce asthma inflammation through up?regulation of CD4+CD25+Foxp3+Treg and TGF?β1 expression.
6.Low-dose ultraviolet B induces sunburn reaction in Nrf2 gene-deficient mice

[Objective] To investigate the roles of Nrf2-Keap1 system in the protection of skin from ultraviolet B (UVB)-induced damage.[Methods] The dorsal surface of ears of 8 wild-type and 8 nrf2-null mutant 8-week-old female mice was exposed to a single dose of UVB irradiation (200 mJ/cm2) by using a FL120SE UV lamp source.Then,the morphology of ears was observed with the measurement of thickness before,as well as on day 1,2,4,7,9,11 and 14 after,the irradiation.Biopsy specimens were taken from the ears 36hours after the irradiation and subjected to hematoxylin and eosin staining as well as terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling (TUNEL) assay.The test Results were recorded and statistically analyzed by rank sum test and t test.[Results] A significant increase was observed in the thickness of mouse ears and number of sunburn cells per high-power field (400 ×) in nrf2-null mutant mice compared with wild-type mice ((0.49 ± 0.22) cm vs.(0.25 ± 0.03) cm,P< 0.01; 17.0 ± 3.9 vs.5.0 ± 1.7,t=13.8,P< 0.01).The number of TUNEL positive cells in the nrf2-null mutant mice was about 5 times that in the wild mice.The sunburn reaction appeared more intense and persistent in nrf2-null mutant mice than in the wild mice.[Conclusion] Nrf2-Keap1 pathway may protect skin against acute UVB damage,including cell apoptosis and oxidative damage.
7.Effect of lipid emulsions on the IL-1βand IL-6 expressions in acute lung injury induced by endotoxin in rats

Objective To compare the effect of three types of lipid emulsions on the IL-1βand IL-6 expressions in acute lung injury induced by lipopolysaccharide (LPS) in rats. Methods One hundred young SD rats were divided into control,LPS,ω-6,ω-9 andω-3 group. Age 29 days, the control group were intravenously injected with saline,ω-6 group injected lipid emu-sion (C14-24),ω-9 group injected long chain fat emusion,ω-3 group injected fish oil fat emusion. Then the control group fol-lowed by intratracheal instillation of saline, and the reamining four groups were sprayed with LPS. Pathologic changes in lung tissue section were observed. The expression levels of IL-1βmRNA and IL-6 mRNA were determined by RT-PCR,and the con-centrations of IL-1βand IL-6 in bronchoalveolar lavage fluid (BALF) were measured by enzyme-linked immune-specific assay. Results Infiltration and bleeding were observed in lung tissue under light microscopy in ALI rats. The lung indexes and patho-logical scores of ALI model groups were significantly higher than those of control group (P<0.005). The expressions of IL-1βand IL-6 mRNA in groupsω-9 andω-3 were significantly higher than those in groups LPS andω-6 (P<0.005). The levels of IL-1βand IL-6 in BALF in groupsω-9 andω-3 were significantly higher than those in groups LPS andω-6 (P<0.005). There was no significant difference (P>0.005) of the expressions of IL-1βand IL-6 mRNA betweenω-9 andω-3 group. Conclusions ω-6 PUFAs can accentuate inflammation by up-regulating the levels of IL-1βand IL-6 whileω-9 PUFAs andω-3 PUFAs can relieve inflammation by down-regulating the levels of IL-1βand IL-6.
8.Hospital Infection:Analysis of 706 Patients

OBJECTIVE To understand the related factors,the situation and dynamics of hospital infection in order to effectively prevent and control it.METHODS From 12306 cases of inpatients who had investigated retrospectively,706 cases were with nosocomial infection monitored from Jan to Dec 2006.RESULTS The hospital infection rate was 5.74% in 2006.The hematological disease and,cancer patients and the elderly,were the high-risk population.The main infection site was the respiratory tract.Fungi accounted for 21.35%,the top was the Candida albicans.CONCLUSIONS To strengthen the training of medical staff,to monitor the key departments of hematology and cancer and respiratory diseases and to attend the disease surveillance and rationally use of antibiotics are the effective measures to reduce hospital-acquired infections.

Objective To explore the puberty and height development in childern with congenital adrenal hyperplasia(CAH) due to 21-hydroxylase deficiency(21-OHD) with the simple virilizing(SV) form.Methods Patients of SV 21-OHD,diagnosed and treated in Tianjin Children's Hospital from Jan.1970 to Jan.2008,were treated with cortisone acetate or hydrocortisone after diagnosed,and blood investigations were performed as part of monitoring,and dosages were adjusted accordingly to obtain normal growth velocity and slow bone age(BA) development.Gonadotropin-releasing hormone agonist(Gn-RHa) was given after the appearance of central precocious puberty(CPP).Forty-one patients(18 females,23 males) had achieved final height before Dec.31st 2008,and their puberty and height development were analyzed in this longitudinal study by contrast the ages and statures of obtaining final height(FH),chronological age(CA) and BA of G2/B2 stage,duration from G2/B2 stage to obtain FH and stature development of different gender during this period and the influences of compliancy on the ages and statures of obtaining FH were contrasted.Results With regarding to the age of testicular volume ≥4 mL,78.26% boys were before 9 years old.As far as the age of breast development was concerned,38.89% girls were before 8 year old and 66.67% menarche occurred before 10 year old.The differences between the duration and stature development of the period from the state of G2/B2 to skeletal maturation of different gender were significant (Pa
10.Reasons and Countermeasures for Different Compliances of Insulin Pump Therapy in Children with Type 1 Diabetes
