Long noncoding RNA ( lncRNA) is a functional RNA molecule longer than 200 nucleotides.They are involved in epigenetic, transcriptional and post-transcriptional modifications, playing an important role in gene expres-sion, chromatin remodeling, histone modification, X chromosome inactivation, genomic imprinting, protein folding and ac-tivity as well as other important biological events.Some studies have proven that lncRNAs are involved in the development of metabolic diseases.The changes of their expression levels may be new diagnostic markers and therapeutic targets.

3. Modified posterior lumbar interbody fusion using unilateral pedicle screw fixation in lumbar spinal fusion

Objective: To compare the clinical efficacies and complications between the posterior lumbar interbody fusion (PLIF) using bilateral pedicle screw fixation and modified PLIF using unilateral pedicle screw fixation. Methods: A total of 96 patients who underwent lumbar spinal fusion operation during Feb. 2007 to May 2009 were divided into 2 groups. Group A included 40 patients (14 males and 26 female, average age being [52.23±9.75] years) receiving unilateral modified PLIF; Group B included 56 patients (22 males and 34 female, average age being [56.02±10.25] years) receiving bilateral PLIF. Oswestry disability index (ODI), visual analog scale (VAS), operating time, blood loss, length of hospital stay, hospitalization cost, fusion rates, complication rates, intervertebral height and medical expenses were all compared between the two groups. Results: The short-term ODI, VAS indices showed no significant difference between the two groups. Blood loss, operating time, and hospitalization cost of group A were significantly less than those in group B (P<0.05). Fusion rate was 100% in both groups 12 months after the operation. Conclusion: Modified PLIF using unilateral pedicle screw fixation is effective for lumbar spinal fusion, and it has the less trauma, risk and hospitalization cost compared with PLIF using bilateral pedicle screw fixation; but its long-term efficacy still needs to be further observed.
4.The effect of neural stem cell transplantation on the hindlimb motor function and caspase-3 expression of motor cortex in spinal cord transected rats

Objective To observe the effect of neural stem cell (NSC)transplantation on the hindlimb motor function and caspase-3 expression of motor cortex (MC) in spinal cord transected (SCT) rats. Methods S prague-Dawley (SD) rats were randomly divided into sham operated group, operation group( T9 transection), and subacute NSC transplantation group. The MC of each group( n= 8 )was harvested 7 days post operation (dpo), then western blot was employed to detect the level of caspase-3( β-actin was used as control ). The left 5 animals of each group were subjected to BBB score evaluation at 16th week,then the animals were sacrificed and the MC was harvested and performed immunostain by using caspase-3 rabbit antibody. Results Following NSC transplanta tion, BBB scores( ( 7.58 ± 0. 99 ) ) increased significantly than seen in the SCT animals( ( 5.16 ± 1.19) ). The expressional level of caspase-3 at 7day post operation was( (0.89 + 0.12)) in MC of SCT rats,while it decreased significantly to( (0.76 + 0.11 ) ) in NSC transplantation rats(P<0.05). The immunoreactive stain of caspase-3 was seen in the cytoplasm of pyramidal neuron in the cortex. Conclusion NSC engraft can downregulate the expression of caspase-3, corresponding to a significant improvement in hindlimb motor function. These findings indicate that NSC transplantation probably regulate the expression of apoptosis genes in MC to promote neurological function recovery in rats subjected SCT.
5.Effects of NF-κB/p65 siRNA on Apoptosis and Bax Expression in Lung Tumour Cell Xenografts of Nude Mouse

Objective To investigate the effects of nuclear factor-kappaB(NF-κB)/p65 signaling transduction on the apoptosis and expressions of apoptosis-related genes Bax in nude mouse lung tumour cell xenografts. Methods The nude mice Lewis lung carcinoma cell xenograft model was established,and then the mice were intraperitoneally injected with NF-κB/p65 small interfering RNA(siRNA). The apoptosis of xenografted tumor cells in nude mice was detected by TUNEL assay. Expressions of Bax mRNA and protein were detected by RT-PCR and Western blotting. Results The result of TUNEL assay demonstrated that p65 siRNA evidently evoked cell apoptosis. Compared to the PBS treatment group or the normal control mice,both mRNA and protein expression levels of Bax in tumor xenografts were significantly up-regulated. There were significant differences among three groups(P<0.05). Conclusion NF-κB/p65 subunit may play an essential role in cell apoptosis of Lewis lung tumor.
6.Establishment and Application of Access Database in Therapeutic Drug Monitoring

OBJECTIVE: To establish Access database to conserve and manage patients’ clinical information in therapeutic drug monitoring (TDM). METHODS: Therapeutic Drug Monitoring Database Management Software was established to analyze the utilization of digoxin and CsA in renal transplantation patients combining with the situation of TDM in our hospital. RESULTS: The database had the advantages of convenient data processing and rapid inquiry. CONCLUSIONS: Access database is convenient for information management of case history and can afford a great support for clinical pharmacists.
7.Immunodiagnostic value of recombinant membrane protein Tetraspanin 2 of Schistosoma japonicum

Objective To study the immunodiagnostic potential of recombinant membrane protein Tetraspanin 2 of Schistosoma japonicum(rSjTsp2) as a diagnostic antigen. Methods The rSjTsp2 was expressed and purified as an antigen used in indirect ELISA to detect specific serum antibody against Schistosoma japonicum of rabbits that at different stages of infection, and the results were compared with those of ELISA using natural antigen (AWA) as antigen. Results The sensitivities of rSjTsp2-ELISA were 75.4% (46/61),77.6% (45/58) and 81.5% (44/54), respectively, 2, 4, 6 weeks after rabbits were infected with Schistosoma japonicum, meanwhile the sensitivities of AWA-ELISA were 68.9% (42/61),86.2% (50/58) and 88.9% (48/54), respectively. The sensitivity difference of two methods was not significant statistically (x2 = 0.652, 1.454,1.174, all P > 0.05).Conclusions The rSjTsp2 is expressed successfully in vitro, and rSjTsp2 recombinant protein expressed in prokaryotic system has a certain application potential in immunodiagnosis.

Objective To investigate whether PSD 93 reduce neurotoxity induced by platelet activating factor (PAF) and study on the mechanism of cell molecularbiology.Methods PAF treated the cultural cortex neurons in wild type and PSD 93 knockout mice and the cells were stained with PI/Calcein AM for apoptosis; neuronal viability was measured by MTT. The expression of PSD93 and PSD95 protein in neurons was tested by Western Blot.Results PSD 93 and PSD 95 expressed in wild type neurons and only PSD 95 expressed in PSD 93 knockout. 9.Neuronal injury induced by platelet activating factor involved in NMDA/PSD_(93) signaling pathway

Objective To study whether neuronal injury induced by platelet activating factor (PAF) was involved in NMDA /PSD 93 signaling pathway.Methods Primary cortex neurons culture were from wild type and PSD 93 knockout mice.Then the neurons were pretreated with 0.3 ?mol/L PAF for 24 hours, 5 ?mol/L BN52021, 10 ?mol/L MK-801, or 60 ?mol/L L-NAMA, respectively. The cells were stained with PI/Calcein for apoptosis detection. Varied protein expressions were also observed in the neurons by western blot. Proteins colocalized on neuraxon were detected by cell immunochemistry and confocol microscopy, and cGMP activity was measured by radioimmunoassay.Results (1) PSD 95, NR2A, and nNos except PSD 93 expressed in the cortex neurons from PSD 93 knockout mice. (2) PSD 93, NR2A and nNos colocalized on neurite. (3) Neurotoxicity and cGMP avtivity induced by PAF decreased in PSD 93 knockout cortex neurons.Conclusion NMDA/PSD 93 signaling pathway is involved in neuronal injury induced by PAF.
10.Research progress of bone marrow-derived mesenchymal stem cells in blood diseases

Bone marrow mesenchymal stem cells (MSC) are the non-hematopoietic cellular component in the bone marrow that have multiple differentiation potency. MSC play a key role in the regulation of bone marrow hematopoietic niche and modulation of immune function through various mechanisms. They are currently recognized as a promising cell source in tissue engineering, a vehicle in gene therapy and a powerful tool in the management of graft-versus-host disease after allogeneic stem cell transplantation. In this review some momentous aspects regarding the current status and potential clinical applications of MSC in hematopoietic stem cell transplantation, aplastic anemia and multiple myeloma were summarized.
