Journal of International Oncology 2012;39(4):301-304
Allogeneic stem cell transplantation is the only treatment which may cure multiple myeloma (MM).The high transplantation related mortality (TRM) limits the wide clinical application of myeloablative allogeneic stem cell transplantation.Non-myeloablative allogeneic stem cell transplantation can reduce the TRM,but the risk of relapse and progress of the disease may get increased. Autononmyeloablative allogeneic stem cell tandem transplantation can reduce both the TRM and the relapse and progress of the disease. At present, there exist many new methods which may improve the effect of allogeneic stem cell transplantation for MM in clinical.

OBJECTIVE:To assess the feasibility of production and effects of Tobramycin eye drops prepared in this hospital for treating ocular infections .METHODS: 77 patients with external ocular infections were randomly divided into two groups with imported Tobrex as control drug to compare the effects.Assessment was made according to Gwon project results.RESULTS: Overall improvement rates were 92.7% in Tobramycin eye drops group and 94.4% in control group without significant difference between two groups(P> 0.05) .CONCLUSION: The technology of Tobramycin eye drops production is simple, therapeutic effect is satisfactory, and the production in hospital is feasible.

OBJECTIVETo explore the diagnostic value of the radiologic characteristics of osteoporotic Kummell's disease.
METHODSTotal 16 patients with pathologically confirmed osteoporotic Kummell's diseases were reviewed from May 2010 to May 2012, including 4 males and 12 females with the mean age of 73.4 years (ranged, 67 to 83 years old). Radiologic imagings of all patients, including X-ray, CT and MRI, were analyzed retrospectively.
RESULTSIntravertebral linear clefts could be seen on the AP and lateral X-ray films of vertebrae. Sagittal and axial CT scans demonstrated the vacuum cleft phenomenon with liquid and air was identified within the vertebral body. Sagittal MRI showed the callapsed vertebral segment and the area of fluid signal with clear and intact border within the vertebral body. The fluid signal was low on T1-weighted images and high on T2-weighted images and stir images, which was corresponding to an intravertebral vacuum cleft.
CONCLUSIONThe radiologic characteristics of Kurmmell's diseases can provide valuable evidences for the early diagnosis.

This study was conducted to evaluate the relationship between serum insulin levels and the production of insulin antibody (IA) in type 2 diabetes (T2DM).A total of 647 T2DM were included.Among them,20.9% patients were IA positive, who were elder and had a longer duration, lower BMI,a higher positive rate of glutamic acid decarboxylase antibody(GADAb) and higher serum insulin levels during an insulin secretion test.More patients were treated with insulin in IA positive group than in IA negative group (65.9% vs 41.0%,P =0.000).Fasting serum insulin level was associated with occurrence of IA in all patients (OR =1.02,P =0.001) and insulin treated patients (OR =1.033,P =0.002).The cut-off point of fasting serum insulin level for predicting IA positive was 17.87 mIU/L (sensitivity 55.1%,specificity 89.0%).Exogenous insulin use is associated with the presence of IA.Fasting serum insulin level can be used as a predictor for the production of IA in insulin-treated patients.

Objective To evaluate the influence of tissue factor pathway inhibitor 2(TFPI-2)gene on the proliferation and invasion of K562.Methods The expression vector pcDNA3.0/TFPI-2 was transfected into human leukemia line K562 cells(K562-T)by using liposome, then the mRNA and protein TFPI-2 were detected by real-time RT-PCR and western blot separately. The growth curve and the colony-forming unit assay were used to measure the ability of cells growth and transwell chamber model was employed to test the ability of cell invasion in vitro. Results Expression of mRNA and protein of TFPI-2 was detected in transfected cells. The growth rate and self-replication ability of K562-T cells were lower than those of the two control groups obviously. The number of K562-T cells to traverse a Matrigel-coated membrane was dramatically decreased compared with that of non-expressing cells. Conclusion The gene of TFPI-2 can inhibit the growth, proliferation and invasion of the K562 cells.

Objective To investigate sequence variations of 12 miRNA genes in multiple myeloma(MM) in order to find whether sequence variations in miRNA genes are associated with tumorigenesis and discuss the clinical significance of MM associated with miRNA genes mutations. Methods The miRNA gene mutations in 20 cases of MM, 4 MM-derived cell lines and 20 controls were detected by the methods of polymerase chain reaction single stranded conformation polymorphism (PCR-SSCP) and silver staining technique. Both clinical features and laboratory results were analyzed simultaneously. Results The electrophoretic patterns showed a total of three variations in miR-19a, miR-19b and miRNA-335,which were observed in 3 MM cells (15 %, 3/20). We also found variations of miRNA-335 in MM-derived cell lines KM-3and RPMI8226. However, no sequence alteration in the miRNA genes was observed in our set of controls. One of the three MM patients died, and two of them were detected mutations at the terminal stage of the disease. Conclusion A relative high frequency of miRNA gene mutation was found in MM and MM derived cell lines, which suggests possibility of a main mechanism underlying tumorigenesis. And, detecting miRNA gene mutations in MM might be benefit to evaluate the progression and prognosis of disease.
