OBJECTIVETo investigate the relationships between erythrocyte membrane n-6:n-3 PUFAs ratio and blood lipids and high sensitivity C-reactive protein (hs-CRP).
METHODSThe observational study consisted of a population-based cross-sectional study of 456 Chinese and a subsequent 1-year follow-up study of 171 subjects with the fasting plasma total cholesterol of 5.13-8.00 mmol/L.
RESULTSIn the cross-sectional analysis, plasma low-density lipoprotein cholesterol (LDL-c) had a significant and negative association with the erythrocyte membrane n-6:n-3 PUFAs ratio (P for trend=0.019) after adjusting for sex, age and total PUFA percentage. In the follow-up study, 171 subjects were categorized into quartiles by the changes of n-6:n-3 ratio in erythrocyte membrane (Δ=month 12-month 0). In the top quartile whose ratios of n-6:n-3 increased by an average of 1.25 during the follow-up, the LDL-c-lowering extent was 3.3 times of that in the lowest quartile whose ratios of n-6:n-3 decreased by an average of 1.13 (-1.07 mmol/L v.s. -0.32 mmol/L). The hsCRP decreased by 0.11 mg/dL in the lowest quartile while increasing by 0.10 mg/dL in the top quartile (P for difference=0.052).
CONCLUSIONOur results suggested that the balance between n-6 and n-3 fatty acids may optimize the cardiovascular benefits from dietary PUFAs.

Objective To evaluate gene transfection in liver , lung and kidney by ultrasound , microbubble and recombinant adenovirus mediated exogenous stromal cell derived factor-1α ( SDF-1α) gene transfer to the heart in rats with acute myocardial infarction( AMI) . Methods Forty AMI SD rats were randomly divided into control and experimental groups:myocardial infarction + ultrasound irradiation group (M+ U/control group , n = 10) ,and 3 experimental subgroups on the basis of pAd-EGFP/SDF-1α ( The biotin recombinant adenovirus expressing enhanced green fluorescent protein and SDF-1α) . Genes transfection length of time:1 day ,2 days and 3 days of transfection ( M +S1+U ,M+S2+U and M+S3+U group) . The expression of EGFP in liver ,lung and kidney were detected by laser scanning confocal microscopy at 7 days after administration . Results There was a little expression of EGFP in the liver ,lung and kidney in the drug delivery group and no expression in the control group .The differences in the expression of EGFP between all the gene delivery groups and the control group were statistically significant ( P <0 .05) . With the increase of the number of medication days , the target gene transfection increased slightly ,but there was no significant difference among the different drug delivery groups . Conclusions When the ultrasound ,microbubble and recombinant adenovirus mediated exogenous SDF-1αgene transfer to the heart in AMI rats ,liver ,lung and kidney tissues will also be transfected . However ,with the increasing of the days of administration , the transfection of target gene in non-target tissue produces only a slight accumulation . The transfection area of target gene in non-target tissue is not linear correlated with the days of administration .

Objective To observe the expression of parathyroid hormone - related peptide (PTHrp) receptor in tibial growth plate and its effects on tibial extension in chronic renal insufficiency rats. Methods Two-week-old male Sprague-Dawley rats were randomly divided into sham group, model group and enalapril group, each with 20 rats. In model group and enalapril group rats had chronic renal insufficiency induced by left ureteral obstruction, and rats were respectively given saline and enalapril by gavage after the operation. In sham group, left ureter was only exposed without ligation, and rats were given saline. The urine was collected 4 weeks after the operation and the total protein content was measured. Then all rats were killed. The concentrations of PTHrp, creatinine and urea nitrogen in intracardiac blood were detected. HE staining and Masson staining were performed on the left kidney to observe pathological changes of glomeruli and renal tubules. The total length of bilateral tibia was measured. The number of columnar cells in the growth plate proliferative zone was measured by safranin O staining and the expression of PTHrp receptor in the growth plate was detected by immunohistochemistry. Results The 24 h urine total protein, creatinine and urea nitrogen in model group were higher than those in sham group (all P<0.05), while these 3 renal functional parameters in enalapril group were lower than those in model group (all P<0.05). In model group and enalapril group rats had higher blood concentrations of PTHrp than that in sham group (all P<0.05), but blood PTHrp in enalapril group was lower than that in model group (P<0.05). HE staining and Masson staining showed that in the model group rats had severe tubular dilation, inflammatory cell infiltration and the tissue fibrosis, while in enalapril group renal tubules slightly dilated and had a few inflammatory cell infiltration and tissue fibrosis. Compared with those in the sham group, in model group the tibia length, the chondrocyte number of column structure in the growth plate proliferative zone and the PTHrp receptor decreased (all P<0.05). But in enalapril group those indexes increased than model group (all P<0.05). Conclusions Chronic renal insufficiency rats had increased PTHrp concentration in the blood but decreased PTHrp receptors expression in tibial growth plate, which lead to their limited tibial extension.
