1.Effect of ginseng Shensong Yangxin capsule and metoprolol sustained release tablet on the curative effect and the levels of BNP and HR in elderly patients with coronary heart disease with arrhythmia

Objective To study the effect of Snsong Yangxin capsule and metoprolol sustained release tablet on the curative effect and the levels of BNP and HR in the elderly coronary heart disease (CHD) arrhythmia patients. Methods It selects 108 patients with coronary heart disease and arrhythmia in elderly patients were divided into observation group (54 cases) and control group (54 cases). The control group was treated with metoprolol sustained-release tablets, and the observation group was treated with Shensong Yangxin capsule and metoprolol sustained-release tablets. The curative effect of two groups and the changes of BNP and HR levels before and after treatment were observed and compared. Results The treatment effect, times of contraction, frequency of cardiac fibrillation, frequency of defect, load, time, blood pressure and BNP of the observation group were significantly better than those of the control group (P<0.05). Conclusion For elderly patients with coronary heart disease arrhythmia, the treatment of Shensong Yangxin capsule combined with metoprolol sustained release tablets is effective. It can significantly improve the patient's heart rate, restore the blood supply to the heart, improve the blood pressure and BNP, with less adverse reactions, safe and reliable, and worthy of promotion.

Objective:To study on effect of Danshao Huaxian Capsule on proliferation of activated hepatic stellate cells(HSCs)in vivo and in vitro.Methods:(1)In vitro experiment:57 male Wistar rats were divided into a normal group,a model group and a treatment group.The rats of model group and treatment group were made hepatic fibrosis model with complex stimulation of CCl_4,drinking alcohol,high lipids and low protein diet for 8 weeks.The treatment group were treated for 8 weeks with intragastric perfusion of Danshao Huaxian Capsule,1g/kg.At the end of experiment,a part of the rats were used for detection of hepatic functions and hepatic fibrosis degrees,and another part of the rats were used to separate HSCs.Cell cycle percentage was detected with flow cytometry.(2)In vivo experiment:prepare Danshao Huaxian Capsule serum of the rat;separate cultured HSCs of the rat and divide into calf serum group,normal rat serum group and Danshao Huaxian Capsule serum group.5%,10% and 20% of the above serum was respectively added into the cultured HSCs.Primary HSCs proliferation was detected with MTT method.Results:(1)In vitro experiment:The hepatic function,hepatic fibrosis level and HSCs proliferation reduced significantly.(2)In vivo experiment: Danshao Huaxian Capstde serum group could significantly inhibit proliferation of HSCs as compared with calf serum group and normal rat serum group at the same content.The action showed dose-dependence.Conclusion:Danshao Huaxian Capsule can obviously inhibit proliferation of HSCs in vivo and in vitro.

5.Analysis of type 2 diabetic nephropathy markers

Objective To investigate the type 2 diabetic patients with glomerular filtration rate and urinary albumin value as renal finction evaluation.Methods A total of 1217 patients with type 2 diabetic nephropathy from August 1,2010 to June 31,2012 were selected.Evaluated glomerular filtration rate (eGFR) was calculated according to the currently recommended simplified modification of diet in renal disease study (MDRD) formula.According to the K/DOQI guidelines for chronic kidney disease,patients were divided into three groups:group A [eGFR ≥90 ml/ (min ·1.73 m2)] of 891 cases,group B [60 ml/(min· 1.73 m2) ≤eGFR < 90 ml/(min· 1.73 m2)] of 256 cases,group C[eGFR < 60 ml/(min· 1.73 m2)]of 70 cases,respectively.Eight-hour urinary albumin excretion rate (UAER) was compared between and among the groups; according to 8-hour UAER,patients were divided into three groups:group a (8-hour UAER <20 μg/min) of 946 cases,group b (20 μg/min≤8-hour UAER <200 μg/min) of 193 cases,group c(8-hour UAER ≥ 200 μ g/min) of 78 cases.eGFR was calculated and compared between and among the groups.Results Eight-hour UAER was (32.71 + 79.62),(57.56 + 130.59),(107.50 + 222.64) μ g/min in group A,B,C,and the difference among three groups was statistically significant (P<0.05).eGFR was (110.78 ±32.75),(108.26 ±45.78),(94.96 ±32.30) ml/ (min ·1.73 m2) in group a,b,c,and the difference between group a and group c,or between group b and group c was statistically significant (P <0.05),but the difference between group a and group b was not statistically significant (P >0.05).Conclusion When evaluating the function of type 2 diabetic nephropathy,two indicators should be considered:glomerular filtration rate and urinary albumin.
6.Relationship between urinary albumin excretion rate and cardiac structure and function in type 2 diabetes mellitus

Objective To study the relationship between urinary albumin excretion rate(UAER)and cardiac structure and function in type 2 diabetes mellitus(T2DM).Methods Five hundred and twenty patients with T2DM were divided into three groups by 8 h UAER:normoalbuminuria group(424 cases),microalbuminuria group(75 cases)and macroalbuminuria group(21 cases).At the same time,50 healthy people were selected as control group.The cardiac structure and function in four groups were evaluated by echocardiographic measurements.Results Left atrial dimensio[(34.39±3.46)mm],left ventricular posterior wall thickness[(10.34±1.05)mm],relative wall thickness(0.46±0.05),interventricular septal thickness[(10.49±1.07)mm],left ventricular mass[(167.37±32.12)g],left ventricular mass index[(87.62±12.16)g/m2]in macroalbuminuria group were significantly higher than those in control group[(32.59±2.71)mm,(9.30±0.77)mm,0.42±0.04,(9.75±0.81)mm,(147.33±27.23)g,(80.14±13.81)g/m2](P<0.05),mean while the ratio of E/A(0.82±0.19)in macroalbuminuria group was significantly lower than that in control group(1.21±0.16)(P<0.05).Conclusions In T2DM patients with albuminuria,the cardiac structure and function have changed.With the increase of UAER,the impairments are more prominent.And it may predict diabetes cardiomyopathy.
7.Study on the severity of diabetic retinopathy and its relationship with serum bilirubin

Objective To investigate the relationship between the severity of diabetic retinopathy and serum bilirubin level. Methods From August 1 2008 to December 31 2009, hospitalized patients of 1062 cases with type 2 diabetes were divided into three groups according to fundus examination: no diabetic retinopathy (NDR) group (457 cases), non-proliferative diabetic retinopathy (NPDR) group (484 cases)and proliferative diabetic retinopathy (PDR) group ( 121 cases). The difference in serum bilirubin level was compared. Results Total bilirubin in PDR, NPDR and NDR group were (9.96 ± 3.84), ( 11.41 ± 3.82),( 13.38 ± 3.98) μ mol/L respectively. There was significant difference among three groups (P < 0.01 ). Logistic multivariate regression analysis showed that a drop in serum total bilirubin level was an independent risk factor of diabetic retinopathy. Conclusions Bilirubin level and the severity of diabetic retinopathy have some relationship. The antioxidant bilirubin and anti-inflammatory effect should be paid attention to.
8.Polymorphism within the interleukin-10 receptor cDNA gene in patients with systemic lupus erythematosus

Objective To assess the association between polymorphism within the interleukin-10 receptor cDNA gene (IL-10R) and Chinese patients with systemic lupus erythematosus (SLE). Methods The IL-10R genotypes of 94 SLE patients and 80 healthy subjects were examined by reverse transcription-polymerase chain reaction-single strand conformation polymorphism method (RT-PCR-SSCP), RT-PCR-restriction fragment length polymorphism (RFLP) and DNA sequencing. Results There were significant differences in the IL-10R2 genotype frequencies of these two groups. The IL-10R2 G520/G520 genotype increased the risk of developing SLE (OR = 0.515, 95% CI 0.414-0.579, P = 0.004) and individuals who had G520/A520 genotype also had a higher susceptibility to SLE (OR = 1.968, 95% CI 0.981-3.949, P = 0.055). There was no significant association between SLE and IL-10R1 genotypes. The risk of developing SLE was detected in the individuals who had the combination of IL-10R1 G241/G241 and IL-10R2 G520/G520 (OR = 0.515, 95% CI 0.444-0.597, P = 0.004). Conclusion The IL-10R2 genotypes of G520/G520 and G520/A520 as well as the combination of genotypes IL-10R1G241/G241 and IL-10R2 G520/G520 may increase the susceptibility to SLE in Chinese people.
9.D-galactopyranosyl zinc phthalocyanines kill hepatocellular carcinoma cells in vitro and in vivo

Objective To explore the therapeutic efficacy of the water soluable photosensitizer D-galactopyranosyl zinc phthalocyanines (T1)-mediated photodynamic therapy (PDT) applied to HepG2 human hepatocellular carcinoma cells in vitro and in vivo,as well as its mechanism.Methods HepG2 cells in their logarithmic growth phase were cultured and divided into different concentrations ofT1 (0 μM,0.06 μM,0.125 μM,0.25 μM,0.5 μM and 1 μM).Methyl thiazolyl tetrazolium (MTT) assays were employed to determine the effect of the T1-PDT on the proliferation of the HepG2 cells.Cell apoptosis and necrosis were measured using a cell analyzer with Annexin VFITC/PI/Hochest33342 triple-staining.The reactive oxygen species (ROS) and the mitochondrial membrane potentials of the HepG2 cells were detected using fluorescence microscopy.Confocal microscopic assays were used to observe T1's subcellular localization on the HepG2 cells.Real-time quantitative polymerase chain reactions (RT-PCRs)were used to detect any apoptosis of Bcl-2-and Bax-related genes.H-22-bearing mice were used to calculate the antitumor rate of T1-PDT,and the expression levels of Bcl-2 and Bax mRNA were detected using RT-PCRs.Twenty-four healthy mice were randomly divided into a control group,a low-dose group,a middle-dose group and a high-dose group,each of 6.Each group was given different doses of T1-PDT and the tumor inhibition rate was calculated.Results The MTT assays showed that T1-PDT could significantly inhibit HepG2 cell growth,but T1 or PDT alone had little effect.The confocal microscope assay showed that T1 was mainly localized in the mitochondria in HepG2 cells with little in the lysosome.Cell analyzer results showed that T1-PDT could induce HepG2 apoptosis.The ROS levels of HepG2 cells increased after T1-PDT.The RT-PCR results showed that T1-PDT could increase the expression of Bax and decrease the expression of Bcl-2.The in vivo experiments demonstrated that T1-PDT significantly inhibited the growth of H-22-xenografied tumors.Conclusions T1-mediated PDT has a significant lethal effect on HepG2 cells in vitro and in vitro.The lethal effect of PDT on cancer cells is shown in the apoptosis and can be attributed to T1's subcellular localization in the mitochondria,increasing ROS levels,and regulating apoptosis-related genes.
10.Levels of Th2 cytokine receptors in the peripheral blood mononuclear cells of the patients with lupus erythematosus and its clinical significance

Objective:To investigate the roles of Th2 cytokine receptors in the pathogenesis of systemic lupus erythematosus(SLE) and its clinical significance.Methods:RT-PCR technique was used to semiquantitatively analyze Th2 cytokine receptors mRNA expression in PBMC from 89 patients with SLE,double antibody sandwich ELISA was applied to determine Th2 cytokines in serum.Results:The positive Th2 cytokine receptors mRNA expression accounted for 100% among active SLE group(group Ⅰ),inactive SLE group(group Ⅱ) and control group(group Ⅲ).The mean levels of IL-4R mRNA expressions in group Ⅰ,Ⅱ and Ⅲ were 0.604?0.147,0.40?0.13 and 0.37?0.07,respecitively.Compared with group Ⅱ and group Ⅲ,the mean level of IL-4R mRNA expressions in group Ⅰ was markedly increased(P=0.00),while there were no statistical differences between group Ⅱ and group Ⅲ(P=0.351).The mean levels of IL-6R mRNA expressions in group Ⅰ,Ⅱ and Ⅲ were 0.902?0.273,0.519?0.11 and 0.573?0.24,respectively.Compared with group Ⅱ and group Ⅲ,the mean level of IL-6R mRNA expressions in group Ⅰ was markedly increased(P=0.00),while there were no statistical differences between group Ⅱ and group Ⅲ(P=0.289).The mean levels of IL-10R mRNA expressions in group Ⅰ,Ⅱ and Ⅲ were 0.87?0.29,0.722?0.209 and 0.684?0.138,respectively.Compared with group Ⅱ and group Ⅲ,the mean level of IL-10R mRNA expressions in group Ⅰ was markedly increased(P=0.00),while there were no statistical differences between group Ⅱ and group Ⅲ(P=0.405).The mean levels of IL-4 in serum in group Ⅰ,Ⅱ and Ⅲ were 105.57?29.17,51.17?16.69 and 23.73?6.24,respectively.Compared with group Ⅱ and group Ⅲ,the mean level of IL(4 in group Ⅰ was significantly higher than those in group Ⅱ and group Ⅲ(P=0.00),while that in group Ⅱ was higher than group Ⅲ(P=0.00).The mean levels of IL-6 in serum in group Ⅰ,Ⅱ and Ⅲ were 71.89?20.02,17.96?6.93 and 0.035?0.035,respectively.Compared with group Ⅱ and group Ⅲ,the mean level of IL-6 in group Ⅰ was significantly higher than those in group Ⅱ and group Ⅲ(P=0.00),while that in group Ⅱ was higher than group Ⅲ(P=0.00).The mean levels of IL-10 in serum in group Ⅰ,Ⅱ and Ⅲ were 32.32?11.51,18.66?6.63 and 0.289?0.867,respectively.Compared with group Ⅱ and group Ⅲ,the mean level of IL-10 in group Ⅰ was significantly higher than those in group Ⅱ and group Ⅲ(P=0.00),while that in group Ⅱ was higher than group Ⅲ(P=0.00).There were positive correlation between IL-4 and IL-4R in active group and inactive group(r=0.622-0.859,P
