The Apolipoprotein E(APOE)gene plays important roles in the pathogenesis of the Alzheimer disease(AD).It is clear that APOE gene is closely related with dementia in the elderly.In recent years,extensive researches regarding the relationship between APOE gene polymorphism and AD has been conducted in both health population and patients with AD using molecular biology technique.This paper reviewed the progress in this field for sharing some fundamental information on the research in AD.

ObjectiveTo investigate the combined effects of one to one whole process companion delivery model on emotional status and postnatal recovery of parturients.Methods350 cases adopted one to one whole process companion delivery model were set as the observation group,another 350 cases who did not received companion delivery model were set as the control group.The emotional state and postpartum recovery (the amount of vaginal bleeding after childbirth,time of the uterus return to normal size,and the incidence of urinary retention 6 h after delivery,colostrum time and fasting time) were compared between the two groups.ResultsIn the observation group,312 cases were mild anxiety(89.1％),24 cases were with moderate anxiety (6.9％),14 cases were with severe anxiety (4.0％).In the control group,32 patients with mild anxiety (9.1％),237 cases were with moderate anxiety(67.7％),81 patients were with severe anxiety(23.2％),the difference was significant.In the observation group,postpartum vaginal bleeding was(48.6 ± 9.7) ml,time length of the size of the uterus return to normal was(4.5 ± 3.1 ) weeks,the incidence of postpartum urinary retention 6 hours after delivery was 1.4％,colostrum time (16.4 ± 5.3) h,and fasting time was (12.4 ± 2.1 ) h.In the control group,postpartum vaginal bleeding was (56.5 ± 11.2) ml,time length of the size of the uterus return to normal was(7.1 ± 4.2) weeks,the incidence of postpartum urinary retention 6 hours after delivery was 6.9％,colostrum time was (23.5 ± 4.7) h,and fasting time was (15.3 ± 2.6) h,the difference was significant.Conclusions One to one whole process companion delivery model helps reduce maternal anxiety,fears,reduce the incidence of postpartum unfavorable situation effectively and promote early resumption of maternal,it is worthy of clinical application.

Objective: To investigate the effects of the exogenous testosterone propionate on apoptosis of rat germ cells and the mechanisms thereof. Methods: Thirty 35-day-old male SD rats were randomly divided into experimental group and the control group. The rats in experimental group were injected (i.m.) testosterone propionate and the control group with an equal volume of saline. By using terminal deoxynueleotidy transferase nediated dUTP nick-end labeling (TUNEL), flow cytometry (FCM), radioimmunoassay (RIA) and electron microscopy, the quantity and quality of apoptosis of germ cells were evaluated. Results:(1) Compared with the control, the apoptotic number of rat germ cells was increased in the experimental group, especially the primary spermatocyte. The apoptotie rate was 11.3% detected by FCM in experimental group,while 3.6% in the control group (P ＜ 0.01). (2) The percentages of 1C were 21.8% in experimental group and 33.8% in control group (P ＜ 0.01).The percentages of 2C were 52.6% in experimental group and 37.1% in control group (P ＜ 0.01). (3) The serum levels of testosterone were (3 486.8±333.3) ng/L in experimental group and (846.9±167.5) ng/L in control group (P ＜ 0.01). The serum levels of follicle-stimulating hormone (FSH) were (2.5±0.8) IU/L in experimental group and (5.2±1.7) IU/L in control group (P ＜0.01). Conclusion: The exogenous testosterone propionate might induce apoptosis of germ cells by retroinhibition of the hypothalamie-pituitary-gonadal axis, thus having contraceptive effects.

Objective:To investigate the effects of titanium surface modified by micro-arc oxidation (MAO)on the collagen secre-tion and the expression of osteogenesis-related genes of MC3T3-E1 cells.Methods:Pure titanium discs were divided into two groups:Titanium with modified surface by MAO(MAO group)and titanium with polished surface(PT group).Tissue culture polysty-rene plates (TC)were used as controls.Surface morphology of the samples was examined by field-emission scanning electron micros-copy(FESEM).Surface roughness of samples was measured by surface roughness meter.MC3T3-E1 cells were cultured on the sur-face of the samples and the collagen secretion on the samples was measured by sirius red-based colorimetric microassay after 12 days of culture.On day 16 of culture,the expression of osteogenesis-related genes was examined by RT-PCR.Results:A porous oxide layer was observed on the surface of MAO treated samples and the surface roughness was more than that of PT group(P<0.01 ). Cells in MAO group showed higher collagen secretion than in PT or TC groups(P<0.05).MAO treated surface induced higher OSX, COL-Iα1 and OPN expression than PT.Conclusion:MAO treated titanium surface may induce more collagen secretion of MC3T3-E1 cells than PT.

Objective To study the effect of different iodine nutrition on thyroid function in adult and pregnant women.Methods A random sampling method was used to select healthy adult and pregnant woman from the communities of coastal city,coastal rural and inland rural areas in Liaoning Province.Drinking water,urine and salt samples were collected to measured urinary iodine (U I),salt iodine (SI) and water iodine content.Fasting venous blood was collected to measured thyroid stimulate hormone (TSH),freethyroxine (FT4),free triiodothyronine (FT3),thyroglobulin antibody (TgAb) and thyroid peroxidase antibody (TPOAb) with the method of immunoassay chemical luminescence.Results A total of 150 salt samples were collected,means of SI was (30.1 ± 6.0)mg/kg.A total of 72 pregnant woman and 271 adults were investigated in iodized salt supplied regions.median UI of pregnant woman and adults were 176.3,203.2 μg/L.Iodine nutrition of pregnant women and coastal region adults was in an adequate level.Means of SI of inland adults(244.4 μg/L) was higher than appropriate level but not reached the excessive level.FT4 of the adults (11.7 pmol/L) and pregnant women (10.7 pmol/L) in inland regions were slightly higher than that of coastal city,rural adults(11.2,8.6 pmol/L) and pregnant women (10.9,9.6 pmol/L).TSH,FT3 and FT4 were not statistically different between regions (all P ＜ 0.05).But UI,FT4 and FT3 levels of pregnant women(176.3 μg/L,9.5 pmol/L,4.3 pmol/L) were significantly lower than that of the adults(203.2 pμg/L,11.3 pmol/L,4.7 pmol/L,all P ＜ 0.05).Hypothyroxinemia(4.4％,10/173) was higher than that of the inland adults (2.0％,2/98,P ＜ 0.05).And all hypothyroxinemia were found in women of childbearing age.Hypothyroxinemia prevalence of pregnant women(16.7％,12/72) was higher than that of adults(4.4％,12/271,P ＜ 0.05),The prevalence of hypothyroidism,hyperthyroidism and subclinical hyperthyroidism between the 3 regions adults and pregnant women were not statistically different (all P ＞ 0.05).Conclusions Under appropriate supply conditions of iodized salt,iodine nutrition and thyroid function are closely related.Pregnant women and women of childbearing age are at risk of iodine deficiency.The thyroid function of these people should be strengthen detect.

Objective To observe the effect of gene gun transduction of K-RAS Antisense gene on the expression of K-RAS P21 protein in human pancreatic carcinoma cells.Methods K-RAS Antisense gene was transduced into pancreatic carcinoma cells by gene gun transduction, the expression of K-RAS P21 protein on BxPC-3、AsPC-1 and MiaPaCa-2 pancreatic carcinoma cells line were examined by western blot and immunocytochemistry staining. Results The expression of K-RAS P21 protein in AsPC-1 and MiaPaCa-2 pancreatic carcinoma cells was obviously lower after the transduction of specific K-RAS Antisense gene,which has little impact on BxPC-3 pancreatic carcinoma cells. Conclusion Gene gun transduction of K-RAS Antisense gene is a potential method for treatment of pancreatic carcinoma.

Objective To study the relationship between the changing law of cytokine gene expression profile and tumor-related fatigue and to verify the preliminary theory conclusion that the pathogenesis of tumor-related fatigue may be due to the up-regulation of the TNF,TGF-?,etc.Methods The changing characteristics of gene expression profile of cytokines were studied in peripheral blood mononuclear cells samples with gene chip.Results The gene expression level of TNF-? and TGF-?1 was up regulated.Conclusions The experimental results conform to the theory research conclusion and show that the pathogenesis of tumor-related fatigue due to the up-regulation of the gene expression level of TNF and TGF-?.

Objective To investigate the expression of radiation-induced IER5 protein and screen its potential interaction proteins that may participate in DNA repair process.Methods HeLa cells were irradiated with 4 Gy ionizing radiation.IER5 protein expression in whole cell lysate and in nuclear fraction were detected by Western blot at different timepoints after irradiation.3 × Flag-IER5 pCMV plasmid was constrcuted and the Flag tagged-IER5 expression was verified by Western blot.293T cells were transfected with 3 × Flag-IER5 pCMV plasmid.After irradiation the cells were collected and proteins were extracted.The IER5 interaction proteins were purified using immunoprecipitation and separated by 12％ SDS-polyacrylamide gel electrophoresis.Then the binding proteins were cut from the gel and analyzed by Mass spectrometry.Results The expression of IER5 protein began to increase 4 hour post-irradiation and its peak level was observed at 12 hour post-irradiation,and it lasted until 48 hour after irradiation.The expression level of IER5 protein in whole cell lysate and nuclear fraction were both increased.With the mass spectrometry analysis,a total of 347 proteins and 256 proteins were identified in irradiated and nonirradiated groups,respectively.Fourty one differential proteins were obtained,where 10 proteins were associated with DNA metabolic process and DNA rapair in the irradiated group and the poly(ADP-ribose) polymerase 1 (PARP1) protein was further confirmed by Western blot.Conclusions IER5 protein is an DNA damage related protein,and it may participate in DNA repair process.

Objective To investigate the expression and relationship of TIP30(HIV-1 Tat interactive protein 2), vascular endothelial growth factor (VEGF) and MVD (detected by CD34) in the angiogenesis of human brain astrocytomas. Methods Expression of TIP30, VEGF and CD34 in 19 cases of normal brain tissue and 71 cases of astrocytoma were immunohistochemically examined with Elivision plus two-step method. Results The positive expression of TIP30 could be seen in cytoplasm of neuroglial cells and neurons of 19 normal brain tissues. The positive expression rate of TIP30 in 71 cases of astrocytoma was 33.80 % (24/ 71). The positive expression rate of TIP30 in astrocytoma of different grades was 52 % for grade Ⅱ, 34.78 % for grade Ⅲ and 13.04 % for grade Ⅳ. The positive expression rate of TIP30 in high grade (Ⅲ+Ⅳ) of astrocytoma was found significantly lower than that in low grade(Ⅱ) (χ2=5.71, P <0.05); The expression of VEGF and MVD detected by CD34 in astrocytomas were higher than that in normal brain tissue and increased as the tumor grade increased; In astrocytoma, the negtive correlation was found between the expression of TIP30 and VEGF (r=-0.428, P<0.05); no correlation was found between TIP30 and MVD(r=-0.065, P 0.05); the positive correlation was found between VEGF and MVD(r=0.684, P<0.01). Conclusion The positive expression rate of TIP30 in normal brain tissue is significantly higher than that in astrocytoma. The positive expression rate of TIP30 significantly decreases as the pathological grade of the astrocytoma increases; The expression of TIP30 and VEGF is negatively correlated in astrocytoma.
