1.Ingenane diterpene ester constituents from Tibetan medicine Euphorbia wallichii

Objective To study the chemical constituents from the acetone extract in the root of Tibetan medicine Euphorbia wallichii. Methods The constituents were separated by column chromatography with silica gel and purified by Sephadex LH-20. Their structures were identified on the basis of spectral analysis such as IR, HRESIMS, HRSIMS, and 1D-and 2D-NMR techniques ( 1H- 1H COSY , HMQC, HMBC). Results Six compounds were isolated from the acetone extract in the root of E. wallichii. Their structures were identified as lanosterol (Ⅰ), ingenol-20-myristinate (Ⅱ), ingenol-3-myristinate (Ⅲ), acid (Ⅳ), 1-O-?-L-arabinofuranosyl-(1→6)-?-D-glucopyranosyl-3, 7-dimethyl-oct-2-en-7-ol (Ⅴ), 1-O-galloyl-?-D-glucose (Ⅵ) . Conclusion Ingenane diterpene esters ingenol-20-myristinate (Ⅱ) and ingenol-3-myristinate (Ⅲ) are new compounds and other compounds are found from this plant for the first time. It is the first time that monoterpene disaccharide glycoside, compound Ⅴ, is isolated from the plants of Euphorbia L.
2.Effect of different hemodialysis modes on serum levels of beta 2 microspheres,leptin and hemoglobin in patients with chronic renal failure and safety analysis

Objective To investigate the influence of different hemodialysis modes on serum levels of beta 2 microspheres , leptin and hemoglobin(Hb) in the patients with chronic renal failure (CRF) and to analyze its safety .Methods Sixty inpatients with CRF in our hospital from January 2014 to January 2015 were selected as the research subjects and divided into the common he‐modialysis group (common HD groups) ,hemodialysis filtration group(HDF group) and the common hemodialysis plus hemoperfu‐sion group(HD+ HP group) according to different treatment methods .The levels of serum leptin ,beta 2 microspheres (β2‐MG) , Hb and erythropoietin (EPO) were measured .The occurrence rates of adverse reactions were observed .The living quality was eval‐uated and conducted the comparative analysis .Results Compared with before hemodialysis ,the levels of serum leptin and β2‐MG levels after the first time of hemodialysis and 6‐month hemodialysis in the three groups were decreased ,moreover the levels in the HD+ HP group were the lowest ,the differences were statistically significant (P<0 .05) ;compared with the common HD group , the levels of Hb and EPO after the first time of hemodialysis and 6‐month hemodialysis in the HDF group and HD+ HP group were increased ,the differences were statistically significant (P<0 .05);the incidence rate of complications after hemodialysis in the HD +HP group was lower than that in the common HD group and the HDF group ,the differences were statistically significant (P<0 .05);the living quality scores of each item after hemodialysis in the HDF group and HD + HP group were superior to those in the common HD group ,moreover the living quality scores in each item in the HD + HP group were higher ,the differences were statisti‐cally significant(P<0 .05) .Conclusion Adopting the HP therapy in the patients with CRF can better reduce the levels of blood leptin andβ2‐MG without affecting the Hb level ,moreover with low occurrence rate of complications ,and the patient′s living quali‐ty is significantly improved after treatment .
3.Application and prospect of optical coherence tomography in the diagnosis and management of ocular surface squamous neoplasia

As a non-invasive examination method, anterior segment optical coherence tomography (AS-OCT) has huge potential in detecting neoplastic epithelial lesion.With AS-OCT, detection and differentiation of ocular surface squamous neoplasia (OSSN) within ocular surface pathologies can be achieved and the diagnosis rate is comparable to that of biopsy.With the development of technology, AS-OCT imaging mode has developed from time-domain OCT to spectral-domain OCT with higher penetrance and resolution, the high-quality images of which are helpful to distinguish the benign and malignant OSSN, enhance the detection rate of OSSN, improve the differentiation of ocular surface tumors, and thus improve the diagnostic accuracy of OSSN.In addition, AS-OCT can help optimize the treatment regimen and effectively monitor the outcome of the disease in the clinical management stage, such as monitoring the recurrence of OSSN and evaluating chemotherapy.In this article, the morphological features of OSSN, the advantages and disadvantages of OSSN auxiliary examinations, the development of AS-OCT, the specific manifestation of OSSN on AS-OCT, the application of OCT in atypical OSSN, as well as the value and limitation of AS-OCT in the diagnosis and management of OSSN were reviewed.
4.Factors influencing the returning of plateletpheresis donors

【Objective】 To explore and analyze on the factors that influence the returning of plateletpheresis donors. 【Methods】 A total of 1 302 plateletpheresis donors, selected from 2014 to 2020 in the blood center, were followed up for 1 year, and their re-donations were recorded. The data of factors were analyzed through Univariate analysis and Logistic regression on statistical software. 【Results】 From 2014 to 2020, there were 1 302 donors participated first-time plateletpheresis donation, including 1070 males and 232 females, aged 18-55 years old, with an average age (28.69±12.90) of years old. As to the education level, 539 cases were junior high school education level and below, 435 cases senior school or technical secondary school education level, and 328 cases junior college education level and above. Within the 1 302 first-time platelet donors, 16 developed adverse reactions to blood donation. 455 donors donated one dose, and 847 cases donated two doses. 506 were public or permanent employed, and 796 were unemployed. 1 267 were ethnic Hans and 35 were ethnic minorities. 21.81%(284/1 302) returned to donate within one year. Univariate and multivariate Logistic regression analysis showed that gender, age, educational level, whether suffered adverse reactions during the first-time donation, doses of donation, ethnicity and occupations all affected the returning of plateletpheresis donors. 【Conclusion】 Plateletpheresis donors with younger age, lower education level, unemployed status, ethnic minorities, no adverse reactions occurring during the first-time donation, and donation history of two doses are more likely to return to donate.
5.Effects of remifentanil and fentanyl on N-methyl-D-aspartate receptor currents in rat spinal cord dorsal horn neurons

Objective To investigate the effect of remifentanil and fentanyl on N-methyl-D-aspartate (NMDA) receptor currents in rat spinal cord dorsal horn neurons.Methods Whole-cell patch-clamp technique was used to record the NMDA receptor currents.The primary cultured E14SD rat spinal cord dorsal horn neurons (DH cells) were randomly divided into 3 groups (n =10 each):remifentanil group (group R),fentanyl group (group F) and control group (group C).DH cells were perfused with 4 nmol/L remifentanil (group R) or 10 μmol/L fentanyl (group F) for 60 min followed by washout.NMDA receptor currents were recorded immediately after administration (T0),at 15,30,45 and 60 min of action of drugs (T1-4),and at 15 and 30 min (T5-6) after washout.Results Compared with group C,no significant change in the peak NMDA receptor current was found at each time point in group F and at T0 and T1 in group R (P > 0.05),and the peak NMDA receptor current was significantly increased at T2-6 in group R (P < 0.01).The peak NMDA receptor current was significantly higher at T2-6 than at T0,while lower at T2-4 and T6 than at T5 in group R (P < 0.01).Conclusion Remifentanil can increase NMDA receptor function in rat spinal cord horn neurons,and the peak effect is reached after washout,but fentanyl dose not have the effect.
6.Using damaged cardiomyocytes onditional induction culture medium to induce the differentiation of human bone marrow mesenchymal stem cells

Objective To investigate the possibility of conditionally induced culture medium (CICM) of oxidative damaged cardiomyocytes(CMs) derived from SD suckling rats to induce the differentiation of human bone marrow mesenchymal stem cells (hMSCs) into cardiomyo-like cells. Methods hMSCs and SD suckling rats' CMs were respectively isolated and cultured in vitro. Beating CMs were damaged by H_2O_2 and the the resultant culture medium wherein the oxidative damaged CMs were euhivated was used as CICM. Passaged hMSCs were induced by CICM for 4 weeks. The morphologies of induced hMSCs were observed by inverted microscope and the special cardio-markers-Cardiac troponin I(cTnI) and desmin were identified by immunocytochemistry method. Results After coincubation with CICM, hMSCs became larger and expressed cTnI and desmin.Conclusion CICM of oxidative injured CMs could induce hMSCs differentiation into cardiomyo-like cells.
7.Preparation process for Tibetan Anshen Pills with supercritical fluid extraction

Objective: To explore the preparation process for Tibetan medicine Anshen Pills. Methods: Comparing the preparation process of Tibetan medicine extracted by SFE CO 2 with that extracted by pertroleum ether and analyzing chemical components of their extracts by GC/MS. Results: The method of SFE CO 2 is superior to extractive method by pertroleum ether. The former has higher extraction ratio and shorter extraction time without residues of chemical solvent. Conclusion: The process of SFE CO 2 is suitable for the preparation of Tibetan medicine Anshen Pills.
8.Induced eruption techniques of maxillary impacted canines with labial medial inclination

Objective To explore induced eruption techniques of the impacted maxillary canine with labial medial inclination. Methods A total of 38 cases with upper impacted canines with labial medial inclination were selected. The impacted canine was connected to the buccal hook of the upper first molar band, moved distantly and uprightly and then drawn to the dental arch after the pressure to the adjacent teeth was removed. Results After average 18 months treatment, 38 patients had good functions. The deeper impacted canines in 5 patients were extracted, and then the dental arch spaces closed. The lateral incisors of root resorption in 4 patients were extracted. The premolars in 19 patients were removed, and 10 patients without extractions. Then the impacted canines were moved into dental arch. There was no aggravated root resorption during the treatment process. Conclusion When the maxillary canine with labial medial inclination is impacted, it is moved distantly and uprightly by elastic line connected to the buccal hook of the maxillary first molar band, and then drawn to the dental arch. The process is smoothly and the neighbor teeth are protected.
9.Role of 5-azacytidine in differentiation of human mesenchymal stem cells into cardiomyocytes in vitro

Objective 5-azacytidine could induce the diffcrentiation of stem cells into cardiomyocytes (CMs). The aim of this study was to screen the optimal condition for 5-azacytidine inducing differentiation of human mesenchumal stem cells (hMSCs) into CMs, and the effect of 5-azacytidine on adherence, cell vigor and chromosome karyotype ofhMSCs. Methods hMSCs were isolated from human bone marrow and cultured in vitro. The phenotypes ofhMSCs were identified by flow cytometric analyses. MTT test was used to investigate the effect of different concentrations of 5-azacytidine on proliferation ofhMSCs. Four weeks after 5-azacytidine induction, semi-quantitative RT-PCR, transmission electron microscopy (TEM), single-cell action potentials, detection ofcardio-cnzyme AST and LDH, cell adherence, cell viability and chromosome karyotype test were performed. Results The typical morphological features of hMSCs were fibroblast-like in shape, hMSCs expressed CD44 and CD105,and did not express CD34, CD45 and CD31. The optimal concentration of 5-azacytidine was 10μmol/L. The shape ofhMSCs treated with 5-Azacytidine changed from fusiform to polygon or astrocyte gradually, and passaged cells were evenly arranged as polarity structure. Induced-hMSCs connected with neighbouring cells, forming myotube-like structures 4 weeks later. It was confirmed that induced hMSCs shaped myotubule-like structure and had some of micro-histologic structures of CMs by TEM. RT-PCR showed that induced hMSCs expressed cardiac specific product BNNP and early cardio-myogenesis specific transcription factor NKX2.5mRNA. Besides, induced-MSCs led to the weak action potential and secreted cardio-enzyme AST and LDH. There was no significant difference in cell adherence and viability before and after induction. Both hMSCs and induced-hNSCs kept stable normal diploid nucleus. Conclusion The optimal condition for inducing effect of 5-azacytidine is 10 μ mol/L and 24-hour incubation; and under this condition, the adherence, vigor and chromosome karyotype ofhMSCs would not be affected.
10.Isolation,cultivation and identification of human embryonic fibroblasts and expressions of cytokines essential for growth of human embryonic germ cells in vitro

Objective To isolate,cultivate and identify the human embryonic fibroblasts(hEFs) derived from the gonadal ridges and dorsal mesenteries of human embryos,and to detect the expression by hEFs of cytokines crucial for the growth of human embryonic germ cells(hEG) in vitro.Methods The hEFs were isolated by enzyme digestion from gonadal ridges and dorsal mesenteries of 5-to 9-week old human embryos.The cells were then cultivated.The biologic characteristics(morphology,growth characteristics and cell cycle) of these cells were also studied.The reverse transcription-polymerase chain reaction(RT-PCR) was used to seek the expressions of a specific fibroblast marker,prolyl 4-hydroxylase ?,and a specific marker of epithelial cells,cytokeratin-4,in the cells.The expressions of cytokines essential for the growth of hEG,namely basic fibroblast growth factor(bFGF) and leukemia inhibitory factor(LIF),were also examined by RT-PCR.Results The hEFs were successfully isolated and cultivated from gonadal ridges and dorsal mesenteries of human embryos.They could be passaged beyond the 25th generation.The biologic characteristics of the cells did not change,even in high-passage cells or frozen-thawed cells.The cells expressed prolyl 4-hydroxylase ?, but not cytokeratin-4,which was similar to the fibroblasts.The cultured cells expressed bFGF and LIF.Conclusion The hEFs derived from gonadal ridges and dorsal mesenteries of human embryos are successfully isolated and cultivated,and the cells express the cytokines essential for the growth of hEG in vitro.
