1.Development of PET agent for cardiac sympathetic nervous system

Many cardiac sympathetic changes occur before clinical symptoms being found in heart disease.PET/CT imaging can sensitively detect abnormal cardiac sympathetic nerve function.The recent development of several 11 C and 18F labeled cardiac sympathetic imaging agents is reviewed in this paper.11C-meta-hydroxyephedrine (11 C-HED) and 6-18F-fluorodopamine (18F-FDA) have been comprehensively studied already.N-11C-CH3-dopamine (11C-MDA) and N-(3-bromo-4-(3-18F-fluoropropoxy)-benzyl)-guanidine (18F-LMI1195) are novel imaging agents with potential for clinical application.
2.Research progress of norovirus

ABSTRACT:Norovirus infection in human and animals can cause acute viral gastroenteritis .With molecular biology ,cell culture and animal models of development ,the different groups of norovirus genome sequenced ,the virus capsid protein ex‐pressed in vitro in eukaryotic ,prokaryotic and plant systems ,and thus the characteristics of the virus with some new under‐standing and perspective .Recently ,an increasing number of studies were focused on the norovirus and related topics along with the research progress all around the world .Here we systematically expound the latest advances in norovirus research from dif‐ferent aspects ,such as genome structure and function ,cell culture ,tissue tropism ,epidemiology ,virus infection mechanism , detection methods and vaccine development .Furthermore ,different viewpoints for norovirus molecular evolution ,detection technology and transmission route will be discussed as well .
3.Control rate of blood glucose and its influencing factors in type 2 diabetes mellitus patients in Yulin City

【Objective】 To investigate the control rate of blood glucose and its influencing factors in type 2 diabetes mellitus patients in Yulin area. 【Methods】 We selected the adult type 2 diabetes patients who visited our hospital from May 2020 to December 2021 as the subjects. Then we collected their basic information (gender, age, household income, type of medical insurance payment, education level, and duration of disease), measured their height and weight, calculated their body mass index (BMI), detected HbA1c, and measured their subcutaneous and visceral fat. The Chi-square test and multivariate Logistic regression methods were used to analyze the influencing factors. 【Results】 The total attainment rate of HbA1c (HbA1c<7%) among 877 adults with type 2 diabetes was 13.34%. The Chi-square test showed that statistical differences in the attainment rate of HbA1c among different ages, family annual income, type of medical insurance, and duration of disease. Further unconditional multivariate Logistic regression analysis model results showed that the HbA1c attainment rate in 18-44 years old group was 0.418 times higher than that in ≥60 years old group (95% CI=0.219-0.799, P=0.008). The HbA1c compliance rate of patients with employees’ medical insurance was 1.744 times that those with residents’ medical insurance (95% CI=1.131-2.782, P=0.013). The HbA1c attainment rate of diabetic patients with an annual family income of 30 000 yuan to 100 000 yuan was 1.873 times (95% CI=1.074-3.266, P=0.027), and with an annual family income of more than 100 000 yuan was 2.649 (95% CI=1.299-5.404, P=0.007) times than that of diabetic patients with less than 30 000 yuan. 【Conclusion】 The blood glucose control rate in adults with type 2 diabetes mellitus in Yulin area is lower compared with the level of the nation and other regions. Age, annual household income, and type of medical insurance payment are independent influencing factors.
4.Isolation,cultivation and identification of human embryonic fibroblasts and expressions of cytokines essential for growth of human embryonic germ cells in vitro

Objective To isolate,cultivate and identify the human embryonic fibroblasts(hEFs) derived from the gonadal ridges and dorsal mesenteries of human embryos,and to detect the expression by hEFs of cytokines crucial for the growth of human embryonic germ cells(hEG) in vitro.Methods The hEFs were isolated by enzyme digestion from gonadal ridges and dorsal mesenteries of 5-to 9-week old human embryos.The cells were then cultivated.The biologic characteristics(morphology,growth characteristics and cell cycle) of these cells were also studied.The reverse transcription-polymerase chain reaction(RT-PCR) was used to seek the expressions of a specific fibroblast marker,prolyl 4-hydroxylase ?,and a specific marker of epithelial cells,cytokeratin-4,in the cells.The expressions of cytokines essential for the growth of hEG,namely basic fibroblast growth factor(bFGF) and leukemia inhibitory factor(LIF),were also examined by RT-PCR.Results The hEFs were successfully isolated and cultivated from gonadal ridges and dorsal mesenteries of human embryos.They could be passaged beyond the 25th generation.The biologic characteristics of the cells did not change,even in high-passage cells or frozen-thawed cells.The cells expressed prolyl 4-hydroxylase ?, but not cytokeratin-4,which was similar to the fibroblasts.The cultured cells expressed bFGF and LIF.Conclusion The hEFs derived from gonadal ridges and dorsal mesenteries of human embryos are successfully isolated and cultivated,and the cells express the cytokines essential for the growth of hEG in vitro.
5.Inhibition of diterpenoid tanshinones from Salvia miltiorrhiza Bge.on xanthine oxidase activity

AIM Inhibitors of xanthine oxidase (XOD) may be potentially useful for the treatment of gout or other XOD inducing diseases, so the inhibition on XOD activity of diterpenoid tanshinones such as cyptotanshinone (CT) and methylenetanshinone (MT) from Tanshen (Salvia miltiorrhiza Bge.) was studied. METHODS The formation rate of uric acid from xanthine was determined by measuring the absorbance increment at 290 nm (ΔA290 nm) in the reactive medium of xanthine/XOD when CT or MT was added. RESULTS It was found that CT and MT inhibited XOD activity. Dixon plots showed that the inhibition mode was competitive type. The Ki values of CT and MT were 17.8 μmol·L-1 and 25.9 μmol·L-1, respectively. Their inhibitory potencies positively correlated with their concentrations and the IC50 values of CT and MT were 70 and 67 μmol·L-1, respectively. The IC50 value of allopurinol, the positive control, was 60 μmol·L-1. CONCLUSION CT and MT have inhibitory effects on XOD activity and may be potentially useful for the treatment of gout or other XOD inducing diseases.
6.Role of apoptosis related gene Bcl-2 and Bax during periodontal ligament remodeling in rats

Objective: To study the influence of occlusal force changes on apoptosis related gene Bcl-2 and Bax in rat periodontal ligament fibroblasts (PDLF). Furthermore, to explore the possible mechanism in remodeling of periodontal ligaments. Methods: Animal model of occlusal force changes was established by extracting the right first, second and third maxillary molars in male SD rats. Rats were sacrificed at 6,12 hours and 1, 2, 3, 5, 7, 14 and 28 days after teeth extraction(n=6), and their alveolar bone tissues were harvested. HE staining was used to observe the morphologic changes of alveolar bone tissues and immunohistochemistry were used to detect Bcl-2 and Bax expressions in PDLF. The rats of normal occlusal forces were used as control(n=6). The results were assigned a mean score based on the expressions and the intensity of cell positively stained for Bcl-2 and Bax. Results: Animal model of occlusal force changes was successfully established. HE staining showed that the control group had more pycnotic periodontal ligament, orderly aligned fibres, and flatter alveolar bone than model group. Immunohistochemical staining showed that expression of Bax reached its peak at 12 h after extracting the teeth(267.00?7.82), significantly higher than that of control group(25.67?4.97,P

【Objective】 To investigate the practice and benefit of national standardized management of type 2 diabetes in Yulin City. 【Methods】 We recruited the adult type 2 diabetes patients who sought medical help at our hospital from May 2020 to October 2022 as subjects. We collected their basic information (sex and age); measured height, weight, waist and hip circumference, and blood pressure; calculated body mass index (BMI); and detected blood glucose, c-peptide, HbA1c, biomarkers, urinary microalbumin, sensory nerve conduction velocity of lower limbs, ABI, and subcutaneous and visceral fat at the time of MMC recruited and the end of six months. T test and Mann-Whitney U rank sum test were used for measurement data and χ2 test or Fisher’s exact probability method for counting data to analyze the data. 【Results】 After 6 months, the levels of fasting blood glucose, postprandial blood glucose, HbA1c, and visceral and subcutaneous fat in all the patients decreased, but the level of fasting c-peptide increased compared with the baseline (all P<0.05). Secondly, compared with the baseline, the control rate of HbA1c (35.21% vs. 13.71% ) and the comprehensive control rate (13.97% vs. 7.26% ) were both significantly increased at six months (P<0.05). Thirdly, after 6 months, the levels of fasting blood glucose, postprandial blood glucose, HbA1c, TG, TC, and UA were decreased more, while the fasting c-peptide and postprandial c-peptide were increased more in the patients of the HbA1c standard group (HbA1c<7% ) than those of the non-standard group. 【Conclusion】 The multiple benefits of blood glucose, blood lipid, uric acid and islet function can be achieved by taking type 2 diabetes patients into MMC. Meanwhile, the rates of HbA1c control and comprehensively reaching the standard are significantly increased. Therefore, MMC can explore a new way for the management of type 2 diabetic patients in this area.
9.Assessment of effects of 7,8-dihydroxyflavone on the striatum in normal cynomolgus monkeys with 99TCm-TRODAT-1 SPECT/CT imaging

Objective To assess the effects of 7,8-dihydroxyflavone (7,8-DHF) on the striatum (ST) in normal cynomolgus monkeys using 99Tcm-TRODAT-1 imaging.Methods A total of six healthy female cynomolgus monkeys were included in this study.Three of them were fed with normal food (control group),and the other three were given oral administration of 7,8-DHF in addition to normal food (experimental group).The SPECT/CT imaging was performed at different time after 99Tcm-TRODAT-1 injection.The ROI of ST was drawn on images of 3 consecutive transverse slices that could be visualized best.The cerebellum (CB) was taken as the background reference area.The radioactivity uptake ratios of ST/CB at 1,3,4 and 5 h were calculated respectively.Paired-t test was used to analyze the data.Results ST radioactive uptake ratios showed continuing increase on the delay images.ST/CB uptake ratios of the control group at 1,3,4 and 5 h were 1.43±0.04,1.82±0.06,2.04±0.12,2.42±0.23,respectively,and those of the experimental group were 1.35±0.08,2.40±0.09,2.74±0.13 and 3.25±0.15 respectively.There was no significant difference between the two groups at 1 h (t =2.57,P>0.05),while ST/CB uptake ratios of the experimental group at 3,4 and 5 h were significantly higher (t values:2.77,2.87 and 2.92,all P<0.05).Conclusion 99Tcm-TRODAT-1 SPECT/CT imaging can be used to assess the DAT activation effect by 7,8-DHF on ST of cynomolgus monkeys.
10.Observation of intra-articular Hydromorphone with Ropivacaine for analgesia after knee arthroscopy

ObjectiveTo assess the effects of intra-articular Hydromorphone with Ropivacaine for postoperative analgesia after arthroscopic knee surgery.Methods 90 patients undergoing arthroscopic knee surgery were randomly divided into 3 groups. Group R: 0.375% Ropivacaine 20ml; group H1: Hydromorphone 0.3 mg and 0.375 % Ropivacaine 20 ml; group H2: Hydromorphone 0.6 mg and 0.375 % Ropivacaine 20 ml. The visual analogue scale (VAS) at rest were recorded at 6, 12, 18 and 24 h after surgery, Duration of analgesia, number of patients and frequency requiring Parecoxib at 24 h after surgery were observed.Results Compared with group R, VAS of group H1 and group H2 were signiifcantly lower at 12 and 18h after the operation, duration of analgesia was much longer, number of patients and frequency requiring Parecoxib was lower in group H1 and H2 (P < 0.05); Compared with group H1, No signiifcant differences of VAS, duration of analgesia and number of patients and frequency requiring Parecoxib of group H2.Conclusions After knee arthroscopic surgery, intra-articular 0.3 mg Hydromorphone can signiifcantly improve the efifcacy of Ropivacaine for postoperative analgesia; the efifcacy of Hydromorphone can’t increased with the increase of dosage.
