1.Effect of self-made atractylodes and spleen decoction combined with quadruple therapy on Hp-positive peptic ulcer and Its effect on wound healing, gastric secretion and recurrence

Objective To observe the effect of self-assembling Atractylodes Rhizome Decoction combined with quadruple therapy on Helicobacter pylori (Hp)-positive peptic ulcer and its effect on wound healing, gastric acid secretion and recurrence.MethodsThis study was based on 70 patients with Hp-positive peptic ulcer treated in our hospital from March 2012 to May 2014.The patients were divided into study group and control group according to the order of admission.(BAO), the maximum gastric acid secretion (MAO), the peak gastric acid secretion (P<0.05).The treatment group is divided into two groups according to the treatment group.(PAO), Hp eradication and recurrence were observed and compared.ResultsAfter treatment, the total effective rate of wound healing study group was 80.00% higher than that 57.14% of the control group (χ2=4.242, P<0.05).The levels of BAO, MAO and PAO in the two groups were significantly lower than those before treatment (P<0.05).The eradication rates of Hp in the study group were significantly higher than those in the control group at 6 months and 1 year follow-up.The recurrence rate was significantly lower in the study group than in the control group (P<0.05),There was no significant difference between the two groups.ConclusionThe clinical effect of self-made Baishui Yipi Decoction combined with quadruple therapy on Hp-positive peptic ulcer patients is effective in promoting wound healing and Hp eradication, and inhibiting gastric acid secretion and reducing the recurrence rate.It is a kind of medicine worthy of clinical application Program.

Objective To examine the clinical factors associated with postoperative outcome in dementia combined with cerebral amyloid angiopathy-related intracerebral hemorrhage (CAA-ICH). Methods The clinical data of 25 surgical patients of dementia combined with CAA-ICH were retrospectively analyzed. The postoperative short-term effects and long-term effects were evaluated at the 30 d and 6 months after operation according to the modified Rankin score (MRS), and the influencing factors were analyzed. Results Among the 25 patients, eusemia was in 6 cases. Age ≥ 75 years, Glasgow coma score (GCS) ≤ 8 scores on admission, hypertension, postoperative anemia, pulmonary diseases, midline shift were the influencing factors of postoperative short-term effects (P < 0.05 or <0.01). Twenty-two patients were followed up for 6 months, and ensemia was in 9 cases. Age≥75 years, GCS ≤ 8 scores on admission, hypertension, postoperative anemia, hypoproteinemia and pulmonary diseases were the influencing factors of postoperative long-term effects (P<0.01 or<0.05). Conclusions Preoperative assessment is important for the patients of dementia combined with CAA-ICH. Age ≥ 75 years, GCS ≤ 8 socres on admission, hypertension, postoperative anemia, pulmonary diseases, midline shift are associated with poor short-term effects; age ≥ 75 years, GCS ≤ 8 scores on admission,hypertension, postoperative anemia, hypoproteinemia and pulmonary diseases are associated with poor long-term effects.

Objective:To investigate if the combined use of CDAⅡ and sodium butyrate can induce demethylation and re-expression of retinoic acid receptor?2(RAR?2)gene in cultured human breast cancer cells MCF7.To explore if the two drugs can inhibit cell growth and induce cell apoptosis synergetically.Methods:MCF7 cell line was treated with CDAⅡ,sodium butyrate,combination of the two drugs respectively.Methylation was assessed by methylation-specific polymerase chain reaction(MSP)for RAR?2 gene.Gene expression was evaluated by reverse transcription polymerase chain reaction(RT-PCR).Apoptosis was detected by terminal deoxynucleotidyl transferase mediated dUTP-biotin nick end labeling(TUNEL)and Hoechst33342/propidiumiodide(PI)staining.Cell growth inhibition was measured by MTT assay.Results:Neither CDAⅡ nor sodium butyrate induced demethylation and re-expression of RAR?2 gene,Combination of the two drugs partially demethylated gene promoter accompanied by re-expression of RAR?2.The apoptotic cells in the double-drug group were obvious following Hoechst33342/PI staining.The percentage of apoptotic cells in the double-drug group was significantly higher than that of the two single-drug group(39.5% vs 5.2%,8.1%)(P
5.Effects of three-month rehabilitation on motor function in hemiplegics after stroke

ObjectiveIn order to study the effect of three-month rehabilitation on motor function in hemiplegics after stroke. Methods125 patients were randomly divided into two groups, rehabilitation group (72 cases)and control group(53 cases).Patients in the rehabilitation group were given clinical treatment,electric stimulation therapy(EST) and regularly physical therapy, while those in control group were given clinical treatment and unguided self-training, and the evaluation was done in pre-treatment and post-three-month respectively. Motor function was assessed in Fugl-Meyer Assessment(FMA).ResultsMotor scores in each group had a more significant difference(P<0.001)before and after treatment, the process of scores in the rehabilitation group was obviously superior to that in the control group(P<0.001). Conclusions Rehabilitation training on hemiplegics after stroke may obviously improve motor function, depressing the disability and increasing the living quality.
6.Clinical effect observation of 125I seed implantation combined with endocrinal therapy for prostate cancer

Objective To retrospectively study the efficacy and side-effect of 125I seed implantation combined with endocrinal therapy in stage T3N0M0 prostate cancer.Methods The study included 22 patients with clinical stage T3 N0 M0 prostate cancer who were treated with transperineal 125I seed implantation guided by transrectal ultrasound,real time TPS and endocrinal therapy.The minimum peripheral doses (MPD) were 140-160 Gy.The median number of seeds was 74(26-90).The activity of each seed was 1.55 × 107 (1.30 × 107-1.85 × 107) Bq.11 patients were treated with orchidectomy,and 11 patients were treated with androgen deprivation therapy.Results All 22 patients completed the seed implantation successfully.The 5-year biochemical progression-free survival was 70.6%,and 5-year overall survival was 81.8%.2 patients were found biochemical failure in 12 months after seed implantation,and another 1 patient failed in 90 months.Endocrinal therapy was followed thereafter.After the seed implantation,the urinary complications of grade 1 and 2 were 54.5% and 9.1% respectively,and the rectum side-effect of grade 1 and 2 were 22.7% and 9.1%.1 patient suffered rectal complication of grade 4.Conclusions Good effect and tolerance are observed in prostate cancer patients of stage T3N0M0 receiving 125I seed implantation plus endocrinal therapy.The treatment can be considered for those who refuse to receive external beam radiotherapy.
7.Application of virtual-reality simulator for the training of ureteroscopy

ObjectiveTo assess the validity of virtual-reality simulator UroMentorTM in skill training of ureteroscopy. MethodsThirty urologists were included and divided into groups A (n =18) and B (n =12 ) based on former ureteroscopy experience ( ≥ 20 or < 20).Participants were assessed on their ability to perform cystoscopy,gnidewire insertion,semirigid ureteroscope advancement and basket extraction of a distal ureteric stone on the simulator.A blinded examiner assessed the subjects' performance using global rating scale (GRS).In addition,computer-generated parameters including time to complete the task,endoscope and instrument trauma,and the number of attempts to insert a guidewire were recorded as pretest.After 2 days of simulator training,they were retested with the same task. ResultsAll participants had reduced time to completion (333 ± 32 s & 228 ± 18 s,P =0.001 ) and improved GRS (24.4 ± 2.1 & 28.1 ±1.2,P =0.010).Differences were significant between the two groups in the time to completion (before 405 ±40 s & 262 ±22 s,P =0.014; after 276 ± 12 s & 179 ±9 s,P =0.000),and GRS (before 19.6 ±2.5 & 29.2 ± 1.3,P =0.009 ; after 25.0 ± 1.1 & 31.2 ± 0.7,P =0.002).Previous ureteroscopy experience was correlated to GRS (before r=0.705,after r=0.756). ConclusionThe UroMentor virtual-reality simulator is an appropriate and useful tool in training and assessing the skills of ureteroscopy.
8.Human Papillomavirus Type 16 Mutant E7 Protein Induces Oncogenic Transformation via Up-regulation of Cyclin A and cdc25A

A new mutant human papiUomavirus type 16 E7 gene, termed HPV16 HBE7, was isolated from cervical carcinoma biopsy samples from patients in an area with high incidence of cervical cancer (Hubei province, China). A previous study showed that the HPVI6 HBE7 protein was primarily cytoplasmic while wild-type HPV16 E7 protein, termed HPV16 WET, was concentrated in the nucleus. With the aim of studying the biological functions of HPV16 HBE7, the transforming potential of HPV16 HBE7 in NIH/3T3 cells was detected through observation of cell morphology, cell proliferation assay and anchorage-independent growth assay. The effect of HPVI6 HBE7 on cell cycle was examined by flow cytometry. Dual-luciferase reporter assay and RT-PCR were used to investigate the influence of HPVI6 HBE7 protein on the expression of regulation factors associated with GI/S checkpoint. The results showed that HPV16 HBE7 protein, as well as HPV16 WE7 protein, held transformation activity. NIH/3T3 cells expressing HPV16 HBE7 could easily transition from G1 phase into S phase and expressed high level of cyclin A and cdc25A. These results indicated HPV16 mutant E7 protein, located in the cytoplasm, induces oncogenic transformation of NIH/3T3 cells via up-regulation of cyclin A and cdc25A.
9.The study on screening the differences of serum proteins in ulcerative colitis

Objective To search for the differences of serum proteins expression in ulcerative colitis (UC) by proteomics method and to preliminary explore the potential biological markers of ulcerative colitis. Method The serum of 30 ulcerative colitis patients and 30 healthy individuals were collected. The equal amounts of proteins in pooled serum were separated by two-dimensional gel electrophoresis (2-DE) and then compared and analyzed by image analysis software to recognize the differences of protein expression. Some spots of proteins with different expression were identified by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF-MS).Result There was no statistic significant in age, weight index, smoking and alcohol taking between UC group and control (P > 0.05 respectively). Thirty-nine proteins with significant different expression between UC patients and healthy individuals were preliminary screened out. Nine of those spots were selected, after analyzed by MALDI-TOF-MS, it was found that the expression of haptoglobin, heat shock factor protein 2, receptor tyrosine kinase, aldehyde reductase,apolipoproteinC-Ⅲ, pericentriol material 1 increased in ulcerative colitis patients, keratinl, filamin A interacting protein 1 and tropomyosin 3 decreased. Conclusions With proteomics 2-DE and spectrometry methods, nine UC associated serum proteins were screened out and identified, which provide new molecular markers for the research of UC biological behavior.
10.The efficacy and security of adenosine combined tirofiban on patients with percutaneous coronary intervention

Objective To compare the egicacy and security of intracoronary administration of tirofiban combined high-dose adenosine during percutaneous coronary intervention (PCI) in patients with acute myocardial infarction (STEMI).Methods Eighty-eight cases with STEMI were randomly divided into observation group(44 cases) who were accepted 2 times intracoronary adenosine(2 mg,10 ml 0.9% NaCl),and control group(44 cases) who were afforded only 10 ml 0.9% NaCl by prospective,double-blind,and random study.The two groups were received10 g/kg tirofiban after aspiration catheter in the culprit lesion distal bolus injection of 3 rain,at the same time,continuous infusion of 0.15 g/(kg · min) for 24 h.The postoperative coronary arteriography and electrocardiogram were evaluated.Meanwhile,the postoperative myocardial blush grade(MBG),thrombolysis in myocardial infarction (TIMI),corrected TIMI frame counts (CTFC),ST-segment elevation resolution (STR) major adverse cardiac events (MACE),and adverse reactions of adenosine were recorded.Results There was no significant difference in terms of postoperative TIMI and STR between two groups (P > 0.05).The CTFC of observation group was (24.4 ± 4.9) frames,significant better than that of control group((21.9 ±3.7) frames;t =2.701,P <0.01).The ratio of MBG in observation group was 24/44,higher than that of control group(14/44 ; x2 =4.632,P < 0.05).There were no significant difference regarding of the ratio of death,MACE,target vessel revascularization,grade of NYHA between observation and control group at followed up for 1 and 12 month (P > 0.05).The ratio of patients with blood pressure decrease ≥ 10 mm Hg,new second degree atrioventricular block in observation group were 15.9% and 20.5%,higher than that in control group (2.3% and 15.9% ; x2 =4.950,7.221 ; P =0.026,0.007).The adverse reaction was transient.Conclusion The intracoronary administration of tirofiban combined high-dose adenosine during PCI in patients with STEMI plays an effective role on improvement of myocardial perfusion.
