1.Effects of FuzhengJiedu decoction on antioxidant enzyme activity and MDA contents of human bronchial epithelial cells exposed to NiSO_4

Objective: To study the effects of pharmaco-serum from rabbits administrated intragastrically with Fuzheng Jiedu decoction(FJD) on injured human bronchial epithelial(16HBE) cells exposed to nickel sulfate(NiSO4).Methods: Cultured 16HBE cells were treated with both NiSO4 and different doses of FJD pharmaco-serum in vitro.The detections of cell activities and viabilities were carried out.Meanwhile,activities of glutathione peroxidase(GSH-Px) and superoxide dismutase(SOD) as well as maleic dialdehyde(MDA) contents were measured.Results: FJD pharmaco-serum could decrease mortalities and increase viabilities of 16HBE cells exposed to NiSO4.In cells co-treated with NiSO4 and FJD pharmaco-serum,the content of MDA was decreased,while GSH-Px and SOD activities were increased at the same time.High dose of FJD pharmaco-serum had the most dramatic effect.Conclusion: This study suggested that FJD could antagonize NiSO4-induced toxicity,which may be involved in its antioxidant function.
2.Preparation and in vitro-targeted study of liquid perfluorocarbons-filled nanocapsules ultrasound contrast agent

Objective To prepare human vascular endothelial growth factor receptor 2 (VEGFR2)-targeted liquid perfluorocarbons-filled nanocapsules ultrasound contrast agent,and to investigate its ability of targeting human umbilical endothelial vein cells(HUEVC) and the peculiarity of the enhancing ultrasound imaging in vitro.Methods Rotary evaporation/emulsion technique was used to prepare biotinylated liquid perfluorocarbons-filled nanocapsules (LNCs).VEGFR2-tageted LNCs (V2-LNCs) was further made through conjugating the ligand (biotinylated anti-VEGFR2 antibody) to the surface of LNCs by biotin-avidin system and its appearance,distribution,size and zeta-potential properties were assessed.Immunofluorescent staining assay was used to identify the combination of ligand with the nanocapsules.The ability of V2-LNCs targeting with HUEVC in vitro was evaluated under the fluorescence microscope.Quantitative and statistical analysis were performed to evaluate the ultrasound contrast enhancement of the nanocapsules in vitro.Results The V2-LNCs are uniform and stabilized with size about (98.45 ± 0.07)nm.The ligation of antiVEGFR2 antibody and nanocapsules was positive in immunofluorescent straining assay.In vitro,targeting ability research showed the V2-LNCs could actively adhere to HUEVC,while the control was negative.At the same time,using anti-VEGFR2 antibody to pre-incubation with HUVEC could effectively block the interaction between V2-LNCs and HUEVC.The ultrasound images proved both V2-LNCs and LNCs can significantly enhanced ultrasound imaging contrast agent effect at a concentration of 5 mg/ml in vitro.Conclusions A stable VEGFR2-targeted liquid perfluorocarbons-filled nanocapsules was prepared successfully with biotin-avidin method and effectively bound to HUVEC specially in vitro.Also the liquid perfluorocarbons-filled nanocapsules can strongly enhance the ultrasound contrast in vitro,which might be taken as a kind of vascular ultrasound contrast agent and to get the basic experimental data for the later research.
3.Effects of water extract of radix salviae miltiorrhizae on renal injury in rats exposed to cadmium

Objective To explore effects of water extract of radix salviae miltiorrhizae on renal injury in rats exposed to cadmium and its possible mechanism.Methods Twenty male and female healthy Wistar rats,respectively,at age of 6 weeks were randomly divided into blank group (n =8),and model group (n =32) after adaptive feeding for 1 week,given intraperitoneal injection of saline for 5 mL/(kg · d) and intraperitoneal injection of cadmium chloride solution for 2.8 mg/(kg · d).Continuously building for 4 weeks,the general situation of rats was observed,and the rat kidney function and kidney tissue pathological changes were detected.After successful molding,the model group was randomly divided into medicine group,negative and positive and blank control groups.The medicine group was given Salvia miltiorrhiza water extract of high dose of 2.7g/(kg · d),Danshen aqueous extract of low dose 1.35 g/(kg · d) by gavage,positive control group was given sodium selenite for 0.05 mg/kg · d by gavage,the negative and blank control groups were given the same mount of saline for 2.7 mL/(kg · d).After 10 weeks of treatment,serun and urine was collected,and kidney tissue was to be detected.Results Salvia miltiorrhiza water extraction liquid of low and high dose group had less 24 h urine volume than negative control group,and serum superoxide dismutases (SOD) increased significantly,Malondialdehyde (MDA) decreased significantly,total antioxidant capacity (T-AOC) increased significantly,blood urea nitrogen,urinary beta β2-MG content and serum creatinine levels was decreased.Pathological section revealed that renal proximal convoluted tubules epithelial cells was turbid,swelling,degeneration and necrosis in the negative control group.Salvia miltiorrhiza water extraction liquid of low and high dose group had lower serum cortical cadmium content than the negative control group,Salvia miltiorrhiza water extraction liquid of low and high dose group had lower apoptosis rate than the negative control group.Conclusion The salvia miltiorrhiza water solution can accelerate the metabolism of cadmium,and effectively interfere with cadmium induced renal injury,delay chronic renal failure,and protect the kidney.
