1.Association between brominated flame retardants and obesity: a mediation analysis through markers of oxidative stress and inflammation.

BACKGROUND:
Recent studies have provided compelling evidence that exposure to brominated flame retardants (BFRs) can adversely affect human health. We aim to explore the potential impact of BFRs on adiposity and central obesity.
METHODS:
Data from the National Health and Nutrition Examination Surveys (NHANES) cycles conducted between 2009 and 2014 was used to study the connections between variables. After filtering, we analyzed a sample of 4,110 adults aged 20 years and above. Our goal was to examine the potential association between BFRs and consequences and investigate the part played by oxidative stress and inflammatory markers as intermediaries. To achieve this, we used advanced statistical methods such as weighted quantile sum (WQS) regression, quantile-based g-computation (QGC), and the Bayesian kernel machine regression (BKMR).
RESULTS:
The findings showed that among the examined chemicals, exposure to PBDE85 (weight: 41%), PBDE100 (24%), and PBB153 (23%) may be the dominant contributors to general obesity risk. Upon controlling for all variables that could impact the results, it was found that the QGC outcomes indicated a positive correlation between exposure to mixtures of brominated flame retardants and the occurrence of abdominal obesity (OR = 1.187, 95% CI: 1.056-1.334, p = 0.004). Significant contributions were made by PBDE85 (52%), PBB153 (27%), and PBDE100 (21%). Mediation analysis shows that lymphatic cells (LC) and albumin (ALB) partially mediate the link between brominated flame retardants and obesity. The results of BKMR are generally consistent with those of WQS and QGC.
CONCLUSION
At a population level, our research has revealed a noteworthy correlation between BFRs and obesity. However, further investigation is required through prospective cohort studies and in-depth mechanistic exploratory studies.

3.Psychological availability of the full-time candidates for master's degree of nursing during their clinical work:a qualitative study

Objective To explore the psychological availability of the full-time candidates for master degree of nursing during the clinical work. Methods A phenomenological approach was adopted in this study. About 11 full-time candidates for master's degree of nursing took part in the semi-structured and in-depth interviews and the acquired data were analysed. Results About 4 themes promoting psychological availability were identified: strong research knowledge and skills, career development confidence, perception of attention from leaders and more satisfactory salary. The four themes hindering psychological availability were:clinical work tasks, lower level of expertise, lack of social support and lower perception of self benefit. Conclusions The full-time candidates for master's degree hold active perception and much expectation about their clinical work but meanwhile they feel passive and disappointed. Nursing administrators should develop distinguishing and definite clinical personnel training and continuing education programs for them, offer them an access to learning chances and emotional support in order to improve the psychological availability and stabilize the nursing team.
4.Clinical, endoscopic, pathologic and prognostic factors of primary gastric lymphoma

Objective To explore the clinical,endoscopic,pathologic and prognostic characteristics of primary gastric lymphoma (PGL) and to improve the level of diagnosis and treatment.Methods Sixtythree patients who were confirmed as PGL with operation and endoscopic biopsy pathology during January 2001 to December 2010 were retrospectively analyzed with respects of clinical,endoscopic and pathologic features.Survival analysis and prognosis were evaluated by kaplan-Meier and Cox proportional hazard model,respectively.Results In 63 PGL patients,the numbers of male and female were 40 and 23,respectively,and the average age was (59.8±13.3)years.The major symptoms were abdominal pain,abdominal distension,and gastrointestinal hemorrhage,accounting for 47.6 % (30/63),17.5 % (11/63),and 17.5 % (11/63),respectively.There were 39 (61.9 %) PGL patients with endoscopic performance for ulcers,34 (54.0 %) cases involved the gastric stomach antrum.The most immunohistochemistry analyses were diffuse large B-cell lymphoma (DLBCL) (71.4 %,45/63),followed by mucosa-associated lymphoid tissue (MALT) lymphoma (22.2%,14/63).The frequency of Helicobacter pylori (H.pylori) positivity was lower in patients with DLBCL than that in patients with MALT lymphoma (37.8%(17/45) vs 10/14,x2 =4.872,P=0.027).The accumulate survival rates of one,three and five years were 74.6%,63.5%,55.6%,respectively,and the average survival time was (41.5±3.0) months (95% confidence interval (CI) 35.7 to 47.4 months) in PGL patients.There was no difference in the average survival time between DLBCL patients treated with surgery combined chemotherapy and those with surgery or chemotherapy alone (38.33±5.21) months vs (50.17±8.98) months vs (41.39±4.40) months,P>0.05).The patients diagnosed as DLBCL with H.pylori positive had longer average survival time than those with H.pylori negative ((51.90±4.30) months vs (33.30±4.50) months,t=-4.004,P<0.01).Conclusions Male patients with PGL are slightly more than female.Abdominal pain is the most frequent symptom.Ulcerative lesions are the most common endoscopic demonstrations mostly at stomach sinus.DLBCL is the most pathologic characteristic.There is no significant difference in the survival rate between patients treated with surgery combined with chemotherapy and those treated with surgery or chemotherapy alone.
5. Study on the dosage of GMDTC for removing cadmium

OBJECTIVE: To explore different doses of sodium(s)-2-(dithiocarboxylato((2R,3R,4R,5R,6R)-2,3,4,5,6-pentahydroxyhexyl) amino)-4-(methylthio) butanoate(GMDTC) for removing cadmium. METHODS: Thirty-five male New Zealand rabbits were randomly divided into blank control group,GMDTC high dose control group,model control group,ethylene diamine tetraacetic acid(EDTA) control group and GMDTC low,medium and high dose groups,five rabbits in each group. The blank control group and GMDTC high dose control group were given 0. 90% normal saline solution intravenously; model control group,EDTA control group and GMDTC low,medium and high dose group were given 2 μmol/kg of cadmium chloride(CdCl_2) and 40 μmol/kg of β-mercaptoethanol mixed solution intravenously,5. 0mL/kg body weight(bw),once a day for five days. On the forty-one day of the experiment(the fist day of GMDTC treatment),the control group and the model control group were injected 0. 90% normal saline solution 250 mL via ear vein,the EDTA control group was given EDTA solution at the dose of 93. 5 mg/kg bw with 250 mL 0. 90% normal saline solution,also via ear vein; the GMDTC high dose control group,and the GMDTC low,medium and high dose groups were given 250 mL GMDTC solution at the concentration of 108.0,12.0,36.0 and 108. 0 mg/kg bw with 0. 90% normal saline by intravenous infusion,once a day,6 times a week for four consecutive weeks. The urine β_2-microglobulin(MG),renal cadmium,blood cadmium,and urinary cadmium before and after the treatment were detected. RESULTS: The body weight of New Zealand rabbits increased with the increasing feed time(P < 0. 01). The levels of β_2-MG before treatment increased in model control group,EDTA control group,GMDTC low,medium and high dose groups than that in the blank control group(P < 0. 01). The levels of renal cadmium after treatment in GMDTC medium and high dose groups decreased compared with those in the blank control group and EDTA control group respectively(P < 0. 05). The blood cadmium after treatment in EDTA control group,GMDTC low,medium and high dose groups were decreased compared with those before treatment in the same group respectively(P < 0. 05),meanwhile decreased than the blood cadmium after treatment in the model control group respectively(P < 0. 05). The blood cadmium after treatment had not a statistically significant difference among the EDTA control group,GMDTC medium and high dose groups(P < 0. 05). At all the time points(1,6,8,13,15,20,22 and 28 days after treatment),the urinary cadmium after treatment in EDTA control group and the three GMDTC dose groups increased compared to the model control group at the same time(P < 0. 05). The urinary cadmium after treatment increased with GMDTC dose increased at the other six time points,expect on 20 and 22 days after treatment(P < 0. 05). The blood cadmium removal rates after treatment were 70. 06%,74. 86% and 78. 05% and the renal cadmium removal rates were 14. 27%,27. 95% and 61. 24% in GMDTC low,medium and high dose groups,respectively. CONCLUSION: The intravenous infusion of GMDTC at the dose of 108. 0 mg/kg bw effectively removed cadmium in cadmium poisoning rabbit. This dose had no obvious toxic effect and was equivalent to human dose of 36. 0mg/kg bw which meets the requirement of new drug property.
6.Genetic characteristics of rubella virus isolated in Sichuan province in 2015

Objective To understand the genetic characteristics of rubella virus in Sichuan province in 2015.Methods Veto/SLAM cells were used for rubella virus isolation and culture.The 739 nucleotides of E1 gene was amplified by Reverse Transcription-Polymerase Chain Reaction (RT-PCR) and subjected to sequence and homological analysis.Results 19 strains of rubella virus were isolated and identified as genotype 2B.The homology of nucleotides and amino acids were 99%-100% and 97.9%-100%,respectively.There were no changes in the important antigenic epitopes.Conclusions The rubella virus genotype circulated in Sichuan province in 2015 was genotype 2B.

SUMMARY Thehumanembryonicstemcells(hESCs)serveasaself-renewable,genetically-healthy, pluripotent and single source of all body cells,tissues and organs.Therefore,it is considered as the good standard for all human stem cells by US,Europe and international authorities.In this study,the standard and healthy human mesenchymal progenitors,ligament tissues,cardiomyocytes,keratinocytes,primary neurons,fibroblasts,and salivary serous cells were differentiated from hESCs.The human cellular health-safety of NaF,retinoic acid,5-fluorouracil,dexamethasone,penicillin G,adriamycin,lead ace-tate PbAc,bisphenol A-biglycidyl methacrylate (Bis-GMA)were evaluated selectively on the standar-dized platforms of hESCs,hESCs-derived cardiomyocytes,keratinocytes,primary neurons,and fibro-blasts.The evaluations were compared with those on the currently most adopted cellular platforms.Parti-cularly,the sensitivity difference of PM2.5 toxicity on standardized and healthy hESCs derived fibroblasts, currently adopted immortalized human bronchial epithelial cells Beas-2B and human umbilical vein endo-thelial cells (HUVECs)were evaluated.The results showed that the standardized hESCs cellular plat-forms provided more sensitivity and accuracy for human cellular health-safety evaluation.
8.Protective effect of the traditional Chinese medicine prescription, Jiu Nao Yi Zhi water extract, on human neuroblastoma SH-SY5Y cell line

Objective To observe the protective effect of the traditional Chinese medicine prescription, Jiu Nao Yi Zhi water extract, on human neuroblastoma SH-SY5Y cell line, its effect on expression of insulin signal transduction pathway, and to explore the related mechanisms.Methods SH-SY5Y cells cultured in vitro, were divided into control group, Jiu Nao Yi Zhi No.1 prescription group and No.3 prescription group.The doses were 0.0625 mg/mL, 0.125 mg/mL, 0.25 mg/mL, 0.5 mg/mL and 1 mg/mL.The thiazolyl blue ( MTT) metabolic rate of each group was determined.The dose of 0.125 mg/mL was chosen for cell immunofluorescence analysis, and to observe the expression of insulin receptor substrates-1 ( IRS-1 ) , cAMP response element binding protein ( CREB ) , and the factors of insulin signal transduction pathway.Results Compared with the control group, MTT metabolic rates of the Jiu Nao Yi Zhi groups were significantly increased (P<0.05), and the cell morphology was much better in those groups, cell body more plump, well-adherent and neurite extensions were observed.The expressions of IRS-1 and CREB were higher than that in the control group.Conclusions The traditional Chinese medicine prescription Jiu Nao Yi Zhi water extract can protect neurons by promoting nerve cell growth, and improving the expression of IRS-1 and CREB, the factors of insulin signal transduction pathway.
9.Expression characteristics of epithelial markers in human embryonic stem cells differentiating into keratinocytes

Objective:To differentiate human embryonic stem cells ( hESCs ) into keratinocytes ( K-hESCs) and analyse the expression characteristics of biomarkers of K-hESCs.Methods: The hESCs of line H9 were seeded on matrigel in mTeSR1 medium.The hESCs were directly differentiated into kerati-nocytes in epithelial differentiation medium with bone morphogenetic protein 4, retinoic acid and N2 sup-plement.The karyotype of K-hESCs was analyzed, comparing the gene expression differences of K-hESCs with human gingival epithelial cells (HGECs), human immortalized oral epithelial cells (HIOECs) and HaCaT by Real-time PCR.Molecular characteristics of the cell differentiation were observed throughout the process by immunocytochemical techniques.Results:H9-hESCs were successfully differentiated into the cells that exhibited characteristics of keratinocytes in epithelial differentiation medium.The karyotype of K-hESCs was 46, XX; and the keratinocyte gene p63 expression in K-hESCs was significantly lower than that in HaCaT ( P<0.05) , but there was no significant difference of p63 expression in K-hESCs, comparing with that in HGECs and HIOECs ( P >0.05 ) .Conclusion: H9-hESCs could be directly differentiated into K-hESCs.The gene expression of K-hESCs was similar to that of epithelial cells in the early stage of monolayer cells differentiation with high proliferative activity.
10.Reliability and validity of the Chinese version of the Functional Assessment of Cancer Therapy-Cognitive Function in Chinese women with breast cancer

Objective To examine the reliability and validity of the Chinese version of the Functional Assessment of Cancer Therapy-Cognitive Function (FACT-Cog) in Chinese women with breast cancer.Methods The Chinese version of the FACT-Cog was obtained by FACT.A total of 202 patients with breast cancer were investigated by convenient sampling survey.The reliability and validity of the scale were analyzed by the principal component analysis and Pearson correlation analysis.Results The Cronbach α of the Chinese version of the FACT-Cog was 0.96, and those of four dimensions were 0.87-0.96.The test-retest correlation coefficients for the Chinese version of the FACT-Cog were 0.45-0.73, P<0.01.The four-factor structure of the Chinese version of the FACT-Cog was confirmed and accounted for 66.63% of the total variance.The criterion-related validity of the Chinese version of the FACT-Cog were significantly related with those of anxiety, depression and fatigue,-(0.36~0.48),-(0.42~0.52), 0.48-0.59,P<0.01.Conclusion The Chinese version of the FACT-Cog is valid, reliable, and appropriate for clinical and research use in Chinese women with breast cancer.
