1.Determination of content of acetyl tyrosine in pediatric compound amino acid injection (19 AA-Ⅰ) by HPLC

Objective To establish a method for HPLC to determine content of acetyl tyrosine in pediatric compound amino acid injection (19AA-Ⅰ).Methods The chromatographic separation was achieved on a Agilent Hypersil ODS column (250 mm ×4.0 mm,5 μm) with Agilent 1200 liquid chromatography system.The mobile phases consisted of 20 mmol/L sodium dihydrogen phosphate solution ( adjusting pH to 2.5 with phosphoric acid)-acetonitrile (90:10) at a flow rate of 1.0 mL/min, the detection wavelength was 210 nm, the column temperature was 25℃.Results Acetyl tyrosine was completely separated from other amino acids.The calibration curves for acetyl tyrosine revealed good linearity in the range of 12.062-120.62μg/mL (r=0.9999).The average recoveries (n=9) of acetyl tyrosine was 100.0%, RSD% (n=9) was 0.9.The limits of quantification (S/N=10) was 0.15μg/mL.Conclusion The methodological validation results indicate that the established method can be applied to quality control of acetyl tyrosine in pediatric compound amino acid injection (19AA-Ⅰ).
2.Inhibitory effect of pulmonary carcinoma by adenovirus-mediated CD/UPRT gene.

The cell killing effects and bystander effects of double suicide gene on pulmonary carcinoma cells were explored. Lung adenocarcinoma cells (A549) were transfected with different titers of adenovirus vector and followed with different concentrations of 5-FC after a recombinant adenovirus vector carrying CD/UPRT gene (Ad-CD/UPRT) was constructed. The cell viability was measured by MTT assay 4 days later. The cell viability was dropped to 30.57 %-8.62 % after 10 MOI of Ad-CD/UPRT transfected and 5-FC (10-1000 microg/mL) administration. Furthermore, Ad-CD/UPRT-infected A549 cells showed a profound neighbor cell killing effect in the same methods. These results suggested that Ad-CD/UPRT/5-FC system can effectively suppress growth of lung adenocarcinoma cells, which may provide a novel and powerful candidate for lung cancer gene therapy strategies.
3.Effect of Electroacupuncture on Learning and Memory Ability of Cerebral Ischemia/Reperfusion Injured Rats

Objective To investigate the effect of electroacupuncture at Shenting (DU24) and Baihui (DU20) on learning and memory ability, as well as the nerve cell and the expression of mRNA of Bcl-2 and Bax in the hippocampus of rats with cerebral ischemia/reperfusion (I/R) injury. Methods 45 male Sprague-Dawley rats were randomly diveded into sham group (n=15), control group (n=15) and electroacupuncture group (n=15). The latter 2 groups were modeled with middle cerebral artery occlusion for 2 h and reperfusion. The electroacupuncture group received electroacupuncture at Shenting and Baihui for 7 days. They were tested with Morris water maze, and then, their hippocampus was observed under Nissl staining and the level of Bcl-2 and Bax mRNA were determined with RT-PCR. Results The escape latency decreased (P<0.001) and the frequence the platform was crossed increased (P=0.001) in the electroacupuncture group compared with the model group, with less damage of the nerve cell and the expression of Bax mRNA, and more of Bcl-2 mRNA (P<0.05). Conclusion Electroacupuncture can prevent learning and memory ability from cerebral ischemia/reperfusion injury in rats, which may associate with adjustment of expression of apoptosis-related gene to inhibit apoptosis of nerve cell.
4.The study of the colorimetric characteristics of the cobalt-chrome alloys abutments covered by four different all-ceramic crowns by using dental spectrophotometer.

OBJECTIVEThis study aims to evaluate the optical data of the different sites of the cobalt-chrome (Co-Cr) alloy abutments covered by four different all-ceramic crowns and the color difference between the crowns and target tab using a digital dental spectrophotometer.
METHODSTen Co-Cr alloy abutments were made and tried in four different groups of all-ceramic crowns, namely, Procera aluminia, Procera zirconia, Lava zirconia (Lava-Zir), and IPS E.max glass-ceramic lithium disilicate-reinforced monolithic. The color data of the cervical, body, and incisal sites of the samples were recorded and analyzed by dental spectrophotometer. The CIE L*, a*, b* values were again measured after veneering. The color difference between the abutments covered by all-ceramic crowns and A2 dentine shade tab was evaluated.
RESULTSThe L* and b* values of the abutments can be increased by all of the four groups of all-ceramic copings, but a* values were decreased in most groups. A statistical difference was observed among four groups. After being veneered, the L* values of all the copings declined slightly, and the values of a*, b* increased significantly. When compared with A2 dentine shade tab, the ΔE of the crowns was below 4.
CONCLUSIONFour ceramic copings were demonstrated to promote the lightness and hue of the alloy abutments effecttively. Though the colorimetric baseline of these copings was uneven, veneer porcelain can efficiently decrease the color difference between the samples and thee target.

5.Impact of Toxoplasma gondii infection on pregnancy outcomes in early preg-nant women

Objective To explore the impact of Toxoplasma gondii infection on pregnancy outcomes in early pregnancy wom-en. Methods Toxoplasma gondii IgM and IgG antibodies in the peripheral blood of 2 993 early pregnant women were detected by using enzyme-linked immunosorbent assay(ELISA). According to the test results,the infected ones were divided into an acute in-fection group,a previous infection group,and an active infection group,and 200 pregnant women without Toxoplasma infection were randomly chosen as a control group,and the pregnancy outcomes of the four groups were followed up and the results were compared. Results There were 286 women infected with Toxoplasma gondii,with the infection rate of 9.56%(286/2 993),in which 43 cases were diagnosed as acute infection,156 were previous cases,and the other 87 were active infection ones. The inci-dences of adverse pregnancy outcomes in the above 3 groups and the control group were 13.95%(6/43),1.92%(3/156),5.75%(5/87)and 1.50%(3/200),respectively. The incidences of adverse pregnancy outcomes in the acute infection group and active in-fection group were both higher than that in the control group,the differences were statistically significant(both P<0.05),while there was no significant difference between the previous infection group and control group(P>0.05). Conclusion Acute and ac-tive Toxoplasma gondii infections are closely associated with the occurrence of adverse pregnancy outcomes in early pregnant wom-en;therefore,Toxoplasma gondii IgM antibody should be included in the routine inspection items of the pre-pregnancy physical examination for child-bearing age women.
6.Influence of CCN3 on differentiation of bone marrow mesenchymal stem cells into endothelial cells

Objective:To explore the influence of nephroblastoma over expressed gene (CCN3)on differentiation of bone marrow mesenchymal stem cells (BM-MSCs)into endothelial cells.Methods:BM-MSCs differentiation into endothelial cells induced by condition medium containing vascular endothelial growth factor (VEGF,50ng/ml)was regarded as positive control group.BM-MSCs differentiation into endothelial cells induced by condition medium con-taining recombined CCN3 protein (100 ng/ml)and VEGF (50 ng/ml)was regarded as CCN3 group,and BM-MSCs incubated in pure complete medium was regarded as negative control group.Immunofluorescence staining and semi-quantitative reverse transcription-polymerase chain reaction method (RT-PCR)were used to measure expression of von Willebrand factor (vWF)after 16d to evaluate endothelial cell differentiation,and semi-quantitative RT-PCR method was used to measured expression of Notch1 gene mRNA before and after induced BM-MSCs differentiation. Results:On 16d after induced BM-MSCs differentiation,some cells of positive vWF fluorescence staining were ob-served in positive control group,those of CCN3 group was significantly more than those of positive control group, and expression of vWF mRNA [ratio of optical density (OD): (0.550±0.090)]of CCN3group was significantly higher than that of positive control group (0.358±0.080),(P<0.01),and no vWF-positive cells were observed in negative control group;Besides,before induced differentiation,expression of Notch1 gene mRNA were low in three groups without significant difference between any two groups (P>0.05 all),on 16d after induced differentiation, expressions of Notch1 gene mRNA in positive control group (0.232±0.047)and CCN3 group (0.352±0.029)were significantly higher than that of negative control group (0.132±0.033),P<0.01 all,it of CCN3 group was signifi-cantly higher that of positive control group (P<0.01).Conclusion:CCN3 can enhance BM-BMCs differentiation into endothelial cells by activating Notch1 signal pathway.
7.Effect of dexmedetomidine postconditioning on acute lung injury induced by lipopolysaccharide in rats

Objective To investigate the effect of dexmedetomidine postconditioning on acute lung injury (ALI) induced by lipopolysaccharide (LPS) in rats.Methods Fifty male Spragne-Dawley rats,aged 7-8 weeks,weighing 200-250 g,were randomly divided into 5 groups (n =10 each) using a random number table:control group (group C),LPS group and postconditioning with 3 different doses of dexmedetomidine groups (LD,MD and HD groups).ALI was induced with LPS 8 mg/kg injected via the caudal vein in LPS,LD,MD and HD groups.Dexmedetomidine 5,10 and 15 μg/kg were injected intraperitoneally in LD,MD and HD groups,respectively,at 1 h after LPS injection.The equal volume of normal saline was injected intraperitoneally in C and L groups.Blood samples were taken from the left ventricle at 6 h after dexmedetomidine administration,then the animals were sacrificed and broncheoalveolar lavage fluid (BALF) was collected.The concentrations of tumor necrosis factor-α (TNF-α) and interleukin-6 (IL-6) in plasma and BALF were detected by ELISA.The lungs were removed for microscopic examination of pathological changes and for determination of wet/dry lung weight (W/D) ratio and expression of Toll-like receptor 4 (TLR4) mRNA (by RT-PCR) in lung tissues.Results Compared with group C,W/D ratio,pathological scores,and the concentrations of IL-6 and TNF-α in plasma and BALF were significantly increased,and the expression of TLR4 mRNA was up-regulated in LPS,LD,MD and HD groups.Compared with LPS and LD groups,W/D ratio,pathological scores,and the concentrations of IL-6 and TNF-α in plasma and BALF were significantly decreased,and the expression of TLR4 mRNA was down-regulated in MD and HD groups.There was no significant difference in the parameters mentioned above between LPS and LD groups,and between MD and HD groups.Conclusion Dexmedetomidine postconditioning can alleviate ALI induced by LPS in rats,and up-regulated TLR4 mRNA expression and reduced inflammatory responses may be involved in the mechanism.
8.Efficiency of three methods for detecting Toxoplasma IgG antibody

Objective To discuss the test efficiency of three methods for detecting Toxoplasma IgG antibody. Methods To-tally 304 specimens were detected parallelly for Toxoplasma IgG antibody by using the gold marked method,indirect hemagglutina-tion test(IHA),and enzyme-linked immunosorbent assay(ELISA),and the sensitivity,specificity and Youden index of these methods were compared. Results The detection sensitivities of gold marked method,IHA,and ELISA for Toxoplasma IgG anti-body were 85.5%,89.8%and 91.9%respectively(χ2=4.12,P>0.05);the specificities were 92.4%,96.6%and 97.5%respec-tively(χ2=4.06,P>0.05). The detection efficiency and Youden index of ELISA were 94.1%and 0.89 respectively,being high-er than those of IHA and gold marked method. Conclusion The sensitivity and specificity of the ELISA method for Toxoplasma IgG antibody are higher,and in addition,it can be automated. Therefore,it is suitable for large-scale Toxoplasma IgG antibody screening.
9.Clinical analysis of 55 cases of primary esophageal small cell carcinoma

Objective To analyze the clinical characteristics,treatment and prognosis of primary esophageal small cell carcinoma (PESC).Methods Fifty-five patients with PESC were analyzed retrospectively.The survival rate was calculated by the Kaplan-Meier method and the Log-rank test using SPSS 17.0 software.Results The median survival time of 55 patients was 12 months.The 6,12,24.36months survival rates of these patients were 87.3 % (48/55),50.9 % (32/55),12.7 % (7/55),7.3 % (4/55),respectively.In multivariate analysis,stage and type of treatment were shown to be independent predictive.Conclusion PESC is a malignant tumor with early metastasis and poor prognosis.Combined therapy may improve the survival time of PESC patients.
10.A PRELIMINARY STUDY OF PECTIN ON THE MANAGEMENT OF DIABETES

In this study, 38 experimental subjects of diabetes mellitus were given pectin 25g extra per day orally under carefully controlled dietary plan for six weeks. Another 38 cases were selected at random as control group.Pre- and posttreatrnent, blood and urine sugar and serum insulin of each subject were measured and compared. The results were: 1) the blood sugar dropped markedly as compared with that of pretreatment (p05). 2) sugar in urine decreased evidently in contrast with control group, but no difference was seen in the volume of urine between the two groups. 3) serum insulin levels dropped significantly when compared with control group (p
