1.Expression and significance of CD146 in peripheral blood of patients with Henoch-Sch(o)nlein purpura

ObjectiveTo detect the expression of CD146 in sera and peripheral blood mononuclear cells (PBMCs)of patients with Henoch-Sch(o)nlein purpura(HSP),and to assess its role in the pathogenesis of HSP.MethodsSixty patients with HSP in acute or recovery stage and 20 normal human controls were included in this study.Enzyme-linked immunosorbent assay (ELISA)was performed to measure the expression levels of serum soluble CD146(sCD146),and real-time fluorescence-based quantitative reverse transcription-PCR to detect the mRNA expression levels of CD146 in PBMCs.The correlation of the above parameters with disease activity was assessed.ResultsThe levels of serum sCD146 and CD146 mRNA in PBMCs in patients with HSP in recovery stage were significantly higher than those in the normal human controls(0.52 ± 0.15) ng/L vs. (0.23 ± 0.08) ng/L,t =8.90,P < 0.05; 14.18 ± 2.49 vs.10.08 ± 2.08,t =6.92,P < 0.05),but lower than those in patients with HSP in acute stage(0.84 ± 0.49) ng/L,t =3.44,P < 0.05; 26.34 ± 2.55,t =18.69,P <0.05).ConclusionCD146 may participate in the pathogenesis of HSP.
2.Comparison and identification of Herba Sedi and Herba Sedi Linearis

Objective: To research identification methods of the two crude drugs from the original plants with close relationship and very similar macroscopic shape,"Chuipencao"(Herba Sedi) and "Fujiacao"(Herba Sedi Linearis).Methods:Microscopic technique and TLC were used to determine the pharmacognostical characters for identification purpose.Results: The microscopic observations showed that the tissue structures of the stems and the microscopic characters of the powders were very similar between the two herbal drugs.But there were significant differences in number of the vessel,shape of the pith,and number of the purplish-red cell.For the samples collected in August,number of the vessel of the former usually was only 35-58,while the latter was 75-150.Pith of the former usually was in the shape of a triangle,while the latter was in the shape of Y.The TLC result had also obvious difference between the two drugs.Besides,there were some differences of the pharmacognostical characters of the samples collected in distinct periods.Conclusion: The results provided scientific basis for the identification of the two crude drugs and suggested that the general microscopic features be not important in the microscopic identification of the crude drugs from the original plants with closer relationship.
3.Expressions of interleukin-27,-35 and Epstein-Barr virus-induced gene 3 mRNA in peripheral blood of patients with psoriasis vulgaris

Objective To detect the expression of interleukin (IL)-27,-35 and their common subunit Epstein-Barr virus-induced gene 3 (EBI3) mRNA in peripheral blood from patients with psoriasis vulgaris (PV).Methods Peripheral blood was collected from patients with psoriasis vulgaris (PV) in progressive (n =22),stationary (n =20) and regressive (n =20) stage as well as 30 healthy human controls.A competitive enzymelinked immunosorbent assay (ELISA) was performed to determine the serum level of IL-27 and IL-35.SYBR Green-based real-time reverse transcription (RT)-PCR was carried out to measure the mRNA expression of EBI3 in peripheral blood mononuclear cells (PBMCs).Results The patients with psoriasis vulgaris showed a higher level of IL-27 and IL-35 compared with the healthy controls.The serum level of IL-27 was significantly higher in the patients with progressive,stationary and regressive psoriasis vulgaris than in the healthy human controls ((226.72 ±71.32) ng/L,(160.80 ± 20.02) ng/L and (181.93 ± 27.36) ng/L vs.(138.66 ± 33.93) ng/L,P< 0.01,0.05,0.01,respectively),and higher in the patients with progressive and regressive psoriasis vulgaris than in those with stationary psoriasis vulgaris (P < 0.01,0.05,respectively).Increased serum level of IL-35 was observed in the patients with regressive psoriasis compared with those with progressive and stationary psoriasis and healthy controls ((355.78 ± 49.82) ng/L vs.(309.36 ± 36.10) ng/L,(299.14 ± 31.98) ng/L and (292.43 ± 36.26) ng/L,all P < 0.01),while no statistical difference was observed between the patients with progressive and stationary psoriasos and healthy controls.The mRNA expression level of EBI3 in the patients with progressive,stationary and regressive psoriasis was 20.15,16.17 and 19.74 times,respectively,higher than that in the healthy controls.Conclusion IL-27,IL-35 and EBI3 may function as anti-inflammatory factors in the development,as well as the reversal of progression,of psoriasis.

5.Comparison of two kinds of magnetic nanoparticles in vivo and in vitro.

This study compared a new type of polysaccharide-coated magnetic nanoparticles (in which the polysaccharide is derived from Angelica sinensis) with the dextran magnetic nanoparticles in terms of preparation, biocompatibility and tissue distribution in vivo and in vitro in order to examine the potential application of Angelica polysaccharide as a novel carrier in magnetic drug targeting (MDT). Magnetic nanoparticles were prepared by chemical co-precipitation. Their physical and chemical properties were determined by using the transmission electron microscope (TEM), laser particle size analyzer (DLS) and vibrating sample magnetometer (VSM), and their purity and structure by using X-ray diffractometer (XRD) and Fourier transform infrared spectroscopy (FTIR). The atomic absorption spectrometric method was performed for quantification of the iron content in different tissues. Histological sections were stained by Prussian blue staining to observe the disposition of magnetic nanoparticles in the liver and kidney. The results showed that both kinds of magnetic nanoparticles possessed small particle size, good dispersion and good magnetic properties. XRD showed the main component of the two magnetic nanoparticles was Fe(3)O(4) crystals, and FTIR proved Fe(3)O(4) was successfully coated by each polysaccharide, respectively. In vivo, Fe(3)O(4)-dextran accumulated in the liver, spleen and lung and Fe(3)O(4)-Angelica polysaccharide only in the spleen and lung. It was concluded that Angelica polysaccharide may be applied as a novel carrier in the preparation of magnetic nanoparticles.
6.Knowledge, attitudes and practices of post abortion family planning services in Tianjin obstetrics and gynecology personnels

Objective To better understand the knowledge, attitudes, practice (KAP) status and relative demand to post abortion family planning service (PAFPS) in medical staffs of obstetrics and gynecology department in Tianjin. Meth?ods By using random, stratified and clustered sampling approach, 290 obstetrics and gynecology personnels in Tianjin were selected. A Self-Administered Questionnaire on PAFPS KAP was employed to survey the general characteristics, aware?ness of PAFPS knowledge, attitudes to PAFPS and the occurrence of unintended pregnancy themselves, and service capabili?ties of PAFPS and request of continuing education. Results The respondents showed defects in PAFPS forms, follow-up ar?rangements and contraceptive method after abortion. The average knowledge score was higher in tertiary medical institution than that of secondary medical institutions. The score was higher in clinical doctors than that of nurses, and the higher the ti?tle, the higher the score. The 95.34%of respondents believed that it was necessary to develop PAFPS widely. But 93.55%of respondents thought that it can be difficult to carry out PAFPS as being required because of unclear service process, shortage of funds, human resource constraints, inadequate facilities and weak sense of services. The 53.36%of respondents, who had sexual life history, occurred unintended pregnancy themselves. The current defects of PAFPS were no standardized service processes, missing of medium-term and long-term follow-up, missing of collective consulting, providing contraceptives not timely and hadn′t pay attention to contraceptive advice. There were 90.68%medical personnels showed demand on relative training. Conclusion It′s necessary to perform continuing education about PAFPS to obstetrics and gynecology personnels via different intervention. At the same time, we should increase investment and make the service processes perfectly, so as to improve the level of PAFPS in obstetrics and gynecology medical staffs.
7.Effects of Shengmai San on oxidative damage in mentally stressed mice

AIM: To investigate the preventive effects of Shengmai San (SMS) on oxidative damage in mentally stressed mice.METHODS: An oxidative stress mouse model was established by moustache-removed. Protein carbonyl and thiobarbituric acid reactive substance (TBARS) formation were determined as the oxidative stress markers.RESULTS: (1)Moustache-cut was founded to significantly enhance the behavioral movements of mice, especially large movements (movement 2 and rearing). SMS pre-administration inhibited the accelerated movements. (2) Protein carbonyl was increased in brain, heart, liver and kidney. TBARS in liver and heart increased in the moustache-cut mice, but SMS pretreatment inhibited the increased protein carbonyl and TBARS.CONCLUSION: SMS has the preventive effects on oxidative damage induced by emotional stress.
8.Human papillomavirus 16 E6 inducing autophagy of the cervical cancer C33A cells by endoplasmic reticulum stress pathway

Objective To investigate the effects of human papilloma virus 16 E6 (HPV16 E6) on endoplasmic reticulum (ER) stress-autophagic response in the cervical cancer C33A cells.Methods Polymerase chain reaction was used for detecting the integration of HPV DNA.The eukaryotic expression vector of HPV16 E6 was constructed and transfected via lipofectamine into C33A cells.Experimental cells were classified into 3 groups:pcDNA3.1--HPV16 E6 group,pcDNA 3.1-group and C33A group.Western blot was used to measure expression of protein of HPV16 E6,Beclin 1,LC3 Ⅱ,IRE1,PERK and ATF6 in transfected cells.Results here was no HPV DNA integration in C33A cells that were confirmed as the intervention cells.Eukaryotic expression vector pcDNA3.1--HPV16 E6 was constructed successfully.The eukaryotic expression vector pcDNA3.1--HPV16 E6 significantly improved the expression of protein of HPV 16 E6 in C33A cells.The protein expression of Beclin 1,LC3 Ⅱ,IRE1,PERK and ATF6 were significantly improved after transfection with vector pcDNA3.1 +-HPV16 E6 (P < 0.05).Furthermore,LC3 Ⅱ protein level was reduced by treatment with ER stress inhibitor.Conclusion HPV16 E6 can improve autophagy through the ER stress pathway,and this response may play an important role in the process of HPV16 E6 inducing cervical cancer,providing one of the new strategies for gene therapy of cervical carcinoma.
9.Protective effect and mechanism of β-CM7 on renin angiotensin system & diabetic cardiomyopathy.

This article aimed at exploring the effects and protective mechanism of β-CM7 on renin angiotensin system (RAS) in diabetic rats myocardial tissue. We divided 32 male SD rats into 4 groups: control group, diabetic model control group, insulin (3.7x10(-8) mol/d) treatment group and β-CM7 (7.5x10(-8) mol/d) treatment group. After 30 days, all rats were decapitated and myocardical tissues were collected immediately. After injection, β-CM7 could decrease the content of Ang II, increase the content of Angl-7. And β-CM7 could improve the mRNA of AT1 receptor and Mas receptor. β-CM7 also could improve the mRNA of ACE and ACE2, enhance the activity of ACE and ACE2. These data confirmed tli β-CM7 could activate ACE2-Angl-7-Mas axis, negative passage in RAS, to inhibit the expression ACE mnRiJA and protein in rat myocardium, alleviate the myocardial tissue damage induced by Ang II. The effect of β-CM7 on inhibiting myocardium damage might be related to ACE/ACE2 passageway.

10.Effect of Astragaloside Ⅳ on hematopoietic growth factor expression in rat bone marrow mesenchymal stem cells

BACKGROUND:Astragaloside Ⅳ is a major component of Huangqi,promoting proliferation and differentiation of bone marrow mesenchymal stem cells;however,the mechanism has been less reported yet.OBJECTIVE:To explore the effect of Astragaloside Ⅳ on expression of multiple hematopoietic growth factors in bone marrow mesenchymal stem cells.METHODS:Bone marrow mesenchymal stem cells were isolated from adult Wistar rats by using the method of adhesive culture and clone,and they were then plated on 96-well plate and separately incubated with 100 uL Astragaloside Ⅳ(25,50,100,200 g/L)for 72 hours.The cells in the control group were cultured with an equal volume of DMEM-LG culture liuquid.Indirect immunofluorescence was used to detect the biological activity,MTT method was used to evaluate the effect of Astragaloside Ⅳ on proliferation and differentiation of bone marrow mesenchymal stem cells,and RT-PCR method was used to measure the expression of hematopoietic growth factors in bone marrow mesenchymal stem cells.RESULTS AND CONCLUSION:The 3~(rd)-passage bone marrow mesenchymal stem cells highly expressed CD44 but lowly expressed CD45.As compared with control group,Astragaloside Ⅳ promoted proliferation of bone marrow mesenchymal stem cells in a time/dosage-dependent manner,in particular,the 200 g/L Astragaloside IV and 72-hour intervention(P< 0.05).SCF expression was significantly increased in the drug group compared with control group(P < 0.01);however,TPO,GM-CSF,and TGF-β1 expressions were not changed significantly(P > 0.05).Moreover,interleukin-3 expression was not found in the bone marrow mesenchymal stem cells.Astragaloside Ⅳ promoted in vitro proliferation of bone marrow mesenchymal stem cells,possibly involving in SCF secretion.
